Total 2,451 sera collected from pig farms nationwide were tested for the detection of porcine reproductive and respiratory syndrome(PRRS) virus antibodies. The results were analyzed between different geographic regions, types of breeding pigs, and different years. The overall seroprevalence of PRRS virus antibodies for 3 years was 32.4%(705/2,451). The seroprevalence of PRRS virus antibodies in years 2000, 2001, 2002, and 2004 was 33.4% (284/850), 38.6%(291/754), 33.3%(155/466), and 17.1%(65/381), respectively. The seropevalence of PRRS virus antibody in sow in years 2000, 2001, 2002 and 2003 was 31.7%, 28.4%, 29.6%, and 13.4%, respectively. The seropevalence of PRRS virus antibody in gilts in years 2000, 2001, 2002 and 2003 was 36.6%, 67.4%, 54.7%, and 33.9%, respectively. The seropevalence of PRRS virus antibody in boars in years 2000, 2001 and 2003 was 45.7%, 36.4%, and 100%, respectively. No boar serum sample was submitted for the diagnosis of PRRS virus antibody in the year 2000. High seroprevalence of the PRRS virus antibody in sow, gilts and boars indicates that the infected breeding pigs are the major source of the PRRS virus infection, and also play an important role in spreading the PRRS virus between fan mates or herds.

It has been reported that ginseng is effective in the central nervous system, immune system, and the strong inflammatory responses. However, there has been no research report yet about the effect of ginseng on allergic hypersensitivity reactivity. To confirm the ginseng effects on the release of mediators(histamine. leukotrienes etc.) which cause the hypersensitivity reactivity and inflammatory response, we used actively sensitized guinea pig airway tissues by utilizing the superfusion technique. In this procedure. the contractile response and mediators released after antigen stimulation of sensitized tissues, and IgG and IgE antibody products were measured in sera of immunized animals. Then the results of the controll group were compared to those of ginseng pretreatment groups. In the total saponin(TS) and panaxatriol(PT) pretreatment, histamine release decreased by $20\%$ in the tracheal tissues after active sensitization by ovalbumin(OVA, 10mg/kg), but in the lung parenchyma, histamine release decreased by $40\%.$ Panaxadiol(PD) significantly decreased histamine release by $40\%$ in the both tissues after active sensitization. TS, PT and PD of ginseng poorly blocked leukotrienes (LTs) and prostagrandin $D_2(PGD_2)$ release(less than $10\%$). Ginseng TS and PT had no effect on the serum IgG antibody production by ovalbumin, whereas PD significantly increased serum IgG antibody contents(approximately by 2 times). However, $IgG_1$ antibody products in the serum of guinea pig actively sensitized with ovalbumin after PD pretreatment were decreased, compared to that with ovalbumin alone. IgE antibody production by passive cutaneous anaphylaxis(PCA) titer in the TS pretreatment increased 3 times more than in the absence of TS(PCA titer by PT was not detected). These studies show that some ginseng saponins can in part act to inhibit mediator release in antigen - induced airway smooth muscle by inducing the IgG antibody production which has been changed in the specificity.
This study was conducted to determine the serum antibodies against toxoplasma in swine from breeding-pig farm, pig farm and abattoir by latex agglutination(LA) test. LA test was carried out with commercial Toxo-MT kit (Eiken chemical co.). The results obtained were summerized as follows : 1. The cut-off titer of positive and negative reactions by Toxo-MT antigen used in this experiment was determined as the serum dilution of 1 ; 32. 2. positive rates of toxoplasma antibodies in 823 swine sera were 17.0%(140 cases) by LA test. 3. The toxoplasma antibody detection rates against 194 swine sera in breeding-pig farm, 273 swine sera in pig farm and 356 swine in abattoir were 46.9%(91 cases), 8.4%(23 cases) and 7.3% (26 cases) , respectively. 4. In LA test serum antibody titers in 823 test sera were shown as 51 cases (36.4%) in 1 : 32, 40(28.6%) in 1;64, 17(12.1%) in 1:128, 14(10.0%) in 1:256, 10(7.1%) in 1:512, 5(3.6%) in 1:1,024, and 3(2.1%) in 1 : 2,048. 5. Positive rates of toxoplasma antibodies in swine sera from each breeding-pig farm were 20.0∼61.9%.
Investigation of leptospiral antibody in Korean cattle and pigs was carried out from February to October, 1971. Ten different living antigens, namely L. icterohaemorrhagiae, L. canicola, L. antumnalis, L. hebdomadis, L. australis A, L. pomona, L. pyrogenes, L. grippotyphosa, L. bataviae and L. javanica, were used. A total of 590 Korean cattle and 460 pig blood samples collected from Seoul Majang-dong slaughterhouse were tested by the rapid microscopic agglutination test. Throughout the studies the following results were obtained and summarized. 1. Of 590 serum samples of Korean cattle 51 were positive(8.64%). 2. Of 460 serum samples of pigs 27 were positive (5.87%). 3. Of 51 positive cattle samples, 29(4.92%) showed antibody to a serotype of L. icterohaemorrhagiae and 18(3.0%) to L. canicola, and 4 (0.68%) to L. pomona. Eight of L. icterohaemorrhagiae positive samples showed a cross reaction to L. canicola. 4. Of 27 positive pig samples, 14(3.04%) showed antibody to L. ictereohaemorrhagiae and 7(1.52%) to L. grippotyphosa. 4(0.87%) to L. canicola, 2(0.43%) to L. pomona. Two of L. canicola positive samples showed a cross reaction to L. grippotyphosa. 5. Serum samples of seven pigs, showing antibody positive to L. grippotyphosa were first observed in Korea. 6. Infection rate of bovine and porcine leptospirosis, in Korea, appeared to be lower than that of Japan, Taiwan, Thailand and the Philippines.
In order to obtain information on murine model for foot and mouth disease virus(FMDV) type Asia 1, we studied whether guinea pig was a suitable model for studying FMDV. Apparently healthy 3 months old albino guinea pigs and unweaned 3 days old Swiss albino mice were used for this study. Total of 8 guinea pigs were divided into the infected(n=5) and control(n=3) groups. The incubation period of FMDV in the guinea pigs were roughly 2 days and the viremia persisted for 3 days in the guinea pigs. Mice inoculated with the plasma from control guinea pigs did not show any sign of viremia. The plasma were titrated by virus neutralization test using suckling mice as an indicator host. The mean virus neutralizing antibody titers of infected guinea pig at 3 DPI, 4 DPI and 5 DPI were log$\_$10/2.16, log$\_$10/ 3.39 and log$\_$10/ 3.44, respectively whereas there was no neutralizing antibody titer in control group. The difference between the mortality pattern and mean virus neutralizing antibody titer of infected and that of control group at day 3, 4, 5 were statistically significant(p<0.0l).
The purpose of this study was survey of porcine reproductive and respiratory syndrome (PRRS) virus antibody in Gyeongbuk province by enzyme-linked immunosorbent assay (ELISA). Total 690 samples collected from 15 pig farms were tested. The overall seroprevalence of PRRS virus antibodies was 63.2% (436/690) and 13 farms of 15 farms had at least one pig with PRRS virus antibodies. The seroprevalence of PRRS virus antibody varied with age. Results in 1 to 30 day old, 31 to 60 day old, 61 to 90 day old, 90 to 120 day old and over 120 day old pig were 58.3%, 36.0%, 68.0%, 84.0%, 80.0% and sow were 61.9% respectively.
We examined the immunological properties of the recombinant hepatitis B surface antigen (r-HBsAg) which was expressed in mammalian cell (C127). The cross-immunity of r-HBsAg and plasma-derived hepatitis B surface antigen (p-HBsAg) were tested using Western blotting and ELISA with guinea pig polyclonal antibody and naturally infected human-derived antibody and the both antigens show the same results in their response pattern and intensity, which indicate they have a good cross-immunity. from the measurement of $ED_{50}$ after formalin- or heat-inactivation, both r-HBsAg and p-HBsAg and p-HBsAg showed $ED_{50}$ of 0.2-0.3 in formalin-inactivaton, while r-HBsAg was 0.05-0.09 and p-HBsAg was 0.03-0.07 in heat-inactivation, which means heat-inactivation method is 3-4 times superior in immunogenicity. In the immunopersistency test performed in guinea pig for the period of 3 months with two different adjuvants, antibody titer was 34.2 with muramyl dipeptide adjuvant, which was 1.8 times greater than the antibody titer of 18.9 with $AIPO_{4}$ adjuvant. the mutagenicity of r-HBsAg has the same cross-immunity with p-HBsAg, and heat-inactivation method and muramyl dipeptide adjuvant allow development of r-HBsAg vaccine with excellent immunogenicity.
Neonatal Fc receptor (FcRn) gene encodes a receptor that binds the Fc region of monomeric immunoglobulin G (IgG) and is responsible for IgG transport and stabilization. In this report, the 8,900 bp porcine FcRn genomic DNA structure was identified and putative FcRn protein included 356 amino acids. Alignment and phylogenetic analysis of the porcine FcRn amino acid sequences with their homologies of other species showed high identity. Tissues expression of FcRn mRNA was detected by real time quantitative polymerase chain reaction (Q-PCR), the results revealed FcRn expressed widely in ten analyzed tissues. One single nucleotide polymorphism (SNP) (HQ026019:g.8526 C>T) in exon6 region of porcine FcRn gene was demonstrated by DNA sequencing analysis. A further analysis of SNP genotypes associated with serum Classical Swine Fever Virus antibody (anti-CSFV) concentration was performed in three pig populations including Large White, Landrace and Songliao Black pig (a Chinese indigenous breed). Our results of statistical analysis showed that the SNP had a highly significant association with the level of anti-CSFV antibody (At d 20; At d 35) in serum (p = 0.008; p = 0.0001). Investigation of expression and polymorphisms of the porcine FcRn gene will help us in further understanding the molecular basis of the antibody regulation pathway in the porcine immune response. All these results indicate that FcRn gene might be regarded as a molecular marker for genetic selection of anti-CSFV antibody level in pig disease resistance breeding programmes.
This studies were carried out to investigate the rearing managements of pig farms and survey on pneumonia of the slaughtered pigs from 5 selected herds located in Jangsu, Jeonbuk. Isolated aerobic microorganisms from pneumonic lung were examined antibiotic susceptibility and tested serological antibody titers of the herd base. Prevalence rate of pneumonia were examined according to rearing and health management conditions of pig farms. Prevalence rate of pneumonia were detected in 78.8% and enzotic pneumonia. In 47.7%, pleuropneurnonia in 31.1%. In serological antibody titers showed the positive reaction with 54.4% in Actinobacillus pleuropneumonia serotype 5, 44.8% in Pasteurella multocida, 36.8% in A pleuropneurnonia serotype 2, 13.6% in Mycoplasmal pneumonia. Isolated aerobic microorganisms were examined antibiotic susceptibility and showed the high activity in gentamicin (58.3%), enrofloxacin (53.3%), norfloxacin (51.6%), cephalothin (41.7%) and low activity in amoxycillin (98.3%), oxytetracycline (98.3%), penicillin G (90.0 %), tetracycline (88.4%), ampicillin (88.3%). The risk of an outbreak of classical swine fever (CSF) was evaluated in relation to the vaccination and seroprevalence of antibodies. Blood samples were collected from 60 piggeries throughout Korea and information regarding CSF vaccination habits was also obtained via in-person interviews with pig farmers. The results of the survey indicated that 51 out of 60 farms were regularly performing CSF vaccinations in reproductive herds. Farmers preferred to vaccinate their reproductive pigs before weaning (41 farms) than on (9 farms) or after weaning (1 farm). In growing herds, however, double vaccinations as recommended were implemented for only 40 farms (66.7%) and vaccine schedules were identified as being frequently ignored. Moreover, many farms (18/40) were found to vaccinate earlier or later than the recommended time. According to antibody seroprevalence levels, only 36 farms (60%) were considered to be safe regarding potential risk for a CSF outbreak. Among the remaining pig farms, 6 were at low-risk (10%), 12 were at medium-risk (20%), and another 6 were at high-risk (10%). Antibody levels were found to be consistent with vaccination status obtained from personal interview surveys. Our findings demonstrate the importance of vaccinations regarding the prevention of a CSF outbreak, suggesting that vaccinations must be promoted toward pig raisers and practitioners.
