Hydrocotyle japonica의 약효성분에 관한 연구(I) (Studies on the Pharmaco-Constituents of Hydrocotyle japonica (I))

  • For the investigation of medicinal resources in Hydrocotyle species, the studies were conducted to evaluate the pharmaco-constituents in Hydrocotyle japonica MAKINO (Umbelliferae), which is used as folk medicine in Korea. From the methyl alcohol extract of the whole plant, $isorhamnetin-3-O-{\beta}-D-galactoside$ ($C_{22}H_{22}O_{12}{\cdot}1/3H_2O$, bright yellow needle crystal, mp $247{\sim}248^{\circ}C$, $[{\alpha}]_D^{28}^{\circ}=-52.27^{\circ}$ in pyridine), one of three flavonol substances in extrat, was isolated and identified by physicochemical properties and spectroscopic evidences (UV, IR, NMR and MS etc.,) in comparison with authentic sample. This flavonoid was appeared from Hydrocotyle japonica MAKINO through phytochemical approaches at the outset.

Antioxidative and Radical Scavenging Properties of the Constituents Isolated from Cosmos caudatus Kunth

  • A phytochemical investigation on the methanolic extract of Cosmos caudatus has led to the isolation of quercetin $3-O-{\beta}-D-arabinofuranoside$ (1), quercetin $3-O-{\beta}-D-rhamnoside$ (2), quercetin $3-O-{\alpha]-D-glucoside$ (3) and quercetin (4). These compounds were shown to be the antioxidative constituents of the plant when evaluated using the ferric thiocyanate (FTC) and thiobarbituric acid (TBA), and radical scavengers based on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assays.

  • Five constituents were isolated from the stem of Acanthopanax senticosus. Their structures were elucidated as (-)-sesamin (1), iso-fraxidin (2), 5-hydroxymethylfurfural (3), syringin (4) and acanthoside D (5) by spectral analysis. Among these compounds, 5-hydroxymethylfurfural (3) was isolated for the first time from this plant.

  • The famous prescription of So Cheong Ryong Tang (小靑龍湯)in Chinese herb medicine, which has been used for the treatment of common cold, influenza, asthmatic bronchitis, and bronchial asthma and, is being commercially produced in the form of decoction. However, the storage problem for this dosage form remains unsettled. The quantitative changes of the major constituents, at different temperature and time course, were examined employing HPLC and GC. Cinnamaldehyde, cinnamic acid, ephedrine, albiflorine, paeoniflorin, benzoic acid, glycyrrhizin, and liquiritin were choosen as phytochemical markers. The content of cinnamaidehyde significantly decreased when stored at increased temperature, while that of benzoic acid has increased. By overall consideration of the present experimental data, it was suggested that most constituents are considerably stable when preserved below freezing temperature.

LC-MS/MS-Based Comparative Investigation on Chemical Constituents of Six Aster Species Occurring in Korea

  • Aster species (Asteraceae) are widely distributed edible and medicinal plants, known to contain various specialized metabolites including polyphenols and saponins. However, systemic analysis on the chemical profiles of these plants have rarely been made. Here we analyzed the phytochemical constituents in leaves of 6 Aster species occurring in Korea, A. ageratoides, A. altaicus var. uchiyamae, A. glehnii, A. hispidus, A. incisus, and A. yomena, by applying a LC-MS/MS-based untargeted metabolomics approach. The analysis revealed that A. ageratoides, A. hispidus, and A. yomena are relatively rich in saponins most of which are expected to be previously unknown.

Apoptotic Potential and Chemical Composition of Jordanian Propolis Extract against Different Cancer Cell Lines

  • Propolis is a resinous substance that is collected by Apis mellifera from plant sources and is used in traditional medicine. To study the phytochemical constituents and apoptotic potential of Jordanian propolis extract against different cancer cell lines, propolis was extracted using methanol, hexane, and ethyl acetate and was fractionated using chromatographic methods. Cytotoxicity was assessed using MTT and LDH assays. The apoptotic potential was investigated using florescence microscopy, multicaspase assay, Annexin-V and dead cell assay, and cell cycle assay. The phytochemical constituents were analyzed using GC-MS. The methanol extract of propolis exhibited cytotoxic potential against all cell lines tested. The IC50 values of the methanol extract were 47.4, 77.8, 91.2, and 145.0 ㎍/ml for HepG2, LoVo, MDAMB231, and MCF7 cell lines, respectively. The IC50 values of the F1 fraction were 31.6 (MDAMB231), 38.9 (HepG2), 36.7 (LoVo) and 75.5 (MCF7) ㎍/ml. On further purification using thin-layer chromatography, the IC50 values of the F1-3 fraction were found to be 84.31(HepG2), 79.2 (MCF7), 70.4 (LoVo), and 68.9 (MDAMB231) ㎍/ml, respectively. The anticancer potential of the F1 fraction was confirmed through the induction of apoptosis and cell cycle arrest at the G0/G1 phase. The GC-MS analysis of the F1 fraction revealed the presence of 3-methyl-4-isopropylphenol (29.44%) as a major constituent. These findings indicate the potential of propolis extract as a cancer therapy. However, further investigation is required to assess the acute and subacute toxicity of the most active fraction.

Wild Mushrooms: A Potential Source of Nutritional and Antioxidant Attributes with Acceptable Toxicity

  • This paper describes in detail proximate composition, nutritional profile, phytochemical constituents, antioxidant activities, antimicrobial potential, and antihemolytic activity (towards human erythrocytes) of various fractions of wild Ganoderma lucidum. Proximate analysis established that wild G. lucidum comprises about $87.02{\pm}5.45%$ of moisture, and the remaining part is a rich source of proteins ($8.59{\pm}0.37%$), crude fiber ($54.21{\pm}1.2%$), and carbohydrate (35.16%) with smaller fat content (3.33 %). Similarly, phytochemical screening revealed the presence of flavonoids ($217.51{\pm}0.30mg/g$), ascorbic acid ($116{\pm}7.32mg/g$), phenolics ($360.72{\pm}34.07mg/g$), ${\beta}$-carotenes ($0.42{\pm}0.04{\mu}g/g$), and lycopene ($0.05{\pm}0.00{\mu}g/g$). Extracts of wild G. lucidum in various solvents provided first line protection against Escherichia coli and Pasteurella multocida in the order of ethyl acetate> ethanol> methanol> n-hexane> water. Furthermore, aqueous and methanolic extracts of wild G. lucidum were found to be safe towards human erythrocytes. Overall, wild mushroom (G. lucidum) was found to be a good source of dietary supplements, antimicrobial and antioxidant agents in the pursuance of its commercial utilization in food and pharmaceutical industries.

  • Phytochemical investigation of the MeOH extract of the aerial parts of Bistorta manshuriensis resulted in the isolation of two cerebrosides, two lactams, six phenolic compounds and seven flavonoids. Their chemical structures were characterized by spectroscopic methods to be pinelloside (1), soyacerebroside I (2), pterolactam (3), 5-hydroxypyrrolidine-2-one (4), vanillic acid (5), caffeic acid methyl ester (6), protocatechuic acid (7), caffeic acid (8), 3,5-di-O-caffeoyl quinic acid methyl ester (9), chlorogenic acid methyl ester (10), avicularin (11), afzelin (12), quercetin (13), isoorientin (14), quercetin 3-O-${\beta}$-D-glucoside (15), quercitrin (16), and luteolin (17). The isolated compounds (1 - 4, 7, 12, 14) were isolated for the first time from this plant source and the compounds 1 - 4, 9 and 10 were first reported from the genus Bistorta. Compound 17 exhibited moderate cytotoxicity and compound 6 exhibited weak cytotoxicity against four human cancer cell lines in vitro using an SRB bioassay.

해동피의 화학성분 및 생리활성 (Chemical Constituents and Biological Activity of Kalopanacis Cortex)

  • The study on phytochemical analysis and the biological activity of Kalopanacis Cortex was carried out in this research. As a phytochemical result, liriodendrin as a lignan glycoside was isolated and characterized. Two subfraction separated from the acidic substance of $CHCl_3$ fraction were saturated and unsaturated fatty acid, respectively. Saturated fatty acid mixture identified from GC-MS tool was as follows: palmitic acid, stearic acid, arachidic acid, heneicosanoic acid, docosanoic acid, tricosanoic acid, tetracosanoic acid, pentacosanoic acid, hexacosanoic acid and octacosanoic acid. Unsaturated fatty acid was found to be linoleic acid on the basis of spectroscopic method. An active principle of liriodendrin exhibited significant antihepatotoxic activity but failed to show a considerable antiedemic activity. In this paper, the result of writhing test on liriodendrin was also described.

Phytochemical Screening, Isolation, Characterization of Bioactive and Biological Activity of Bungkang, (Syzygium polyanthum) Root-bark Essential Oil

  • Bungkang (Syzygium polyanthum) is a medium to tall plant which produces medicinal root-bark, the plant is normally found along inland river bank and produces small white flowers and fruits. Essential oils are among the most interesting components of the plant extracts consisting mostly of monoterpenoid or sesquiterpenoids. They are used as therapeutic agents in ethno, conventional, and complementary alternative medicines. Investigation and evaluation of the essential oil of Syzygium polyanthum as well as the antibacterial, antioxidant and antifungal activity was ascertained. The experiment was performed. 100 chemical constituents were obtained and two pure compound was isolated as Eugenol (1) and Farnesol (2). Significant growth inhibition of Staphylococcus aureus, (ATCCⓒ25923) Klebsiellia pneumonia (ATCCⓒ19155), Salmonella typhi (ATCCⓒ14028) and Escherichia coli (ATCC©25922) and the fungal strains Aspergillus flavin, Aspergillus niger, Candida, tropicalis, and Fusarium oxysporium was observed from the essential oil at concentration of 500 ㎍/mL. Antioxidant potential was observed to be strong of 18.42 ㎍/mL when compared to the control of 15.23 ㎍/mL. The result indicated that the oil obtained from root-bark of Syzygium polyanthum can be considered as an agent for antioxidant, antibacterial and antifungal in pharmaceutical food and cosmetic industries trails.
