미생물반응을 이용한 생리활성 물질의 합성 (Microbial Synthesis of Physiologically Active Compounds)

  • 지구상에서 생합성된 천연물은 완급의 차이는 있겠으나 궁극적으로는 CO$_{2}$ H$_{2}$O, NH$_{3}$ 등으로 다시 산화분해된다. 이 분해과정은 미생물의 효소반응으로 이루어지며 생물권의 항상성도 일차적으로는 미생물의 이 산화분해작용으로 유지된다고 볼 수 있다. 미생물이 영위하는 이러한 효소반응을 생리활성물질의 공업적합성에 이용한 초기의 예로는 부현피질호르몬의 합성과정에서 Rhizopus nigricans를 사용한 수산화반응과 ascorbic acid 합성과정에서 Acetobacter xylinum을 사용한 탈수반응이 유명하며 이러한 반응은 아직도 이용되고있다. 특히 지난 십수년간의 응용미생물학의 발전은 대단한 것이며 여러가지 항생물질, 당질, amino산, 핵산관련물질들이 생리활성물질과 함께 발효법으로 생산되고 있다. 그러나 이글에서 다루는 미생반응은 발효과정에서 생산되는 미생물자체의 일차적 대사산물 (primary metabolite)이나 이차적대사산물 (secondary metabolite)를 대상으로 한것이 아니며 어디까지나 외부에서 공급되는 화학물질에 대한 균체의 효소반응산물을 목적물로 하고있다. 또한 근래 활발히 이용되는 균체고정및 효소고정법을 이용한 효소공학적수법도 제외하고 배양과정의 균체 또는 배양후의 군체를 이용한 미생물반응에 한정코저 한다.

Selective Mercuration of 2-Hydroxy Nile Red and Its Application towards Chemodosimetric Hg2+-selective Signaling

  • Selective mercuration at the 1,6-positions of 2-hydroxy derivative of Nile Red and its application towards $Hg^{2+}$-selective signaling was investigated. The 2-hydroxy Nile Red exhibited a selective UV-vis absorption and fluorescent signaling behavior towards $Hg^{2+}$ ions over common coexisting physiologically important metal ions in aqueous environment. $^1H$ NMR studies revealed that the mercuration was selectively effected at the 1,6-positions of 2-hydroxy Nile Red, which is quite different from that at the 6,8-positions for the parent Nile Red.

  • Airway mucus provides the protective functions such as lubrication, barrier, disposal of trapped materials, and humidification. In the normal state, the mucus do not interfere with Bas transport and the other vital functions of lung. In diseases such as asthma, bronchitis, and cystic fibrosis, the mucus hypersecretion was physiologically developed in the response of multiple neurohumoral mechanism system. And regardless of the mechanism, many clinical sequelae result from mucus hypersecretion: atelectasis, infection, increased airway resistance, increased work of breathing, increased cough with its resultant complication. And the condensation of mucus tv mucus hypersecretion can make the mucus plug by which bronchial obstruction is developed. We have experienced a 7 Pear-old male patient with recurrent pneumonic symtom, which the bronchial obstruction was developed by the impacted mucus plug on the bronchoscopic finding. We report this case with the review of literature.

Structural Studies of G Protein-Coupled Receptors

  • G protein-coupled receptors (GPCRs) constitute the largest and the most physiologically important membrane protein family that recognizes a variety of environmental stimuli, and are drug targets in the treatment of numerous diseases. Recent progress on GPCR structural studies shed light on molecular mechanisms of GPCR ligand recognition, activation and allosteric modulation, as well as structural basis of GPCR dimerization. In this review, we will discuss the structural features of GPCRs and structural insights of different aspects of GPCR biological functions.

  • The glycosylation pattern of Erythropoietin (EPO), produced by recombinant CHO cells, was studied using the simple and rapid technique of 'Lectin-blotting'. In this experiment we used three different kinds of lectins, MAA(Maackia amurensis agglutinine), RCA(Ricinus communis agglutinine), and DSA(Datura stramonium agglutinine), which bind to the terminal sialic acid, galactose, and the N-acetyllactosamine chain respectively. The lectin-blotting technique was used to analyze the carbohydrate structure of EPO produced in the presence of two physiologically active chemical compounds, ammonium and chloroquine. The effect of the ammonium ion on the glycosylation of EPO was studied because it accumulated in the medium mainly as a by-product of glutamine matabolism. Ammonium chloride significantly inhibited the sialylation of the terminal galactose residue at concentrations of 8mM or more. Chloroquine, a potent inhibitor of glycosylation, inhibited terminal sialylation at concentrations of 100 and 200 $\mu$M, and at a concentration of 300 $\mu$M, also inhibited Nacetyllactosamine chain synthesis.

Metabolic Characterization of Lactic Acid Bacterium Lactococcus garvieae sk11, Capable of Reducing Ferric Iron, Nitrate, and Fumarate

  • A lactic acid bacterium capable of anaerobic respiration was isolated from soil with ferric iron-containing glucose basal medium and identified as L. garvieae by using 16S rDNA sequence homology. The isolate reduced ferric iron, nitrate, and fumarate to ferrous iron, nitrite, and succinate, respectively, under anaerobic $N_2$ atmosphere. Growth of the isolate was increased about 30-39% in glucose basal medium containing nitrate and fumarate, but not in the medium containing ferric iron. Specifically, metabolic reduction of nitrate and fumarate is thought to be controlled by the specific genes fnr, encoding FNR-like protein, and nir, regulating fumarate-nitrate reductase. Reduction activity of ferric iron by the isolate was estimated physiologically, enzymologically, and electrochemically. The results obtained led us to propose that the isolate metabolized nitrate and fumarate as an electron acceptor and has specific enzymes capable of reducing ferric iron in coupling with anaerobic respiration.

수도성장 및 수량예측을 위한 동적모형 SIMRIW의 적용 (Application of Dynamic Model SIMRIW for Predicting the Growth and Yield of Rice)

  • A simplified physiologically-based dynamic model, SIMRIW was selected for predicting the growth and yield of rice. The applicability of the model to the rice cultivars and weather conditions in the Republic of Korea was evaluated. Parameters of the model were calibrated using actual rice yields in Suweon region and an optimization scheme, Constrained Rosenbrock Algorithm. The simulated results from the calibrated model were in good agreement with the field data. The model with parameters calibrated for Suweon was applied to other five regions for the evaluation of transferability, but the simulated results fell short of satisfaction. However, the model is found to be applied to real-time prediction of the growth and yield of rice crop, which is believed to be useful for timely rice crop management, agricultural policy making, and optimal irrigation water management.

건강기능성 수산식품소재의 개발 (Development of functional food products with natural materials derived from marine resources)

  • Recently demand for safer and healthier food has augmented with advancements in health conditions. Food ingredients with yet to be known safety and functionality, are being investigated for their safety or detrimental effects. The Ministry of Food and Drug Safety has introduced "health functional food" by the "Health Functional Food Act" to evaluate bio-functional and safety properties of raw materials using standard methods including in-vitro and in-vivo testing before human consumption. Despite recent growth in net worth of domestic functional food market, most of the raw materials are not from local Korean industries with own research and development, and mostly terrestrial not marine resources. Geographically, Korea has access to diverse marine bio-resources that need to be managed and utilized sustainably. Recently, diverse novel physiologically active substances have been reported from marine organisms. Hence, the development of functional foods from marine bio-resources is considered as an inevitably important task.

  • Fruits of Hovenia dulcis T. (Rhamnaceae) was called ‘jiguja’ in oriental medicine which has been used for diuresis, remove of hangover and leaves has been used for detoxified the alcohol. In order to investigate the efficacy of antioxidative activity from leaves of Hovenia dulcis, the activity guided fraction and isolation of physiologically active substance were performed. Its 20%, 40%, 60%, 80%, 100% MeOH, H2O, Acetone fractions were examined antioxidative activity by DPPH method and TBARS assay. (omitted)

  • Seeds of Euryale ferox have been used for disorder of kidney, hysterorrhea of female and a tonic. In this study, in order to investigate the efficacy of antioxidant activity, the bio-activity guided fraction and isolation of physiologically active substance were performed. Roots, stems, flowers(seeds) were extracted with MeOH and each fractions were examined antioxidant activity by DPPH method. It was revealed that flowers(seeds) fration has significantly antioxidant activity. From flowers(seeds) frction, H$_2$O, 30%, 60%, 100% MeOH and acetone fractions were examined antioxidant activity by DPPH method. (omitted)
