뇌파 및 심전도 복합 생체신호를 이용한 실시간 감정인식 인터페이스 연구 (Research of Real-Time Emotion Recognition Interface Using Multiple Physiological Signals of EEG and ECG)

  • 뇌파 및 심전도 생체신호를 복합적으로 이용한 감정인식을 통한 실시간 사용자 인터페이스를 제안한다. 기존에 뇌파를 통한 감정인식의 문제점이었던 낮은 정확도를 개선하기 위해 뇌파의 Theta, Alpha, Beta, Gamma의 상대파워 값과 심전도의 자율신경계 비율을 혼합하는 복합 생체신호 감정 인식 시스템을 개발했다. 기쁨, 공포, 슬픔, 즐거움, 화남, 혐오에 해당하는 6가지 감정을 인식하기 위해 사용자별 확률 값을 저장하는 데이터 맵을 생성하고, 채널에 대응하는 감정 인식의 정확도를 향상시키기 위해 가중치를 갱신하는 알고리즘을 제안한다. 또한 뇌파로 구성된 단일 데이터와 뇌파/심전도 생체신호 복합 데이터의 실험 결과를 비교한 결과 23.77%의 정확도 증가를 보였다. 제안된 인터페이스 시스템은 높은 정확도를 통해 게임 및 스마트 공간의 제어에 필요한 인터페이스로 기기에 활용이 가능할 것이다.

ENVI-Met 시뮬레이션을 통한 도로 방향별 가로수 식재 형태에 따른 여름철 열환경 개선 효과 분석 (Analysis of Thermal Environment Modification Effects of Street Trees Depending on Planting Types and Street Directions in Summertime Using ENVI-Met Simulation)

  • 컴퓨터 시뮬레이션 프로그램인 ENVI-met을 활용하여 여름철 도로 방향과 가로수 식재형태에 따른 도시공간의 열환경 개선 효과를 분석하였다. 수목 식재 시 낮 시간 동안의 기온과 풍속은 감소하고 상대습도는 증가하였다. 평균복사온도와 인간 열환경지수 PET(physiological equivalent temperature), UTCI(universal thermal climate index)는 수목 식재 시 감소하는 경향을 보였다. 수목 식재 시 가장 큰 변화를 보인 기상요소는 평균복사온도로, PET와 UTCI 변화량 또한 평균복사온도와 유사한 패턴을 보였다. 가장 큰 저감효과를 보인 수목식재방법은 수고가 낮고, 수관폭이 넓고, 엽면적지수가 높은 수목을 좁은 간격으로 식재하는 시나리오(LWDN)였다. 평균복사온도, PET, UTCI는 태양의 고도 및 방위와 건물의 위치에 따른 그림자 형성 조건에 따라 큰 차이를 보였으며, 건물에 의한 태양복사에너지 차단이 클수록 수목에 의한 영향이 줄어들었다. 특히, 동측과 서측 보도에서는 시간대별 저감량에서 큰 차이를 보였다. 가장 저감효과가 큰 수목식재 시나리오인 LWDN으로 식재하였을 때 서측 방면에 위치한 북서, 서, 남서측 보도는 오전 시간대인 10:00에 수목이 없는 경우에 비해 PET 8.6-12.3℃, UTCI 4.2-4.5℃, 동측 방면에 위치한 북동, 동, 남동측 보도는 오후 시간대인 16:00에 PET 8.1-11.8℃, UTCI 4.4-5.0℃로 큰 저감효과를 보였다. 반면, 가장 저감효과가 적었던 수고가 높고, 수관폭이 좁으며, 엽면적지수가 낮은 수목을 넓은 간격으로 식재하였을 때 최대 저감량은 동측 방면 보도에서 PET 1.8℃, UTCI 0.9℃ 이하, 서측 방면 보도에서 PET 3℃, UTCI 0.9℃ 이하로 적은 저감효과를 보였다. 또한, 건물에 의한 영향으로 인해 수목영향이 적은 시간대에는 수목식재 시나리오 간 평균복사온도, PET, UTCI 저감효과의 차이가 크지 않았다. 이 연구결과는 가로수 식재 시 여름철 도시 열환경 저감을 위한 모델 개발에 기초자료로서 활용가능할 것이다.

Acetylation of Retinoblastoma Like Protein2 (Rb2/p130) in Tumor Tissues

  • The activity of Rb proteins is controlled by post-translational modifications, especially through phosphorylation. Acetylation of Rb2/p130 was reported recently in NIH3T3 cells but its physiological relevance in cell cycle control and tumorigenesis is still unknown. Efforts are underway to investigate possible interplay between Rb2/p130 phosphorylation and acetylation. Here we hypothesized that Rb2/p130 acetylation, like p53 acetylation, may play a role in development of the tumor phenotype. The proposed hypothesis regarding acetylation of Rb2/p130 in tumor VS normal cells was found to be true in our case study of 36 tumor samples. Statistical analysis of results suggest strong correlation among Rb2/p130 acetylation and cancer phenotype.

Study on the variation of cellular physiology of Escherichia coli during high cell density cultivation using 2-dimensional gel electrophoresis

  • Physiological changes of Escherichia coli during the fed-batch fermentation process were characterized in this study. Overall cellular protein samples prepared at the different stage of fermentation were separated by 2-dimensional gel electrophoresis (2-DE), and differently expressed 15 proteins, Phosphotransferase enzyme I, GroEL, Trigger factor, ${\beta}$ subunit of ATP synthase, Transcriptional regulator KDGR, Phosphoglycerate mutase 1, Inorganic pyrophosphatase, Serine Hydroxymethyl-transferase, ${\alpha}$ subunit of RNA polymerase, Elongation factor Tu, Elongation factor Ts, Tyrosine-tRNA ligase, DnaK suppressor protein, Transcriptional elongation factor, 30S ribosomal protein S6 were identified using matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF MS). When bacterial cells grow to high cell density, and IPTG-inducible heterologous protein is produced, expression level of overall cellular proteins was decreased. According to their functions in the cell, identified proteins were classified into three groups, proteins involved in transport process, small-molecule metabolism, and synthesis and modification of macromolecules.

Use of DNA Methylation for Cancer Detection and Molecular Classification

  • Conjugation of the methyl group at the fifth carbon of cytosines within the palindromic dinucleotide 5'-CpG-3' sequence (DNA methylation) is the best studied epigenetic mechanism, which acts together with other epigenetic entities: histone modification, chromatin remodeling and microRNAs to shape the chromatin structure of DNA according to its functional state. The cancer genome is frequently characterized by hypermethylation of specific genes concurrently with an overall decrease in the level of 5-methyl cytosine, the pathological implication of which to the cancerous state has been well established. While the latest genome-wide technologies have been applied to classify and interpret the epigenetic layer of gene regulation in the physiological and disease states, the epigenetic testing has also been seriously explored in clinical practice for early detection, refining tumor staging and predicting disease recurrence. This critique reviews the latest research findings on the use of DNA methylation in cancer diagnosis, prognosis and staging/classification.

  • Nitric oxide (NO) mediates various physiological and pathological processes, including cell proliferation, differentiation, and inflammation. Protein S-nitrosylation (SNO), a NO-mediated reversible protein modification, leads to changes in the activity and function of target proteins. Recent findings on protein-protein transnitrosylation reactions (transfer of an NO group from one protein to another) have unveiled the mechanism of NO modulation of specific signaling pathways. The intracellular level of S-nitrosoglutathione (GSNO), a major reactive NO species, is controlled by GSNO reductase (GSNOR), a major regulator of NO/SNO signaling. Increasing number of GSNOR-related studies have shown the important role that denitrosylation plays in cellular NO/SNO homeostasis and human pathophysiology. This review introduces recent evidence of GSNO-mediated NO/SNO signaling depending on GSNOR expression or activity. In addition, the applicability of GSNOR as a target for drug therapy will be discussed in this review.

Regulation of a Novel Guanine Nucleotide Binding Protein Tissue Transglutaminase ($G{\alpha}_n$).

  • Tissue transglutaminase (TGII, $G{\alpha}h$) belongs to a family of enzymes which catalyze post-translational modification of proteins by forming isopeptides via $Ca^{2+}$-dependent reaction. Although TGII-mediated formation of isopeptides has been implicated to play a role in a variety of cellular processes, the physiological function of TGII remains unclear. In addition to this Tease activity, TGII is a guanosine triphosphatase (GTPase) which binds and hydrolyzes GTP It is now well recognized that the GTPase action of TGII regulates a receptor-mediated transmembrane signaling, functioning as a signal transducer of the receptor. This TGII function signifies that TGII is a new class of GTP-binding regulatory protein (G-protein) that differs from "Classical" heterotrimeric G-proteins. Regulation of enzyme is an important biological process for maintaining cell integrity. This review summarizes the recent development in regulation of TGII that may help for the better understanding of this unique enzyme. Since activation and inactivation of GTPase of TGII are similar to the heterotrimeric G-proteins, the regulation of heterotrimeric G-protein in the transmembrane signaling is also discussed.

Oxidative Modification of Human Ceruloplasmin by Methylglyoxal: An in vitro study

  • Methylglyoxal (MG) is an endogenous physiological metabolite which is present in increased concentrations in diabetics. MG reacts with the amino acids of proteins to form advanced glycation end products. In this in vitro study, we investigated the effect of MG on the structure and function of ceruloplasmin (CP) a serum oxidase carrier of copper ions in the human. When CP was incubated with MG, the protein showed increased electrophoretic mobility which represented the aggregates at a high concentration of MG (100 mM). MG-mediated CP aggregation led to the loss of enzymatic activity and the release of copper ions from the protein. Radical scavengers and copper ion chelators significantly prevented CP aggregation. CP is an important protein that circulates in plasma as a major copper transport protein. It is suggested that oxidative damage of CP by MG may induce perturbations of the copper transport system and subsequently lead to harmful intracellular condition. The proposed mechanism, in part, may provide an explanation for the deterioration of organs in the diabetic patient.

  • As a result of studing on the "Joa-gui Em", we could reach conclusions as follows. 1. "Joa-gui Em" shows in vivo effect of that it supplement Kidney-Um essence and nomalize a physiological function of Kidney-Qi. 2. "Joa-gui Em" supplement Kidney-Um essence, it does not use in The Rise of Insubstantial Fire(음허화왕) body condition. 3. "Joa-gui Em" use in many ways women disease, in case of menstruation and meonpause disorder. 4. "Joa-gui Em" and "Joa-gui Hwan" are modification prescription of "Uk-mi-ji-hwang-hwan. 5. "Joa-gui Em" is a pure prescription in supplement Kidney-Um essence, Roast Glycyrrhizae Radix of prescription has effect of supplement Sam-cho Qi. 6. "Joa-gui Hwan" is similarly "Joa-gui Em." Colla Cornus Cervi and a Colla Cornus Glemmys of prescription have effect of supplement DUMAI and RENMAI CHONGMAI.

Aberrant phosphorylation in the pathogenesis of Alzheimer's disease

  • The modification of proteins by reversible phosphorylation is a key mechanism in the regulation of various physiological functions. Abnormal protein kinase or phosphatase activity can cause disease by altering the phosphorylation of critical proteins in normal cellular and disease processes. Alzheimer' disease (AD), typically occurring in the elderly, is an irreversible, progressive brain disorder characterized by memory loss and cognitive decline. Accumulating evidence suggests that protein kinase and phosphatase activity are altered in the brain tissue of AD patients. Tau is a highly recognized phosphoprotein that undergoes hyperphosphorylation to form neurofibrillary tangles, a neuropathlogical hallmark with amyloid plaques in AD brains. This study is a brief overview of the altered protein phosphorylation pathways found in AD. Understanding the molecular mechanisms by which the activities of protein kinases and phosphatases are altered as well as the phosphorylation events in AD can potentially reveal novel insights into the role aberrant phosphorylation plays in the pathogenesis of AD, providing support for protein phosphorylation as a potential treatment strategy for AD.
