In Vitro에서 Retrovirus Vector System을 이용한 인간 성장 호르몬 유전자의 발현 (Expression of Human Growth Hormone Gene using Retrovirus Vector System In Vitro)

  • Human growth hormone (hGH), one of the most important hormones in medicine, is secreted from anterior pituitary gland. Its broad physiological function includes body growth, cell regeneration, increasement of muscle volume, bone density, body fat reduction, and so on. Due to the wide range of therapeutic effects, the hGH produced from E. coli has been commercialized already. In this study, we asked whether it is possible to produce recombinant hGH efficiently from various cultured mammalia cells. To meet this purpose, we chose a retrovirus vector system for transfer and expression of the hGH gene in various mammalian cells. Analyses of RT-PCR, ELISA, and Western blot to determine expression of the hGH gene showed the highest production of the hGH was determined from chicken embronic fibroblast (CEF) cells with the concentration of 8.58 ${\mu}g$/ml. The biological activity of the hGH was similar to the commercially available counterpart. These results suggest that mass production of hGH is possible not only in the E. coli but also in the various mammalian cells.

  • Bacterial strains which degrade Arabian Light crude oil were isolated by enrichment culture from oil-spilled soil. The strain A54 was finally selected after testing emulsifying activity and oil conversion rate. Strain A54 was identified as a Acinetobacter sp. based on the morphological, biochemical and physiological characteristics. It appears to be highly specialized for growth on Arabian Light crude oil in minimal salts medium since it showed preference for oil or degradation products as substrates for growth. It was found that it could grow on at least fifteen different hydrocarbons. The optimum cultural and environmental conditions were as follows; 25$\circ$C for temperature, 7,5 for pH, 2.0% for NaCl concentration and 2.0% for crude oil concentration. Additionally, the optimal concentration of NH$_{4}$NO$_{3}$, and K$_{2}$HPO$_{4}$, were 12.5 mM and 0.057 mM, respectively. Cell growth and emulsifying activity as a function of time were also determined. Crude oil degradation and the reduction of product peaks were identified by the analysis of remnant oil by gas chromatography. Approximately 63% of crude oil were converted into a form no longer extractable by mixed organic solvents.

  • Acute or repeated physical exercise affects a large number of physiological parameters including hemodynamics, respiration, pH, temperature, gastrointestinal function and biotransformation, which determine the pharmacokinetics of drugs and chemicals. The rate and the amount of a chemical reaching the active site are altered by physical exercise, which results in significant changes in pharmacolosical/toxicological activity of the chemical. This aspect of physical exercise has vast implication in therapeutics and in safety evaluation, particularly for chemicals that have a low margin of safety. However there appears to be a wide inter- and intraindividual variation in the effects of physical exercise depend-ing on the duration, intensity and type of exercise, and also on the properties of each chemical. It is suggested that more studies need to be done to determine which factor(s) plays a major role in the disposition of chemicals in human/animals performing physical exercise. Certain chemicals induce severe toxicity due to metabolic conversion to reactive intermediate metabolites. it is suggested that repeated exercise may enhance the free radical scavenging system by increasing the activity of antioxidant enzymes. This area of research remain to be explored to elucidate the interaction of exercise and chemical on the antioxidant system.

Total Phenolic Contents, Radical Scavenging Capacities and Inhibitory Effects on Lipid Peroxidation and LDL Oxidation of Prunus persica Branch

  • This study was undertaken to elucidate the antioxidant activity of the ethanol (EEPB) and water (WEPB) extracts of Prunus persica branches. The extracts contained a high phenolic content and revealed a potent hydrogen donating activity in DPPH scavenging assay. Compared to $\alpha$-tocopherol, EEPB (p < 0.001) and WEPB (p < 0.05) significantly inhibited $FeCl_2$-ascorbic acid-induced lipid peroxidation, and also exhibited potent antiradical activities against hydroxyl radical, superoxide anion, nitric oxide and peroxynitrite. In copper- and AAPH-mediated human low-density lipoprotein (LDL) oxidation systems, the extracts demonstrated a strong antioxidant function by metal chelating, rather than direct scavenging, action. Furthermore, EEPB at 5 ${\mu}g/mL$ concentration showed 80.77% inhibition of the electrophoretic mobility of LDL, compared to 77.69% for ascorbic acid and 76.92% for BHT. These results suggest that PB branch extracts may protect against oxidative stress-induced diseases.

  • PET/CT combines the functional information from a positron emission tomography (PET) exam with the anatomical information from a computed tomography (CT) exam into one single exam. A CT scan uses a combination of x-rays and computers to give the radiologist a non-invasive way to see inside your body. One advantage of CT is its ability to rapidly acquire two-dimensional pictures of your anatomy. Using a computer these 2-D images can be presented in 3-D for in-depth clinical evaluation. A PET scan detects changes in the cellular function - how your cells are utilizing nutrients like sugar and oxygen. Since these functional changes take place before physical changes occur, PET can provide information that enables your physician to make an early diagnosis. The PET exam pinpoints metabolic activity in cells and the CT exam provides an anatomical reference. When these two scans are fused together, your physician can view metabolic changes in the proper anatomical context of your body. PET/CT offers significant advantages including more accurate localization of functional abnormalities, and the distinction of pathological from normal physiological uptake, and improvements in monitoring treatment. A PET/CT scan allows physicians to measure the body's abnormal molecular cell activity to detect cancer (such as breast cancer, lung cancer, colorectal cancer, lymphoma, melanoma and other skin cancers), brain disorders (such as Alzheimer's disease, Parkinson's disease, and epilepsy), and heart disease (such as coronary artery disease).

Distinct Repressive Properties of the Mammalian and Fish Orphan Nuclear Receptors SHP and DAX-1

  • It has been suggested that the structure and function of nuclear receptors are evolutionally conserved. Here, we compare the molecular functions of the nile tilapia (Oreochromis niloticus) small heterodimer partner (nSHP/NR0B2) and the Dosage-sensitive sex reversal AHC critical region on X chromosome gene 1 (nDAX-1/NR0B1) with those of human SHP and DAX-1 (hSHP and hDAX-1, respectively). We found that, upon transient cotransfection of human cells, nDAX-1 repressed the activity of tilapia SF-1 (nSF-1) but not that of human SF-1, although the physical interaction with human SF-1 was retained. Similarly, nSHP repressed the activity of nSF-1, whereas hSHP did not, pointing to divergent evolution of SHP/SF-1 in fish and human. We thus propose that the repressive functions of SHP and DAX-1 have been conserved in fish and mammals although with different transcriptional targets and mechanisms. These differences provide new insights into the physiological diversification of atypical orphan nuclear receptors during vertebrate evolution.

비기허(脾氣虛)로 인한 내습(內濕)의 발생과 사군자탕(四君子湯) (Study on the Endogenous Dampness Caused by Gi Deficiency of the Spleen and Sagunja-tang)

  • The spleen is the source of gi, blood, body fluid and plays a vital role in maintaining life. The function of the spleen is to transform food nutrients and water, and to transport them to the heart and the lung. The movement of splenic gi is marked by elevation. The spleen governs the activity of elevating the lucid. The function of transportation and transformation is usually disturbed in the state of Gi deficiency of the spleen. The main clinical manifestations of gi deficiency of the spleen can be divided into as followers: anorexia, loose stool if the digesting and absorbing functions are disturbed; phlegm and edema if dampness and water are retained due to unhealthy water metabolism. Sagunja-tang can be applied for gi deficiency syndrome of the spleen. Ingredient bakchul(Rhizoma Atractylodis Macrocephalae) and bokryeung(Poria) can be used as monarch drug to eliminate dampness and strengthen the spleen.

  • Chrysanthemum zawadskii Herbich var. latilobum Kitamura (Compositae), colloquially known "Gujulcho" in Korea, has been used in traditional medicine for the treatment of various diseases, including cough, common cold, bladder-related disorders, gastroenteric disorders, hypertension, and inflammatory diseases, such as pneumonia, bronchitis, pharyngitis, and rheumatoid arthritis (RA) However, the effect of Chrysanthemum zawadskii var. latilobum on breast cancer invasion is unknown. In this study, we investigated the inhibitory effect of Chrysanthemum zawadskii var. latilobum extract (CZE) on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced matrix metalloproteinase-9 (MMP-9) expression and cell invasion, as well as the molecular mechanisms involved in MCF-7 cells. CZE were not cytotoxic up to 100 ${\mu}g/ml$ concentration in the MCF-7 cell line. CZE decreased MMP-9 expression. TPA substantially increased NF-${\kappa}B$ DNA binding activity. Pre-treatment with CZE inhibited TPA-stimulated NF-${\kappa}B$ binding activity and NF-${\kappa}B$ related protein expression. To identify invasion ability of MCF-7 cells decreased by CZE, we used martrigel invasion assay. As a result, it is significantly decreased cell invasion. These results indicate that CZE-mediated inhibition of TPA-induced MMP-9 expression and cell invasion involves the suppression of the NF-${\kappa}B$ pathway in MCF-7 cells. Chrysanthemum zawadskii var. latilobum may have potential value in restricting breast cancer metastasis.

참나무 (Quercus Mongolica)로부터 수용성 식이섬유소의 제조 및 기능성 검증 (Preparation of Soluble Dietary Fiber from Oak Wood (Quercus Mongolica) and Its Physiological Function in Rat Fed High Cholesterol Diets)

  • 본 연구에서는 참나무(oak wood)류중 신갈나무(Querrus mongolica)로부터 수용성 식이섬유소의 제조와 제조된 수용성 식이섬유소의 생리적 기능성을 검정하고자 하였다. 신갈나무로부터 수용성 식이섬유소의 분리는 전처리 기술로써 폭쇄처리를 하였으며 최적 폭쇄전처리 조건은 25kgf/$\textrm{cm}^2$의 압력으로 6분간 처리하였다. 폭쇄시료를 탈리그닌 처리하기 위해 가장 효과적인 방법중 1% NaOH 용액으로 수회여과 처리 하였다. 앞에서 탈리그닌된 시료를 효소가수분해 처리하기 위 해 사용된 효소로는 Onnozok R-10보다 Cellusoft 효소의 가수분해율이 효과적이었다. 제조된 수용성 식이섬유소의 분자량 분포는 약 1,200-348 사이에 존재하는 oligomer 형태의 소당류로 분포되어 있었다. 제조된 수용성 식이섬유소의 생리적 기능성을 검정하기 위해 실험동물은 Sprague-Dawley종 수컷을 이용하여 정상군과 고콜레스테롤식이 실험군으로 나눈 후 고콜레스테롤 실험군을 다시 섬유소 종류와 공급수준에 따라 섬유소를 공급하지 않은 무섬유식이군 (FF군), 시판 식이섬유소를 5%공급한군 (5P군) 10%공급한군 (l0P군) 제조 식이섬유소를 5% 공급한 군 (5M군) 10% 공급한 군(10M군) 등 각 10마리씩 6군으로 나누어 4주간 사육하였다. 식이섭취량은 섬유소공급군이 FF군에 비해 증가되었으며 체중증가량은 섬유소공급군에서 FF군에 비해 유의적 (p<0.05)으로 낮았다. 식이효율은 FF군에 비해 모든 식이섬유소군에서 유의적 (p<0.05)으로 낮았으며 특히 10% 공급군에서 가장 낮았다. 간 무게근 FF군에 비하여 식이섬유소 공급군에서 유의적 (p<0.05)으로 감소되었으며 소장 및 맹장무게는 정상군과 무섬유식이군에 비하여 식이섬유소 공급군에서 증가되었다. 혁청 GOT 활성은 식이섬유소 공급군이 FF군에 비해 유의적 (p<0.05)으로 저하되었다. GPT 활성은 정상군에 비해 무섬유식이군인 FF군에서 증가되었으며 섬유소 공급관들은 다소 감소되는 경향이었다. 간조직 중 GST 활성은 FF군에 비해 식이섬 유소 공급군 모두 활성이 증가되었다. 그리고 이러한 생리적 기능은 제조된 수용성 섬유소군이 시판 수용성 섬유소와 차이가 없었다. 결론적으로 제조된 수용성 식이섬유소와 시판되는 수용성 식이섬유소가 생리져 기능이 거의 비슷하고 무독성이 관찰됨으로써 신갈나무로부터 제조된 수용성 식이섬유소의 제조 방법이 우수하다고 볼 수 있다.

Deletion of cg1360 Affects ATP Synthase Function and Enhances Production of L-Valine in Corynebacterium glutamicum

  • Bacterial ATP synthases drive ATP synthesis by a rotary mechanism, and play a vital role in physiology and cell metabolism. Corynebacterium glutamicum is well known as an industrial workhorse for amino acid production, and its ATP synthase operon contains eight structural genes and two adjacent genes, cg1360 and cg1361. So far, the physiological functions of Cg1360 (GenBank CAF19908) and Cg1361 (GenBank CAF19909) remain unclear. Here, we showed that Cg1360 was a hydrophobic protein with four transmembrane helices (TMHs), while no TMH was found in Cg1361. Deletion of cg1360, but not cg1361, led to significantly reduced cell growth using glucose and acetic acid as carbon sources, reduced F1 portions in the membrane, reduced ATP-driven proton-pumping activity and ATPase activity, suggesting that Cg1360 plays an important role in ATP synthase function. The intracellular ATP concentration in the ${\Delta}cg1360$ mutant was decreased to 72% of the wild type, while the NADH and NADPH levels in the ${\Delta}cg1360$ mutant were increased by 29% and 26%, respectively. However, the ${\Delta}cg1361$ mutant exhibited comparable intracellular ATP, NADH and NADPH levels with the wild-type strain. Moreover, the effect of cg1360 deletion on L-valine production was examined in the L-valine-producing V-10 strain. The final production of L-valine in the $V-10-{\Delta}cg1360$ mutant reached $9.2{\pm}0.3g/l$ in shake flasks, which was 14% higher than that of the V-10 strain. Thus, Cg1360 can be used as an effective engineering target by altering energy metabolism for the enhancement of amino acid production in C. glutamicum.
