A Study on the Variation of Phytase Activity in the River Environment (하천환경에서의 phytsae 활성변화에 관한 연구)

  • From August 2000 to August 2001, 9 variables of physicochemical factors and phytase activity were investigated at 4 sites in the River Yungpyung and the influences of Physicochemical factors to Phytase activity were analyzed. Phytase activities of Site 1, Site 2, Site 3, and Site 4 varied between N.D ∼566 nmol/ ι /hr, N.D \" 434 nmol/ ι /hr, N.D ∼557 nmol/ ι /hr, and N.D ∼723 nmol/ ι /hr, respectively. The activities of summer season were higher than those of other season. But the activities were not detected on the winter season. The phytase activity and temperature showed high correlation. The correlation coefficients of Site 1, Site 2, Site 3, and Site 4 were 0.82(p<0.01).0.92(p<0.01),0.87(p<0.01), and 0.88(p<0.01), respectively. The phytase activity and NOI₃/sup -/ ion showed negative relation(r=-0.59, p<0.05) at Site 1. And the phytase activity had relation with Zn/sup 2+/at Site 2(r=().57, p<0.05) and Site 3(r=0.7E, p<7.07).

Cloning of Rod Opsin Genes Isolated from Olive Flounder Paralichthys olivaceus, Japanese Eel Anguilla japonica, and Common Carp Cyprinus carpio

  • G Protein-coupled receptors (GPCRs) mediating wide ranges of physiological responses is one of the most attractive targets for drug development. Rhodopsin, a dim-light photoreceptor, has been extensively used as a model system for structural and functional study of GPCRs. Fish have rhodopsin finely-tuned to their habitats where the intensity and the wavelength of lights are changed depending on its water-depth. To study the detailed molecular characteristics of GPCR architecture and to understand the fishery light-sensing system, genes encoding rod opsins were isolated from fishes living under different photic environments. Full-length rod opsin genes were obtained by combination of PCR amplification and DNA walking strategy of genomic DNA isolated from olive flounder, P. olivaceus, Japanese eel, A. japonica, and Common carp C. carpio. Deduced amino acid sequences showed a typical feature of rod opsins including the sites for Schiffs base formation (Lys296) and its counter ion (Glu113), disulfide formation (Cys110 and Cys187), and palmitoylation (Cys322 and Cys323) although Cys322 is replaced by Phe in Japanese eel. Comparison of opsins by amino acid sequence alignment indicated the closest similarity between P. olivaceus and H. hippoglossus (94%), A. japonica and A. anguilla (98%), and C. carpio and C. auratus (95%), respectively.

FA/Mel@ZnO nanoparticles as drug self-delivery systems for RPE protection against oxidative stress

  • Drug self-delivery systems can easily realize combination drug therapy and avoid carrier-induced toxicity and immunogenicity because they do not need non-therapeutic carrier materials. So, designing appropriate drug self-delivery systems for specific diseases can settle most of the problems existing in traditional drug delivery systems. Retinal pigment epithelium is very important for the homeostasis of retina. However, it is vulnerable to oxidative damage and difficult to repair. Worse still, the antioxidants can hardly reach the retina by non-invasive administration routes due to the ocular barriers. Herein, the targeted group (folic acid) and antioxidant (melatonin) have been grafted on the surface of ZnO quantum dots to fabricate a new kind of drug self-delivery systems as a protectant via eyedrops. In this study, the negative nanoparticles with size ranging in 4~6 nm were successfully synthesized. They could easily and precisely deliver drugs to retinal pigment epithelium via eyedrops. And they realized acid degradation to controlled release of melatonin and zinc in retinal pigment epithelium cells. Consequently, the structure of retinal pigment epithelium cells were stabilized according to the expression of ZO-1 and β-catenin. Moreover, the antioxidant capacity of retinal pigment epithelium were enhanced both in health mice and photic injury mice. Therefore, such new drug self-delivery systems have great potential both in prevention and treatment of oxidative damage induced retinal diseases.
