홍화자와 두충 혼합 추출물이 MG-63 조골세포의 Alkaline Phosphatase 활성에 미치는 영향 (Effects of Alkaline Phosphatase Activity on the Extract of Carthami Semen and Eucommiae Cortex in Human Osteoblast-like MG-63 Cell Line)

  • Carthamus tinctorius L. and Eucommia umoides Oliver are often used in traditional herbal medicines for reducing damage to the liver, kidney, bone and muscle. In the present study, we investigated cell viability and alkaline phosphatase activity in the human osteoblast-like MG-63 cell line with methanol extracts of Carthami Semen (CS) and Eucommiae Cortex (EC) alone or in a mixture (CS+EC). Osteoblast cell viability was evaluated using the MTS assay and alkaline phosphatase activity assays. The cell viability and alkaline phosphatase activity significantly increased in MG-63 osteoblast cells treated with the CS+EC mixture. These findings suggest the CS+EC mixture may have beneficial effects on bone health through the proliferation of osteoblast cells.

  • The present investigation has been undertaken to understand the mechanism of implantation process, by demonstrating the role of ovarian steroids in connection with phosphatase activity in the differentiation of uterine endometrium for implantation. The results obtained are as followings: The differentiation of the uterine endometrial tissue was closely influenced by the ovarian steroid hormones; at first, 17${\beta}$-estradiol initiated the differentiation of the uterine luminal and glandular epithelial cells, and then progesterone induced differentiation of stromal cells, and thereby two steroids maintain decidualization of the uterine tissues. We observed that the phosphatase activities seem to be dependent upon the ovarian steroids; that is the activity showed higher level in progesterone treated group than in estradiol treated one, and the highest activity was found in the group treated with both estradiol and progesterone. Acid phosphatase showed the highest activity whereas alkaline phosphatase showed the lowest in the rat uterine endometrium during early pregnancy.

광자극에 대한 Amoeba 의 반응양상과 phosphatase 의 특성 및 활성도 변화 (The Characterization and Activity Changes of Phosphatases in Amoeba sp. to the Light Stimuli and Its Response Pattern)

  • Amoeba sp. was cultured under the light and the dark conditions, and the activity of phosphatases was investigated. There was a linear correlation between the early reaction time and the activity of phosphatases when phosphatases were incubated at 30℃. Then the activity of acid phosphatase was about 2 times higher than that of alkaline phosphatase. The activity of phosphatase was optimal at pH 5.0 in acidic part and at pH 8.0 in alkaline part, respectively. The optimal temperature of phosphatases was near the 40℃. The isozyme patterns of cytoplasmic acid phosphatase were compared with those of membraneous one. Both the isozyme patterns were shown to bo polymorphic on the polyacyamide gel, but different band patterns were observed in the isozymes of the cytoplasmic and the membraneous acid phosphatases. The number of Amoeba sp. under the light stimulus for 48 hours decreased negative exponentially from the illumination. The activity of acid and alkaline phosphatases under the illumination of light incresed 1.7 and 1.5 times higher, respectively, than the activity of those under the dark condition. This result apperars to be related to the mechanism of the autophosphorylation.

저출력 레이저 조사가 치은섬유아세포의 증식과 염기성 인산분해효소의 활성에 미치는 영향 (The effect of low level laser irradiation on proliferation and alkaline phosphatase activity of human gingival fibroblast in vitro)

  • This study was performed to identify the proliferation and to measure the alteration of alkaline phosphatase activity in human gingival fibroblasts cultured. For the present study, the authors cultured the human gingival fibroblasts oriented from the sound interdental gingiva, and used third passage. It was used methyl $[^3H]$ Thymidine to identify the proliferation in human gingival fibroblasts and used 410nm of the spectrophotometer to measure the alteration of the alkaline phosphatase activity in human gingival fibroblasts. The results were as follows: 1. There was a statistically significant increase in the proliferation of gingival fibroblasts following low level laser irradiation at 24 hour(p<0.05). 2. There was a statistically significant increase in activity of alkaline phosphatase compared to control group at 5-day laser irradiation after in laser irradiation groups(p<0.05). And there was a statistically significant increase in activity of alkaline phosphatase compared to control group at 7-day laser irradiation after in the I-minute laser irradiation group(p<0.05), but there was a statistically significant decrease in activity of alkaline phosphatase compared to 1minute laser irradiation group at 7-day laser irradiation in the 2-minute laser irradiation group after(p<0.05). The results, within the limits of the present experiments, suggest that, the low level laser irradiation accelerates the proliferation of gingival fibroblasts and alters the alkaline phosphatase activity until the restricted period.

히스티딘 에시드 포스파테이즈(Histidine Acid Phosphatase) 계열 인간 파이테이즈(Phytase)의 일반적 특성규명 (General Enzymatic Properties of Human Histidine Acid Phosphatase-Phytase)

  • 산업용 균주인 Pichia pastoris에서 재조합 발현된 당화된 인간유래 MINPP (multiple inositol polyphosphate phosphatase) 효소는 기질 피틴태인(InsP6)에 대한 파이테이즈 효소활성을 나타내었고 탈당화시 그 효소활성이 30% 감소되었다. 그 효소의 기질 $InsP_6$에 대한 최적 pH 활성은 중성범위인 pH 7.4였다. 기질특이성 측면에서 인간 MINPP 효소는 para-nitrophenylphosphate (pNPP), ATP, ribose-1-phosphate (R-1-P)와 같은 유기인산화합물(organic phosphate conjugates)의 분해활성은 매우 낮은 대신, 기질 $InsP_6$를 효과적으로 분해하였고 특히 화학적 자극제인(chemical stimulator)인 1 mM 2-phosphoglycolate (2-PG)의 첨가에 따른 기질 2,3-bisphosphoglycerate (2,3-BPG)에 대한 효소활성이 2-PG를 첨가하지 않을때 보다 1.2배 증가되었지만 기질 $InsP_6$에 대한 효소활성에는 영향을 주지 않았다. 또한 2 mM $Mg^{2+}$ 이온과 100 mM $Cl^-$ 이온의 첨가는 MINPP 효소의 기질 2,3-BPG에 대한 효소활성에 역시 영향을 주지 않았다.

백서(白鼠) 간조직(肝組織)에 미치는 적하수오(赤何首烏)와 백하수오(白何首烏)의 효능에 관한 비교(比較) 연구(硏究) (A Comparative Study on the Effects of Polygoni Radix and Cynanchi Radix on Rat Livers Intoxicated with Carbon Tetrachloride)

  • The Polygoni Radix and Cynanchi Radix have been used to potentiate the liver functions in clinic of Oriental Medicine. The water extracts of Polygoni Radix and Cynanchi Radix were administered orally to rats intoxicated with carbon tetrachloride and then this experiment have been performed by observing liver fatty degeneration and activities of enzymes such as cytochrome oxidase (CYO), adenosine triphosphatase (ATP), acid phosphatase (ACP), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP). By oral administration of water extracts of the radices between 1 and 10 days, the following results were obtained. 1. The group given Cynanchi Radix extract showed recovery of the fat liver in 4 days, whereas that given Polygoni Radix extract did the recovery in 8 days. 2. In cytochrome oxidase activity, the group given Cynanchi extract showed normal activity in 6 days, whereas that given Polygoni Radix extract did the activity in 8 days. 3. In adenosine triphosphatase activity, the groups given Cynanchi Radix and Polygoni Radix extracts showed normal activities in 2 and 8 days, respectively. 4. In acid phosphatase activity, the groups given Cynanchi Radix and Polygoni Radix extracts showed recovery of the activities in 2 and 4 days, respectively. 5. In lactate dehydrogenase activity, the group given Cynanchi Radix and Polygoni Radix extracts showed recovery of the activities in 6 and 10 days, respectively. 6. In alkaline phosphatase activity, the group given Cynanchi Radix extract showed normal activity in 2 days, whereas that given Polygoni Radix extract showed slight recovery between 4 and 6 days followed by decrease of the activity in 8 and 10 days. From the above-mentioned results, it was found that both of the water extracts of Polygoni and Cynanchi Radix possessed the recovery action of liver function as intoxicated with carbon tetrachloride in rats. It is also noted that the extract of Cynanchi Radix showed more potent activity than that of Polygoni Radix.

  • 대두 발아종 오존수 처리에 의한 배유세포와 산성 인산호효소의 변화에 관해서 전자현미경으로 조사한 결과는 다음과 같다. 산성 인산화효소의 활성은 발달하고 있는 종자 배유의 여러 소기관에 나타났는데, 효소의 활성은 0.5 ppm 오존수를 처리한 후 12시간 배양중에 나타났다. 오존수를 처리한 후 배유가 분화함에 따라 산성 인산화효소의 반응산물은 액포내에 축적되는 것으로 나타나는데, 이것은 산성 인산화효소가 세포간 저장물질의 분해와 이동에 관여하는 것으로 여겨진다.

  • Fibricola seoulensis의 성체와 피낭유충에서 acid(Acpase)와 alkaline phosphatase (AIPase)의 분포와 동위효소유형 (유형)의 변화를 추구하고자 효소조직화학적 방법과 전기영동법을 이용하여 성체에서 Acpase는 pH 5가 최적의 활성이 나타났고. 분자량이 95 kDa. 85 kDa. 73 kDa. 62 kDa인 4종류의 동위효소가 동정되었다. 피낭유충에서 A쳬ase는 활성이 약하거나 나타나지 않았고. 분자량이 62 kDa인 1종류의 동위효소가 동정되었다 성체와 피낭유충에 AIPase는 pH 8에서 최적의 활성이 나타났고, 피낭유충의 생식원기에서 강한 활성이 나타났다.

  • 著者는 白色레그혼의 種卵을 孵化시켜 얻은 鷄胎中 賢과 後賢에 있어서의 alk. p., PAS-陽性物質 및 燐脂質의 消長과 局在位置의 變異를 組織化學的 方法으로 追究하여 다음과 같은 結果를 얻었다. 1. 分泌尿細管의 器官形成에 앞서 未分化 間葉組織內에 强한 alk. p. 活性이 出現하였으며 同活性은 分泌尿細管의 分化와 함께 消失하였다. 2. 中賢 및 後賢 分泌尿細管의 分化와 함께 alk. p. 및 PAS-陽性物質은 分泌尿細管의 刷子綠에 强陽性反應을 보였다. 3. 中賢 및 後賢 分泌尿細管의 細胞質과 刷子綠은 强한 燐脂質 陽性反應을 띠었다. 4. 造賢組織의 分化中 未分化 間葉組織에 alk. p. 活性이 出現하였다가 組織의 分化와 함께 消失된 事實에서 보아 alk. p. 는 組織分化에 關與하는 것으로 생각된다. 5. 中賢 및 後賢 分泌尿細管의 刷子  에 있어서 alk. p. 와 PAS-陽性物質 및 燐脂質의 變異와 分布狀態가 一致한 것으로 보아 alk. p. 는 炭水化物의 再吸收 및 燐脂質代謝에 關與하는 것으로 思料된다.

  • Light membrane vesicles were isolated from the zucchini hypocotyl by floatation on ficoll density gradients and the proteins were solubilized with Triton X100. Three ATP-hydrolyzing enzymes were partially purified by ion-exchange and gel filtration chromatography and isoelectric focusing. There are plasma membrane-type ATPase whose activity was inhibited by vanadate but not by nitrate, tonoplast-type ATPase which was sensitive to nitrate but insensitive to vanadate and one having a phosphatase activity with a pI value different from that of an acid phosphatase. A fraction was obtained after DEAE-ion-exchange chromatography crossreacting with polyclonal antibodies against Ca2+ -ATPase from human erythrocytes.
