흰쥐 뇌하수체 전엽세포에서 PKC나 cAMP에 의한 LH 분비 및 $LH{\beta}$ Subunit mRNA 증가에 미치는 $Ca^{++}$의 영향 (Effects of Extracellular $Ca^{++}$ on PKC or cAMP-stimulated Increases in LH Release and $LH{\beta}$ Subunit mRNA Levels in Rat Anterior Pituitary Cells)

  • 흰쥐 뇌하수체 전엽배양세포에서 PKC나 cAMP에 의한 LH 분비와 $LH{\beta}$subunit mRNA 증가과정에서 세포외 $Ca^{++}$의 역할을 검증하기 위하여 phorbol ester와 forskolin에 의해 촉진된 LH 분비와 $LH{\beta}$ subunit mRNA 수준에 미치는 EGTA와 verapamil의 영향을 조사하였다. Forskolin에 의해 촉진된 LH 분비와 $LH{\beta}$ subunit mRNA 수준은 $Ca^{++}$ chelator인 EGTA나 $Ca^{++}$ 채널차단제인 verapamil의 처리로 세포외부로부터 $Ca^{++}$의 이동을 억제시켰을때 모두 감소하였다. PMA에 의해 유도된 LH 분비는 EGTA와 verapamil 처리에 의해 영향을 받지 않았으나 PMA에 의해 촉진된 $LH{\beta}$ subunit mRNA는 억제되었다. 따라서 본 연구에 의하면 PKC 활성화나 세포내 cAMP 농도 증가는 $LH{\beta}$ subunit mRNA 수준을 증가시키며 이러한 과정은 세포외 $Ca^{++}$ 의존적인 것으로 생각된다. 그러나 PKC 활성화나 cAMP 증가에 의한 세포외 $Ca^{++}$의 역할이 서로 다른 양상을 보인다. PKC 활성화에 의한 LH 분비는 세포외 $Ca^{++}$에 비의존적이나 cAMP에 의해 촉진된 LH 분비는 세포외 $Ca^{++}$ 유입의 영향을 받는 것으로 사료된다.

플로레틴(Phloretin)의 혈관내피수축 융합효과와 관련기전 연구 (The Convergence Effect of Phloretin Existent in Plants on Vascular Contractility)

  • 이 실험에서 뇌졸중, 심혈관계 질환 등에 대해 치료 활성이 기대되는 플로레틴(phloretin) - 사과나무 잎과 개살구의 주성분 - 에서의 쥐의 대동맥에서 ROCK(Rho연관 인산화효소) 불활성화를 통해 혈관수축을 억제한다는 기전을 확인하고자 한다. 개체에서 분리되고 내피가 유지되거나 손상된 동맥의 고리 조각은 2개의 금속 고리로 근운동기록기(myograph) 수조 안에 현수되었고 아래쪽 고리는 수조바닥에 고정되었고 위쪽 고리는 등장력 변환기에 연결되었고 등장력 변환기의 전기적 신호는 생리측정기에 표시되었다. 재미있게도 플로레틴은 수축약 (fluoride, phorbol ester)에 의한 혈관 수축을 억제하여 ROCK 또는 MEK(마이토겐 활성화 단백질 키나제 키나제) 비활성화 같은 경로가 혈관이완에 관여할 수 있음을 보였다. 따라서 phloretin은 내피가 완전한 혈관에서의 내피에 의존적인 작용 외에 추가적으로 내피에 비의존적으로 평활근에서 ROCK 또는 MEK 활성 감소에 참여하여 결과적으로 평활근에서 액틴-미오신 상호작용을 억제하여 혈관을 이완하는 것으로 관찰되었다. 또한 phloretin은 thromboxane A2 유도수축을 억제하고 ROCK 및 MEK 억제를 포함하는 기전있음을 주장합니다.

  • Serotonin (5-hydroxytryptamine (5-HT))는 신경계의 세포-세포 간의 신호전달의 주요한 신경전달물질이다. 세포막에 존재하는 serotonin transporter (SERT)는 연접간격에 존재하는 5-HT를 세포 내로 재흡수 하여 세포외부의 5-HT 농도를 조절하지만 그 기전은 아직 밝혀지지 않았다. 본 연구에서는 yeast two-hybrid system을 사용하여 SERT의 C-말단이 protein kinase C-$\varepsilon$ (PKC-$\varepsilon$)과 특이적으로 결합함을 알았다. PKC-$\varepsilon$는 PKC의 isotype으로 calcium 비의존적이며 phorbol ester/diacylglycerol 민감성 serine/threonine kinase이다. $Na^+/Cl^-$ 의존성 SLC6 gene family의 다른 수송체는 PKC-$\varepsilon$과 결합하지 않았다. Deletion mutant들을 사용하여 SERT는 PKC-$\varepsilon$의 C-말단부위와 결합함을 알았으며, 또한 이 단백질간의 결합을 GST pull-down assay로 확인하였다. PKC-$\varepsilon$는 in vitro에서 SERT의 N-말단의 펩티드를 인산화시켰다. 이러한 결과들은 PKC-$\varepsilon$에 의한 SERT의 인산화가 세포막에 존재하는 SERT의 활성을 조절하는 역할을 할 가능성을 시사한다.

혈관수축에 대한 감초산 Glycyrrhizic acid의 효과 (The Effect of Glycyrrhizic acid on Vascular Contractility)

  • 평활근 수축에 대한 감초산(glycyrrhizic acid)의 효과를 연구하고자 하였고 관련 상세 기전을 결정하기 위해 glycyrrhizic acid가 agonist-sensitive하게 평활근 수축한다는 가설을 제시하였다. S/D rats의 평활근을 수조 내 준비하였고 신호 변환기에서 평활근에 의한 운동 신호가 전기적 신호로 변환되어 생리기록기에 표시되는 데이터는 일원배치 분산분석으로 분석하였다. 재미있게도, 감초산은 불화물, 트롬복산 유사체 등 굵은 미세섬유성 조절성 수축제에 의한 평활근 수축을 중등도로 억제하였고 (농도 0.01, 0.03, 0.1 mM 불화물에서 p=0.113, 0.008, 0.004 (Student's t-test), p=0.113, 0.008, 0.004 (One way ANOVA), 그리고 0.01, 0.03, 0.1 mM 트롬복산 유사체에서 p=0.156, 0.004, 0.003 (Student's t-test), p=0.156, 0.004, 0.003 (One way ANOVA)) 포볼 에스테르 등 가는 미세섬유성 조절성 수축제에 의한 평활근 수축에 대해 효과가 소실되었다 (0.01, 0.03, 0.1 mM 포볼 에스테르에서 p=0.392, 0.086, 0.065 (Student's t-test), p=0.392, 0.086, 0.065 (One way ANOVA)). 이러한 결과는 내피 기능이 억제된 평활근에서 감초산은 평활근 내피에서 EDRF (NO) 생성 효과 외에 주로 ROCK 활성 감소 등 평활근에 대한 직접적 효과를 통해 최종적으로 평활근에서 악틴-미오신 상호작용을 제한하여 평활근을 이완시키는 것으로 생각된다.

Induction of the Nuclear Proto-Oncogene c-fos by the Phorbol Ester TPA and c-H-Ras

  • TPA is known to cooperate with an activated Ras oncogene in the transformation of rodent fibroblasts, but the biochemical mechanisms responsible for this effect have not been established. In the present study we used c-fos promoter-luciferase constructs as reporters, in transient transfection assays, in NIH3T3 cells to assess the mechanism of this cooperation. We found a marked synergistic interaction between TPA and a transfected v-Ha-ras oncogene in the activation of c-fos promoter and SRE. SRE has binding sites for TCF and SRF. A dominant-negative Ras (ras-N17) inhibited the TPA-Ras synergy by blocking the PKC-MAPK-TCF pathway. Dominant-negative RhoA and Rac1 (but not Cdc42Hs) inhibited the TPA-Ras synergy by blocking the Ras-Rho-SRF signaling pathway. Constitutively active $PKC{\alpha}$ and $PKC{\varepsilon}$ showed synergy with v-Ras. These results suggest that the activation of two distinct pathways such as Ras-Raf-ERK-TCF pathway and Rho-SRF pathway are responsible for the induction of c-fos by TPA and Ras in mitogenic signaling pathways.

Effect of Platycodin D on Airway MUC5AC Mucin Production and Gene Expression Induced by Growth Factor and Proinflammatory Factor

  • In this study, we tried to investigate whether platycodin D significantly affects MUC5AC mucin production and gene expression induced by epidermal growth factor (EGF), phorbol ester (PMA) and tumor necrosis factor-$\alpha$ (TNF-$\alpha$) from human airway epithelial cells. Confluent NCI-H292 cells were pretreated with varying concentrations of platycodin D for 30 min and then stimulated with EGF, PMA and TNF-$\alpha$ for 24h, respectively. MUC5AC mucin gene expression and mucin protein production were measured by RT-PCR and ELISA. The results were as follows: (1) Platycodin D was found to inhibit the production of MUC5AC mucin protein induced by EGF, PMA, and TNF-$\alpha$, respectively. (2) It also inhibited the expression of MUC5AC mucin gene induced by the same inducers. These results suggest that platycodin D can regulate mucin gene expression and production of mucin protein, by directly acting on human airway epithelial cells.

Enhanced Uptake of Modified Low-Density Lipoprotein by Eicosapentaenoic Acid-Treated THP-1 Macrophages

  • Animal and clinical studies as well as epidemiological data have provided convincing evidence that n-3 polyunsaturated fatty acids can protect against atherosclerosis. However, the effects of the fatty acids on atherogenesis are contradictory. This discrepancy could derive from great susceptibility of the fatty acids to oxidation. We investigated the effect of eicosapentaenoic aced(EPA) on cellular atherogenesis via the scavenger receptor of THP-1 derived macrophages. THP-1 cells were fully differentiated into macrophages by incubating with phorbol 12-myristate 13-acetate for seven days. Atherogenic features of EPA were compared by subsitituting for linoleic acid (LA). Macrophages were also incubated without treatment of the fatty acids as controls. EPA (5-50 nmol/mL) was not cytotoxic and did not measurably induce cellular oxidation compared to bovine serum albumin (BSA) vehicle or identical doses of LA. EPA increased macrophage uptake and degradation of acetylated LDL(AcLDL) up to 14% and 88%, respectively. EPA increased markedly total cellular sterol synthesis and heparin-releasable lipoprotein lipase activity of macrophages, indicating that EPA may enhance accumulation of cellular cholesteryl ester and possibly facilitate formation of foam cells. These results demonstrate that EPA promotes the modified LDL-triggered atherosclerotic process by the modulation of the scavenger receptor and the activation of LPL in macrophages.

Anti-Inflammatory Activity of Elsholtzia splendens

  • Elsholtzia splendens Nakai has been used in North-East Asia as an ingredient of folk medicines for treating cough, headache and inflammation. The present investigation was carried out to establish its in vivo anti-inflammatory activity using several animal models of inflammation and pain. The 75% ethanol extract of the aerial part of E. splendens significantly inhibited mouse croton oil-induced, as well as arachidonic acid-induced, ear edema by oral administration (44.6% inhibition of croton oil-induced edema at 400 mg/kg). This plant material also showed significant inhibitory activity against the mouse ear edema induced by multiple treatment of phorbol ester for 3 days, which is an animal model of subchronic inflammation. In addition, E. splendens exhibited significant analgesic activity against mouse acetic acid-induced writhing (50% inhibition at 400 mg/kg), while indomethacin (5 mg/kg) demonstrated 95% inhibition. E. splendens ($5-100{\;}{\mu}g/mL$) significantly inhibited $PGE_2$ production by pre-induced cyclooxygenase-2 of lipopolysaccharide-treated RAW 264.7 cells, suggesting that cyclooxygenase-2 inhibition might be one of the cellular mechanisms of anti-inflammation.

적포도의 주 항산화물질, 레스베라트롤의 항암작용: 아폽토시스 의한 인체 암세포 사멸 유도 (Anticarcinogenic Activity of Resveratrol, a Major Antioxidant Presnet in Red Wine : Induction of Apoptosis in Human Cancer Cells)

  • Resveratrol (3,5,4'-trihydroxy-trans-stilbene) has been considered to be as one of major antioxidants present in grapes responsible for beneficial effects of red wine consumption on coronary heart disease. This triphenolic stilbene has been suggested as a potential cancer chemopreventive agent based on its striking inhiitory effects on diverse cellular events associated with tumor initiation, promotion, and progression. The compound has strong antioxidative and anti-inflammatory activities which amy contribute to its chemopreventive/chemoprotective properties. In the present work, we have found that resveratrol reduces viability and DNA synthesis capability of cultured human promyelocytic leukemia (HL-60) cells. Likewise, the viability of human breast cancer cell line, MCF-7 was reduced by resveratrol treatment. The growth inhibitory and antiproliferative properties of resveratrol appear to be associated with its induction of apoptotic cell death as determined by morphological and ultrastructural changes, agarose gel electrphoretic analysis of internucleosomal DNA fragmentation, and in situ terminal end-labeling of fragmented DNA (TUNEL). This compound also inhibited the phorbol ester-induced expression of cyclooxygenase-2 (COX-2) protein in immortalized human mammary epithelial MCF-10A cells. These results suggest that resveratrol has the promising cancer therapeutic/chemopreventive potential.

Requirement of EGF Receptor Kinase for Signaling by Calcium-Induced ERK Activation and Neurite Outgrowth in PC12 Cells

  • Membrane depolarization in PC12 cells induces calcium influx via an L-type voltage-sensitive calcium channel (L-VSCC) and increases intracellular free calcium, which leads to tyrosine phosphorylation of epidermal growth factor (EGF) receptor and the associated adaptor protein, She. This activated EGF receptor complex then can activate mitogen-activated protein (MAP) kinase, as in nerve growth factor (NGF) receptor activation. In the present study, we investigated the role of EGF receptor in the signaling pathway initiated by membrane depolarization of PC12 cells. Prolonged membrane depolarization induced phosphorylation of extracellular signal-regulated kinase (ERK) within 1 min in undifferentiated PC12 cells. Pretreatment of PC12 cells with the calcium chelator EGTA abolished depolarization-stimulated ERK phosphorylation, but NGF-induced phosphorylation of ERK was not affected. The chronic treatment of phorbol ester, which down-regulated the activity of protein kinase C (PKC), did not affect the phosphorylation of ERK upon depolarization. In the presence of an inhibitor of EGF receptor, neither depolarization nor calcium ionophore increased the level of ERK phosphorylation. These data imply that the EGF receptor is functionally necessary to activate ERK and neurite outgrowth in response to the prolonged depolarization in PC12 cells, and also that PKC is apparently not involved in this signaling pathway.
