Enhancement of Antioxidant Activities of Curcuma longa Leaves by Ultra High Pressure Extraction (초고압 공정을 이용한 강황 잎 추출물의 항산화 활성 증진)

  • This study was designed to evaluate antioxidant activity of Curcuma longa L. leaves treated by ultra high pressure extraction. Curcuma longa L. leaves was subjected to 5,000 bar for 5 and 15 min at $25^{\circ}C$ The highest phenolics and flavonoids content was observed in the treatment of high pressure extraction for 15 min ($308.28{\mu}g/mg$, $124.33{\mu}g/mg$). The DPPH scavenging activity was 82.34% at $1.0mg/m{\ell}$ of Curcuma longa L. leaves treated by ultra high pressure process for 15 min. The highest SOD-like acitivity of Curcuma longa L. leaves ($1.0mg/m{\ell}$) was observed at ultra high pressure extraction for 15 min (67.54%). The high pressure extraction significantly increased the contents of phenolics and flavonoids and also enhanced the antioxidant activity. These results provide useful information for enhancing biological properties of Curcuma longa L. leaves.

Antioxidant and Antimicrobial Activities of Various Solvent Fractions of Fine Ginseng Root

  • This study was carried out to investigate the changes of yield, total phenolics, saponin content and composition, antimicrobial, and antioxidant activities of various fractions of fine ginseng root (Panax ginseng C.A. Mayer) by maceration method in the order of increasing polarity (hexane, chloroform, ethyl acetate, butanol, and water). Butanol fraction showed the highest total saponin content compare to other fractions. Hexane fraction could harvest significantly high ginsenoside Rg2, Rg1, and Rf (p<0.05). And the contents of ginsenoside Rh1, Rg3, and Rg1 showed relatively higher in the fraction of ethyl acetate than other fractions. The system of hexane-chloroform-ethyl aceate-butanol showed relatively high content of ginsenoside Re, Rd, Rc, Rb3, and Rb1. However, the last fraction of water still remained lots of Rb2 content. The fraction of water was the highest phenolics. The 1,1-diphenyl-2-picryhydrazil, superoxide, and hydroxyl radical scavenging activity of water fraction was higher than the other fractions. In antimicrobial activity, the fraction of hexane showed relatively high antimicrobial activity against Pseudomonas aeruginosa, Salmonella typhimurium, Staphylococcus aureus, Bacillus cereus, and Escherichia coli. And the fractions of the chloroform and ethyl acetate showed higher antimicrobial activities than the other samples in against P. aeruginosa and S. typhimurium.

  • The objective of this study was to evaluate the antioxidant and physiological The objective of this study was to evaluate the antioxidant and physiological activities of organic and inorganic coffee based on flavonoid composition. Organic and inorganic coffee varieties were prepared with Brazil arabica bean and Columbia arabica bean, respectively. Color values (Lightness, redness, and yellowness), mineral contents, total phenol content, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, individual flavonoids, and sensory characteristics were then measured in the espresso of each coffee bean. The pH, Brix, and color difference between organic coffee and inorgainc coffee did not correlate with the harvest method. Coffee varieties were more influential on physiological properties, and were markedly more influential than the mineral content. Total phenolics had a higher correlation with DPPH radical scavenging activity ($r^2$=0.832). Depending on the harvesting method, significant differences (p<0.05) were shown in aroma, color, and overall acceptability in sensory evaluation.

  • This study aimed to investigate the biological and antibacterial activities of phenolics from Sambucus sieboldiana var. pendula leaf extract. The phenolic contents of the S. sieboldiana var. pendula extracted with water and ethanol was 11.60 and 12.39 mg/g, respectively. The inhibitory effects of the water and ethanol extracts on the angiotensin converting enzyme (ACE) were 71.94-92.08 and 48.42-78.33%, respectively, and on xanthine oxidase (XOase) were 42.11-100 and 35.61-100%, respectively, at a phenolic concentration of 50-200 ㎍/mL. All these effects were found to be concentration-dependent. Additionally, the leaf extracts exhibited excellent antibacterial activities against Streptococcus mutans, Staphylococcus aureus, and Propionibacterium acnes. Hence, S. sieboldiana var. pendula was confirmed to have excellent antihypertensive, antigout, and antimicrobial properties.

Effect of particle size of naked oat flours on physicochemical and antioxidant property (쌀귀리 가루의 입도별 이화학적 특성 및 항산화 활성)

  • This study was carried out to investigate the effect of particle size of naked oat flour (NOF) on physicochemical property and antioxidant activity. The NOF was passed through $250{\mu}m$ and $160{\mu}m$ size sieves to obtain three fractions (fraction A: $250{\mu}m$ or more, fraction B: $160-250{\mu}m$, and fraction C: $160{\mu}m$ or less). Moisture, crude protein, crude fat, and crude ash contents of NOF were 8.4, 15.7, 10.0, and 1.8%, respectively, and these contents were increased as the particle size of NOF decreased. The mineral and free amino acid contents of NOF also had a similar tendency. The contents of total starch, amylose, starch damage, total dietary fiber, ${\beta}$-glucan, total phenolics, and flavonoids in NOF were 56.4%, 21.4%, 11.7%, 11.0%, 4.7%, $237.8{\mu}g/g$ and $90.9{\mu}g/g$, respectively. As the particle size of NOF decreased, total starch, amylose, and starch damage contents increased, whereas total dietary fiber, ${\beta}$-glucan, total phenolic and flavonoid contents decreased. Also, three antioxidant activities of NOF were closely correlated with their total phenolics and flavonoids contents, showing high correlation coefficient values ($R^2=0.87$ and 0.81, respectively).

Changes in nutritional components and antioxidant activities from soybean leaves containing high isoflavone contents according to different storage temperatures and periods (고 이소플라본 함유 콩잎의 저장 온도와 기간에 따른 영양학적 성분과 항산화 활성 변화)

  • This study investigated that change of the nutrients (including fatty acids, amino acids, and minerals) and total phenolic (TP), total flavonoid (TF), and isoflavone contents and antioxidant activities during the storage of soybean leaves containing high isoflavone contents at 5, 25, and 55 ℃ for 180 days. The contents of free amino acids were increased from 1313.81 mg/100 g at 0 day to 1776.15, 1693.93 and 1551.18 mg/100 g at 5, 25, and 55 ℃ storage for 180 days, respectively, but the values of fatty acids were little unchanged. The contents of minerals were detected from 51.65 mg/100 g (0 day) to 49.93 (5 ℃), 50.20 (25 ℃), and 61.21 (55 ℃) mg/100 g at 180 days, respectively. The levels of total isoflavones did not change during the storage periods. In case of storage at 55 ℃, the contents of glycosides (1347.78→2195.13 ㎍/g) and aglycones (342.79→480.61 ㎍/g) increased during storage, while the levels of malonylglycosides (2209.22→1289.48 ㎍/g) decreased. Also, the TP and TF contents were slightly increased from 9.31 and 8.61 mg/g at 0 day to 9.97 and 9.3 mg/g at 180 days, corresponding to the radical scavenging activities of 2,2-diphenyl-1-picrydrazyl, 2,4,6-azino-bis (3-ethylbenzthiazoline-6-sulphnoic acid), and hydroxyl increased from 30.91, 55.98 and 23.27% from 37.10, 62.54, and 30.95%, respectively.

The Effect of Plant Ethanolic Extracts on Oxidation of Soybean Oil (다류원류 식물류의 에탄올 추출물이 대두유의 산화에 미치는 영향)

  • This study was planned to investigate the effect of 40 plant ethanolic extracts on antioxidant activities in vitro. The total phenolics, $\beta$-carotene, $\alpha$-tocopherol and selenium contents were also determined . Antioxidant activities fo the ethanolic extracts(0.02%, w/w) in the soybean oil were measured both by determining the peroxide value (POV) during 35 days of storage at 4$0^{\circ}C$ in a forced draft air-incubator and by determining changes in conductivity at 11$0^{\circ}C$(Rancimat method.). Soybean oil without any additives was used as a control and that treated with 0.02% BHT was used as a positive control. Based on the POV determination, green tea extract was found to be the most effective in stabilizing soybean oil, then followed by long tea, which both of them showed higher antioxidant activities compared to the BHT treatment. The antioxidant activities of them showed higher antioxidant activities compared to the BHT treatment. the antioxidant activities of coffee, cinnamomi cortex, acanthopanacis cortex, black tea, orange peel , instant coffee, peony and crni fructus extracts were stronger compared to the control .By the Rancimat method, green tea leaf and oolong tea leaf, foxglove, acanthopanacis cortex and peony extracts. Compared to other extracts, green tea leaf, black tea leaf, foxglove, acanthopanacis cortex and peony extracts had stronger antioxidative effects in both the POV and Rancimat methods used in this study. ethanolic extracts which showed the stronger antioxidative effect also has the higher contents of total phenolics, $\beta$-carotene, and/or $\alpha$-tocopherol. The antioxidative effect of ethanolic extracts was found to be due to the combined effect of various antioxidants.

Sensuous Characteristics and Physiological Activity of Cheongtaejeon Tea Produced with the Inoculation of Microbial Strains (균주를 접종하여 제조한 청태전 차의 관능적 특성과 생리활성 효과)

  • This study was conducted to restore our traditional Cheongtaejeon tea and to develop the special products. We inoculated Cheongtaejeon tea with lactobacilli (Lactobacillus plantarum CHO 25) and the mixed microbial strains (L. plantarum CHO 25 + Saccharomyces cerevisiae + Bacillus amyloliquefaciens CHO 104). We also examined the sensuous characteristics and physiological activity of Cheongtaejeon tea which was produced by the inoculation of microbial strains. The external appearance of Cheongtaejeon teas were not significant among the teas which were produced with or without the inoculation of L. plantarum CHO 25 and the mixed microbial strains. The taste of the tea increased most in Cheongtaejeon tea which was produced without the inoculation of microbial strains. The taste and liking of Cheongtaejeon tea which was inoculated with Aspergillus oryzae and Aspergillus niger decreased significantly, and it was not suitable to drink. Total phenolics compound contents, total flavonoid contents and DPPH ($\alpha,\alpha$-diphenyl-$\beta$-picryl-hydrazyl) radical scavenging activity of Cheongtaejeon tea extracts increased much more in the order of that produced with the inoculation of L. plantarum CHO 25, control and that with the mixed microbial strains. However, nitrite radical scavenging activity in 1,000 mg/L Cheongtaejeon tea hot water extracts were in the order of the control (94.4%), the inoculation of L. plantarum CHO 25 (93.6%) and the mixed microbial strains (91.1%). Overall results indicated that the sensuous characteristics increased most in Cheongtaejeon tea which was produced without the inoculation of microbial strains, and those physiological activities in tea with the inoculation of L. plantarum CHO 25.

Antioxidant Activity and Cytotoxicity of Different Taraxacum Species in Korea (국내 자생 민들레의 종류별 항산화성 및 세포독성 연구)

  • Contents of phenolics and flavonoids, antioxidant activity and cytotoxicity were investigated in the methanol extracts of three different $Taraxacum$ species, $Taraxacum$ $coreanum$, $Taraxacum$ $mongolicum$, and $Taraxacum$ $officinale$. Total phenolics content at $1000mg\;kg^{-1}$ was more present in shoot parts than in roots, and was highest in $T.$ $mongolicum$ shoot and root extracts (76.8 and $40.0mg\;kg^{-1}$, respectively), followed by $T.$ $coreanum$ and $T.$ $officinale$ ($p$ < 0.05). Total flavonoid level had same tendency to total phenolics among $Taraxacum$ species, showing lower amounts ($6.5{\sim}36.4mg\;kg^{-1}$) than total phenolics. The antioxidant activity of the methanol extracts from all the species dose-dependently increased. DPPH free radical scavenging activity at $1,000mg\;kg^{-1}$ was highest in shoot and root extracts from $T.$ $mongolicum$ by 89.6 and 83.4%, respectively. According to MTT assay, cell viability of Calu-6 (human pulmonary carcinoma) was lowest in the $T.$ $mongolicum$ shoot and root extracts ($IC_{50}$ values=83.4 and $66.4mg\;kg^{-1}$, respectively), and followed by $T.$ $coreanum$ and $T.$ $officinale$ (lowest). Calu-6 was more sensitive to the extracts than SNU-601 (human gastric carcinoma). Antioxidative and anticancer activities in three different $Taraxacum$ species was more correlated with total phenolics content ($r^2$=0.0097 to 0.6213) than with total flavonoids level ($r^2$=0.0027 to 0.4627). The results showed total phenolics content and total flavonoids level were highly correlated with anticancer activity and antioxidant activity, and their content and activities were different depending on species.

Effect of Hot Water Boiling and Autoclaving on Physicochemical Properties of American Ginseng (Panax quinquefolium L.)

  • This study evaluates changes in the chemical composition and bioactivities of American ginseng (Panax quinquefolius L.) processed by boiling in water, $75^{\circ}C$ for 10, 20, 30, and 40 min, and autoclaving at high temperatures, $115^{\circ}C$ for 30 and 60 min and $130^{\circ}C$ for 90 and 120 min. Total ginsenoside contents of boiled ginseng remained relatively unchanged, whereas the contents of autoclaved ginseng samples significantly decreased with an increase of both time and temperature. Compared to unheated ginseng (control), the color of both boiled and autoclaved ginseng decreased in lightness and increased in redness. The acidic polysaccharide contents, the total phenolic contents and the antioxidant capacity of boiled and autoclaved ginseng were higher than the untreated ginseng, with the highest values being exhibited by the autoclaved samples. In particular, the antioxidant capacity of unheated ginseng increased about 2.5 times ($285.7{\pm}14.03\;mg$/100g to $777.2{\pm}26.4\;mg$/100g) when ginseng was autoclaved at $130^{\circ}C$ for 120 min as compared to the control. It was concluded that as American ginseng was processed at a high temperature, especially steam-heated in an autoclave, its chemical constituents changed and, in particular, acidic polysaccharides, total phenolics and antioxidant capacity were considerably increased.
