  • The aqueous ethanolic leaf extract of Ailanthus altissima was found to contain the new natural product, $luteolin\;7-O-{\beta}-(6"-galloylglucopyranoside)$, 13, along with fourteen known phenolic metabolites (1-12, 14 and 15). Structures of all compounds (1-15) were established by conventional methods of analysis and confirmed by FAB-MS, $^1H-\;and\;^{13}C-NMR$ spectral analysis.

  • Physiological characteristics of brown rice and black rice were investigated to provide the fundamental information of physiological property of rice and to show the potential of rice as a functional ingredient. Bioactive compounds were extracted from brown and black rices with aqueous solvents like 80% ethanol. Total phenolics, flavonoids and antioxidative capacity of brown and black rices' extracts were determined. Brown rice showed much higher amount of total phenolics and flavonoid contents as well as antioxidative capacity than those of milled rice indicating that most of bioactive compounds are located in the bran layer. Black rice showed higher total phenolics and flavonoid contents and antioxidative capacity than those of brown rices. The highest antioxidative capacity was obtained from Heugjinju followed by Heugseol, Sinnongheug-chal, Hopum and Samkwang. This result indicated that antioxidative capacity is affected by total phenolics and flavonoid contents. Both brown and black rices contained higher amount of ferulic acid than that of p-coumaric acid.

  • Improvements in the analytical methodology used in the gas chromatographic/mass spectral analysis of phenolic compounds from cigarette smoke are described. For the direct analysis of crude samples, pyridine extraction and the glass capillary column GC was used for the separation of phenolics as trimethylsilyl derivativatives. The separations of cigarette smoke on Carbowax 20M and SE-54 wall coated open tubular columns are given. Improved methodology for the routine quantitation of the identified components using the computer-controlled multiple ion selection technique of MS presented. Considerations pertaining to routine analyses of a multitude of complex smoke samples are also discussed.

Antithrombotic Phenolics from the Stems of Parthenocissus tricuspidata Possess Anti-inflammatory Effect

  • In the course of our program to search for antithrombotic and anti-inflammatory agents from plants, twelve phenolics (1-12) were isolated from the stems of Parthenocissus tricuspidata. Their structures were elucidated on the basis of spectroscopic (1D and 2D NMR, and MS) data analyses, and comparison with published data. At the concentration of $100{\mu}g/ml$, compounds 2, 4, 6 and 10 possessed potential effects on anti-blood coagulation, with inhibitory percentage of 216, 174, 148 and 225%, respectively; while aspirin used as positive control showed 181% inhibition at the same concentration. Furthermore, the anti-inflammatory activity of isolated compounds (1-12) was investigated on lipopolysaccharide (LPS)-induced murine macrophage cells (RAW264.7). Compounds 2, 4 and 6 also potential inhibited the production of nitric oxide, with $IC_{50}$ values of $11.9{\pm}0.3$, $2.9{\pm}0.2$ and $29.0{\pm}0.6{\mu}M$, respectively. Celastrol, the positive control used, gave an $IC_{50}$ value of $1.0{\pm}0.1{\mu}M$.

Phenolic compounds and antioxidant activity of berries produced in South Korea

  • Berries are rich sources of phenolic compounds, which are known to have health-promoting effects. In this study, phenolic compounds of seven popularly consumed berries were analyzed by HPLC-ESI-MS/MS. In addition, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and nitrite scavenging activities were investigated for comparison of the biological activity of the berry extracts. Chlorogenic acid was the major phenolic acid, which had the highest content in aronia, followed by blueberry. The rutin and isoquercetin contents were the highest in mulberry, followed by aronia, black raspberry, and blueberry. The anthocyanin content was the highest in black raspberry. Aronia showed the highest DPPH free radical and nitrite scavenging activities with the highest contents of total phenolics and proanthocyanidins.

Amino Acid and Phenolic Contents in lilfected Leaves of Rice in Relation to Adult - Plant Resistance to Leaf Blast (잎도열병에 대해 성체식물저항성을 지닌 벼의 감염엽에서 아미노산과 페놀화합물의 함량)

  • Ethanol-soluble amino acids and phenolics in healthy and blast-infected leaves of the susceptible rice cultivar Nakdong and adult-plant-resistant cultivar Dobong were quantitatively analyzed, At the 3 days after inoculation, the levels of soluble amino acids and phenolics in the infected fifth and eighth leaves of the two cultivars ere similar to those of comparable healthy controls. As blast lesions appeared on the leaves at the 5 days after inoculation, the soluble amino acids and phenolics began to increase. At the 7 days after inoculation, the levels of amino acids and phenolics were about 1.5-3 times more than those in healthy controls at the five-and eight leaf stages. The adult-plant-resistant cultivar Dobong showed higher amounts of soluble amino acids and phenolics in both healthy and infected fifth and eighth leaves than did the susceptible cultivar Nakdong, although Dobong was less infected by Pyricularia oryzae than Nakdong, The pronounced increases in amino acids and phenolics in rice leaves of the cultivar Dobong during the blast infection may play an important role in the expression of adult-plant resistance to blast.

Beauty Food Activities of Isolated Phenolic Compounds from Tetragonia tetragonioides (번행초(Tetragonia tetragonioides)로부터 추출한 Phenolic Compounds의 미용 식품 활성)

  • This study examined the beauty food activities of water and ethanol extracts from Tetragonia tetragonioides. Content of phenolic compounds extracted with water and 50% ethanol extracts were 3.29 mg/g and 4.14 mg/g, respectively. 1,1-Diphenyl-2-picrylhydrazyl free radical scavenging activities of water and ethanol extracts were 98.45% and 91.20%, respectively, at $200{\mu}g/mL$ of phenolics. 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical decolorization activity was 97.28% for water extracts and 97.83% for ethanol extracts at $100{\mu}g/mL$ of phenolics. Antioxidant protection factor (PF) was 1.77 PF for water and ethanol extracts at $200{\mu}g/mL$ of phenolics. Thiobarbituric acid reactive substances of water and ethanol extracts were 94.77% and 95.64%, respectively, at $100{\mu}g/mL$ of phenolics. Tyrosinase inhibitory activity, which is related to skin-whitening, was confirmed to be 34.96% for ethanol extracts at $200{\mu}g/mL$ of phenolics. Elastase inhibitory activity and anti-wrinkle effect of 50% ethanol extracts were 78.9% at $200{\mu}g/mL$ of phenolics. Collagenase inhibitory activity of ethanol extracts was 61.29% at $200{\mu}g/mL$ of phenolics. Astringent effect was not detected in water extracts but was 7.82% for 50% ethanol extracts at $200{\mu}g/mL$ of phenolics. Hyaluronidase inhibitory activity as a measure of anti-inflammation was confirmed to be 81.04% for water extracts at $200{\mu}g/mL$ of phenolics. Based on these results, Tetragonia tetragonioides extracts can be used as a functional material and functional beauty food with antioxidant effects.

Determination of Phenolic Compounds in Adzuki bean (Vigna angularis) Germplasm

  • The aim of this study was to evaluate 30 phenolic compounds in adzuki bean germplasm. Adzuki 21653 had the highest content of total phenolics compounds ($6597\;{\mu}g\;g^{-1}$) while 104372 had the lowest concentration. The average total phenolic content of Japanese ($2432\;{\mu}g\;g^{-1}$) adzuki beans was higher than that of Korean ($2256\;{\mu}g\;g^{-1}$) adzuki beans. The average total phenolic contents were $2507\;{\mu}g\;g^{-1}$ in small sized adzuki beans from Japan and $2459\;{\mu}g\;g^{-1}$ in those from Korea. In large sized adzuki beans, the average total phenolic contents were $1315\;{\mu}g\;g^{-1}$ in Japanese seeds and $1232\;{\mu}g\;g^{-1}$ in Korean seeds. The average total phenolic contents in medium seeds were $2369\;{\mu}g\;g^{-1}$ in Japanese adzuki beans and $1397\;{\mu}g\;g^{-1}$ in Korean ones. In small seeds, the total phenolic contents of adzuki beans varied from $524\;{\mu}g\;g^{-1}$ to $6597\;{\mu}g\;g^{-1}$ in Japanese ones and from $375\;{\mu}g\;g^{-1}$ to $6569\;{\mu}g\;g^{-1}$ in Korean ones. Japanese and Korean adzuki beans were divided into landraces and wild adzuki beans. In this study, the wild adzuki beans showed higher contents of total phenolics than the native varieties. Specifically, the wild adzuki beans from Korea had the highest concentration of phenolics ($3403\;{\mu}g\;g^{-1}$). All adzuki bean germplasms were measured for their color and were classified into four groups accordingly: A; L < 30, +a, +b; B; L < 30, +a, -b, C; L > 50, +a, +b, D; L > 50, +a, -b. Especially, group B had the highest concentration of total phenolic compounds ($2827\;{\mu}g\;g^{-1}$), whereas group C had the lowest concentration ($1882\;{\mu}g\;g^{-1}$).

Effects of aging on the phenolics content and antioxidant activities of rose flower (Rosa hybrida L.) extracts (숙성조건이 장미꽃 추출물의 페놀화합물(phenolics) 함량과 산화방지 활성에 미치는 영향)

  • Rose flower is widely used in the preparation of tea and contains a large variety of phytochemicals, including phenolics such as catechin, quercetin, and rutin. The effects of aging on rose (Rosa hybrida L.) flower extracts (RFE) were examined under conditions of varying temperature and relative humidity. The total phenolic content, antioxidative activity, and catechin levels were measured to evaluate the effects of temperature and relative humidity on the aging process. Performing the aging process at $30^{\circ}C$ under 60% or 90% relative humidity for 24 h significantly increased the total phenolic content and the antioxidant activities of RFE (p<0.05). Additionally, an aging process performed at $30^{\circ}C$ and 60% relative humidity for 24 h maximized the extraction rate of phenolics such as catechin and consequently led to increased antioxidative activity of RFE. In summary, this study indicates that the extraction rate of physiologically active phenolic compounds in rose flower can be increased by performing an aging process under optimized temperature and relative humidity conditions.

Beauty Food Activities of Isolated Phenolic Compounds from Ulmus pumila (유근피(Ulmus pumila)로부터 분리한 페놀성물질의 미용식품활성)

  • Phenolic compounds of $17.9{\pm}1.0mg/g$ were extracted from Ulmus pumila with 70% ethanol. The elastase inhibitory activity related with forming wrinkle was shown an excellent wrinkle improvement effect in 70% ethanol extracts as 55.5-69.5% at phenolic concentration of $50-200{\mu}g/mL$. The tyrosinase inhibitory activity related with skin-whitening was 24% in 70% ethanol extracts at phenolic concentration of $200{\mu}g/mL$. The astringent activity of 70% ethanol extracts was shown activity of 71% at phenolic concentration of $200{\mu}g/mL$ therefore it is judged that there is a high effect on pores reduction of the skin. The hyaluronidase inhibitory activity of U. pumila extracts was confirmed anti-inflammation effect of 80% at phenolic concentration of $50{\mu}g/mL$. Antimicrobial activity of U. pumila water extracts was shown each 8.7, 10.0, 11.1 and 11.8 mm clear zones on Propionebacterium acnes at phenolic concentration of $50-200{\mu}g/mL$. The stability of the multi-functional cosmetic (lotion) added U. pumila extracts was very stable for 28 days without changing of pH and viscosity also it's stable on temperature and sun lights. As the concentration of extracts was increased, the color of lotion was getting dark, but the sensory evaluation was high at score of 8.5.
