The stem barks, heartwoods, and leaves of Acer tegmentosum (Aceraceae) are widely used in Korea to treat hepatic or cerebral disorders mainly due to alcohol poisoning. This study was aimed to analyze phenolic substances in A. tegmentosum. Quantitative analysis of the three phenolic substances (salidroside, (+)-catechin and scopoletin) was performed by HPLC and the identification of volatile phenolic substances were done by GC-MS. The contents of the three compounds in the three MeOH extracts were higher in the stem bark (salidroside: 80.22 mg/g, (+)-catechin: 23.31 mg/g, and scopoletin: 9.45 mg/g) compared to the heartwoods and leaves. And GC-MS analysis of the stem bark extract demonstrated that p-tyrosol is a main substance of twenty-one compounds identified.
This study was performed to determine the composition of phenolic substances contained in the leaves of Cirsium setidens (Compositae), validate the high-performance liquid chromatography (HPLC) method, and determine the in vivo sedative effect of the main component pectolinarin. Six phenolic compounds isolated from C. setidens were spectroscopically identified as chlorogenic acid (1), hyperoside (2), 3,4-di-O-caffeoylquinic acid (3), caffeic acid methyl ester (4), linarin (5), and pectolinarin (6) and then used as standard compounds for HPLC analysis. HPLC proved to be precise, accurate, and sensitive for the simultaneous analysis of the phenolic substances. In particular, six compounds showed good regression ($R^2$ > 0.999) within test ranges and recovery was in the range of 95.4 - 104.8%. The content of pectolinarin was considerably higher (156.48 mg/g) than those of other phenolic substances including the other flavone glycoside, linarin (18.99 mg/g). The contents of other phenolic substances, in order, were chlorogenic acid (8.41 mg/g), 3,4-di-O-caffeoylquinic acid (5.74 mg/g), hyperoside (4.33 mg/g), and caffeic acid methyl ester (0.51 mg/g). Oral administration with compound 6 (10 and 20 mg/kg) enhanced the sleeping time induced by pentobarbital in mice, indicating that it has a sedative effect.
The antioxidant activity and the pharmacologically active constituents such as nucleosides and phenolic substancs of Panax ginseng C.A. Meyer (white ginseng powder) treated with gamma irradiation (Rl group) and ethylene oxide fumigation (EO group) were compared with those of non-treated ginseng (control group). High performance liquid chromatograms of nucleosides in three groups were similar with each other. The bathochromic effect of phenolic substances at W absorption region tends to be higher for EO group than Rl group. This tendency was also observed in heat treatment of each group. However, the antioxidant activity may have some effects on the labile property of phenolic substances. In summary the gamma-irradiated ginseng was more stable than the ethylene oxide-treated ginseng, but both ginseng samples gave the similar antioxidant activity.
A on-line SPE (solid phase extraction)-HPLC preconcentration method was developed for the determination of phenolic compounds at trace levels in environmental water sample. XAD-4 and Dowex 1-X8 were used as sorbent in the on-line SPE-HPLC method for the selective enrichment of nine phenolic compounds, which are included in the priority pollutants list of the US EPA. Also alumina prefiltering considerably reduced the amount of interfering peaks due to humic substances that could accumulated due to the preconcentration step and prevent quantification of polar phenolic compounds in environmental water samples. This method was used to determine the phenolic compounds in tap and river water and superiority to the US EPA 625 method in its enrichment factor, pretreatment time, recoveries, and detection limit. The limits of detection were in the range of $0.3-0.9\;{\mu}g/L$ in tap water sample.
This experiment was conducted to detect the presence of allelopathic substances in the leaves of Ginkgo biloba L. Water extracts from G. biloba leaves which collected at different season markedly inhibited the germination and growth of O. sativa, E. crus-galli, D. sanguinalis, and L. sativa, indicating the presence of biological substances. Linolenic and palmitic acid were the major fatty acids of G. biloba leaves. The biochemical substances such as salicylic arid, p-coumaric acid, catechol, hydroquinone, orchinol, ferulic acid, phloroglucinol, and umbelliferone etc., belonging to the phenolic, compounds were, detected in a large amount, which may be responsible for exhibition inhibitory effects. The common phenolic compounds were detected in the early-harvested and late-harvested G. biloba leaves were salicylic and p-coumaric acid. All these compounds were related to the allelopathic activities in G. biloba leaves.
Phenolic compounds from 7 species of naturalized invader species and Korean wild plants were analyzed by high performance liquid chromatography. Eleven phenolic compounds including benzoic acid were identified. The extract of naturalized plants was significantly more inhibitory to seed germination and seedling growth of the both naturalized and Korean wild plants. The content of total phenolic compounds in each extract were 43.5 mg/l in Ailanthus altissima as the maximum amount and 25.5 mg/l in Phytolacca americana as the minimum. Phytotoxic substances of ethanol extracts was investigated for antifungal activity against 23 selected fungus species. The antifungal activity of Phytolacca americana showed the greatest clear zone of 23 mm in Aspergillus awamori and its activity had an effect against 6 fungus species. Ailanthus altissima formed the greatest clear zone of 26 mm in Erwinia carotovora sub. sp. carotovora and had an effect agsinst 2 fungus species.
Phenolic compounds such as gallic acid, ferulic acid, p-hydroxybenzoic acid, vanillic acid, salicylic acid, tannic acid, and hydroquinone were identified from the aqueous extracts and volatile substances of tomato plant by paper chromatography, high performance liquid chromatography and gas chromatography. The seed germination and seedling growth of the experimental species, lettuce and egg plant, were severely inhibited in 5$\times$10-3M of phenolic reagents identical to those identified from tomato plant. Germination and growth rate of test species in 5$\times$10-4M and 5$\times$10-5M were higher than that of 5$\times$10-3M. Therefore, 5$\times$10-3M of phenolic compounds would be assumed to be threshold concentration for allelopathic effects.
KDICical substances from Artemisia scoparia were analyzed by gas chromatography. Seven phenolic compounds and thirty nine terpenoids were identified. Most abundant within each group were cinnamic acid and camphor, respectively. The KDICicals were prepared as aqueous extracts and then used for germination, growth, and chlorophyll content tests. The extracts were inhibitory to germination and seedling growth of the receptor lants. This inhibitory effect was dependent on concentration. When the effect of the aqueous extract on chlorophyll content was assayed, both chlorophylls a and b were shown to be reduced. The reduction in seedling elongation and growth in dry weight paralleled the reduction in chlorophyll concentration. These KDICical substances, including phenolic compounds and terpenoids, from Artemisia scoparia were responsible for the growth inhibition of the selected species.
This study was conducted to investigate the presence of biologically active substances such as phenolic acids, fatty acids and organic acids in the medical plants like Kalopanax pictum and Acanthopanacis cortex. Alcohol extracts of K. pictum and A. cortex showed complete inhibition of lettuce seed germination, indicating that these plants contained the biologically active substances. Eleven phenolic acids including protocatechuic acid were identified from K. pictum and A. cortex by GLC, and the contents of total phenolic acid were 1.7917mg/g in K. pictum, and 0.9567mg/g in A. cortex. Polyphenols such as neochlorogenic acid, chi orogenic acid, scopoletin, rutin and kaempferolglycoside which were not detected by GLC were anayzed by HPLC, and among phenolic acids indentified chi orogenic acid seemed to be the major acid in both K. pictum and A. cortex presented in amount of 23.7 and 13.0ppm, respectively. K. pictum contained 5.26mg/g of fatty acids and 27.69mg/g of organic acids, and A. cortex possessed 3.22mg/g of fatty acids and 9.80mg/g of organic acids, linoleic and oxalic acid appeared to be the major fatty and organic acids, representing more than 50% of total fatty acids and 80% of total organic acids.
Lactuca raddeana (Compositae) is used to treat obesity and complications due to diabetes. The five phenolic compounds including chlorogenic acid, chicoric acid, luteolin 7-O-glucoside, luteolin 7-O-glucuronide, luteolin were qualitatively identified by LC-ESI-MS analysis. The contents were quantitatively determined by HPLC, under the condition of a Capcell Pak C18 column ($5{\mu}m$, $250mm{\times}4.6mm\;i.d.$) and a gradient elution of 0.05% trifluoroacetic acid (TFA) and 0.05% TFA in $MeOH-H_2O$ (60 : 40). The contents of chicoric acid (100.99 mg/g extract) and luteolin 7-O-glucoside (101. 69 mg/g extract) were high, while those of other three phenolic substances were very low. The 3T3-L1 adipocyte cells treated with chicoric acid and luteolin 7-O-glucuronide significantly suppressed the accumulation of fat, suggesting they are effective against obesity. Since high level of peroxynitrite (ONOO) causes cardiovascular disease in obese patients, its scavenging activity was also studied.
