Taxonomic Position and Affinities of Isopyrum mandshuricum within Korean Isopyroideae (Ranunculaceae) Based on Molecular Data

  • To examine the taxonomic position and affinities of Isopyrum mandshuricum (Ranunculaceae) and related taxa, genetic analysis were carried out on the basis of isozyme patterns and ITS sequences. Molecular data, both isozyme patterns and ITS sequences suggest that I. mandshuricum is closely related to Enemion raddeanum than to Semiaquilegia adoxoides. The estimation of genetic identities by isozyme analysis reveals that I. manshuricum is genetically distant from E. raddeanum. The phylogenetic tree based on molecular data is rather congruent with the phenogram based on quantitative morphological characteristics, but not consistent with one based on qualitative morphological characteristics. Incongruencies between molecular and qualitative morphological data provide clues to re-evaluate several morphological features.

Genetic variation and relationship of Artemisia capillaris Thunb.(Compositae) by RAPD analysis

  • Randomly Amplified Polymorphic DNA (RAPD) was performed to define the genetic variation and relationships of Artemisia capillaris. Fifteen populations by the distributions and habitat were collected to conduct RAPD analysis. RAPD markers were observed mainly between 300bp and 1600bp. Total 72 scorable markers from 7 primers were applied to generate the genetic matrix, and 69 bands were polymorphic and only 3 bands were monomorphic. The genetic dissimilarity matrix by Nei's genetic distance (1972) and UPGMA phenogram were produced from the data matrix. Populations of Artemisia capillaris were clustered with high genetic affinities and cluster patterns were correlated with distributional patterns. Two big groups were clustered as southern area group and middle area group. The closest OTUs were GW2 and GG1 in middle area group, and GB1 from southern area group was clustered with OTUs in middle area group. RAPD data was useful to define the genetic variations and relationships of A. capillaris.

제주도산 감귤속 식물의 성분 분류학적 연구 (Chemotaxonomic Studies on the Citrus Plants cultivated in Je Ju Island)

  • A thin-layer chromatographic study was made of the chloroform-soluble and flavonoid fractions from the fruit peels of 16 species, 2 varieties and 5 formas of the Citrus plants cultivated in Je Ju Island for their interspecific relatinships. In addition, 3 hybrids and 9 native plants were also studied for their taxonomic position. Three phenograms were developed from these chromatographic data after cluster analysis via the unweighted paired group method using rithmatic average by Sneath and Sokal. These plants were grouped into 5 alliences based on the phenogram obtained from the chloroform-soluble fracitons, which were nearly identical to the subgenus rank by Tanaka, and rutinoside and neohesperidoside groups by Horowitz. Those from the flavonoid and methanol-soluble fractions were not able to evaluate the morphological classification except for a few cases.

Genetic Relationships and Phylogeny of the Asplenium antiquum Makino (Aspleniaceae) and its relative species based on RAPD Analysis

  • This study characterized the genetic variations of 13 populations of Asplenium antiquum and its relative species using randomly amplified polymorphic DNA (RAPD) markers. A total 88 scorable RAPD bands were generated by the 12 random oligo primers and were analyzed by Nei and Li's genetic distance. High genetic variability was detected between A. antiquum and A. nidus, with the range from 0.568 to 0.682. And slightly low genetic variations showed within the populations of same species. Seven populations of A. antiquum showed slight differences (0.000-0.216), and five populations of A. nidus showed similar low genetic variations (0.114 to 0.171). Two individuals from Sup-seom Island which are growing in might be the regenerated one from abroad. A. antiquum were clustered as two groups (Group I, Group II) by UPGMA phenogram. And five populations of A. nidus were clustered as two groups correlated with geographical distribution. The RAPD data was very useful to define the genetic variations and to discuss the phylogenetic relationships among A. antiquum and the related species..

Genetic relationships among penicillium species by characterizing RAPD markers

  • Random amplified polymorphic DAN markers were characterized for three taxonomically problematic Penicillium species : P. aurantiogriseum var. Aurantiogriseum, P. verrucosum and P. puberulum, as well as for 25 species of mono, bi-, and terverticillate Penicillia. The relationships among mono, bi-, and terverticillate Penicillium species were determined from these RAPD markers. Eight species from mono-, eight from bi-, and nine from terverticilate Penicillia were examined. With 14 randomly chosen 10-mer primes, a 310 character by 25 species matrix was generated. Phenetic analysis separated the 25 species into three genetically distinct groups that correspond to the different arrangements of penicilli (mono-, bi-, and terverticillate). The results of this study suggest that P. aurantiogriseum var. aurantiogriseum, P. VERRUCOSUM, AND P. puberulum represent genetically distinct species, and that P. vulpinum should be included in terverticilate Penicillia. Phenogram branching patterns indicated that biverticillate species are genetically more similar to monoverticilate species than they are to terverticillate species.

RAPD를 이용한 고추냉이의 유연관계 분석 (Intraspecific Genetic Relation of Wasabia japonica Matsum. Based on RAPD Analysis)

  • 국내에서 육성 또는 외국에서 도입되어 재배되고 있는 고추냉이 (Wasabia japonica Matsum.) 7품종의 10개체와 울릉도 자생 1개체, 총 11개체에 대한 유연관계 분석을 위하여 RAPD분석을 실시하였다. RAPD분석에 사용한 random primer는 총 50종류였으며 screen 결과 21종류만이 11품종 전체에서 반응을 보였다. 21종류의 primer로부터 관찰된 밴드는 총 144개였으며 이중 다형화 (polymorphism)를 보이는 앤드는 68개 (47.2%)로 primer 한 개당 평균 3.2개의 다형화 밴드를 보였다. Primer별 밴드의 수는 $2{\sim}13$개로 다양하게 나타났고 평균 밴드의 수는 6.8개였다. 유집분석 결과 phenogram은 유사도지수 $0.81{\sim}0.96$의 높은 범위 내에서 11개체들이 단계통군으로 유집되었으나 품종내 개체간에는 뚜렷하게 유집되지 않았다. 울릉도에 자생하는 개체의 경우는Sayatori와 Himenisiki 품종을 제외한 나머지 8개체를 위한 자매군으로 유집되었다. 이러한 결과는 neighborjoining 분석에서도 같은 경향을 보였다. 따라서 고추냉이의 종내 유연관계 분석을 위한 방법으로 RAPD법은 유용하지 않은 것으로 나타났으며 좀더 정확한 유연관계분석을 위해서는 AFLP방법이나 계통분류에 널리 이용되는 핵, 업록체 DNA의 유전자 염기서열 비교와 같은 정밀한 분자계통학적 연구가 수행되어져야 할 것으로 사료된다.

Wood anatomy of Korean Symplocos Jacq. (Symplocaceae)

  • Despite poorly documented species delimitation and unresolved taxonomic nomenclature, four species of Symplocos (S. coreana, S purnifolia, S sawafutagi, and S. tanakana) have been described in Korea. In this study, we carried a comparative wood anatomy analysis of all four species of Korean Symplocos to understand the wood anatomical variations among them. The results of this study indicated that Korean Symplocos are comparatively indistinguishable in terms of their qualitative wood features, except for exclusively uniseriate rays present in S. purnifolia instead of uniseriate to multiseriate in other three species. Nevertheless, differences are noticed in quantitative wood variables such as the vessel density, vessel size, and ray density. The vessel density of S. purnifolia is more than twice as high as those of S. sawafutagi and S. tanakana. In contrast, the vessel circumference and diameter on both plants of S. sawafutagi and S. tanakana is nearly twice as large as those of S. purnifolia. Symplocos coreana has characteristic intermediacy between these two groups in terms of vessel features and is closer to S. purnifolia in terms of its ray density level. A cluster analysis based on a paired group (unweighted pair-group method with the arithmetic mean, UPGMA) algorithm using the Euclidean similarity index clearly differentiates S. purnifolia from the remaining species, representing the first branch of the phenogram.

Aroclor 1254 May Induce Common DNA Effects in Developing Paralichthys olivaceus Embryos and Larvae

  • Polychlorinated biphenyls (PCBs) are persistent pollutants in aquatic environments, often causing the decline or disappearance of wild populations. In this study, we used a random amplified polymorphic DNA (RAPD) assay to evaluate the effects on the genomic DNA of olive flounder embryo and larval stages of exposure to Aroclor 1254 at concentrations of 1, 5, 10, 20, and $40{\mu}g/L$. We compared RAPD fingerprints of exposed and non-exposed samples. Polymorphisms were revealed as the presence and/or absence of DNA fragments between the two samples. A dose-dependent increase in the number of polymorphic bands was observed with Aroclor 1254 treatment. Also, RAPD profiles of animals exposed to Aroclor 1254 exhibited an increase in the frequency values (FV) compared to the control. A phenogram constructed using neighbor-joining method indicated that genomic template stability in developing embryo and larval stages was significantly affected at ${\geq}5{\mu}g/L$. This study suggested that DNA polymorphisms detected by RAPD analysis could be used as an investigative tool for environmental toxicology and as a useful biomarker in early life stages for the detection of potential genotoxicants.

Genetic Variations and Phylogenetic Relationships of Tribe Forsythieae (Oleaceae) Based on RAPD Analysis

  • RAPD analysis was performed to discuss the taxonomic status and phylogenetic relationships among the tribe Forsythieae and related groups. Two hundred and eighteen scorable polymorphic bands were detected from fourteen oligonucleotide primers. From the results of RAPD analysis by Nei and Li's genetic distance, each individuals of Abeliophyllum distichum showed high genetic relationships with ranging from 0.085 to 0.301, also the genus Forsythia showed from 0.042 to 0.655 among the species and populations. But, Abeliophyllum and Forsythia showed distinct dissimilarity, ranging from 0.610 to 1.258. And genetic differences among the population of Forsythia were 0.042 in F. koreana, 0.275 in F. saxatilis, 0.275 in F. ovata, 0.279 in F. nakaii, and 0.249 in F. viridissima. The UPGMA phenogram of tribe Forsythieae based on the results of RAPD analysis were presented that Abeliophyllum is distinct genus different from Forsythia. NJ tree which applied as the outgroups Fontanesia and Jasminum was derived, and it showed that tribe Forsythieae might be a monophyletic group. The genus Fontanesia was showed as sister group of tribe Forsythieae. Among the populations of taxa in Forsythia, F. koreana and F. saxatilis were more closely related, and F. ovata and F. nakaii were very closely related to F.japonica. And Fontanesia was the sister group of tribe Forsythieae.

Analysis of the Genetic Relationship among Mulberry (Morus spp.) Cultivars Using Inter-Simple Sequence Repeat (ISSR) Markers

  • Mulberry (Morus spp. family: Moraceae) has prime importance in the sericulture industry, and its foliage is the only natural feed of the silkworm Bombyx mori L. Traditional classification methods using morphological traits were largely unsuccessful in assessing the diversity and relationships among different mulberry species because of environmental influences on the traits of interest. For these reasons, it is difficult to differentiate between the varieties and cultivars of Morus spp. In the present study, inter-simple sequence repeat (ISSR) markers were used to investigate the genetic diversity of 48 mulberry samples genotyped using nine ISSR primers. The ISSR markers exhibited polymorphisms (53.2%) among mulberry genotypes. Furthermore, similarity coefficient estimated for these ISSR markers was found to vary between 0.67 and 0.99 for the combined pooled data. The phenogram drawn using the UPGMA cluster method based on combined pooled data of the ISSR markers divided the 48 mulberry genotypes into seven major groups. No genetic association was found in the collection area, and there was a mixed pattern between the mulberry lines. The hybridization between different mulberry species is highly likely to be homogenized due to natural hybridization.
