A Further Study on the Inhibition of Tumor Growth and Metastasis by Red Ginseng Acidic Polysaccharide (RGAP)

  • We have recently reported that red ginseng acidic polysaccharide (RGAP), isolated from Korean red ginseng (Panax ginseng C. A. Meyer), showed immunomodulatory antitumor activities, mainly mediated by nitric oxide (NO) production by macrophage. In this study, we examined the effect of RGAP on anticancer activity using lung carcinoma 3LL, sarcoma 180 and adenocarcinoma JC tumor cells transplanted into mice as well as antimetastatic activity using B16-F10 melanoma. When RGAP (300 mg/kg) were treated to mice implanted with one of the three kinds of tumor cells, the tumor weight significantly decreased compared with control mice. Tumor inhibition ratios of RGAP (300 mg/kg) in mice transplanted with lung carcinoma 3LL, sarcoma 180 and adenocarcinoma JC cells were 26.8%, 29.3% and 31.6%, respectively. Hundred mg/kg of RGAP did not cause a significant decrease in tumor weight compared with control group. When RGAP was administered i.p. with the dose of 100 and 300 mg/kg in B16-F10 melanoma-bearing mice, lung metastasis were reduced significantly in mice. Corrected phagocytic index was also remarkably increased by RGAP. These results suggest that stimulation of phagocytic activity of macrophages may be a mechanism for in vivo anticancer and antimetastasis activities of RGAP.

Enzyme Hydrolysates of Ginseng Marc Polysaccharides Promote the Phagocytic Activity of Macrophages Via Activation of TLR2 and Mer Tyrosine Kinase

  • Although ginseng marc is a by-product obtained during manufacturing of various commercial ginseng products and has been routinely discarded as a waste, it still contains considerable amounts of potential bioactive compounds, including saponins and polysaccharides. Previously, we reported that ginseng oligosaccharides derived from ginseng marc polysaccharides by enzymatic hydrolysis exert immunostimulatory activities in macrophages and these activated macrophages are in turn able to inhibit the growth of skin melanoma cells by inducing apoptosis. In the present study, a more detailed investigation of the immunostimulatory activity and underlying action mechanisms of an enzymatic hydrolysate (GEH) containing these oligosaccharides derived from ginseng marc polysaccharides was performed. The levels of proinflammatory cytokines and anti-inflammatory cytokines were measured in GEH-stimulated RAW264.7 macrophages using RT-PCR analysis and ELISA. The expression levels of Toll-like receptor 2 (TLR2) and TLR4, Dectin-1, and MerTK were measured by RT-PCR analysis or western blot analysis, and the phagocytic activities of GEH-challenged bone marrow-derived macrophages toward apoptotic Jurkat cells were assayed using fluorescence microscopy. GEH induced the production of both proinflammatory cytokines $TNF-{\alpha}$ and IL-6, and anti-inflammatory cytokine IL-10 in RAW 264.7 cells. The expression of the TLR2 and MerTK mRNAs was increased upon GEH treatment. Phagocytosis of apoptotic Jurkat cells was enhanced in GEH-treated macrophages. Based on the results, this enzymatic hydrolysate (GEH) containing oligosaccharides exerts immunostimulatory effects by maintaining the balance between M1 and M2 cytokines, facilitating macrophage activation and contributing to the efficient phagocytosis of apoptotic cells. Therefore, the GEH could be developed as value-added, health-beneficial food materials with immunostimulatory effects.

Nuclear factor kappa-B- and activator protein-1-mediated immunostimulatory activity of compound K in monocytes and macrophages

  • Background: Compound K (CK) is a bioactive derivative of ginsenoside Rb1 in Panax ginseng (Korean ginseng). Its biological and pharmacological activities have been studied in various disease conditions, although its immunomodulatory role in innate immunity mediated by monocytes/macrophages has been poorly understood. In this study, we aimed to elucidate the regulatory role of CK on cellular events mediated by monocytes and macrophages in innate immune responses. Methods: The immunomodulatory role of CK was explored by various immunoassays including cell-cell adhesion, fibronectin adhesion, cell migration, phagocytic uptake, costimulatory molecules, reactive oxygen species production, luciferase activity, and by the measurement of mRNA levels of proinflammatory genes. Results: Compound K induced cell cluster formation through cell-cell adhesion, cell migration, and phagocytic activity, but it suppressed cell-tissue interactions in U937 and RAW264.7 cells. Compound K also upregulated the surface expression of the cell adhesion molecule cluster of differentiation (CD) 43 (CD43) and costimulatory molecules CD69, CD80, and CD86, but it downregulated the expression of monocyte differentiation marker CD82 in RAW264.7 cells. Moreover, CK induced the release of reactive oxygen species and induced messenger RNA expression of proinflammatory genes, inducible nitric oxide synthase, and tumor necrosis factor-alpha by enhancing the nuclear translocation and transcriptional activities of nuclear factor kappa-B and activator protein-1. Conclusion: Our results suggest that CK has an immunomodulatory role in innate immune responses through regulating various cellular events mediated by monocytes and macrophages.

새우 껍질에서 추출한 키토산의 항암 및 면역활성 (Antitumor and Immunologic Activity of Chitosan Extracted from Shell of Shrimp)

  • 수산물 폐기물을 이용하기 위하여 대롱 새우껍질에서 추출하여 만든 키토산을 마우스에 이식한 sarcoma 180을 이용하여 in vivo와 in vitro에서 항암 효과 및 면역기능에 미치는 영향에 대하여 실험하였다. 키토산의 sarcoma-180에 대한 고형암 성장 저지율을 40mg/kg투여시 63.84%의 저지율을 나타내었다. 수명 연장효과는 대조군에 비하여 키토산을 40mg/kg 투여하였을 때 26.84%로 그다지 높지 않았다. In vivo에서의 항암효과를 근거로 하여 in vitro에서는 sarco-ma-180에 대한 직접적인 세포의 독성 작용은 거의 없었다. 키토산은 30mg/kg과 40mg/kg을 투여시 혈중 백혈구수와 총복강세포수가 대조군에 비하여 다소 증가 하였고 장기인 간, 비장 및 흉선의 무게도 투여한 용량에 따라 증가하는 용량 의존성 반응을 보였다. 항체 유도염증 반응인 항체매개형 과민반응과 지연형 과민반응은 족척의 두께가 키토산에 의해 거의 정상 수준까지 회복되었다. Macrophage의 식작용에 미치는 영향은 phagocytic index 와 corrected phagocytic index에서 거의 차이가 없었고, 용혈성 반응 세포수(plaque forming cell, PFC)는 대조군에 비하여 PFC/$10^{7}$ spleen의 경우 18.88%와 31.83%로 다소 증가하였다.

경구투여 $\beta$-glucan이 잉어와 넙치의 비특이적 면역활성에 미치는 영향 (Effects of $\beta$-glucan from Schizophillum commune on Non-specific Immune Parameters in Common Carp (Cyprinus carpio) and Flounder (Paralichthys olivaceus) by Oral Administration)

  • [ $\beta$ ]-glucan을 시판의 배합사료에 $0.1\%$ 첨가한 다음 담수어인 잉어 (평균체중 1.0g 및 68.7g)와 해산어인 넙치 (평균 체중, 12.1 g 및 54.0g)에 2주간 첨가사료, 1주간 무첨가사료, 2주간 첨가사료 투여의 투여방법으로 5주간 경구투여한 다음 인위감염에 대한 생잔율을 조사하였다. 또 투여기간중의 말초혈액의 백혈구수와 혈중리소자임의 활성과 5주간 투여후에 채취한 백혈구의 식작용능을 조사하였다. $\beta$-glucan첨가사료 투여군에 $1\times10^6$cells의 Aeromonas hydrophila를 접종한 평균체중 68.7g의 잉어군과 $1\times10^5$cells의 Edwardsiella tarda를 접종한 평균체중 68.7g의 넙치군에서 $\beta$-glucan첨가사료 투여군이 무첨가투여군에 비해 생존율이 높았고 (P<0.05),그 외의 접종군에서는 무첨가투여군과 차이가 없었다. $\beta$-glucan첨가사료 투여기간 동안 말초혈액중의 마크로파아지와 호중구수는 투여기간에는 증가, 무투여기간에는 감소하는 경향이었으며 특히 마크로파아지의 증가가 현저하였다. 혈중 리소자임은 잉어에서는 $\beta$-glucan첨가사료 투여기간에는 상승하고 무투여기간에는 감소하였지만, $\beta$-glucan첨가사료를 다시 투여한 후 급격히 상승하여 높은 활성을 유지하였다 그러나 넙치는 잉어보다는 활성의 증가는 현저하지 않았고 무투여기간에도 변동이 없었지만 무첨가투여군보다는 항상 높은 활성을 나타내었다.

Conjugated Linoleic Acid를 급여한 돼지의 호중구 탐식능 증강 (Supplement of Conjugated Linoleic Acid Increases Neutrophil Phagocytosis in Pigs)

  • CLA를 급여시킨 돼지에 있어 말초 혈액 세포의 변화와 탐식세포의 탐식반응을 조사하였다. 100일령, 평균체 중 80kg인 거세 수퇘지에게 10t-l2c CLA와 CLA mixture를 각각 사료 중에 5% 첨가하여 총 4주간에 걸쳐 자유급여 시켰다. 급여 전 (0주), 급여 2주후, 4주후 그리고 급여 중지 후인 5주차에 각각 혈액세포 수치(PCV 백혈구수 및 백혈구 감별계산치)의 변화와 탐식세포의 탐식활성을 측정하였다. CLA의 급여 따른 PCV의 수치에 있어서는 아무런 변화가 없었다. 그러나 10t-l2c CLA 또는 CLA mixture가 함유된 사료를 급여한 돼지에 있어서 CLA를 급여하지 않은 돼지에 비해 백혈구수 특히 호중구 수가 현저히 증가하였다. 말초혈액 단핵구세포(mononuclear cells; MNC)와 다형핵백혈구(polymorphonuclear cells; PMN)의 탐식능를 유세포분석기로 분석한 결과, 10t-l2c CLA 또는 CLA mixture 급여와 상관없이 MNC와 단구세포들(monocyte-rich cells)의 탐식능에는 변화가 없었다. 그러나 PMN의 탐식 활성은 CLA 무급여 대조돼지들에 비해 CLA 급여군은 2주에서 5주차에 걸쳐 증가된 탐식능를 나타내었다. 이상의 결과로부터 돼지에 CLA 급여는 호중구수 증가와 호중구 탐식능 증가를 유도할 수 있음을 제시하였으며 향후 생체방어 증강을 위해 CLA를 임상적으로 투여할 수 있을 것으로 보인다.

Inhibitory Effect of Ginsenoside-Rp1, a Novel Ginsenoside Derivative, on the Functional Activation of Macrophage-like Cells

  • Ginsenoside Rp1 (G-Rp1) is a ginseng saponin derivative with chemopreventive and anti-cancer activities. In this study, we examined the regulatory activity of G-Rp1 on the functional activation of macrophages. G-Rp1 remarkably inhibited TNF-$\alpha$ production, LPS-induced cell cytotoxicity, NO production, ROS generation, and phagocytic uptake from lipopolysacchride (LPS)-activated RAW264.7 cells. According to structural feature study using several G-Rp1 analogs, two carbohydrates (glucose-glucose) at R1 position were observedto be highly effective, compared to other structural derivatives. Although the inhibitory activities of G-Rp1 on macrophage functions were not remarkable, several points that G-Rp1 was known to be safe, and that this compound was orally effective, suggest that G-Rp1 may be beneficial in treating macrophage-mediated immunological diseases.

Effects of Non-Saponin Red Ginseng Components (NSRG) on Functions of Macrophages Isolated from Young and Aged Mice

  • Macrophages play an important role in the first line of immunologic effects against tumor cells. The effects of nonsaponin red ginseng (NSRG) components on macrophage functions like tumoricidal activity, phagocytic activity, and NO production in young (8-weeks-old) and aged (82-weeks-old) male C57BL/6 mice were assessed in vitro, respectively. The treatment of tumor cells (melanoma B16 cells) with the supernatants of NSRG-treated macrophages resulted in an increase of cytotoxicity at 300 $\mu$g/ml in the aged mice, whereas the supernatants did not have a cytotoxic effect in the young mice. It was observed that the supernatants induced the increase of tumor cell proliferation at 150 $\mu$g/ml in the young mice, suggesting that the supernatants contain growth factors rather than cytotoxic molecules. In addition, NSRG alone had a direct cytotoxic effect on the B16 tumor cells. NSRG had no effect on the NO production by the macrophages in the young mice, while it significantly increased the level of NO release in the aged mice. There was no difference in the phagocytic activities of the macrophages by NSRG in both groups of mice. These results suggest that NSRG has differential effects on the macrophage functions in young and aged mice.

흰쥐의 복강내 세포, 특히 비만세포의 식작용 및 복굴절성에 관한 분석 (Analysis of Phagocytosis and Birefringence in the Peritoneal Cells of the Rat, with Special Regard to the Mast Cells)

  • 1. 10種染料 (neutral red, toluidine blue, pyronin, methylene blue, alcian blue, trypan blue, carmine, orange G, aniline blue, Janus green B, and India ink)로 處理된 흰쥐의 腹腔內細胞 特히 肥滿細胞의 식作用 및 複屈折性의 有無를 位相差顯微鏡法 및 偏光顯微鏡法으로 分析하였으며 附隨的으로 cytomorphic change 도 觀察하였다. 2. 腹腔內 肥滿細胞는 大部分 染料에 對하여 아무런 식作用을 보이지 않았으며 India ink 群에서 輕微한 식作用을 보였다. 大식細胞와 各種 白血球는 大體로 旺盛한 식作用을 나타내었다. 3. 腹腔內 肥滿細胞는 大部分의 染料를 處理하여도 아무런 複屈折性을 보이지 않았으며 trypan blue 와 aniline blue 處理群에서 매우 輕微한 反應을 보였을 뿐이었으며 白血球는 全然 反應을 보이지 않았다. 4. 各種染料에 對한 肥滿細胞의 細胞形態學的 反應은 一定하지 않았으며 大體로 輕微한 變化를 일으켰다. 白血球의 同染料에 對한 反應은 特히 trypan blue, orange G, India ink群에 서 顯著하였다.

보양약류(補陽藥類)의 면역약리학적(免疫藥理學的) 고찰(考察) (Immunopharmacologic studies about Drugs for Tonifying Yang)

  • To understand immunopharmacologic effects on Cervi Pantotrichum Cornu, Morindae Officinalis Radix, Cistanches Herba, Curculginis Rhizoma, Epimedii Herba, Eucommiae Cortex, we investigated chinese experimental documents, and we could reach conclusions as follows : 1. The effects on cell-mediated immune system were as follows. 1) The effects on macrophage (1) The herbal medicines promoting to increase the number of WBC in the peripheral blood were Morindae Officinalis Radix, Epimedii Herba and that promoting to reinforce the phagocytic functions of neutrophil was Curculginis Rhizoma. (2) The herbal medicines promoting the phagocytic functions of mononuclear, macrophage were Cervi Pantotrichum Cornu, Cistanches Herba, Eucommiae Cortex. 2) The herbal medicines stimulating the activities of T lymphocytes were Cervi Pantotrichum Cornu, Curculginis Rhizoma, Epimedii Herba, Eucommiae Cortex. 2. The effects on humoral immune system were as follows. 1) The herbal medicines increasing the activity of complement receptor were Cervi Pantotrichum Cornu, Curculginis Rhizoma. 2) The herbal medicines reinforcing immunity of spleen cells were Cervi Pantotrichum Cornu, Cistanches Herba, Epimedii Herba. 3) The herbal medicines promoting proliferation of spleen cells that produce antibody after having been immunized by SRBC were Cervi Pantotrichum Cornu, Cistanches Herba, Epimedii Herba. 3. The herbal medicines, reinforcing immunity on delayed type hypersensitivity were Cervi Pantotrichum Cornu, Cistanches Herba, Eucommiae Cortex. As you know in the many bibliological documents, the studies on the effects of Drugs for Tonifying Yang were started along right lines. Recently the studies on those were accomplished more rapidly and applied many immune diseases. We thought that Drugs for Tonifying Yang could be important immunopotentiators. Therefore we can apply those herbal medicines not only to immune diseases but also inflammatory diseases, senile infirmity and all sorts of tumor.
