Anti-Helicobacter pylori Activity of Bifidobacterium spp.

  • The inhibitory effects of different Bifidobacterium spp. on the growth of Helicobacter pylori (HP) were investigated. A significant suppression of HP growth occurred only when HP was inoculated onto a petri dish containing 0.1 mg/ml of Bifidobacterium spp. When HP was separately cultured with B. breve K-110, B. catenulatum K-309, B magnum K-311, B. magnum K-321, and B. cuniculi K-513, the urease activity was also inhibited by these Bifidobacterium spp. Therefore, it appears that these Bifidobacterium spp. excrete a heat-labile inhibitory component for HP growth into the culture medium. Although most organic acids produced by the Bifidobacterium spp. inhibited the growth of HP, the HP growth was not inhibited by the physiological concentrations of organic acids produced in bifidobacteria-cultured media. Accordingly, these results suggest that some Bifidobacterium spp. may produce antibiotic-like compounds (bacteriocins).

  • An image processing algorithm was developed for a robot system which was used in gene study. The robot system achieved a job of colony picking. The colony included DNA of an organism. The robot picked up the colony in petri-dish, which included the cultivated colony in medium, by a picking pin, and moved the colony to wellplates. The vision system consisted of an image acquisition system which acquired the image information of colony, an illumination device which irradiated the object once when it got the image of it, a computer and so on. The image processing algorithm distinguished the colony and detected colony positions. Performance test of the developed algorithm showed that the distinguishing success rate of colony and detecting success rate of colony positions were over 96%.

Ecological Characteristics of Alatae and Apterae of the Green Peach Aphid, Myzus persicae(Sulzer) (Homoptera : Aphididae), on Tobacco Plants (복숭아혹진딧물 유시충과 무시충의 생태적 특성)

  • The green peach aphid was relatively the majority(36.9-72.3% ) among all alate aphids collected from tobacco plants. Density of glatae was relatively lower on varieties Tl 1112 and Br 21 than on Va 115 or Y S A among seven varieties tested. However, no considerable difference was observed in general biology of nymphs produced by alatae on the tobacco varieties. About 55% of alate green peach aphid collected from tobacco fields produced progency, and over 80% of the nymphs became adults on cut tobacco leaves in petri dish. Adult longevity of both alate and apteral green peach aphid was shorter, and numbers of nymphs produced by both types were lower in summer than in autumn. Apterae produced more nymphs than alatae, but the longevity of apterae was shorter.

  • In the conventional biology, the most of cell studies was carried out by culturing cells in the Petri dish and by investigating cellular behavior under the diverse bio-molecule (cell signalling materials, drugs or etc.) conditions. However, in vivo environments, diverse stimulations including chemical, mechanical and topological environments involved in the proliferation, differentiation and migration of cells and it is almost impossible to provide these conditions with traditional method. We have developed the methods to provide the well defined chemical and mechanical stimulations using microfluidic devices and applied these approaches to the study of environmental effect on cells. In this paper, we will introduce our microfluidic chips to provide microenvironment and its applications using several cells.

Culture Characteristics of Streptomyces spp. on Improved Polyacrylamide Gel and Agar Media

  • Application of polyacrylamide gel (PAG) instead of agar to solid cultures of Streptomyces spp. was studied. The improved media were prepared by 1) gelling 20 ml of 5% acrylamide in a glass petri dish at room temperature, 2) washing by running water for more than 8 hr to remove residual reaction reagents, 3) drying at 5$0^{\circ}C$ for 12 hr to make a gel film, 4) autoclaving at 121$^{\circ}C$ for 15 min, and 5) swelling gel for about 4 hr by adding sterile liquid medium. In PAG media there were no differences from the observation of morphological characteristics showing during the cellular differentiation on agar media, whereas the ability to utilize carbohydrates differed somewhat from agar media. Agar media thus were little favorable for biochemical tests which the growth was determined depending on the formation of colony, but washed PAG was superior to serve as a solidifying agent.

Effects of Aphelenchus avenae on Suppression of Soilborne Diseases of Ginseng (Aphelenchus avenae에 의한 인산 토양병의 억제효과)

  • The monoxenic culture of the fungivorous nematode, Aphelenchus avenae, was applied for the control of soil-borne ginseng pathogens such as Fusarium solani and Rhizoctonia solani. Fungivorous nematode populations were measured in a field to examine relationships between the nematode populations and suppression of ginseng root diseases. Inoculation of A. avenae (5000 nematodes per petri-dish) reduced the colonization of the Fusarium mycelium on root discs of ginseng and carrot by 80.0% and 60.5%, respectively. A. avenae also significantly reduced the occurrence of damping-off of ginseng by R. solani pathogenic to ginseng, and no plant damage by the nematode was noted. In a 3-year-old ginseng field infested with Cylindrocarpon destructans, plant missing caused by root rot positively correlated to the density of potato rot nematode, Ditylenchus destructor, but it was reduced with the population of A. avenae, suggesting that A. avenae might inhibit the occurrence of ginseng root rot.

  • A colony counting on a petri-dish is a laborious task in microbiology fields that automated counting systems are needed. But lots of such systems are high price that majorities of labs rely on the manual counting and it is a time consuming and laborious job. In this study, an attempt was made to select the relative atmospheric correction method for the water quality factors extraction from the satellite image data. And also, the time-series analysis of the water quality factors was performed by using the multi-temporal image data. The result shows that the proposed method can be commercialized with low prices.

Plant Regeneration from Turnip (Brassica rapa ssp. rapifera) Organs

  • Shoot induction system was developed in the recalcitrant plant species, Brassica rapa ssp. rapifera by using optimum selection of profit organ, phytohormone combination, seedling age and kind of culture container. Out of in vitro cultured leaf segment, petiole, hypocotyl, and cotyledon with petiole, only cotyledon with petiole derived from 4 day-old seedlings induced multiple shoot. The optimum combination of auxin and cytokinin for the multiple shoot induction was MS medium containing 5mg/L BA and 0.5mg/L NAA. The major factors for multiple shoot propagation were part of plant organ, age of seedling, and ratio of auxin and cytokinin. In addition, shoot regeneration was promoted in the 100ml Erlenmeyer flask compared with the $90mm{\times}20mm$ Petri-dish. The induced shoots formed roots easy on MS medium containing 0.1mg/L IBA and the whole plants were successfully cultivated in soil.

Airborne Microorganisms of Some Underground Shops in Pusan (부산시 일부 지하상가 공기중 부유 균주)

  • This study was investigated to evaluate the distribution of airborne microorganisms at underground shops in Seomyun and Nampodong, Pusan. The number of bacterial colonies on the nutrient agar plate plates were calculated by the open petri dish method for 30 minutes in indoor air of underground shops at every seasons in a year. There was no statistically significant difference between Seomyun and Nampodong in mean colony counts. Isolation rates of Gram positive rods was highest, and Gram positive cocci and Gram negative rods were followed. In Nampodong underground shops, Enterobacteriaceae strains was isolated. Mean colony counts according to seasons was higher at summer and autumn in Seomyun, and spring and winter in Nampodong. In near future, a study on the distribution of bacteria causing respiratory infection should be followed.
