Characterization of Symptom and Determination of Tissue Critical Concentration for Diagnostic Criteria in 'Maehyang' Strawberry (Fragaria$\times$ananassa Duch.) as Influenced by Magnesium Concentrations in the Fertigation Solution ('매향' 딸기의 마그네슘 영양진단을 위한 결핍증상 및 식물체 내 한계농도)

  • This study was carried out to investigate the effect of magnesium (Mg) concentrations in nutrient solution on growth of and nutrient uptake by 'Maehyang' strawberry. Tissue analysis based on dry weight and petiole sap were also conducted to determine the threshold level in plants when Mg deficiency disorders developed in strawberry plants. In the Mg deficient plants, the spotted yellowing or yellowing area developed on the interveinal area of mature leaves. The dark brown color also developed on the interveinal area of mature leaves with marginal browning or marginal necrosis. The response in dry weight production of 'Maehyang' strawberry to elevated Mg concentrations in nutrient solution was quadratic and the equation is y=6.84+1.7533x-$0.9278x^2$ $(R^2=0.1081^{***})$. But the Mg contents in tissue increased lineally with the equation of y=0.1764+0.1275x $(R^2=0.8307^{***})$. The trends of fresh weight production and Mg concentrations in petiole sap were also quadratic (y=24.127+7.3565x-$1.125x^2$, $R^2=0.2314^{^***}$) and linear (11.954+5.793x, $R^2=0.6869^{***}$), respectively. To prevent growth suppression, the Mg concentrations based on dry weight of above ground tissue and in petiole sap should be in the range of 0.30 to 0.65% and 19 to $40mg{\cdot}kg^{-1}$, respectively, fer the commercial production of 'Maehyang' strawberry.

Effect of Plant Growth Regulators, Media and Celling Agents on In Vitro Microtuber Production of Pinellia ternata Breit (1식물생장조절제, 배지와 고형지지물이 반하의 기내 소괴경 생산에 미치는 영향)

  • The study was carried out to establish in vitro microtuber production from leaf and petiole explant cultures of Pinellia ternata. Culture conditions were optimized by investigating the effect of plant growth regulators, different media, and gelling agents on the efficiency of microtuber indurtion. Among the different combinations of plant growth regulators tested, the combination of 0.1 mg/l 2,4-Dichlorophenoxyacetic (2,4-D) and 0.5 mg/l 6-benzylaminopurine (BAP) showed the highest yield fer microtuber production from leaf (3.9) and petiole (4.7) explant culture on MS medium far 6 weeks. SH(Schenk and Hildebrandt) medium was the most effective medium for microtuber induction and the half strength SH medium was better than SH medium for microtuber production from both leaf and petiole culture. Gelrite was better than agar in the formation of microtubers and 4% Gelrite showed the highest number of microtubers per explant frome leaf (5.9) and petiole (7.8) culture. Germination rate of microtubers after cold storaged for one months long was 86% from in vitro culture and 43% from autoclaved soil.

Rapid Nutrient Diagnosis of Tomato by Test Strips and a Chlorophyll Meter (Test strip과 chlorophyll meter를 이용한 토마토의 신속한 영양진단)

  • This study was performed to develop a more rapid and simple nutrient diagnosis method of plants than the conventional leaf analysis method. tomato (Lycopersicon esculentum Mill. cv. super momotaro) was planted in the mixed media produced by mixing perlite and rock wool at 1 . 1 (v/v) ratio. The Yamazakki nutrient solutions for cucumber and tomato were supplied to the media using the micro-drip irrigation system. Experimental plots produced consisted of no fertilization, deficient fertilization, adequate fertilization and surplus fertilization for N, P and K, respectively. Each experimental plot was replicated four times. Specific color different sensor values (SCDSV) measured by the chlorophyll meter were closely related to total-N concentrations in the leaves measured by the conventional method. Nitrate, $PO_4$ and K concentrations in petiole sap measured by test strips #(Reflectoquant^{\circledR},\;Merck,\;Germany)$ showed a significant relationship with total-N, p and K concentrations in leaves. The linear regression equations between $NO_3,\;PO_4$ and K concentrations in petiole sap and total-N, p and K concentrations in the leaves were prepared. The optimum levels of $NO_3,\;PO_4$ and K in petiole sap were obtained by plugging the optimum concentrations of total-N, p and K in the leaves by other researchers into the equations. In conclusion, the SCDSV by the chlorophyll meter and concentrations of NO3, p and K in petiole sap measured by the test strips would be suitable for rapid estimation of plant nutrient status.

Plant Regeneration from the Segments of Petioles of Cacalia firma (병풍쌈의 엽병 조직 절편으로부터 식물체 재분화)

  • Cacalia firma recently has been used increasingly as leaf vegetables but endangered in natural forest. In this work, we established the plant regeneration via adventitious shoot formation from petiole segments of seedling and in vitro plantlets. Wounding of seed coats and $GA_3$ treatments were effective to induce in vitro germination of seeds, whereas, seed did not germinate at all without these treatment. When cotyledon, leaf, petiole, and root segments of seedling were cultured on medium with 2 $mg{\cdot}L^{-1}$ benzyl adenine (BA) and 0.5 $mg{\cdot}L^{-1}$ naphthaleneacetic acid (NAA), petiole segments showed highest number of shoots per explant among the other segments. Among the various kinds of cytokinins, BA, isopentyl adenine (2-ip), kinetin, zeatin, thidiazuron (TDZ), TDZ and BA treatments were effective to induce high frequency of adventitious shoot formation from petiole segments of in vitro propagated plants. NAA stimulated the frequency of adventitious shoot formation but not for number of adventitious shoots per explants compared to TDZ or BA treatment alone. Most of adventitious shoots were developed directly from surfaces of explants. Adventitious shoots were transferred on medium with IBA for root formation, thereafter the plantlets were successfully transferred to soil.

Breeding of ‘Gommany’ Cultivar of Gomchwi with Disease Resistant, High Quality, and Yield (고품질 내병 다수성 곰취 신품종 ‘곰마니’ 육성)

  • A new Gomchwi cultivar ‘Gommany’ was bred by crossing between Gomchwi (Ligularia fischeri (Ledeb.) Turcz.) and Handaeri-gomchwi (Ligularia fischeri var. spiciformis Nakai). The selection and investigation of growth and yield characteristics were conducted from 2006 to 2011 in field and greenhouse of Highland Agriculture Research Institute, NICS, Rural Development Administration. The color of petiole ear was purple. Petiole trichome existed, but light of leaf back did not. Density of leaf vein was 5 degree in a newly developed cultivar ‘Gommany’. Plant height, leaf length, leaf width and petiole length were 47.7, 16.3, 21.2, and 31.4 ㎝, respectively in the 3 year of growth characteristics. Plant size was also higher than that of ‘Gondalbi’. Bolting and flowering time were Aug. 5 and Sept. 10, respectively. In contrast, Bolting time of ‘Gondalbi’ showed 25 days quite earlier bolting time compared to ‘Gommany’, Which led to earlier flowering time. ‘Gommany’ showed higher number of leaves (138) per plant compared to ‘Gondalbi’ (130). Furthermore, yield was 97% higher in ‘Gommany’ (1,575 g/plant) than in ‘Gondalbi’ (798 g/plant). ‘Gommany’ showed higher leaf thickness (0.69㎜) than ‘Gonalbi’ (0.46 ㎜), and consequently showed more hardness in leaf characteristics (24.3 ㎏/㎠) compared to ‘Gondalbi’ (23.0㎏/㎠). ‘Gommany’ showed higher resistance in the susceptibility of powdery mildew compared to ‘Gondalbi’. ‘Gommany’ cultivar was registered as a novel variety with number 90 in The Minister of the Korea Forest Service on December 2015.

The Variation of Leaf Form of Rare Endemic Berchemia berchemiaefolia Populations (희귀수종(稀貴樹種) 망개나무 자생집단(自生集團)의 엽형변이(葉型變異))

  • 7 populations of rare endemic Berchemia berchemiaefolia were analysed using multivariate analysis for 9 characteristics of leaf morphology. The aim of this study was to examine geographic variation to support a genetic resource conservation plan of B. berchemiaefolia in Korea. In the morphological characters, nine characters of leaf were 10.25 cm (blade length), 4.10 cm (maximum width), 2.52 (blade length/maximum width), 3.22 cm (upper 1/3 width), 3.42 cm (lower 1/3 width), 0.95 (upper 1/3 width/lower 1/3 width), 1.24 cm (petiole length), 8.91 (blade length/petiole length), 8.16ea (vein number), respectively. Nested anova showed that were statistically significant differences among populations as well as among individuals within populations in all 9 quantitative characters. In 7 of 9 characters, variance components among individuals within populations were higher than those among populations. Cluster analysis using complete linkage method showed two groups (Chungbuk and Gyeongbuk districts) to Euclidean distance 1.2. Among principal components, primary 3 principal components appeared to be major variables because of the loading contribution of 87.3%. The first contribution was blade length, blade length/maximum width and blade length/petiole length; the second one was maximum width, upper 1/3 width and lower 1/3 width; the third one was petiole length, respectively.

A Study on the Method of Identifying for Landscape Architecture (조경용 수목의 형태적 식별방법에 관한 연구)

  • A number of illustrated floras are generally used when we want to idenufy a name of a certain tree. This method, however, has not been efficient because it has taken a great deal of time and effort for persons who do not have much knowledge on the group of the plant. In contrast to this method, the use of taxonomical keys is very efficient for finding out the group of a certain plant. But this method Is very dirtficult for non-specialists to understand the taxonomical ternunology and thus has not been of general use. In thins context, this study was conducted to present the efficient way of the group of a certain tree through the computer database by using the brief forms of plant organs. The database consists of effect fields such as form of trunk and evergreen or deciduous, types of leaf and leaf arrangement, existence of price and hair on twig, form of leaf margin, leaf form and venation, existence of hair on leaf surface and petiole, length of leaf and petiole. The 222 species of tree used for landscape architecture were sorted by items of the eight fields. As a result, the 222 species were divided into 185 groups and the one group contains 7species, 2 groups contain 3 species, 27 groups contain 2 species and 155 groups contain only one species. Therefore it is suggested that the use of computer database and illustrated floras is very easy and efficient in identifying a tree.

High frequency plant regeneration from transverse thin cell layers in Indian mustard (Brassica juncea L.)

  • An efficient and reproducible plant regeneration system was established using transverse thin cell layers (tTCLs) in five cultivars of Brassjca juncea L. The effects of medium conditions, explant types (tTCLs of hypcotyl and cotyledonary petiole) on shoot regeneration were examined in this study. The maximum shoot regeneration frequency was obtained in Murashige and Skoog (MS) medium supplemented with 4 mg/L 6-benzylaminopurine (BA) and 0.2 mg/L 1-naphthaleneacetic acid (NAA). The hypocotyls derived tTCL explants had more shoot regeneration frequency (52%) than the cotyledonary petiole derived tTCL explants. Shoot induction was further improved by the addition of silver nitrate ($AgNO_3$) in the regeneration medium. A significant genotypic effect was also observed between the five cultivars; Rai-5 displayed higher capacities to produce shoots than other cultivars. Regenerated shoots were rooted on MS basal medium without PGRs which induced 90% of roots. The plantlets established in greenhouse conditions with 99% survival, flowered normally and set seeds. The regenerated plants were fertile and identical to source plants.
