산마늘의 아미노산 함량과 생리활성 효과 (Amino Acid Contents and Various Physiological Activities of Allium victorialis)

  • This study was conducted to gather the basic data on the alpine leek (Allium victorialis) for the expand of consumption and the production of its manufactured goods. Amino acid content in alpine leek leaves and various physiological activities were examined. Seventeen component amino acids and 38 free amino acids from alpine leek leaves were analyzed, and the total contents were 2,693.28 mg/100g for component amino acids and 535.39 mg/100g for free amino acids. Total phenolic compounds in the leaves of alpine leek showed the highest level from the methanol extract (37.7 mg/l), and followed by ethanol extract (31.9 mg/l) and hot water extract (25.4 mg/l). Total flavonoid contents in 1,000 mg/l extract was the highest in the methanol extract (22.2 mg/l). DPPH radical scavenging activity at 1,000 mg/l extract was high in the order of ethanol extract (51.6%), methanol extract (47.3%) and hot water extract (37.2%). nitrite radical scavenging activity Methanol extract from Allium victorialis leaves was the highest nitrite radical scavenging activity (79.5%). Hyperplasia suppression of lung cancer cells (Calu-6) and gastric cancer cells (SNU-601) by the methanol extract from the bulb of alpine leek were 99.9% in the extracting concentration of over 200 mg/l. No significant difference in antimicrobial activity among the 3 different solvents and extract concentrations was observed, and the inhibition zones against the gram-positive and negative microorganisms were ranged from 8.23 to 10.15 mm. It was concluded that physiological activities in a human body could be improved by the intake of alpine leek as a pharmaceutical material, and that it would be useful for the prevention of health risk such as lung and gastric cancers.

  • The study was conducted to obtain the genetic informations for some useful characters in tobacco breeding. The eight parents and a set of 28 crosses of F1's and F2's were used as materials, and planted on two different cultivated systems, i. e., oriental's and burley systems, during 1986-1988 at taegu Experiment Station, Korea Ginseng & Tobacco Research Institute. The observed characters were six agronomic characters which were plant height, number of leaves per plant, leaf length, leaf width, days to flowering, yield and five chemical components, nicotine, total nitrogen, protein nitrogen, total volatile base and pet, ether extract. The results obtaining are summarized as follows: 1. The higher heritabilities were found for days to flowering yield and nicotine in both generations, but values for number of leaves per plant, leaf length, total nitrogen and protein nitrogen were lower than other characters. 2. Genotypic correlation coefficients among all pairs of characters were slightly higher than the corresponding phenotypic correlations in F1's and F2's on two different cultivated systems. The relationship between leaf length and leaf width was the positive correlation, but that between number of leaves per plant and leaf width was negative. 3. From the genotypic correlations between yield and other characters, a conclusion that the yield was highly correlated with plant height, leaf length, leaf width and days to flowering was given. 4. Quality was positively correlated with number of leaves per plant and nicotine, but negatively with the other agronomic characters and chemical components.

수박 시설 재배에서 정식 전 심경로타리 처리가 토양 이화학성, 생육 및 과실 특성에 미쳐는 영향 (Effects of Deep Tillage before Planting on Physicochemical Properties of Soil, Growth and Fruit Characteristics in Cultivation of Watermelon under Plastic Film House)

  • 본 연구는 전북 고창의 수박 시설 농기에서 '우리꿀'을 대상으로 토양의 이화학성, 생육 및 과실품질에 대한 정식 전 심경로타리 처리의 효과를 알아보고자 수행하였다. 정식 전에 비해 재배 및 수확 후 토양 pH 변화는 거의 없었으나 토양 EC는 크게 떨어졌다. 유기물 및 유효인산 함량은 두 처리 모두에서 증가하였는데, 유효인산의 누적 함량이 심경로타리 처리구에서 적었다. 치환성 양이온도 Mg을 제외하고는 모두 감소한 경향이었는데 심경로타리 처리구에서 K의 감소가 뚜렷하였다. 수박의 정식 후 27일째 생육 특성 중 일반로타리 처리구에 비해 심경로타리 처리구에서 초장, 10절까지의 줄기 길이, 엽폭에서 유의하게 길었고, 1번 및 2번화의착과 절위가 낮았다. 그리고 육안 관찰 시 식물체 위조 증상 정도이 적었다. 수확기에는 일반로타리 처리구에 비해 심경로타리 처리구에서 30절까지의 줄기 길이가 유의하게 길었고, 육안 관찰 시 잎의 황변 현상이 적었다. 수확된 과실의 길이, 직경, 과피경도, 그리고 무게는 심경로타리 처리구에서 유의하게 높았다.

차광정도가 산마늘의 생육 및 엽록소 함량에 미치는 영향 (Effect of Shading Levels on the Growth and Chlorophyll Contents of Allium victorialis L. var. platyphyllum Makino)

  • 본 연구는 쌈채 및 강장제로 이용되고 있는 산마늘을 조경용 지피식물로 활용하고자 수행하였다. 차광 정도를 30%, 50%, 80%로 처리하여 생육 특성 및 엽록소 함량 등을 조사하였다. 엽수는 처리구별로 유의성이 인정되지 않았으나 엽장과 엽폭은 80% 차광 처리구가 각각 16.8cm, 10.1cm로 무처리구의 13.4cm, 7.3cm에 비해 우수하였다. 생체중은 무처리구가 차광 처리구에 비해 우수한 것으로 나타났으나 지상부 생체중은 무처기구가 8.5g인데 비해 80% 차광 처리구가 0.4g로 양호하였다. 엽록소 함량은 50% 차광 처리구가 42.8, 80% 차광 처리구가 40.5로 무처리의 36.7에 비해 높았다. 따라서 차광 수준에 따른 산마늘의 생육 특성은 80% 차광 처리구에서 가장 우수하여 음지성 지피식물로서 가치가 있는 것으로 나타났다.

Detection of viral infections in wild Korean raccoon dogs (Nyctereutes procyonoides koreensis)

  • Wild raccoon dogs (Nyctereutes procyonoides koreensis) may play a role transmitting several pathogens to humans and pet animals. Information concerning the incidence of rabies, canine distemper virus (CDV), canine parvovirus (CPV), canine adenovirus type 2 (CAdV-2), canine parainfluenza virus type 5 (CPIV-5), and canine herpesvirus (CHV) is needed in wild raccoon dogs. In total, 62 brain samples of raccoon dogs were examined for rabies virus (RABV) and CDV, and 49 lung samples were screened for CDV, CAdV-2, CPIV-5, and CHV. No RABV, CAdV-2, CPIV-5, or CHV was identified, but nine CDV antigens (8.1%, 9/111) were detected. Moreover, 174 serum samples from wild raccoon dogs were screened for antibodies against the five major viral pathogens. The overall sero-surveillance against CDV, CPV, CAdV-2, CPIV-5, and CHV in wild raccoon dogs was 60.3%, 52.9%, 59.8%, 23.6%, and 10.3%, respectively. Comparisons of the sero-surveillance of the five pathogens showed that raccoon dogs of Gyeonggi province have slightly higher sero-positive rates against CDV, CPV, and CHV than those of Gangwon province. These results indicate high incidences of CDV, CPV, and CAdV-2 in wild raccoon dogs of two Korean provinces and a latent risk of pathogen transmission to companion and domestic animals.

SSR 마커를 이용한 산양삼의 유전적 다양성 분석 (Genetic Diversity Analysis of Wood-cultivated Ginseng using Simple Sequence Repeat Markers)

  • Background: Panax ginseng C. A. Meyer is wood-cultivated ginseng (WCG) in Korea which depends on an artificial forest growth method. To produce this type of ginseng, various P. ginseng cultivars can be used. To obtain a WCG similar to wild ginseng (WG), this method is usually performed in a mountain using seeds or seedlings of cultivated ginseng (CG) and WG. Recently, the WCG industry is suffering a problem in that Panax notoginseng (Burk.) F. H. Chen or Panax quinquefolium L. are being sold as WCG Korean market; These morphological similarities have created confusion among customers. Methods and Results: WCG samples were collected from five areas in Korea. After polymerase chain reaction (PCR) amplification using the primer pair labeled with fluorescence dye (FAM, NED, PET, or VIC), fragment analysis were performed. PCR products were separated by capillary electrophoresis with an ABI 3730 DNA analyzer. From the results, WCG cultivated in Korea showed very diverse genetic background. Conclusions: In this study, we tried to develop a method to discriminate between WCG, P. notoginseng or P. quinquefolium using simple sequence repeat (SSR) markers. Furthermore, we analyzed the genetic diversity of WCG collected from five cultivation areas in Korea.

Characteristics of Spontaneous Combustion of Various Fuels for Coal-Fired Power Plant by Carbonization Rank

  • Spontaneous combustion propensity of various coals of carbonization grade as a pulverized fuel of coal-fired power plant has been tested from an initial temperature of $25^{\circ}C$ to $600^{\circ}C$ by heating in an oven with air to analyze the self-oxidation starting temperature. These tests produce CPT (Cross Point Temperature), IT (Ignition temperature), and CPS (Cross Point Slope) calculated as the slope of time taken for a rapid exothermic oxidation reaction at CPT base. CPS shows a carbonization rank dependence whereby wood pellet has the highest propensity to spontaneous combustion of $20.995^{\circ}C/min$. A sub-bituminous KIDECO coal shows a CPS value of $15.370^{\circ}C/min$, whereas pet coke has the highest carbonization rank at $2.950^{\circ}C/min$. The nature of this trend is most likely attributable to a concentration of volatile matter and oxygen functional groups of coal surface that governs the available component for oxidation, as well as surface area of fuel char, and constant pressure molar heat.

A report of the second chloroplast genome sequence in Veronica nakaiana (Plantaginaceae), an endemic species in Korea

  • Veronica nakaiana Ohwi (Plantaginaceae) is an endemic taxon on Ulleungdo Island, Korea. We report the second complete chloroplast genome sequence of V. nakaiana. Its genome size is 152,319 bp in length, comprising a large single-copy of 83,195 bp, a small single-copy of 17,702 bp, and a pair of inverted repeat regions of 25,711 bp. The complete genome contains 115 genes, including 51 protein-coding genes, four rRNA genes, and 31 tRNA genes. When comparing the two chloroplast genomes of V. nakaiana, 11 variable sites are recognized: seven SNPs and four indels. Two substitutions in the coding regions are recognized: rpoC2 (synonymous substitution) and rpl22 (nonsynonymous substitution). In nine noncoding regions, one is in the tRNA gene (trnK-UUU), one is in the intron of atpF, and seven are in the intergenic spacers (trnH-GUG~psbA, trnK-UUU, rps16~trnQ-UUG, trnC-GCA~petN, psbZ~trnG-GCC, ycf3~trnS-GGA, ycf4~cemA, and psbB~psbT). The data provide the level of genetic variation in V. nakaiana. This result will be a useful resource to formulate conservation strategies for V. nakaiana, which is a rare endemic species in Korea.

우유 대체 식물성 기반 우유 유사체 개발에 관한 현황과 미래 (Development of Plant-Based Milk Analogues as Alternatives to Cow Milk: Current Status and Future Prospects)

  • Following the COVID-19 pandemic, many people are increasingly becoming interested in health and environmental issues. Therefore, the sale of vegan or vegetarian products has been increasing over the last few years, as well as interest in non-dairy plant-based milk that can replace cow's milk. Furthermore, the global food industry has developed an interest in such products, considering the recent changes in consumer trends. In Korea, various products are being launched annually due to the increasing interest in non-dairy plant-based milk. However, research with regard to the quality and type of products produced in Korea is still at the preliminary stage when compared to those in the United States and Europe. Therefore, the present review has summarized non-dairy plant-based milk analogues based on the following key aspects. First, the types of non-dairy plant-based milk analogues and their production technologies (in the order of almond milk > cocoa milk > coconut milk > hemp milk > kidney bean milk > oat milk > peanut milk > rice milk, and soy milk). Second, the current status and future prospects for non-dairy plant-based milk analogues. Third, recent trends and future challenges associated with the production and quality improvement of non-dairy plant-based milk analogues. Fourth, the current status and outlook of the non-dairy plant-based milk analogue market in Korea. In conclusion, the present review could provide the food industry with valuable information regarding non-dairy plant-based milk analogues to facilitate the development of related products. Data were obtained from previously published studies.

An Antioxidant Capacity Assay Using a Polyvinyl Alcohol-Based DPPH Pellet

  • To assay for antioxidant capacity of natural products considered important in producing human health benefits, a practical and economical method using pellet techniques was developed. A standard visualizing reagent, 1,1diphenyl-2-picryl-hydrazyl (DPPH), was mixed with a water-miscible polyvinyl alcohol (PVA), serving as a solid phase support for the DPPH reagent. A DPPH pellet was prepared by dropping a small volume of the DPPH solution onto PET film, and drying in an oven. The PVA-based DPPH pellet was dissolved into water, in which the water-miscible PVA plays as a non-ionic surfactant to help the DPPH reagent to be dissolved into the solvent. Using the DPPH assay, the antioxidant capacity of water-soluble extracts of black soybean, barley, green tea, and green gram was examined. Among the natural products tested, green tea showed the highest antioxidant capacity. This PVA-based DPPH antioxidant assay can be further applied in the natural food, raw plant material, and health product inspection field.
