자외선 조사에 의한 고춧가루 중 잔류농약 제거효과 연구 (Removal effect of residual pesticides in red pepper powder by UV irradiation)

  • 고춧가루에 잔류되어 검출 빈도가 높은 농약인 Endosulfan(${\alpha}$-, ${\beta}$-체) Cypermethrin, Fenitrothion, Hexaconazole, EPN을 고춧가루에 인위적으로 오염시킨 뒤 365, 254 nm 파장에서 조사거리와 조사시간을 달리하여 잔존률을 조사하여 자외선 조사로 인한 농약의 제거효과를 알아보았다. 365 nm의 경우 조사거리 20 cm, 조사시간이 5분일 때 ${\alpha}$-Endosulfan 73.4%, ${\beta}$-Endosulfan 69.6%, Cypermethrin 60.8%, Fenitrothion 92.7%, Hexaconazole 73.8%, EPN 90.5%의 잔존률을 나타내었으며, 254 nm의 경우 조사거리 5 cm, 조사시간 36시간 일 때 ${\alpha}$-Endosulfan 74.6%, ${\beta}$-Endosulfan 64.5%, Cypermethrin 71.1%, Fenitrothion 79.1%, Hexaconazole 79.4%, EPN 64.7%의 잔존률을 나타냈다.

  • 현재까지의 농약 검출용 바이오센서는 화학 센서, immunoassay, 화학 테스트 킷과 같은 다른 잘 알려진 분석 방법들과 경쟁적으로 연구되어 지고 있다. 바이오센서가 농약을 증명하는 간단하고 저렴한 방법으로 chromatography 방법들을 대체할 잠재력을 가지고 있음에도 불구하고 정확한 정량적 분석 방법이 아직도 미비하다. 안정하고 강력한 바이오센서의 발전을 위해서 유전자 조작을 이용한 효소 개량을 통해 좀 더 민감하고 반응속도가 빠른 생촉매와 특이성이 높은 항체의 개발이 요구되고 있다. 바이오센서의 안정성 개선과 transducer 표면으로부터의 빠른 신호 전달을 위해 새로운 고정화 방법이 탐구되어져야 한다. 비록 약간의 방법들이 시료의 전처리를 필요로 하지 않을 지라도 센서의 안정성은 또 다른 개념으로 접근해야 한다. 그래서 현장 적용을 위해, 보다 간편한 시료의 전처리과정 혹은 직접적인 분석 방식이 동일시되어 개선되어야 한다. 향상된 fabrication 기술을 이용한 소형화 센서 혹은 일회용 킷의 개발은 개인용 및 산업용, 의약용 등의 여러 분야에서 실시간으로 분석이 가능하게 할 것이다. 실제 샘플의 빠르고 자동화 및 소형화된 분석 시스템의 구축을 위해 장차 매우 선택적인 다중 검출 바이오센서의 설계에 더 많은 중점을 둘 필요가 있다. 앞으로 잔류농약 검출을 위한 휴대형 바이오센서의 개발과 상용화를 위해서, 무엇보다 현재의 GC, LC (혹은 GC/MS, LC/MS) 분석을 위해 이루어지고 있는 샘플 전처리 방법의 경우, 다량의 샘플로부터 유기용매 등을 이용한 추출방법으로 진행되고 있기 때문에 샘플 전처리를 간소화하고 간단한 측정방법으로 전체의 측정결과를 대변할 수 있는 방안을 구축하고 잔류농약의 법적 허용기준과 적용이 가능한 방법을 찾아내는 노력이 절실히 요구되는 상황이다.

농약살포액의 이화학적 특성과 생물활성 변화 (Changes in Physicochemical Properties and Bioactivity of Pesticide Spray Solutions)

  • 환경오염을 최소화하고 병해충 방제효과를 증진시킬 수 있는 희석제농약의 적정사용기술을 확립하기 위하여, 희석용수의 수질이 살포액의 물리성과 약효에 미치는 영향 등을 검토한 결과, 국내 6대강의 하천수와 52지점의 지하수에 대한 경도는 $5{\sim}324$(평균 90) ppm, 전기전도도는 $0.038{\sim}l.078$(평균 0.265) dS/m의 범위로 전반적으로 농약의 희석용수로 양호한 수준이었다. 농약살포액의 pH는 주로 희석용수의 pH 변화에 좌우되었으며, 표면장력은 시간이 지날수록 낮아져 부착면에서는 유리할 것으로 판단되었다. 희석제농약의 유화성, 현수성 등의 물리성은 용수의 경도, 염농도의 영향을 받는 것으로 나타났으며, 농약주성분의 경시적 분해율도 조제 3일 후 대부분 5% 이내로 안정하였다. 유제와 수화제 농약의 혼용순서에 의한 살포액의 물리성은 수화시간을 제외한 모든 면에서 동일하였으며, 단지 수화제(WP>WG>SC)>유제>액제 순으로 희석하는 것이 살포액의 조제작업 측면에서 보다 용이하였다. 조제 3일 후 농약살포액의 각각의 적용병해충에 대한 방제효과는 조제 당일에 비해 95% 이상의 높은 방제효과를 유지하였다.

Evaluation of the Synergistic Effect of Mixed Cultures of White-Rot Fungus Pleurotus ostreatus and Biosurfactant-Producing Bacteria on DDT Biodegradation

  • DDT (1,1,1-trichloro-2,2-bis(4-chlorophenyl) ethane) is one of the organic synthetic pesticides that has many negative effects for human health and the environment. The purpose of this study was to investigate the synergistic effect of mixed cutures of white-rot fungus, Pleurotus ostreatus, and biosurfactant-producing bacteria, Pseudomonas aeruginosa and Bacillus subtilis, on DDT biodegradation. Bacteria were added into the P. ostreatus culture (mycelial wet weight on average by 8.53 g) in concentrations of 1, 3, 5, and 10 ml ($1ml{\approx}1.25{\times}10^9$ bacteria cells/ml culture). DDT was degraded to approximately 19% by P. ostreatus during the 7-day incubation period. The principal result of this study was that the addition of 3 ml of P. aeruginosa into P. ostreatus culture gave the highest DDT degradation rate (approximately 86%) during the 7-day incubation period. This mixed culture combination of the fungus and bacteria also gave the best ratio of optimization of 1.91. DDD (1,1-dichloro-2,2-bis(4-chlorophenyl) ethane), DDE (1,1-dichloro-2,2-bis(4-chlorophenyl) ethylene), and DDMU (1-chloro-2,2-bis(4-chlorophenyl) ethylene) were detected as metabolic products from the DDT degradation by P. ostreatus and P. aeruginosa. The results of this study indicate that P. aeruginosa has a synergistic relationship with P. ostreatus and can be used to optimize the degradation of DDT by P. ostreatus.

  • Recently, we reported an improved technology for the degradation of organophosphate nerve agents using whole cells of genetically engineered Escherichia coli that anchored and displayed the enzyme organophosphorus hydrolase on the cell surface. In this paper we report the immobilization of these cells on highly porous sintered glass beads and the subsequent application of the immobilized cell in a continuous-flow packed bed bioreactor for the biodetoxification of a widely used insecticide, coumaphos.

A New Variable Selection Method Based on Mutual Information Maximization by Replacing Collinear Variables for Nonlinear Quantitative Structure-Property Relationship Models

  • Selection of the most informative molecular descriptors from the original data set is a key step for development of quantitative structure activity/property relationship models. Recently, mutual information (MI) has gained increasing attention in feature selection problems. This paper presents an effective mutual information-based feature selection approach, named mutual information maximization by replacing collinear variables (MIMRCV), for nonlinear quantitative structure-property relationship models. The proposed variable selection method was applied to three different QSPR datasets, soil degradation half-life of 47 organophosphorus pesticides, GC-MS retention times of 85 volatile organic compounds, and water-to-micellar cetyltrimethylammonium bromide partition coefficients of 62 organic compounds.The obtained results revealed that using MIMRCV as feature selection method improves the predictive quality of the developed models compared to conventional MI based variable selection algorithms.

토양의 질 지표 개발 동향과 논의 (Review and Discussion on Development of Soil Quality Indicators)

  • The heavy dependence of modern science-based agriculture on chemicals such as fertilizers and pesticides, and heavy machinery gave rise to questions about long-term sustainability of agriculture in relation to degradation of soil quality. The research achievements and trends in developing soil quality indicators were introduced and discussed in this report. Organization for Economic Cooperation and Development (OECD) established 13 agri-environment indicators including soil quality indicator in 1994, collected the soil data and methodologies for development of soil quality indicators in OECD member countries responded to OECD questionnaires and published the OECD reports, Environmental Indicators for Agriculture Volume 1, 2, and 3. Leading countries such as USA, Canada and New Zealand collected national data and evaluated the data in aspect of soil quality. They developed the various methods for selecting a minimum data set (MDS), scoring the soil properties and calculating soil quality index integrating the score of each soil property.

농업 생태계에 대한 잔류농약의 영향 평가 (Evaluation on the effects of pesticide residues to agroecosystem in Korea)

  • Pesticide residues in soil could be affected to the growth of micro organisms and the activity of enzymes directly, and successively to the soil properties as pH, Eh and nitrogen metabolism. However, residues are diminished by degradation of soil microorganisms, run-off, leaching, volatilization, photodecomposition and uptake through crops. In this paper research results published in Korea were summarized about translocation of soil residues into crops, fates of residues in soil, effects to the activity of soil microorganisms and metabolic pathways of some pesticides. Generally speaking, pesticide residues in soil were not much affected to the agro-ecosystem except few chemicals. So it should be needed more further researches in this field, continuously.

Investigating the Impact of Best Management Practices on Nonpoint Source Pollution from Agricultural Lands

  • Abstract Over the last several decades, crop production in the United States increased largely due to the extensive use of animal waste and fertilizers as plant nutrient supplements, and pesticides for crops pests and weed control. Without the application of animal waste best management, the use of animal waste can result in nonpoint source pollution from agricultural land area. In order to increase nutrient levels and decrease contamination from agricultural lands, nonpoint source pollution is responsible for water quality degradation. Nonpoint source pollutants such as animal waste, ferilizers, and pesticides are transported primarily through runoff from agricultural areas. Nutrients, primarily nitrogen and phosphorus, can be a major water quality problem because they cause eutrophic algae growth. In 1985, it was presented that Watershed/Water Quality Monitoring for Evaluation BMP Effectiveness was implemented for Nomini Creek Watershed, located in Westmoreland County, Virginia. The watershed is predominantly agricultural and has an aerial extent of 1505 ha of land, with 43% under cropland, 54% under woodland, and 3% as homestead and roads. Rainfall data was collected at the watershed from raingages located at sites PNI through PN 7. Streams at stations QN I and QN2 were being measured with V-notch weirs. Water levels at the stream was measured using an FW-l Belfort (Friez FWl). The water quality monitoring system was designed to provide comprehensive assessment of the quality of storm runoff and baseflow as influenced by changes in landuse, agronomic, and cultural practices ill the watershed. As this study was concerned with the Nomini Creek Watershed, the separation of storm runoff and baseflow measured at QNI and QN2 was given by the master depletion curve method, and the loadings of baseflow and storm runoff for TN (Total Nitrogen) and TP (Total Phosphorus) were analyzed from 1987 through 1989. The results were studied for the best management practices to reduce contamination and loss of nutrients, (e.g., total nitrogen and total phosphorus) by nonpoint source pollution from agricultural lands.

ROLE OF COMPUTER SIMULATION MODELING IN PESTICIDE ENVIRONMENTAL RISK ASSESSMENT

  • It has been estimated that the equivalent of approximately $US 50 billion has been spent on research on the behavior and fate of pesticides in the environment since Rachel Carson published “Silent Spring” in 1962. Much of the resulting knowledge has been summarized explicitly in computer algorithms in a variety of empirical, deterministic, and probabilistic simulation models. These models describe and predict the transport, degradation and resultant concentrations of pesticides in various compartments of the environment during and after application. In many cases the known errors of model predictions are large. For this reason they are typically designed to be “conservative”, i.e., err on the side of over-prediction of concentrations in order to err on the side of safety. These predictions are then compared with toxicity data, from tests of the pesticide on a series of standard representative biota, including terrestrial and aquatic indicator species and higher animals (e.g., wildlife and humans). The models' predictions are good enough in some cases to provide screening of those compounds which are very unlikely to do harm, and to indicate those compounds which must be investigated further. If further investigation is indicated a more detailed (and therefore more complicated) model may be employed to give a better estimate, or field experiments may be required. A model may be used to explore “what if” questions leading to possible alternative pesticide usage patterns which give lower potential environmental concentrations and allowable exposures. We are currently at a maturing stage in this research where the knowledge base of pesticide behavior in the environmental is growing more slowly than in the past. However, innovative uses are being made of the explosion in available computer technology to use models to take ever more advantage of the knowledge we have. In this presentation, current developments in the state of the art as practiced in North America and Europe will be presented. Specifically, we will look at the efforts of the ‘Focus’ consortium in the European Union, and the ‘EMWG’ consortium in North America. These groups have been innovative in developing a process and mechanisms for discussion amongst academic, agriculture, industry and regulatory scientists, for consensus adoption of research advances into risk management methodology.
