Rice Plant Growth Promotion and Induced Systemic Resistance Against Rice strip tenuivirus by a Selected PGPR, Bacillus amyloliquefaciens (PGPR균 EXTN-1 처리에 의한 벼의 생육촉진 및 벼줄무늬잎마름병(RSV)에 대한 유도저항성 발현)

  • In previous reports, the treatment of Bacillus amyloliquefaciens strain EXTN-1 showed a broad diseasecontrolling spectrum to the plant diseases caused by viral, bacterial, and fungal pathogens as well as the promotion of plant growth. In mechanisms of EXTN-1, treatment of EXTN-1 increased oxidative burst in early stage and induced the expression of resistance genes, PR-1a, PDF1.2. Mechanism involved in induced systemic resistance by EXTN-1 was revealed as simultaneous activation of SA and JA or ethylene metabolic pathways. The purpose of this study was to determine whether B. amyloliquefaciens EXTN-1 has a similar effect on rice plant against Rice stripe tenuivirus (RSV) under greenhouse conditions. When rice seeds were soaked in B. amyloliquefaciens strain EXTN-1, rice plants showed significant systemic resistance against RSV as well as promoted growth. In the case of plant growth, in 30-day old plants treated with B. amyloliquefaciens EXTN-1, the heights, weights, and lengths of roots increased by 12.6%, 9.8%, and 16.0%, respectively confirming the effects of PGPR. When the induced systemic resistance to RSV was examined, in 20-day old plants were treated with B. amyloliquefaciens EXTN-1, the heights, weights, and lengths of roots increased by 8.4%, 10.9%, and 4.8%, respectively compared to the control. Induced systemic resistance was more prominent in susceptible cultivars - Chucheong and Ilpum compared to the resistant cultivar, Nakdong.

Insecticide resistance monitoring in Korean local populations of diamondback moth (Plutella xylostella) (I)

  • Various chemical pesticides are used to control diamondback moths, Plutella xylostella, which are agricultural pests that occur in cruciferous crops worldwide and cause economic losses. However, due to pesticide misuse, resistance to P. xylostella is consistently reported domestically and internationally. Therefore, we aimed to monitor and map regional resistance to devise efficient and economical control methods for P. xylostella in Korea. This study selected eight highly used insecticides among those registered against P. xylostella. P. xylostella were collected from three cities in the Gyeonggi and Yeongnam Provinces to evaluate insecticide resistance. As a result of experiments with populations collected from Yeoju, Gyeonggi Province, resistance ratios were 114.88, 54.75, 119.00, and 64.00 times higher than the susceptible population with methoxyfenozide, indoxacarb, cyantraniliprole, and fluxametamide, respectively. The resistance ratios of the Yongin population in Gyeonggi Province were 166.33 times with cyantraniliprole and 195.25 times with fluxametamide higher than the susceptible population. The Pocheon population in Gyeonggi Province showed a resistance ratio 283.23 times higher than methoxyfenozide. As a result of experiments with populations collected from Gimhae and Sangju, Yeongnam Province, the resistance ratios of the Gimhae population were 80.97, 138.00, and 89.50 times higher than the susceptible population with methoxyfenozide, cyantraniliprole, and fluxametamide, respectively. Meanwhile, the resistance ratios of the Sangju population were 85.83, 224.67, and 303.25 times higher than the susceptible population with methoxyfenozide, cyantraniliprole, and fluxametamide, respectively. The Yeongnam Province Tongyeong population showed a resistance ratio 367.28 times higher to methoxyfenozide.

Pest Control Effect and Optimal dose by Pesticide Dispersion Spray Method in the Paprika Cultivation (파프리카 시설재배지에서 약제 살포방법에 의한 해충방제 효과와 최적 살포함량)

  • We studied on pesticide residue and pest control effect when using various types of sprayers on paprika cultivation. Additionally, a test was conducted to optimize chemical content per unit area in condition of optimum pest control. Two types of sprayer were tested (three times) on paprika cultivation which was divided into seven sections. Blind spots were also examined using a water sensitive paper when spraying chemical pesticide, remote controlled sprayer were confirmed to be not effective in terms of its spraying capacity. However, a U-shaped sprayer was confirmed that it sprayed enough on all the parts of a plant in green house including the blind spots. Additionally, it does not exceed the minimum residue limits on the all parts of pesticides residue conditions. When using remote controlled sprayer, water sensitive paper were changed to blue color (82.5% and 81.2%) in terms of controlling Bemisia tabaci and Aphis gossypii based on the two spraying manners. 53.0% and 42.6% of control effect were shown on the fair parts of the plants. However, on the poor parts on which pesticides were not well-sprayed, thus, not-remained, more number of pests increased. Meanwhile, on farming that only one type of pesticide has been used, resistance pests present with very low control effect, even though sufficient amount of pesticide was well-sprayed. On the test of the optimum amount of spraying per a unit area, which shows no differences in the two cases of using 5L and 2.5L of chemical pesticides on 9 plants of paprika that has 81.8% and 84.5% control effect, respectively.

Rapid diagnosis and control of sulfonylurea resistant Monochoria korsakowii (Sulfonylurea계 제초제 저항성 물옥잠의 조기진단과 방제)

  • Sulfonylurea(SU)-resistant Monochoria korsakowii has resently been found in rice fields in Korea. A quick, practical and accurate test of confirming herbicide resistance is necessary to take timely management decision. This article describes a rapid reliable assay to detect SU-resistant biotype of Monochoria korsakowii. Up to now, the resistance to SU has been usually checked by application seedlings with herbicide. This application technique is time consuming and not practical. Therefore, we have developed efficient, rapid and practical diagnosis which allow easy detection of the SU-resistant Monochoria korsakowii by survival rate and regenerated plant length to herbicide application after cutting plants $0.5\sim1cm$ from the planted surface. This new rapid diagnosis can determine the SU resistance of the Monochoria korsakowii within 7 days at least. If the resistance of Monochoria korsakowii is identified by the rapid diagnosis, the selection of herbicide according to the stages of plants for the effective control is very important. The resistant biotype which treated with SU herbicide-based mixtures survived from the fields could effectively be controlled by soil application of butachlor+pyrazolate GR or by foliar application of the mixtures of bentazone SL and 2,4-D SL.

Resistance of Saccharomyces cerevisiae to Fungicide Chlorothalonil

  • The toxicity of chlorothalonil on the growth of yeasts was investigated using several yeast strains. An alcohol tolerant yeast, Saccharomyces cerevisiae F38-1, was the most chlorothalonil-tolerant. The glutathione content and the glutathione S-transferase activity were related to the chlorothalonil-tolerant phenotype. Several thiol compounds affect the dissipation of chlorothalonil. However, there was no significant difference on the effects of chlorothalonil dissipation among the thiol compounds tested. The growth of yeast cells was arrested by chlorothalonil. It took about 13 h to dissipate 1 mg/l of chlorothalonil, and the growth was restored as the chlorothalonil content decreased. The glutathione content and glutathione S-transferase are suggested to be among the most important factors of yeast resistance to chlorothalonil.

  • Changes in weed floras and development of plant resistance to herbicides seemed to be closely related with increased and repeated use of herbicides. Herbicide use increased from 5% of the total consumption of pesticide in 1950 to 45% in 1976 in world basis. About 200 herbicides have been introduced to agriculture so as to control about 206 weed species which have been recorded important to human beings. In Korea, there was about 351 times in increased use of herbicides from 1966 to 1982. Interspecific selection by herbicide is mainly responsible for changes in weed floras and resulted in varying tolerance or susceptibility to herbicides, together with the changes of agricultural practices. The present trend toward continuous cereal cultivation throughout world will lead to type of changes in weed floras favorable to therophyte which can survive under unfavorable conditions as seeds rather than the types of geophyte which can survive unfavorable seasons as buds placed below soil surface. However, geophyte such as Sagitaria pygmaea, and Scirpus jurtcoides, and Cyperus rotundus and Cynodon dactylon in temperate warm climate become severe paddy weeds, presumably because of the removal of annual weeds by herbicides. Since differential tolerance to 2,4-D was firstly reported in Agrostis stolofera, about 30 species of weeds in 18 genera are presently known to have developed resistance to triazine herbicides. Resistance of weed biotypes to triazine herbicide is not mainly due to limited absorption and translocation or to the difference in metabolism, but is the result of biochemical changes at the site of metabolic activity, such as a loss of herbicide affinity for triazine binding site in the photosystem II complex of the chloroplast membrane. Genetical study showed that plastid resistance to triazine was wholly inherited through cytoplasmic DNA in the case of Brassica campestris. Plant tissue culture method can be utilized as an alternate mean of herbicide screening and development of resistance variants to herbicides as suggested by Chaleff and Parsons. In this purpose, one should be certain that the primary target process is operational in cell culture. Further, there are a variety of obstacles in doing this type of research, particularly development of resistance source and it's regeneration because cultured cells and whole plants represent different developmental state.

Property of action of new insecticide, flupyrazofos against diamondback moth, Plutella xylostella (신규 살충제 flupyrazofos의 배추좀나방에 대한 작용특성)

  • Flupyrazofos (O,O-diethyl-O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl)thiophosphoric acid ester) is a new organo-phosphorous insecticide. Flupyrazofos has exhibited excellent activity against diamondback moth (Plutella xylostella, DBM), and it is highly activity against rice armyworm(Pseudaleta separata) and cotton caterpillar(Palpita indicae). Flupyrazofos has then revealed outstanding both rapidity and residual action for DBM, although no systemic actions were observed and no cross-resistances were found to the resistance strains (Op-R, Py-R, IGR-R). Also, susceptibilities of five local strains to flupyrazofos were similar to those of the susceptible strain. These results indicate that flupyrazofos has considerable potential for controlling diamondback moth, rice armyworm and cotton caterpillar in field.

Statuses and Perspectives of Herbicides Development Against Herbicide-Resistant Weeds in Paddy Field of Korea (논 제초제 저항성 잡초 발생에 따른 제초제 개발 현황과 방향)

  • The widespread and diverse sulfonylurea (SU) resistance problem has found in Korea, where one-shot-treatment herbicides such as pylazosulfuron/molinate and bensulfuron/molinate have been used continuously since 1989. The SU-resistant weeds of 7 annual weeds and 3 perennial weeds as of 2008 have confirmed in paddy fields in Korea. An effective management to SU-resistant weeds requires an integrated approach toward the weed control system, in particular, as to the drastic changes of herbicides development. Recent trend of new paddy herbicides in Japan has been developing to maximize the management of SU-resistant weeds. In the future, it is expected that the development of paddy herbicides in Korea is likely to be shifted toward the new "one-shot-treatment" included with herbicides of over 3-ways to maximize the control of resistant weeds. Bromobutide and carfentrazone are effective against sedges and broad-leaved weeds, respectively, and benzobicyclone and pyrimisulfam are effective against sedges and broad-leaved weeds.

Physiological Diversity between Morphological Phenotypes of Botrytis cinerea (잿빛곰팡이병균(Botrytis cinerea) 형태형 간의 생리적 다양성)

  • Botrytis cinerea isolates obtained from infected plants of cucumber, tomato, and strawberry were divided into three groups (sporing, sclerotial, and mycelial types). Of which sclerotial types were the major group. There were no correlations between morphological phenotypes and responses to benzimidazole and dicarboximide fungicides. External structure of conidia of three phenotypes by scanning electron microscope was the same with verrucose surface. Mycelial type was the most virulent on fruits of eggplants. Comparative tests were carried out to examine correlations between the virulence and production of fungal enzymes such as phenol oxidases, pectin methyl esterases (PME), amylases, cellulases, ureases, ${\beta}-glucosidases$, and proteinases. There was no correlation among the phenotypes in production of phenol oxidases and ${\beta}-glucosidases$. However, there were significantly different from each other in PME, amylase, cellulase, urease, and protease activity.

Studies on Infectivity of Cordyceps, Paecillomyces japonica, on the Domestic Silkworm, Bombyx mori. (눈꽃 동충하초균의 누에감염에 관한 연구)

  • This study was carried out to investigate infection process, symptoms and $LD_{50}$ values of the entomopathogenic fungus cordyceps, Paecillomyces japonica, on the domestic silkworm, Bombyx mori. Susceptibility of infection in the silkworms by cordyceps, Paecillomyces japonica, was 100% in $10^{7}$ block, 96% in $10^{6}$ block, 76% in $10^{5}$ block, 44% in $10^{4}$ block, 28% in $10^{3}$ block and 8% in $10^{2}$ block. Cordyceps, Paecillomyces japonica, was highly infectious to the silkworms. A pathogenicities of cordyceps, Paecillomyces japonica, may be highly virulent because of the low resistance or high susceptibility of the silkworms. Dosage of the pathogen of $LD_{50}$ was $3.78{\times}10^{3}\;spores/mm^{3}$.
