Siberian Tiger (Panthera tigris altica)01서 Actinomycotic Mycetoma에 의한 복막염의 발생 증례보고 (Actinomycotic mycetoma in the peritoneal cavity of a Siberian tiger (fgH1Arrg rigrir gfric6))

  • A case of peritonitis caused by Actinomycotic spp is reported in a 12-year-old male Siberian tiger. Grossly, the mesentery was markedly thickened and contained numerous 1 to 3 mm diameter, white to yellowish foci. Fibrous adhesion showing tumorous thickening was also noted between the mesentery and abdominal organs. Histologicallyi the thickened mesentery and masses consisted of necrotic center with bacterial colonies surrounded by eosinophilic club (Splendore-hoeppli), neutrophils, macrophages, a few Iymphocytes and fibrosis. The bacterial colonies stained positvely with Gram's stain but were negative on acid-fast and periodic acid-Schifr method. Howeverr since the bacterial culture was not availablei the definitive causative agent was not able to specified.

  • 혈관종증(hemangiomatosis)은 하나 혹은 여러 개의 장기에서 다발성으로 발생하는 혈관종(hemangioma)이다. 혈관종증은 주로 간에서 나타나며 대망(greater omentum)과 장간막(mesentery)에 발생하는 경우는 매우 드문 경우로 보고된 예가 적다. 저자들은 특별한 증상이 없는 62세 남자 환자에서 간을 포함한 대망과 장간막에 우연히 발견된 혈관종증의 컴퓨터 단층촬영, 자기공명영상 소견을 병리 소견과 함께 보고자 한다.

  • To find antitumor components from the higher fungi of Korea, the mycelia of Favolus alveolarius (Fr.) Quelet were cultured in a liquid medium. The cultured mycelia were extracted with hot water twice, and a high molecular weight fraction was obtained by adding two volumes of ethanol to the extract. Two grams of Fraction A were obtained by dialyzing it. It was further separated into four fractions by gel filtration with Sepharose CL-4B, and they were designated Fractions B, C, D, and E. The results of the antitumor test showed that Fractions A, B, C, D and E had tumor inhibition ratios of 92.3, 78.5, 59.6, 77.4 and 62.2%, respectively. Anthrone test was carried out to determine the contents of total polysaccharide of the five fractions, and they had 46.3, 27.3, 65.3, 64.6, and 46.1%, respectively. The contents of the total protein of the five fractions were 29.4, 13.9, 14.3, 14.3, and 29.1%, respectively. Monosaccharide subunits of each fraction were analyzed with gas chromatography, and glucose, xylose, mannose, galactose and fucose were identified. Fraction A was examined for immunological effects. It increased the count of hemolytic plaque forming cells 12.8 times to that of the control group, and the population of macrophage in peritoneal cavity 3.2 times to that of the control group.

행인 제제의 독성 및 항암효과의 자연살해세포 활성에 미치는 효과 (Study on toxicity, anti-cancer and NK cell activity of Lateril oil)

  • In this experiment, we examined the the effect of Laetrile Oil to the immune system and tried to disclose the anti-cancer mechanism of this. The result is listed as below. 1. After per-os, sub-cutaneous and into the peritoneal injection of Laetrile Oil to the SPF mice, the LD50 is above 5000mg/kg at even group. 2. Mean survival days of mice treated with laetril oil, after S-180 cells transplantation into the peritoneal cavity decreases 1.5 days(-6.8%) compared with the Mean survival days of the control group(22days.) 3. Effects of laetril oil on natural killer cell activity at Effector/Target Cell Ratio with 100:1, 50:1, 10:1 into methotrexate-pretreated mice is like this.: Compared with $29.22\pm12.7\%$ Cytotoxicity of the control group, sample group's Cytotoxicity had $38.83{\pm}12.5%$ of meaningful acrease. At 100:1 Effecr/Target Cell Ratio. At 50:1 Effector/Target Cell Ratio, control group has $20.02{\pm}9.6%$ Cytotoxiciy and sample group had $31.53{\pm}13.4%$ Cytotoxicity. At 25:1 Effector/Target Cell Ratio control group has $13.60{\pm}6.6%$ Cytotoxicity and sample group had $20.81{\pm}9.8%$ Cytotoxicity According to the above results, the Laetrile Oil represents nontotoxic to a SPF mice. non-effective to transplantable Sarcoma 180 tumors, and activaion in NK cell activity.

Ginsenoside Rg3 promotes inflammation resolution through M2 macrophage polarization

  • Background: Ginsenosides have been reported to have many health benefits, including anti-inflammatory effects, and the resolution of inflammation is now considered to be an active process driven by M2-type macrophages. In order to determine whether ginsenosides modulate macrophage phenotypes to reduce inflammation, 11 ginsenosides were studied with respect to macrophage polarization and the resolution of inflammation. Methods: Mouse peritoneal macrophages were polarized into M1 or M2 phenotypes. Reverse transcription-polymerase chain reaction, Western blotting, and measurement of nitric oxide (NO) and prostaglandin $E_2$ levels were performed in vitro and in a zymosan-induced peritonitis C57BL/6 mouse model. Results: Ginsenoside $Rg_3$ was identified as a proresolving ginseng compound based on the induction of M2 macrophage polarization. Ginsenoside $Rg_3$ not only induced the expression of arginase-1 (a representative M2 marker gene), but also suppressed M1 marker genes, such as inducible NO synthase, and NO levels. The proresolving activity of ginsenoside $Rg_3$ was also observed in vivo in a zymosan-induced peritonitis model. Ginsenoside $Rg_3$ accelerated the resolution process when administered at peak inflammatory response into the peritoneal cavity. Conclusion: These results suggest that ginsenoside $Rg_3$ induces the M2 polarization of macrophages and accelerates the resolution of inflammation. This finding opens a new avenue in ginseng pharmacology.

오른쪽 서혜부 탈장에 의해 이차적으로 발생한 대망의 염전 및 경색 (Omental Torsion and Infarction Secondary to Omental Hernia in the Right Inguinal Canal)

  • 대망의 서혜부 탈장에 의한 이차성 대망 염전은 급성 복통의 원인으로써 드물게 보고된 바 있다. 그러나 만성 복통의 원인으로써 이차성 대망 염전이 섬유성 벽을 가진 거대하고 단단한 종괴로 발견되는 것은 이전까지 보고되지 않았기에 이를 보고하고자 한다. 74세 남자 환자가 한 달간 지속된 만성적인 하복부 복통과 우하복부에 만져지는 종괴를 주소로 내원하였다. 시행한 서혜부 초음파상에서 양쪽 서혜부 탈장이 관찰되었다. 컴퓨터단층촬영상에서 오른쪽 서혜관으로 대망의 일부가 빠져나갔고 그 축을 중심으로 대망 염전이 있었다. 염전 줄기의 원위부 대망은 우하복부와 골반강에 걸쳐 단단한 섬유성 벽을 가진 약 30 cm 정도의 거대한 종괴를 형성하였다. 환자는 복강경하 장막 절제술 및 양쪽 탈장 수술을 시행 받은 뒤 퇴원하였다.

  • Background: The role of macrophages in tumor angiogenesis is known to be the production of angiogenic cytokines and growth factors including TNF-${\alpha}$. Recently, macrophage also can produce the INF-${\gamma}$ that is being studied to be involved in angiogenic inhibition. Thus, the importance of macrophages in tumor angiogenesis is might being an angiogenic switch. Thus, the hypothesis tested here is that TNF-${\alpha}$ can modulate the INF-${\gamma}$ production in the macrophages from tumor environment as a part of tumor angiogenic switch. Methods: Macrophages in tumor environment were obtained from the peritoneal cavity of C57BL/6 mice injected with B16F10 melanoma cell line for 6 or 11 days. $Mac1^+$-macrophages were purified using magnetic bead ($MACs^{TM}$; Milteny Biotech, Germany) and cultured with various concentrations of TNF-${\alpha}$ for various time points at $37^{\circ}C$. The supernatants were analyzed for IFN-${\gamma}$ or VEGF by ELISA kit (Endogen, Woburn, MA). Results: Residential macrophages from the peritoneal cavity did not respond to LPS or TNF-${\alpha}$ to produce INF-${\gamma}$. However, the cells from tumor environment produced IFN-${\gamma}$ as well as VEGF and upregulated by the addition of LPS or TNF-${\alpha}$. RT-PCR analysis revealed the external TNF-${\alpha}$-induced IFN-${\gamma}$ gene expression in the macrophages from tumor environment. Conclusion: The overall data suggest that the macrophages in tumor environment might have an important role not only in angiogenic signal but also in anti-angiogenic signal by producing related cytokines. And TNF-${\alpha}$ might be a key cytokine in tumor angiogenic switch.

In vivo verification of regional hyperthermia in the liver

  • Purpose: We performed invasive thermometry to verify the elevation of local temperature in the liver during hyperthermia. Materials and Methods: Three 40-kg pigs were used for the experiments. Under general anesthesia with ultrasonography guidance, two glass fiber-optic sensors were placed in the liver, and one was placed in the peritoneal cavity in front of the liver. Another sensor was placed on the skin surface to assess superficial cooling. Six sessions of hyperthermia were delivered using the Celsius TCS electro-hyperthermia system. The energy delivered was increased from 240 kJ to 507 kJ during the 60-minute sessions. The inter-session cooling periods were at least 30 minutes. The temperature was recorded every 5 minutes by the four sensors during hyperthermia, and the increased temperatures recorded during the consecutive sessions were analyzed. Results: As the animals were anesthetized, the baseline temperature at the start of each session decreased by $1.3^{\circ}C$ to $2.8^{\circ}C$ (median, $2.1^{\circ}C$). The mean increases in temperature measured by the intrahepatic sensors were $2.42^{\circ}C$ (95% confidence interval [CI], 1.70-3.13) and $2.67^{\circ}C$ (95% CI, 2.05-3.28) during the fifth and sixth sessions, respectively. The corresponding values for the intraperitoneal sensor were $2.10^{\circ}C$ (95% CI, 0.71-3.49) and $2.87^{\circ}C$ (1.13-4.43), respectively. Conversely, the skin temperature was not increased but rather decreased according to application of the cooling system. Conclusion: We observed mean $2.67^{\circ}C$ and $2.87^{\circ}C$ increases in temperature at the liver and peritoneal cavity, respectively, during hyperthermia. In vivo real-time thermometry is useful for directly measuring internal temperature during hyperthermia.

한우 송아지의 선천성 전신수종 (Congenital Dropsy in Korean-native Calf)

  • 선천성 전신수종과 함께 체간의 형성부전을 특징으로 하는 한우 기형 송아지를 해부학적 및 혈청학적으로 검사하여 다음과 같은 결과를 얻었다. 기형의 송아지는 정상 송아지의 2/3 정도 크기 (체장 82cm, 무게 25kg) 이었다. 태자는 외관상 목과 몸통의 구분이 어려웠고 두부에서 포는 보이지 않았으며, 목에 해당하는 부분에 혓바닥을 물고 있는 입이 있었다. 사지의 형태는 유지하고 있었으나 체간의 형성부전으로 기형을 나타내었고 얼굴을 비롯 몸통 전체의 피하에 심한 수종에 의해 파동성을 보였다. 부검시 흉강과 복강내에 맑은 장액성 액상물질이 가득 차 있었고 피하조직내에도 같은 물질의 저류로 인해 피부와 피하의 구별이 되지 않았다. 더불어 이들 수종성 피하에는 연황색의 과립양 물질들이 다수 혼재되어 있었다. 내부장기는 거의 식별하기 어려웠고 흉강과 복강이 부분적으로 개통되어 간의 일부가 흉강쪽으로 빠져 나와 있었다. 폐는 크기가 32$\times$49 mm로 심장(56$\times$45 mm) 보다도 더 작게 위축되어 있었다. 복강에서는 두 개의 수종성 좌측 신장 (15$\times$21cm, 13$\times$18cm)과 1 개의 우측신장 (13$\times$9cm) 이 있었고 이들의 할단면에서의 실질부위는 대부분이 낭포로 채워진 낭종성 기형을 나타내었다. 간은 담황색 변성과 종대를 보였고, 비장은 부종, 충혈 및 종대가 관찰되었으며, 부종성 소장과 대장은 장관의 형태는 유지하고 있었으나 장간막에 황색 결절이 미만성으로 형성되어 있었다. 한편, 이 기형 송아지의 발생 원인체 중 하나로 모우에서 64 배 이상의 높은 중화 항체가를 보인 Akabane 바이러스가 추정되었다.

소양인(小陽人) 인동등지골피탕(忍冬藤地骨皮湯)이 중소합병증(中消合倂症)에 미치는 영향(影響) (Effecs of Soyangin Indongdeungjikolpitang on the Complication of Diabetes)

  • 소양인(少陽人) 인동등지골피탕(忍冬藤地骨皮湯)이 중소(中消) 합병증(合倂症)에 미치는 영향(影響)을 살펴보고자 하였고 결과(結果)는 다음과 같다. 1. 장관운동(腸管運動)을 억제(抑制)하였다. 2. Acetic acid에 의한 writhing syndrome을 억제(抑制)하였다. 3. Evans blue에 의한 모세혈관투과성(毛細血管透過性)을 억제(抑制)하였다. 4. Histamine에 의한 급성족부종(急性足浮腫)을 억제(抑制)하였다. 5. Cotton pellet에 의한 육아종(肉芽腫) 형성(形成)을 억제(抑制)하였다. 6. 흉선세포(胸線細胞) 및 비장세포(脾臟細胞)의 세포생존율(細胞生存率)을 증가(增加)시켰다. 7. 흉선세포(胸線細胞)로부터 ${\gamma}-IFN$ 및 IL-2의 분비(分泌)를 억제(抑制)하였다. 8. 비장세포(脾臟細胞)로부터 IL-4의 분비(分泌)는 증가(增加)시켰으나, $TNF-{\alpha}$$IL-1{\beta}$의 분비(分泌)는 억제(抑制)하였다. 9. 복강(腹腔) 대식세포(大食細胞)로부터 $TNF-{\alpha}$, $IL-1{\beta}$ 및 nitric oxide의 분비(分泌)를 억제(抑制)하였다. 10. 복강(腹腔) 대식세포(大食細胞)의 lucigenin chemiluminescence 및 FITC-conjugated E. cole engulfment를 촉진(促進)하였다. 이상의 실험(實驗) 결과(結果) IJTE는 장관운동(腸管運動)을 억제(抑制)하였으며, 진통(鎭痛) 및 항염작용(抗炎作用)을 나타냈고, 이들 작용은 면역세포(免疫細胞)로부터 $TNF-{\alpha}$, $IL-1{\beta}$ 및 nitric oxide의 생성(生成)을 억제(抑制)하기 때문인 것으로 추정된다. 따라서 이러한 기전(機轉)에 의하여 인동등지골피탕(忍冬藤地骨皮湯)은 중소(中消) 합병증(合倂症)의 하나인 염증(炎症)을 억제(抑制)하는데 유효(有效)한 것으로 사료(思料)된다.
