Sodium Carboxymethylcellulose를 이용한 개의 자궁 유착 방지 (Prevention of Uterine Adhesion by Sodium Carboxymethylcellulose in Dogs)

  • Efficacy of a 1% solution of sodium carboxymethylcellulose (SCMC) infu7ed into the peritoneal cavity of dogs was evaluated for prevention of intraperitoneal adhesion, resulting from operations of the reproductive tract. Saline-treated deles (n = 5) were controls that underwent ventral midline celiotomy, and adhesions were cleated by incision and scraping about 5 cm segment of each uterine horn. Saline (7 ml/kg of body weight) was then infused into the peritoneal cavity. Others (n = 5) were treaded similarly to the saline-treated dogs. except that 1% SCMC :solution (7mH/kg of body weight) was infused into the peritoneal cavity. This group was studied to determine whether SCHC would prevent the adhesions in the peritoneal cavity, especially in the uterine horn model. Abdominal adhesions were evaluated and an adhesion severity score was assigned to each dog on the basis of severity of adhesions. At the time of necropsy. the mean adhesion score in the saline treated group was $2.65{\pm}0.22.$ In contrast, adhesion formation in the SCMC treated group was less ($mean score =1.70{\pm}0.26$). Statistic71 analysis was performed using the grouped t-test and paired t-test. A significanlty lower adhesion score was observed in dogs given SCMC than in the saline treated group (P<0.01). In summary, SCMC significantly reduced adhesion formation in the dog uterine horn model. The results of this study suggest that application of 1% SCMC solution, following various reproductive pelvic surgery, will present the adhesions.

  • Background : Peritoneal metastasis is one of the maj or types of the stomach cancer recurrence and the role of the adhesion molecules is thought to be very much important in this event. Retinoic acid (RA) has been known to induce the growth inhibition and differentiation of various malignancies, and apoptpsis and the change of expression of adhesion molecules have been reported to be involved in the action of RA. Methods : We studied the adhesion abilities of SNU-1, SNU-5, and SNU-6 cells to the peritoneal endothelial cells as well as the expression of the adhesion molecules (CD44, ICAM-1) in Western blot analysis. And also we studied the expression of apoptosis and the change of expression patterns of the various isoforms of CD44 and the change of the adhsion abilities of the cell line cells after RA treatment. Results: CD44 was expressed in SNU-5 and -16, together with an isoform in SNU-16. ICAM-1 was not expressed in any of the cell line cells tested. After the treatment of RA in the concentration range of $1-5{\times}10^{-5}M$ to three stomach cancer cell lines, growth inhibition, apoptosis and the change of expression of the CD44 were noted. After RA treatment, the expression of CD44H was weakly increased in SNU-1, and was markedly increased in SNU-5. In SNU-16, the expression of CD44H was decreased while that of CD44E were markedly increased. The adhesibility of cells to peritoneal cells was increased in relation with the increase of the CD44H expression, which shows the fact that the adhesibility of tumor cells to peritoneal mesothelial cells is mediated by CD44H recognizing hyaluronic acid. Conclusion : RA induces growth inhibition of stomach cancer cell line cells and increase the adhesiblity of stomach cancer cell line cells to peritoneal mesothelium. It is believed that RA decreases the metastatic ability of stomach cancer cells by upregulating the CD44H expression.

랫드에서 고농도의 Carboxymethyl Chitosan이 복벽 유착 예방에 미치는 영향 (Effect of high concentrations of carboxymethyl chitosan on preventing formation of peritoneal adhesion in rats)

  • The objective of this study was to determine the effective concentration of carboxymethyl chitoaan(CMC) solution on preventing intraperitoneal adhesions. In this study, 60 rats were divided into four groups : an untreated control group and three experimental groups that were treated with 3 ml of 3, 5, and 7% CMC solution. Adhesions were induced by suturing both the ileal serosa and peritoneum scraped until petechial bleeding occurred. CMC solutions were instilled intraperitoneally immediately before closure of the peritoneomuscular layer. The tensile strength of formed adhesions was measured by using a computerized tensiometer. Histopathological changes of adhesions were evaluated by hematoxylin-eosin for light microscopic examination and Massons's trichrom staining for collagen fibers. All of CMC solutions reduced significantly the tensile strength of the adhesions. CMC inhibited the increase of inflammatory cell infiltration and damage of adhesion spot, and accelerated the collagen synthesis and neovascuiarization. No significant differences were observed among all of CMC concentration at inhibiting adhesion formation. Therefore, we could conclude that 3% CMC, the lowest concentration adopted in this study, was appropriate to prevent the formation of peritoneal adhesion.

  • 이 실험의 목적은 개에서 poloxamer/sodium alginate 혼합물(PX/SA)의 수술 후 복강 유착 방지 효과를 알아보는 것이다. 잡종견 15마리를 무처치군, 2% Sodium carboxymethylcellulose(SCMC)처치군, PX/SA 처치군으로 각각 5마리씩 배치하였다. 유착은 돌창자의 창자간막 대측에 1 ${\times}$ 1 cm 크기로 미세 점상 출혈 찰과상을 일으켜 유도하였다. SCMC 용액은 카테터를 통하여 복강내 주입하였고, PX/SA는 찰과상 부위에 단순히 도포하였다. 수술 전 1일과 수술 후 1일, 4일, 7일, 14일에 정맥에서 혈액을 채취하여 적혈구수, 총백혈구수, 섬유소원농도를 분석하고, 수술 후 21일에 유착정도를 computerized tensiometer를 이용하여 분석하였다. 적혈구수, 총백혈구수, 섬유소원농도는 각 군간에 유의성은 없다. 유착부 분리에 필요한 평균 장력은 무처치군에서 173.05 ${\pm}$ 113.48 gf, SCMC 처치군에서 111.42 ${\pm}$ 38.25 gf, PX/SA 처치군에서 69.00 ${\pm}$ 45.07 gf였다. PX/SA 처치군의 평균 분리 장력이 다른 두 군에 비하여 유의성 있게 낮았다 (p < 0.05). 본 실험 결과로 PX/SA는 SCMC용액과 비교하여 복강유착을 감소시키는 것으로 보아 PX/SA를 개에서 복강 수술 후 유착 방지에 적용 가능한 것으로 생각한다.

Evaluation of sodium carboxymethylcellulose for prevention of adhesion in intestinal anastomosis in dogs

  • Adhesions are the most common postoperative complication in intestinal surgery in dogs. This study was aimed at verify to the efficacy of sodium carboxymethylcellulose (SCMC) in the adhesion prevention during healing of intestinal anastomosis. Twenty three healthy dogs were taken with average 4.17 years and weight was 5.68 kg and divided into 4 groups. The dogs of group III and IV, a pedicle of greater omentum was wrapped around the suture line. In the animals of group II and IV, 1% SCMC was infused (5 ml/kg, IP) at just before closure of the abdominal cavity. Two weeks after surgery, animals were reoperated and the adhesions were evaluated and graded. We found various degrees of intraabdominal adhesions in animals of all the groups. The significantly (P<0.05) lower adhesion score was observed in group IV than that of other groups. Nevertheless, varying intra-peritoneal adhesions, healing of anastomotic site was normal in all groups. It could be concluded that intra-peritoneal administration of SCMC solution reduces postoperative adhesions without any adverse effects on healing, and a synergistic beneficial effect can be obtained by supplementation with omental graft for intestinal anastomosis.

쥐에서 Sodium Carboxymethylcellulose 및 Dextran 70의 유착방지 효과 (Effects of Sodium Carboxymethylcellulose and Dextran 70 on Prevention of Adhesion in Rats)

  • The 2% Sodium carboxymethylcellulose(CMC) and 10% Dextran 70 solutions were evaluated for the prevention of postoperative adhesion formation in rats. The artificial injuries(2cm longitudinal incision and suture) were performed on the colon and uterine horns in the rats. 2% CMC and 10% dextran solutions were applied to the injured surfaces and peritoneal cavity in the rats. Ten days later, there were significant difference(p<0.01) in the prevention of adhesion formation between control and treated groups(2% CMC and 10% dextran 70), but the,$\boxUl$ was no significant difference between 2% CMC and 10% dextran 70 treated groups. The increment rate of body weight in the 10% dextran 70 treated group was lower than 2% CMC and control groups(p<0.05.). A positive correlation was found between the severity of adhesion formation and the plasma fibrinogen concentration in blood.

Focus on Anti-Oxidative and Free Radical Scavenging Activity of Ganoderma lucidum

  • Present review is built on base of research work on Ganoderma lucidum in our laboratory. A great deal of experimental evidence has suggested that the pharmacological activities of Ganoderma lucidum (Lingzhi) are related to anti-oxidative and free radical scavenging activity. The anti-oxidative and free radical scavenging effects of polysaccharides and triterpenoids isolated from Ganoderma lucidum in different oxidative injury models including tert-butylhydroperoxide (tBOOH)- damaged mice peritoneal macrophages, alloxan-induced diabetes, experimental liver injury models induced by carbon tetrachloride (CCl4), D-galactosamine (DGal) and Bacillus Calmette-Guerin (BCG) plus lipopolysaccharides (LPS) were investigated. It is also demonstrated that Lugu lingzhi, one of Ganoderma product, significantly inhibited LDL oxidation mediated by endothelial cells and decreased monocyte adhesion to endothelial cell (EC) induced by Oxidative low-density lipoprotein (ox-LDL) and advanced glycation endproducts (AGE). Lugulingzhi-treated serum could markedly inhibit the expression of intercellular cell adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-l) induced by ox-LDL and AGE.

시츄견에서 Polypropylene mesh를 이용한 gossypiboma에 의한 복막 결손 수복 증례 (Using of Polypropylene Mesh for Peritoneal Defect induced Gossypiboma in a Shih-Tzu Dog)

  • 10세령의 중성화된 암컷 시츄가 하부 복부의 왼쪽 부분의 종창으로 인해 건국대학교 수의과대학 부속동물병원에 내원하였다. 축주는 본원에 내원하기 두달전에 환축의 하부 복부의 종창과 촉진되는 덩어리를 확인하였다. 환축을 방사선과 초음파검사를 실시하고, 수술 중 방광과 소장 사이에 있는 복강내 gossypiboma로 인해 복강내 유착이 심한 것을 확인할 수 있었다. 그 gossypiboma를 복강내에서 제거되었지만, 복막을 수복하기에 결손이 커서 polypropylene mesh를 이용하였다. 수술 후 하부 복부의 종창 등은 확인할 수 없었으며, 6개월동안 아무런 합병증이 발견되지 않았다.
