RNA aptamer 발현을 통한 CD4+ peripheral blood lymphocytes에서의 인간 면역결핍 바이러스의 증식 억제 (Inhibition of HIV-1 Replication in CD4+ Peripheral Blood Lymphocytes by Intracellular Expression of RNA Aptamer)

  • 제1형 인간 면역결핍 바이러스 (human immunodeficiency virus type 1, HIV-1)의 Rev 단백질에 대하여 야생형보다 10배 더 잘 결합하도록 시험관에서 선택된 RRE40라 명명된 RNA aptamer가 과연 임상적으로 유용한지 알기 위하여 인체의 CD4^+ peripheral blood lymphocytes 세포에서 레트로바이러스 벡터를 이용하여 RRE40 RNA를 발현한 후에 그 세포에서의 HIV-1 증식 현상을 관찰하였다. 그 결과 대조군인 tRNA를 발현하는 유전자가 전달된 세포에 비해 RRE40 RNA를 발현하는 세포에서 보다 더 효과적으로 HIV-1의 증식이 억제되었다. 그러나 바이러스의 증식이 완전히 억제되지는 못 하였고 일시적 또는 감소된 형태로 바이러스 증식이 억제되었다. 이러한 결과는 RRE40 RNA가 decoy로서 세포에서의 HIV-1 증식 억제에 유용함을 시사하지만 RNA decoy를 HIV-1 감염 환자의 치료에 이용하기 위해선 보다 효과적인 유전자 전달방법 및 보다 개선된 RNA decoy의 개발 등이 필요할 것이다.

Zinc Enhances Neutrophil Extracellular Trap Formation of Porcine Peripheral Blood Polymorphonuclear Cells through Tumor Necrosis Factor-Alpha from Peripheral Blood Mononuclear Cells

  • Neutrophil extracellular trap (NET) formation is an immune response for the invasion of microbes. The purpose of this study is to examine the effect of zinc on NET formation of porcine peripheral blood polymorphonuclear cells (PMNs). The NET formation of PMNs was measured by fluorescence microplate reader. The production of tumor necrosis factor (TNF)-α in the culture supernatants from zinc-treated peripheral blood mononuclear cells (PBMCs) was measured by enzyme-linked immunosorbent assay (ELISA). Zinc itself did not have no effect on NET formation. However, the NET formation of PMNs was increased by culture supernatants from PBMCs treated with zinc. Also, the NET formation of PMNs was increased by recombinant porcine (rp) TNF-α. The production of TNF-α in PBMCs culture supernatants was shown to increase upon zinc treatments. These NET formations of PMNs increased by either culture supernatant from PBMCs treated with zinc or rpTNF-α were inhibited by treatment of anti-rpTNF-α polyclonal antibody (pAb). These results suggested that zinc has an immunostimulating effect on the NET formation of PMNs, which is mediated by TNF-α released from zinc-treated PBMCs. Therefore, zinc may play an important role for NET formation in the defense of porcine inflammatory diseases.

Diffusion-Enhanced Modified Hemodialyzer

  • Flow mismatch between blood and dialysate is invariably encountered during conventional hemodialysis, and this deteriorates diffusive mass transfer. A modification of a conventional dialyzer was conceived to prevent this mismatch. The modified dialyzer includes two independent blood flow regions (central and peripheral regions), which were achieved by redesigning the dialyzer cap. Resultantly, the blood stream was divided into two concentric dialyzer regions. Solutes clearances obtained using the modified dialyzers were compared with those of conventional dialyzers. Solutes clearances by conventional dialyzers were uniform, but solutes clearances by modified dialyzers were found to be dependent on the simulated blood split into dialyzer central and peripheral regions. Maximal clearances using the modified dialyzer were improved by up to approximately 7.6% for urea and 7.3% for creatinine, as compared with those of conventional dialyzers. More optimizations are required for clinical applications, but the finding that blood flowrates through central and peripheral fiber bundles can be easily regulated is encouraging.

Ethyl Acetate Extract from Tissue-Cultured Mountain Ginseng Adventitious Roots Inhibits In Vitro Platelet Aggregation in Whole Human Blood and Augments Peripheral Blood Flow in Mice

  • We previously reported that in vitro anti-platelet activity of tissue-cultured mountain ginseng (TCMG) ethanol extracts show improved efficacy when compared with commercial ginseng products such as Korean red ginseng and Panax ginseng. However, information on the anti-platelet activity of the ethyl acetate fraction from TCMG adventitious roots is limited. Therefore, in this study, we further investigated the effects of an ethyl acetate extract of TCMG (EA-TCMG) adventitious roots on in vitro antiplatelet activity in whole human blood and its effect on peripheral blood flow in mice. We found that EA-TCMG inhibited platelet aggregation with $IC_{50}$ values of 271, 180, and 147 ${\mu}g$/mL induced by collagen, adenosine-5'-diphosphate, and arachidonic acid, respectively. Among the three agonists used, thromboxane $A_2$ formation induced by arachidonic acid was markedly suppressed. Furthermore, EA-TCMG improved the peripheral circulatory disturbance by improving vascular blood flow. In conclusion, these results suggest that ethyl acetate extracts from TCMG adventitious roots might inhibit vascular platelet aggregation and thrombus formation.

Temporal expression profiling of long noncoding RNA and mRNA in the peripheral blood during porcine development

  • Objective: We investigated the temporal expression profiles of long noncoding RNA (lncRNA) and mRNA in the peripheral blood of pigs during development and identified the lncRNAs that are related to the blood-based immune system. Methods: Peripheral blood samples were obtained from the pigs at 0, 7, 28, and 180 days and 2 years of age. RNA sequencing was performed to survey the lncRNA and mRNA transcriptomes in the samples. Short time-series expression miner (STEM) was used to show temporal expression patterns in the mRNAs and lncRNAs. Gene ontology and Kyoto encyclopedia of genes and genomes analyses were performed to assess the genes' biological relevance. To predict the functions of the identified lncRNAs, we extracted mRNAs that were nearby loci and highly correlated with the lncRNAs. Results: In total of 5,946 lncRNA and 12,354 mRNA transcripts were identified among the samples. STEM showed that most lncRNAs and mRNAs had similar temporal expression patterns during development, indicating the expressional correlation and functional relatedness between them. The five stages were divided into two classes: the suckling period and the late developmental stage. Most genes were expressed at low level during the suckling period, but at higher level during the late stages. Expression of several T-cell-related genes increased continuously during the suckling period, indicating that these genes are crucial for establishing the adaptive immune system in piglets at this stage. Notably, lncRNA TCONS-00086451 may promote blood-based immune system development by upregulating nuclear factor of activated T-cells cytoplasmic 2 expression. Conclusion: This study provides a catalog of porcine peripheral blood-related lncRNAs and mRNAs and reveals the characteristics and temporal expression profiles of these lncRNAs and mRNAs during peripheral blood development from the newborn to adult stages in pigs.

폐결핵환자의 말초혈액에서 Activated T Cell의 변화 (Immunocytochemical Study on the Change of the Activated T Cells in Peripheral Blood of the Pulmonary Tuberculosis Patients)

  • 연구배경: 결핵감염시 세포매개성 면역반응이 관여하여 말초혈액내 조력 T 세포가 감소하는 것으로 알려저왔다. 그러나 말초혈액내 표지물질을 가진 세포의 수적 감소가 세포매개성 면연반응의 저하라고 할 수있지 여부가 앞으로 해결해야 될 문제인 것으로 지적되고 있다. 이에 저자들은 폐결핵 환자들의 말초혈액에서 활성화된, 또는 활성화 되고 있은 세포의 수적변화를 관찰하기 위하여 본 연구를 시행하였다. 방법: 객담 결핵균 양성인 폐결핵 환자 22명을 대상으로 말초혈액에서 IL-2R, VLA-1, TLiSAI의 활성화 표지에 대한 단세포군 항체를 면역조직 화학법으로 측정하였다. 결과: 1) 폐결핵 환자의 말포혈액에서 $T_1$(+) 세포 및 그 아형들의 단위 부피당 절대수는 $T_4$(+) 세포는 유의하게 감소하였고 $T_8$(+) 세포는 증가하였다(p<0.05). 2) 폐결핵 환자에서 전체 T임파구의 비율은 감소되어 있으며 $T_4$(+) 세포, $T_8$(+) 세포비율은 각각 유의하게 감소, 증가하였다. 또한 $T_4(+)/T_8(+)$ 비율도 유의한 감소가 있었다(p<0.05). 3) 폐결핵 환자에서 activated T cell의 단위 부피당 절대수는 대조군에 비해 모두 유의한 증가를 보였다(p<0.05). 4) 폐결핵 환자에서 activated T cell 비율은, IL-2R, VLA-1, TLiSAI, 각각 6.45+1.56%, 7.64+1.34%, 10.45+1.16%로 모두 대조군에 비해 모두 유의한 증가를 보이며 특히 TLiSAI 항체가 가장 많이 관찰 되었다. 결론: 폐결핵 감염시 말초혈액 임파구의 일부만이 활성화되어 세포매개성 면역반응에 참여하는 것으로 사료된다.

말초혈액 조혈모세포 채혈 및 이식 후 생착에 관한 연구 (A Study of Peripheral Blood Stem Cell Collection and Bone Marrow Engraftment after Peripheral Blood Stem Cell Transplantation)

  • Peripheral bood stem cell collection (PBSCC), including peripheral blood stem cell transplantation (PBSCT), has been utilized worldwide as a very beneficial treatment method instead of allogenic Bone Marrow Transplantation (BMT) because it has many advantages such as rapid bone marrow engraftment and hematopoietic recovery, easy and safe accessibility and lower risk of rejection compared with allogenic BMT. In order to identify most the observable parameter in PBSCC, we analyzed various hematological parameters before and after PBSCC, and evaluated the correlation between the time of bone marrow engraftment and the number of CD34+ cells. Thirteen patients, who underwent 54 PBSCCs from January, 2003 to August, 2004 at Chungnam National University Hospital due to various systemic neoplasms, were analyzed in aspects of various hematological parameters including CD34+ cells using by Flow Cytometry (FCM). PBSCC harvests are described below: Mononuclear cells (MNC) $2.3{\pm}1.4{\times}10^8/kg$ and CD34+ cells $0.63{\pm}0.35{\times}10^6/kg$ on average, respectively. There was a statistical significance in Hb and Hct before and after PBSCC, but not in WBC and platelet counts. The period to reach the hematological bone marrow engraftment was 13.4(10~21) days and 19.5(11~38) days according to the criteria of absolute neutrophile counts (ANC) ${\geq}500/uL$ and platelet counts ${\geq}50,000/{\mu}L$ in peripheral blood, respectively. There was a significant correlation between the numbers of CD34+ cell and ANC (p<0.05), and a borderline significance between MNC and ANC (p=0.051). We found that a group of patients, who were infused with CD34+ cells more than $3.5{\times}10^6/kg$, reached more rapidly the period of bone marrow engraftment in platelet counts (p=0.040). This present study suggested that Hb and Hct were the most useful parameters and should be closely monitored before and after PBSCC, that a PBSCT with the dosage of more than $3.5{\times}10^6/kg$ of CD34+ cells was needed to perform successful bone marrow engraftment, and additionally that platelet counts could be more useful in indicating bone marrow engraftment than ANC.

복식호흡이 조기진통 임부의 불안, 혈압, 말초 피부온도와 산소 포화도에 미치는 효과 (Effects of Abdominal Breathing on Anxiety, Blood Pressure, Peripheral Skin Temperature and Saturation Oxygen of Pregnant Women in Preterm Labor)

  • Purpose: This study was done to examine the effects of abdominal breathing on VAS-Anxiety (VAS-A), blood pressure, peripheral skin temperature and saturation oxygen in pregnant women in preterm labor. Method: The study design was a matched control group interrupted time series. Forty-six women matched to gestational age were assigned to either the experimental group (26) or control group (20). Data were collected between March 2007 and May 2008. For the experimental treatment the women performed abdominal breathing 30 times, which took 5 minutes, and did one set of 5-minute abdominal breathing daily for three days. Data collection was done before and after the abdominal breathing to measure VAS-A, blood pressure, peripheral skin temperature and oxygen saturation. Descriptive, $X^2$, Mann-Whitney U tests were used to analyze the data with the SPSS/PC+Win 15.0 program. Result: For the experimental group there were significant decreases in VAS-A (Z=-4.37, p=.00), systolic blood pressure (Z=-3.38, p=.00), and an increase in skin temperature (Z=-4.50, p=.00) and oxygen saturation (Z=-3.66, p=.00). Conclusion: These findings suggest that abdominal breathing in pregnant women in preterm labor results in decreases in anxiety(VAS-A) including biological evidences such as systolic blood pressure, and increases in peripheral skin temperature and oxygen saturation. Further longitudinal study is needed on the lasting effects and obstetric and neonatal outcomes following abdominal breathing.

Gene Expression Changes in Peripheral Blood Mononuclear Cells from Cynomolgus Monkeys Following Astemizole Exposure

  • Surrogate tissue analysis incorporating -omics technologies has emerged as a potential alternative method for evaluating toxic effect of the tissues which are not accessible for sampling. Among the recent applications, blood including whole blood, peripheral blood lymphocytes and peripheral blood mononuclear cells (PBMCs) was suggested as a suitable surrogate tissue in determining toxicant exposure and effect at the pre- or early clinical stage. In this application, we investigated transcriptomic profiles in astemizole treated Cynomolgus monkey's PBMCs. PBMCs were isolated from 4-6 years old male monkeys at 24 hr after administration45 Helvetica Light (10 mg/kg, 30 mg/kg). Gene expression profiles of astemizole treated monkey's PBMCs were determined using Affymetrix $GeneChip^{(R)}$ Human Genome U133 plus 2.0 arrays. The expression levels of 724 probe sets were significantly altered in PBMCs at 10 or 30 mg/kg after astemizole administration following determination of paired t-test using statistical criteria of ${\geq}$$1.5-fold changes at P<0.05. Gene expression patterns in PBMCs showed a considerable difference between astemizole 10 and 30 mg/kg administration groups in spite of an administration of the same chemical. However, close examination using Ingenuity Pathway Analysis (IPA) software revealed that several gene sets related to cardiotoxicity were deregulated at astemizole 10 and 30 mg/kg administration groups. The deregulation of cardiac hypertrophy related genes such as TXN, GNAQ, and MAP3K5 was observed at 10 mg/kg group. In astemizole 30 mg/kg group, genes involved in cardiotoxicity including cardiac necrosis/cell death, dilation, fibrosis, and hypertrophy were also identified. These results suggest that toxicogenomic approach using PBMCs as surrogate tissues will contribute to assess toxicant exposures and identify biomarkers at the pre-clinical stage.

고양이 말초혈액 탐식세포의 탐식능에 있어서 계난백유래물질의 효과 (Effect of Egg White Derivatives on Phagocytic Response of Feline Peripheral Blood Phagocytes)

  • The immunostimulating effects of egg white derivatives (EWD) on the phagocytic response of feline peripheral blood polymorphonuclear cells (PMN) as well as mono- nuclear cells (MNC) were examined. The phagocytic activity was analyzed by a flow cytometry system. The EWD showed directly an enhanced effect on the phagocytic response of MNC but not PMN. The phagocytic activity of MNC was enhanced by culture supernatant from MNC and PMN treated with EWD, respectively. Similarly, the phagocytic activity of PMN was enhanced by culture supernatant from MNC but not PMN treated with EWD. It was, therefore, indicated that the enhanced phagocytic activity of feline PMN could be mainly mediated by humoral factor(s) released from MNC treated with EWD. These results suggested that EWD could enhance the phagocytosis of feline peripheral blood phagocytes.
