${\omega}-3$계와 ${\omega}-6$계 고도 불포화 지방산의 혼합 비율이 다른 유지가 흰쥐의 지질대사에 미치는 영향(II) -혈청과 간 조직을 중심으로- (The Effect of Fat and Oil Differently Composed of ${\omega}-3$ and ${\omega}-6$ Polyunsaturated Fatty Acids on Lipid Metabolism of Rats -Centered on Serum and Liver Tissues-)

  • ${\omega}-3$계와 ${\omega}-6$계 PUFA가 혈청과 간장의 지질대사에 미치는 영향을 규명하기 위하여 ${\omega}-6$계 linoleic acid를 약 64% 함유하고 있는 고추종자기름과 ${\omega}-3$계 linolenic acid 함유비율이 약 60%인 들깨기름을 몇가지 비율로 혼합하여 흰쥐에게 급여하여 얻어진 결과는 다음과 같다. 혈청중 총 cholesterol 농도는 들깨기름 10%를 급여받은 1군이 고추씨기름의 함량이 높은 4군과 5군에 비하여 유의적으로 낮았으며, HDL-cholesterol 농도는 ${\omega}-6$계 linoleic acid의 비율이 높아짐에 따라 점차로 증가하는 경향이었고 ${\omega}-3$계 linolenic acid 함유율이 높아짐에 따라 유의성있게 저하하는 경향이었다. 총 cholesterol에 대한 HDL-cholesterol 농도는 10% 고추종자기름을 급여한 5군에서 가장 높았고 5% 들깨유와 5% 고추종자기름을 혼합 급여한 3군의 순이었다. 혈청중의 중성지질과 인지질 농도는 7.5% 들깨기름과 2.5% 고추종자기름의 혼합 식이군인 2군이 2.5% 들깨유와 7.5% 고추종자유 혼합 식이군인 4군에 비하여 유의적으로 낮았다. 간장중의 총 cholesterol 농도는 고추씨 기름 10% 단독 식이군에서 가장 높았으며 들깨유와 고추씨 기름의 혼합 식이군 (2, 3, 4 군)에서 낮았다. 간장중의 중성지방 농도는 10% 들깨유군 및 10% 고추씨기름의 단독 식이군에서 높은 반면 혼합 투여군에서 감소하는 경향을 보였으며 인지질 농도는 각 군간에 유의적인 차이는 없었다.

들기름과 미강유 혼합 식용유의 지방산 조성, 토코페롤 및 식물성 스테롤 및 산화안전성 측정 (Fatty Acid Composition, Contents of Tocopherols and Phytosterols, and Oxidative Stability of Mixed Edible Oil of Perilla Seed and Rice Bran Oil)

  • 들기름 및 미강유를 베이스로 개발한 혼합 식용유는 오메가-3 지방산인 리놀렌산 함량이 약 20% 정도로 매우 높다. 이는 기존의 일반 식용유 중 오메가-3 지방 함량이 매우 높은 콩기름과 카놀라유의 리놀렌산 함량이 각각 6% 및 10% 내외인 점을 감안하면, 이들 식용유에 비하여 새로 들기름과 미강유를 혼합한 식용유는 2~3배 높은 비율로 오메가-3 지방산을 제공할 수 있어서, 오메가-3 지방산 섭취가 부족한 현대인의 식단에서 중요한 오메가-3 지방산 급원으로서의 역할을 할 수 있을 것이다. 뿐만 아니라, 토코페롤 함량도 약 46.77 mg/100 g oil 정도인데, 가장 주목할 일은 토코페롤 동족류 중 비타민 E의 활성이 가장 강한 알파-토코페롤(31.71 mg/100 g oil)이 가장 많이 존재하여, 총 토코페롤의 64%를 차지하고 있어서 비타민 E로서의 기능이 높다. 혈중 콜레스테롤 함량을 낮추는 기능성이 알려진 파이토스테롤 함량이 들기름과 미강유를 혼합한 식용유에 약 7,200 ppm 함유되어 있어, 일반적인 식용유의 3,000 ppm의 약 2.3배에 달한다. 본 연구에서 개발한 들기름과 미강유를 혼합한 식용유는 일반적인 식용유인 콩기름과 거의 유사한 산화안정성을 갖는 것으로 확인되어, 일반 들기름에 비하여 현저히 높은 산화안정성이 확보되었다.

정제들깨기름의 산화안정성에 미치는 흑미 추출물의 영향 (Effect for Oxidation Stability of Refined Perilla Oil Use in Extract of Black Rice (Oryza sativa L.))

  • Proximate compositions of black rice and antioxidative effects of its methanol extract on refined perilla oil were investigated. Black rice seeds were composed of water 12.4%, crude protein 7.9%, crude fat 2.2%, crude fiber 1.0%, ash 1.4% and N-free extract 75.1%. The hexane and 80% methanol fractions in yields were higher than 80% ethanol and chloroform. In AOM(active oxygen method) test, antioxidative effects of black rice added to the refined perilla oil was increased with dose-dependent fashion in concentration of the black rice extracts. Among several extracts tested, 80% aqueous methanol extract showed the highest antioxidative activity, followed by hexane, chloroform, in that order. The peroxide value and TBA value for their antioxidation stability were also lower than that of control. This result was similar to that of AOM test.

Analysis of Volatile Oil Components and Identification of Chemotypes in Jaso (Perilla frutescens) Collected in Korea

  • Volatile oil components were analysed in Perilla frutescens accessions collected from different regions in South Korea and identified chemotypes based on the major volatile oil components. Major components out of 30 compounds identified were limonene, perillaldehyde, perillaketone, isoegomaketone, beta-caryophyllene, beta-farnesene, myristicin, and dillapiole. P. frotescens collections were classified into four chemotypes : PA type (57.7% limonene and 19.8% perillaldehyde), PK type (89.8% perillaketone), ST type (82.4% sesquiterpene, as 54.5% beta-caryophyllene and 27.9% beta-farnesene) and PP type (40.3% phenylpropenes as 13.6% myristicin and 26.7% dillapiole) and 37.8% sesquiterpenes. The majorities of P. frutescens collections in this study belong to PA type (41.9%) and PK type(38.8%).

들깨유로부터 $\alpha$-Linolenic Acid의 순수분리 (Isolation of Pure $\alpha$-Linolenic Acid from Perilla Seed Oil)

  • Low-temperature crystallization method silver nitrate-impregnated silicic acid column chromatography were applied for the isolation of pure $\alpha$-linolenic acid(ALA) from perilla seed oil. ALA or 78% in purity(HALA; yield, 83%) was obtained from the fatty acid mixture(ALA, 65.7%) derived from perilla oil by the low-temperature crystallization method, when the mixture was frozen at -8$0^{\circ}C$ for 210min. ALA over 90% in purity(yield, 71%) was also obtained from HALA ethyl esters(ALA, 78%) by the silver nitrate-impregnated silicic acid column(100cm$\times$10cm, i.d.) chromatography. In addition, the silver nitrate-impregnated silicic acid could be semipermanently used for isolation of ALA, because $Ag^{+}$ ion was not dissociated from the stationary phase.

불포화 지방산과 마그네슘, 칼슘과의 상호작용이 3-Hydroxy-3-Methylglutaryl Co-A reductase 활성에 미치는 영향 (Interaction of dietary Mg(II), Ca(II) and polyunsaturated fatty acids on 3-Hydroxy-3-Methylalutaryl Co-A reductase activity)

  • Interaction of dietary Magnesium, Calcium and Polyunsaturated fatty acid(vegetable oils)on 3-Hydroxy-3-Methylglutaryl Co-A reductase activity was studied for a period of 30 days using isocalories and isonitrogenous as a basal diet . The subject rabbits were divided into 18 feeding groups. The results are summarized as follows: 1. The ratio of ${\alpha}_1-lipoprotein$ par lipalbumin is 0.34 for control group, 0.38 the highest group fed 0.1 Mg(II) 10ml plus perilla oil and basal diet, the lowest 0.25 group fed 0.1M Mg(II) 5ml plus sesame oil and basal diet. 2. The ratio ratio of ${\alpha}_2-lipoprotein$ per lipalbumin is 0.64 for control group. 0.95 as the highest for the group fed 0.1M Ca(II) 15ml plus sesame oil and basal diet, 0.1M Ca(II) 5ml plus perilla oil and basal diet. 3. The ratio of ${\beta}-lipoprotein$ per lipalbumin is 0.71 for control group, the highest 0.81 for the groups fed 0.1M Mg(II) 10ml plus sesame oil and basal diet, the lowest 0.37 for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 4. In serum triglyceride, control group was 129.5mg%, the highest 155.4mg% for the group fed 0.1M Ca(II) 5ml plus sesame oil and basal diet, the lowest 85.7mg% for the group fed 0.1M Mg(II) 10ml plus soybean oil and basal diet. 5. In serum cholesterol, control group was 96.7mg%, the highest 152.5mg% for the group fed 0.1M Ca(II) 10ml plus sesame oil and basal diet, the lowest 80.5mg% for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 6. In case of HMG-CoA reductase activity, control group was 0.95, the highest 0.98 for the group fed 0.1M Ca(II) plus soybean oil and basal diet. 7. Interaction between metal(II) ions and polyunsaturated fatty acid(vegetable oil) are soybean oil>sesame oil>perilla oil, for Mg(II). soybean oil>perilla oil>sesame oil, for Ca(II). Therefore, it is invetigated that the interaction between metal ion and polyunsaturated fatty acid is the higher, the cholesterol level is the lower, and HMG-CoA reductase activity is increased.

Suppression of Fatty Acid Synthase by Dietary Polyunsaturated Fatty Acids is Mediated by Fat itself, not by Peroxidative Mechanism

  • This study examined the effect of dietary polyunsaturated fatty acids (PUFA) that were supplemented with vitamin E on lipid peroxidation, glutathione-dependent detoxifying enzyme system activity, and lipogenic fatty acid synthase (FAS) expression in rat liver. Male Sprague-Dawley rats were fed semipurified diets containing either 1% (w/w) corn oil or 10% each of beef tallow, corn oil, perilla oil, and fish oil for 4 wk. Alpha-tocopherol was supplemented in perilla oil (0.015%) and fish oil (0.019%). Hepatic thiobarbituric acid reactive substances, an estimate of lipid peroxidation, were not significantly different among the dietary groups. The glutathione peroxidase, glutathione reductase, and glutathione S-transferase activities were all elevated by the polyunsaturated fats, especially fish oil. The activity of FAS was reduced in the polyunsaturated fat-fed groups in the order of fish oil, perilla oil, and corn oil. The mRNA contents decreased in rats that were fed the 10% fat diets, particularly polyunsaturated fats, compared with the rats that were fed the 1% corn oil diet. Similarly, the inhibitory effect was the greatest in fish oil. These results suggest that lipid peroxidation can be minimized by vitamin E; PUFA in itself has a suppressive effect on lipogenic enzyme.

어유의 Eicosapentaenoic Acid의 식이섭취가 성숙쥐의 혈청 및 간의 지질조성에 미치는 영향 (Effect of Dietary Eicosapentaenoic Acid on Serum and Liver Lipids Patterns of Male Rat)

  • The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

키토산을 첨가한 황석어젓 섭취가 혈액학적 변화에 미치는 영향 (The Effects of Intake of Whangseoke-Jeot with Chitosan on the Hematological Changes)

  • This study was carried out to investigate the effects of intake of whangseoke-jeot with chitosan on the hematological changes. Twelve male students were divided into two groups and have been dieted for 3 days with whangseoke-jeot supplemented with/without 10% of chitosan(50,000 dalton) into seasoning. In order to decrease fishy smell and increase palatable taste effectively, minced ginger, garlic and onion as well as oil and roasted powder of perilla were included into the seasonings. Blood pressure was monitored everyday and serum was prepared following blood collection before and after 3 day diet. Chitosan was shown to decrease systolic pressure and total serum protein. On the other hand, albumin was decreased in the control group. It is noteworthy that seasoned whangseoke-jeot including perilla oil and powder seemed to decrease creatinine and total cholesterol and increase HDL. In conclusion, chitosan and perilla oil and powder might be proposed as good additives into whangseoke-jeot to relieve its highly salty effects on diet.

N-3계 및 N-6계 지방산 식이가 스트레스에 노출된 흰 쥐의 혈당과 혈청 유리지방산 및 지방산 조성변화에 미치는 영향 (The Effect of Dietary n-3 and n-6 Polyunsaturated Fats on changes in Glucose, Non Esterified Fatty Acid and Fatty Acid Compositions in Serum of Rat Exposed to Stress.)

  • This study was designed to investigate the changes in energy substrates, glucose and non-esterified fatty acid(NEFA), and fatty acid compositions in serum, following physiolgical stress in rats fed diets containing various fatty acids. Forty two Sprague-Dawley strain male rats, weighing 108$\pm$2.1g, were fed 3 different experimental diets for 4 weeks. The diets were composed of 105 fat(w/w) of either corn oil(CO;18:2 n6:57%), plant perilla oil(PO;18:3 n3:59%), or tuna fish oil(FO;20:5 n3:17%%, 22:6 n3:19%). After 4 weeks of feeding, each group wa subdiveided into (a) control, (b) 2 min swim in ice-cold water. Animals wer decapitated 20min after commencing the swim; trunk blood, brain, liver and epididymal fat pad were obtained. The levels of serum corticosterone, glucose, NEFA, triglyceride, fatty acid compositions, brain serotonin and 5-hydroxyindoleacetic acid were determined. Basal levels of corticosterone na NEFA of serum were significantly lower in fish oil fed animals than those of any other oil fed animals. Compared to either perilla oil-fed or corn oil-fed rats, cold swim stress in fish oil fed rats produced significantly smaller NEFA and larger corticosterone responses. However, there was no significant difference in basal levels of serum glucose. Stress increased serum glucose levels slightly, and the amount of increment was larger in fish oil rats than those of any other oil fed rats than those of any other oil fed rats, although all the values were normal level. Dietary fats and stress did not affect serotonin metabolism. In additions, the composition of fatty acids in serum was significantly affected by the dietary compostion of fatty acids and stress. Stress induced decreases in monounsaturated fatty acid and non-polyunsaturated fatty acid concentration in either perilla oil fed or fish group, but did not in corn oil fed group. Stress resulted in changes in fatty acid metabolism similar to that associated with essential fatty acid(EFA) dificiency, when feeding animals n-3 fatty acids in diet. In conclusion, feeding fish oil was more effective to decrease NEFA in serum than feeding perilla oil or corn oil and improved lipid metabolism, when the rats were maintained in normal or exposed to stressful environment. However, the fact that feeding diet containing n-3 fatty acids decreased EFA status under stress suggests that the requirement of n-6 PUFA should be increased in these groups.
