Plant Growth Promotion and Gibberellin A3 Production by Aspergillus flavus Y2H001 (Aspergillus flavus Y2H001의 식물생육촉진과 Gibberellin A3의 생산)

  • Perilla frutescens var. japonica Hara was collected from farmland in Seongju-gun. Fifteen endophytic fungal strains with different colony morphologies were isolated from roots of P. frutescens. Waito-c rice seedlings were treated with the concentrated culture filtrates (CF) of endophytic fungi for observation of their plant growth-promoting activities. In the results, the CF of Y2H001 fungal strain promoted the growth of the waito-c rice seedlings. The phylogenetic tree of Y2H001 strain was analyzed by the combined sequences of the partial internal transcribed spacer region (ITS) and partial betatubulin gene. Molecular and morphological studies identified the Y2H001 strain as belonging to Aspergillus flavus. In gas chromatography mass spectrometry (GC/MS) analysis of the CF of Y2H001 strain, gibberellic acid (GA) was detected and quantified. Therefore, we describe Y2H001 strain as a new $GA_3$-producing A. flavus based on morphological, molecular characteristics and analysis of secondary metabolite.

Effect of Electric Conductivity and Potassium Level of the Culture Solution on the air Pollution Sensibility of Perillar Frutescens Suwon 16 (양액의 전기전도도 및 칼륨 수준이 들깨 수원 16호의 대기오염 지표성에 미치는 영향)

  • This experiment was conducted to investigate the effect of electric conductivity (EC) and potassium (K) concentration level in the culture solution on the growth and the air pollution monitoring capability of Perilla frutescens Suwon 16 which was recognized as a possible bioindicator for air pollution. The plants were exposed to $NO_2$, $O_3$, $SO_2$ and $NO_2+SO_2$ within a phytotron at $25^{\circ}C$ (day) or at $20^{\circ}C$(night) with 70% of relative humidity and evaluated the effect of EC and K level on the injury of the plants, The highest dry weights were gained as 16.3 g/plant at 0.5 dS/m of EC and as 32.3 g/plant at 100 and 200mg/l of K concentration, respectively. The diffusive resistances appeared as low values at 0.5 dS/m of EC and at 50, 100mg/l of K concentration. The increasing of the visible injury with increasing the dose of air pollutants could be taken at 0.5. 1.0 ds/m of EC and at 50, 100mg/l of K. The recommendable level of EC and K of the culture solution were 0.5-1.0 dS/m of electric conductivity. 50-100mg/l of K for the plant as an air pollutant biomonitor, when the 7th or 8th leaf was developed.

  • This experiment was conducted to verify the responses of major crop species to saline stress.To determine the saline tolerance of crop species, sevral crop species were cultivated under sand-culture system using Hoagland's nutrient solution whit 200mM NaCl. The crop species showing saline tolerance were cotton(Gossypium indicum), maize(Zea mays), barely(Hordeum vulgare), and wheat(Triticum aestivum) but perilla (Perilla frutescens) and leguminous crops, mung bean(Phaseolus radiatus), azuki bean(Phaseolus angularis) and soy bean(Glycin max) showed very por tolerance. One the typical symptom was the darkening of leaf color due to increase of chlorophyll concentration.Among of the plant families, Fabaceae was the most susceptible but crop species belonging to Poaceae were more proper for cultivating on reclaimed tidal land in the course of desalinazation. It was suggrsted that the crop species belonging to Fabaceae, a sensitive family to soil salinity, must be cultivated when the soil salinity decreased below 10ds/m. To know the critical salinity level for crop growth,salinity of saline soil collected from reclaimed tidal land was adjusted from 10.0ds/m tp41.7ds/m with tap water. It was suggested that the ECs of the soil in which the plant height of each crop spicies was reduced to 50% of control plant were 22.6 and 21.7 ds/m in rice, barley, corn, mung bean, and soy bean,respectively.

Characterization of broad bean wilt virus 2 isolated from Perilla frutescens in Korea (국내 잎들깨에서 발생한 잠두위조바이러스2의 특성 구명)

  • Broad bean wilt virus 2 (BBWV2) is a species in the genus Fabavirus and family Secoviridae, which is transmitted by aphids and has a wide host range. The BBWV2 genome is composed of two single-stranded, positive-sense RNAs, RNA-1 and RNA-2. The representative symptoms of BBWV2 are mosaic, mottle, vein clearing, wilt, and stunting on leaves, and these symptoms cause economic damage to various crops. In 2019, Perilla fructescens leaves with mosaic and yellowing symptoms were found in Geumsan, South Korea. Reverse-transcription polymerase chain reaction (RT-PCR) was performed with specific primers for 10 reported viruses, including BBWV2, to identify the causal virus, and the results were positive for BBWV2. To characterize a BBWV2 isolate (BBWV2-GS-PF) from symptomatic P. fructescens, genetic analysis and pathogenicity tests were performed. The complete genomic sequences of RNA-1 and RNA-2 of BBWV2-GS-PF were phylogenetically distant to the previously reported BBWV2 isolates, with relatively low nucleotide sequence similarities of 76-80%. In the pathogenicity test, unlike most BBWV2 isolates with mild mosaic or mosaic symptoms in peppers, the BBWV2-GS-PF isolate showed typical ring spot symptoms. Considering these results, the BBWV2-GS-PF isolate from P. fructescens could be classified as a new strain of BBWV2.

  • The screening of anti-cancer activity for the MeOH extracts of 163 natural sources, which were registered as edible plants by Korea Food & Drug Administration, exhibited 9 extracts to have significant inhibitory effects on farnesyl-protein transferase (FPTase) and phosphatase of regenerating liver-3 (PRL-3). In order to confirm the inhibitory activity of these active extracts, the activity assay was repeated for some fractions obtained from the active extracts using Medium Pressure Liquid Chromatography (MPLC). Some fractions of Carya illinoensis, Chlorella vulgaris, Panicum miliaceum, Perilla frutescens, Rosmarinus officinalis showed over 50% inhibitory activity on FPTase as well as those of Capsella bursa-pastoris, C. illinoensis, C. vulgaris, Coix lacrymajobi, Myristica fragrans, P. miliaceum, R. officinalis did over 50% inhibitory activity on PRL-3.

Host Range, Occurrence and Developmental Characteristics of Ceroplastes pseudoceriferus (Homoptera : Coccidae) on Persimmon Trees (감나무에 기생하는 뿔밀깍지벌레의 기주범위, 발생소장, 각 태별 생육 특성)

  • This study investigated host range, seasonal occurrence and developmental characteristics of Ceroplastes pseudoceriferus Green on persimmon trees. The host plants covered 34 families and 66 species, including newly recorded species of Chenopodum album var. centrorubrum, Glycine max, Begonia semperflorens, Pharbitis nil, Perilla frutescens var. japonica and Erigeron canadensis. There was one generation of C. pseudoceriferus a year with larval stage expanding from mid-June to mid-October. Average number of eggs laid per female was 1,073$\pm$177.3. Hatching rate was 97.3% and was not affected by temperatures and photoperiods. Stadium varied as 23.4 days in eggs, 128.3 days in larvae and 213.3 days in female adults including overwintering periods at natural condition.

  • Isoegomaketone (IK) was isolated from Perilla frutescens, which has been widely used as a food in Asian cuisine, and evaluated for its biological activity. We have already confirmed that IK induced the HO-1 expression via Nrf2 activation in RAW264.7 cells. In this study, we investigated the effect of IK on the mechanism of HO-1 expression. IK upregulated HO-1 mRNA and protein expression in a dose dependent manner. The level of HO-1 mRNA peaked at 4 h after $15{\mu}M$ IK treatment. To investigate the mechanisms of HO-1 expression modulation by IK, we used pharmacological inhibitors for the protein kinase C (PKC) family, PI3K, and p38 MAPK. IK-induced HO-1 mRNA expression was only suppressed by SB203580, a specific inhibitor of p38 MAPK. ROS scavengers (N-acetyl-L-cysteine, NAC, and glutathione, GSH) also blocked the IK-induced ROS production and HO-1 expression. Furthermore, both NAC and SB203580 suppressed the IK-induced Nrf2 activation. In addition, ROS scavengers suppressed other oxidative enzymes such as catalase (CAT), glutathione S-transferase (GST), and NADH quinone oxidoreductase (NQO-1) in IK-treated RAW264.7 cells. Taken together, it can be concluded that IK induced the HO-1 expression through the ROS/p38 MAPK/Nrf2 pathway in RAW264.7 cells.

  • New herbal formula (NHF) is the ethanol extract mixture of Puerariae radix, Artemisia capillaries and Perilla frutescens. We have Investigated the effects on anti-inflammation by NHF and attempted acetic acid induced writhing to verify the analgesic effect. Macrophages and chondrocytes were obtained from mouse and rabbit. Inflammation was induced bγ interleukin-1, tumor necrosis $factor-{\alpha}$, $interferon-{\gamma}$, and lipopolysaccharide. NHF showed strong inhibitory efficacy against cytokine-induced proteoglycan degradation, $PGE_2$ production, NO production, and MMP-9 expression in rabbit articular chondrocyte. In the writhing test, NHF exhibited a dose-dependent inhibition of writhing. Futhermore, NHF increased the activity of SOD. NHF have anti-inflammatory and analgesic activities, and could be a good herbal medicine candidate for curing of osteoarthritis.

  • The experiments were conducted to isolate the etiogical agent and to survey the distribution of Nosema disease in honey bees. The results obtained were as follows: 1. The etiological agent of the so-called "crawling disease" in honey bees characterized by the symptoms of crawling, diarrhea, and enteritis etc. was first isolated and identified with Nosema apis (Zander 1909) in Korea. 2. 455 colonies were randomely sampled and surveyed in 4,766 bee colonies out of 56 apiaries and 51 colonies (11.2%) out of 455 bee colonies were infected with N, apis. 3. Infection rates according to the period of honeyflow as follows: Brassica napus (Apr.): 25/130 colonies (18.4%) Rohinia pseudoacacia (May) : 8/55 colonies (14.%) Trifolium repels(Jun.): 15/99 colonies (13.6%) Castanea crenate (Jul.): 3/46 colonies (6.5%) Lespedeza bicolor(Aug.): 0/60 colonies (-) Fagopyrumesculentum(Sept.) & Perilla frutescens(Oct.) 0/65 colonies (-) 4. The typical clinical signs of Nosema disease were appeared on loth day after N. apis was orally administered with the level of $16{\times}10^4$ spores/ml to the healthy adult bees. Spores could be harvested with the level of $121{\sim}236{\times}10^4$ spores/ml on 10th day and $392{\sim}429{\times}10^4$ spores/ml on 15 days after infection. 5. In adult honey bees infected with N. apis artificially the 50% lethal day of life-span was 9 to 10 days and 100% lethal day was 16 to 19 days. However, in the control 50% lethal day was 19 to 23 days and 100% lethal day was 31 to 33 days.

Anti-Adipogenic Effects of Ethanol Extracts Prepared from Selected Medicinal Herbs in 3T3-L1 Cells

  • Obesity is a major risk factor for various metabolic diseases such as cardiovascular disease, hypertension, and type 2 diabetes mellitus. In this study, we prepared ethanol extracts from Agastache rugosa (ARE), Chrysanthemum zawadskii (CZE), Mentha arvensis (MAE), Perilla frutescens (PFE), Leonurus sibiricus (LSE), Gardenia jasminoides (GJE), and Lycopus coreanus (LCE). The anti-oxidant and anti-adipogenic effects were evaluated. The $IC_{50}$ values for ascorbic acid and LCE against 2,2-diphenyl-1-picrylhydrazyl radicals were $246.2{\mu}g/mL$ and $166.2{\mu}g/mL$, respectively, followed by ARE ($186.6{\mu}g/mL$), CZE ($198.6{\mu}g/mL$), MAE ($337.1{\mu}g/mL$), PFE ($415.3{\mu}g/mL$), LSE ($548.2{\mu}g/mL$), and GJE ($626.3{\mu}g/mL$). In non-toxic concentration ranges, CZE had a strong inhibitory effect against 3T3-L1 adipogenes (84.5%) than those of the other extracts. Furthermore, the anti-adipogenic effect of CZE is largely limited in the early stage of adipogenesis, and we revealed that the inhibitory role of CZE in adipogenesis is required for the activation of Wnt signaling. Our results provide scientific evidence that the anti-adipogenic effect of CZE can be applied as an ingredient for the development of functional foods and nutri-cosmetics for obesity prevention.
