The biochemical change during regeneration of perilla callus were investigated by comparing total protein and lipid contents, protein band pattern in SDS-PAGE, and fatty acid composition in the calli cultured for various period(0, 1, 3, 5 and 6 weeks) Calli were induced from cotyledon and hypocotyl explants of peplants of perilla on perilla on MS medium containing BA(0.5 mg/L) and NAA(0.5mg/L). The protein contents reached the peak at 3 weeks after induction of calli, and then was decreased. Total lipid contents was decreased as the culture period increased. The band pattern of polypeptides showed that 30KD and 45KD polypeptides and 22KD and 45KD polypetides were major proteins in the cotyledon and hypocotyl explants, respectively. However increase of culture period only 30KD protein was highly accumulated.
The antioxidative activity of various enzymatic extracts from leaves of Perilla frutescens var. japonica was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the leaves were enzymatically hydrolyzed by 8 carbohydrases (Dextrozyme, AMG, Promozyme, Maltogenase, Termamyl, Viscozyme, Celluclast, and BAN) and 9 proteases [Flavourzyme, Neutrase, Protamex, Alcalase, PP-trypsin (trypsin from porcine pancreas), papain, pepsin, $\alpha$-chymotrypsin, and BP-trypsin (trypsin from bovine pancreas)]. The DPPH radical scavenging activities of Promozyme and Alcalase extracts were the highest, and the $IC_{50}$ values were 77.25 and $109.66\;{\mu}g/mL$, respectively. All enzymatic extracts of the leaves scavenged hydroxyl radical, and the $IC_{50}$ values of Celluclast and pepsin extracts which were the highest activity were 243.34 and $241.86\;{\mu}g/mL$, respectively. The BAN and $\alpha$-chymotrypsin extracts showed the highest scavenging activities, and the $IC_{50}$ values were 21.13 and $33.23\;{\mu}g/mL$, respectively. The pepsin extracts from the leaves showed protective effect on $H_2O_2$-induced DNA damage. In addition, the pepsin extracts decreased cell death in PC-12 cells against $H_2O_2$-induced oxidative damage. The findings of the present study suggest that enzymatic extracts of the leaves possess antioxidative activity.
The cytotoxic and antitumor activity of Perilla frutescens extract on cultured 3T3 fibroblast and skin melanoma cells were evaluated by tetrazolium MTT (MTT) and neutral red (NR) colorimetric assay methods. Lactate dehydrogenase activity was also measured. The light microscopic study was carried out to observe morphological changes of cultured mouse fibroblast and skin melanoma cells. The results were as follows: 1. Water and ether extracts showed a significant cytotoxicity in 3T3 fibroblast and all extracts exhibited a significant anti-tumor activity in skin melanoma cells. Methanol, ethyl acetate and ethanol extracts showed low cytotoxic effects, but exhibited a high anti-tumor activity. 2. The MTT absorbance in 3T3 fibroblast was significantly decreased by treatment with ether, water, chloroform and ethanol extracts and skin melanoma cells was significantly decreased by treatment with all extracts. The difference in MTT absorbance in two cell types was most remarkable when treated with methanol and ethanol extracts. 3. Methanol and ethyl acetate extracts showed the strongest effect in growth inhibition of melanoma cells. These results indicated that methanol extract possessed a low cytotoxicity and a strong anti-tumor activity.

BACKGROUND/OBJECTIVES: Perilla frutescens (L.) Britton var. (PF) sprout is a plant of the labiate family. We have previously reported the protective effects of PF sprout extract on cytokine-induced ${\beta}-cell$ damage. However, the mechanism of action of the PF sprout extract in type 2 diabetes (T2DM) has not been investigated. The present study was designed to study the effects of PF sprout extract and signaling mechanisms in the T2DM mice model using C57BL/KsJ-db/db (db/db) mice. MATERIALS/METHODS: Male db/db mice were orally administered PF sprout extract (100, 300, and 1,000 mg/kg of body weight) or rosiglitazone (RGZ, positive drug, 1 mg/kg of body weight) for 4 weeks. Signaling mechanisms were analyzed using liver tissues and HepG2 cells. RESULTS: The PF sprout extract (300 and 1,000 mg/kg) significantly reduced the fasting blood glucose, serum insulin, triglyceride and total cholesterol levels in db/db mice. PF sprout extract also significantly improved glucose intolerance and insulin sensitivity, decreased hepatic gluconeogenic protein expression, and ameliorated histological alterations of the pancreas and liver. Levels of phosphorylated AMP-activated protein kinase (AMPK) protein expression also increased in the liver after treatment with the extract. In addition, an increase in the phosphorylation of AMPK and decrease in the phosphoenolpyruvate carboxykinase and glucose 6-phosphatase proteins in HepG2 cells were also observed. CONCLUSIONS: Our results sugges that PF sprout displays beneficial effects in the prevention and treatment of type 2 diabetes via modulation of the AMPK pathway and inhibition of gluconeogenesis in the liver.

BACKGROUND/OBJECTIVES: The accumulation of amyloid-${\beta}$ ($A{\beta}$) in the brain is a hallmark of Alzheimer's disease (AD) and plays a key role in cognitive dysfunction. Perilla frutescens var. japonica extract (PFE) and its major compound, rosmarinic acid (RA), have shown antioxidant and anti-inflammatory activities. We investigated whether administration of PFE and RA contributes to cognitive improvement in an $A{\beta}_{25-35}$-injected mouse model. MATERIALS/METHODS: Male ICR mice were intracerebroventricularly injected with aggregated $A{\beta}_{25-35}$ to induce AD. $A{\beta}_{25-35}$-injected mice were fed PFE (50 mg/kg/day) or RA (0.25 mg/kg/day) for 14 days and examined for learning and memory ability through the T-maze, object recognition, and Morris water maze test. RESULTS: Our present study demonstrated that PFE and RA administration significantly enhanced cognition function and object discrimination, which were impaired by $A{\beta}_{25-35}$, in the T-maze and object recognition tests, respectively. In addition, oral administration of PFE and RA decreased the time to reach the platform and increased the number of crossings over the removed platform when compared with the $A{\beta}_{25-35}$-induced control group in the Morris water maze test. Furthermore, PFE and RA significantly decreased the levels of nitric oxide (NO) and malondialdehyde (MDA) in the brain, kidney, and liver. In particular, PFE markedly attenuated oxidative stress by inhibiting production of NO and MDA in the $A{\beta}_{25-35}$-injected mouse brain. CONCLUSIONS: These results suggest that PFE and its active compound RA have beneficial effects on cognitive improvement and may help prevent AD induced by $A{\beta}$.
자소엽의 연중생산에 필요한 파종전처리 기술을 확립하기 위하여, 자소종자의 $GA_3$ 처리시 적정 처리농도, 침지시간 및 고온발아에 미치는 영향을 조사하였고, 만기파종시 포장에서 $GA_3$ 처리, 복토깊이 및 왕겨피복 등이 출아에 미치는 상호작용을 비교하였다. $GA_3$ 처리농도에 따른 발아특성은 100 ppm에서 가장 좋았으며, 100 ppm $GA_3$ 처리시 종자의 침지시간에 따른 발아특성은 12시간 이상에서는 유의한 차이가 없었다. 발아온도에 따른 100 ppm $GA_3$ 처리효과는 온도가 높을수록 발아속도와 발아일수가 빨라졌으며, 11일째에는 발아온도에 관계없이 95%이상의 높은 발아율을 보였으나, 무처리구는 $20^{\circ}C$ 이상 온도가 높을수록 발아율이 낮았다. 만기파종시 출아율은 파종시기에 관계없이 100 ppm $GA_3$용액에 침지파종후 5 mm의 복토와 10 mm 왕겨피복구에서 가장 높았다.
Perilla frutescens var. acuta Kudo (PF) is a traditional Korean medicinal herb for allergic reaction regulation. In the present study, we investigated the effect of 30% ethanol extract powder of PF (EPPF) and rosmarinic acid (RA), the active compound of EPPF on various allergic reactions using in vivo and in vitro models. EPPF and RA significantly inhibited compound 48/48-induced systemic anaphylactic reaction and histamine release (P < 0.05). In addition, EPPF and RA significantly inhibited passive cutaneous anaphylaxis (PCA) in a dose-dependent manner (P < 0.05). These effects were stronger than those of disodium cromoglycate, the reference drug tested. EPPF and RA also significantly inhibited production of inflammatory cytokines, tumor necrosis factor-a interleukin (IL)-6, and vascular endothelial growth factor on the PCA reaction and phorbol 12-myristate 13-acetate and calcium ionophore A23187-stimulated human mast cell line, HMC-1 cells (P < 0.05). Moreover, EPPF and RA showed an inhibitory effect on lipopolysaccharide (LPS)-induced IL-4 production from whole spleen cells. Finally, EPPF and RA significantly decreased IL-4-dependent IgE production by LPS-stimulated whole spleen cells (P < 0.05). In conclusion, these results indicate that EPPF has potent anti-allergic activities.
The purpose of this study was to evaluate the protective effect of the Perilla frutescens cv. Chookyoupjaso mutant water extract (PFWE) on gamma ray-induced oxidative stress in mice. Gamma-ray is one of the sources for inducing oxidative stress. The study was divided into 6 groups with 6 mice for each treatment. Groups I and II were treated with saline (vehicle) only, groups III, IV, V, and VI were pretreated with PFWE 10, 20, 50, $50mg\;kg^{-1}$ respectively for 2 weeks before gamma radiation. And then groups II, III, IV, V were irradiated. We found that the activities of aspartate transaminase (AST) and alanine transaminase (ALT) were increased and the activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and glutathione reductase (GR) were decreased by irradiation in mice. However, treatment of PFWE attenuated the activities of AST and ALT in a dose-dependent manner in irradiated mice. Furthermore, treatment of PFWE significantly increased the activities of SOD, GPx, and GR in a dose-dependent manner in irradiated mice, except for the CAT. Interestingly, the activities of GPx and GR were significantly increased by PFWE treatment. Taken together, PFWE could be effective in protecting on gamma ray-induced oxidative stress in mice.
자소는 여러 질환을 치료하는 식물로 알려져 있는데, 본 연구는 자소 추출물을 이용하여 항산화 활성 및 항염증 활성을 알아보았다. 자소 추출물을 이용한 DPPH, FRAP 실험 결과, 농도에 따른 항산화 활성을 확인할 수 있었으며, 자소자 추출물의 경우 고온/고압 추출시 활성이 증가하는 것을 알 수 있었다. 마우스 동물모델을 이용한 항아토피활성 결과 귀와 상피의 비후를 감소시키고, 면역세포의 침투현상을 억제하는 것으로 나타났다. 이상의 결과로, 자소는 항산화 및 염증완화 소재로 이용할 수 있음을 확인하였다.

The contents of functional components in the organically agricultural perilla leaves were compared with the perilla leaves grown in conventional condition. Perilla leaves used in the experiment were purchased or harvested from each three organic farm houses and conventional farm houses in Miryang city. The analyzed components included total phenol, total flavonoid, vitamin C, vitamin E, ${\beta}$-carotene, GABA, caffeic acid, rosmarinic acid, Total N, Zn, Fe, Ca, Mg, Na, K, and P. Organically grown perilla leaves had vitamin E 49% greater than the conventional perilla leaves, and 21% for TN, 29% for Ca, and 73% for Mg, while conventionally grown perilla leaves contained K 16% higher than organic ones. Other components were not showed the differences.
