The Autophagy Protein CsATG8 is Involved in Asexual Development and Virulence in the Pepper Anthracnose Fungus Colletotrichum scovillei

  • Autophagy serves as a survival mechanism and plays important role in nutrient recycling under conditions of starvation, nutrient storage, ad differentiation of plant pathogenic fungi. However, autophagy-related genes have not been investigated in Colletotrichum scovillei, a causal agent of pepper fruit anthracnose disease. ATG8 is involved in autophagosome formation and is considered a marker of autophagy. Therefore, we generated an ATG8 deletion mutant, ΔCsatg8, via homologous recombination to determine the functional roles of CsATG8 in the development and virulence of C. scovillei. Compared with the wild-type, the deletion mutant ΔCsatg8 exhibited a severe reduction in conidiation. Conidia produced by ΔCsatg8 were defective in survival, conidial germination, and appressorium formation. Moreover, conidia of ΔCsatg8 showed reduced lipid amount and PTS1 selectivity. A virulence assay showed that anthracnose development on pepper fruits was reduced in ΔCsatg8. Taken together, our results suggest that CsATG8 plays various roles in conidium production and associated development, and virulence in C. scovillei.

Characterization and Pathogenicity of New Record of Anthracnose on Various Chili Varieties Caused by Colletotrichum scovillei in Korea

  • The anthracnose disease caused by Colletotrichum species is well-known as a major plant pathogen that primarily causes fruit rot in pepper and reduces its marketability. Thirty-five isolates representing species of Colletotrichum were obtained from chili fruits showing anthracnose disease symptoms in Chungcheongnam-do and Chungcheongbuk-do, South Korea. These 35 isolates were characterized according to morphological characteristics and nucleotide sequence data of internal transcribed spacer, glyceraldehyde-3-phosphate-dehydrogenase, and ${\beta}$-tubulin. The combined dataset shows that all of these 35 isolates were identified as C. scovillei and morphological characteristics were directly correlated with the nucleotide sequence data. Notably, these isolates were recorded for the first time as the causes of anthracnose caused by C. scovillei on pepper in Korea. Forty cultivars were used to investigate the pathogenicity and to identify the possible source of resistance. The result reveals that all of chili cultivars used in this study are susceptible to C. scovillei.

Resistance to Anthracnose Caused by Colletotrichum acutatum in Chili Pepper(Capsicum annuum L.)

  • Pepper fruit anthracnose, caused by Colletotrichum acutatum, results in serious yield loss and affects crop quality in many Asian countries, making it a disease of economic consequence. A source resistant to C. acutatum was identified by the AVRDC within the line Capsicum chinense Jacq. PBC932. The resistant breeding line C. annuum AR is the $BC_3F_6$ generation derived from C. chinense Jacq. PBC932. The inheritance of resistance to C. acutatum was analyzed in segregating populations derived from the two crosses HN 11$\times$AR and Daepoong-cho$\times$AR. Detached mature green fruits were inoculated using microinjection method. The disease response was evaluated as the disease incidence at 7 DAI. The segregation ratios of resistance and susceptibility to C. acutatum in the $F_2$ and $BC_R$ populations derived from the two crosses fit significantly to a 1:3 Mendelian model. This indicates that the resistance of AR to C. acutatum is controlled by a single recessive gene.

Antagonistic activity of Streptomyces halstedii and S. violaceusniger In pepper anthracnose fungus Colletotrichum gloeosporioides

  • More than 1200 microorganisms were isolated from soil samples collected from various sources and localities. Among the isolates, 2 actinomyces (TH-04 and BA313) and 1 Bacillus sp. (CJ3) were selected as antagonists to pepper anthracnose fungus Colletotrichum gloeosporioides. These 3 isolates inhibitied mycelial growth of C gloeosporioides and the inhibition rates were over 70% on PDA. When the isolates were co-cultured with conidia of C. gloeosporioides in potato dextrose broth, conidial germination was severely inhibited and the inhibition rates of TH-04, BA313, and CJ3 at 24 hours were 75%, 72%, and 68%, respectively. The inhibition rates at n hours incubation were not much different from the rates at 24 hours. To check the activity on the plant, each isolate was mixed with equal volume of conidial suspension of C. gloeosporioides and wound-inoculated on green pepper fruit. After 6 days, the anthracnose lesions on the fruits inoculated with the mixture were much smaller than the lesions caused by the C. gloeosporioides itself. The lesion areas of TH-04 or BA313 treated pepper were less than 30% of the check. TH-04 and BA313 also showed antagonistic activity to Phytophthora spp. and Botrytis cinerea. By scanning electron microscopy and fatty acid analyses (MIDI), TH-04 and BA313 were identified to Streptomyces halstedii and S. violaceusniger, repectively.

  • Fruit ripening represents a genetically synchronized system that involves developmental process unique to plant species, The phenomenon of ripening includes changes in color, texture, respiration rate, flavor, and aroma. Ripe fruits generally exhibit increased susceptibility to pathogen infection. However, fruits as a reproductive organ have their own protection mechanism against pathogens to maintain their integrity during seed maturation. In several nonclimacteric fruits, such as cherry, grape, and pepper, that do not have an ethylene burst during ripening, resistance against phytopathogens increases during ripening. Colletotrichum gloeosporioides is a causal agent of anthracnose disease in pepper plants (Capsicum annuum). We have established that C. gloeosporioides has susceptible and resistant interactions with pepper fruits during pre- and post-ripening stages, respectively. And we have interested in looking for a molecular mechanism that would explain the fungal resistance during ripening of nonclimacteric pepper fruit. In this presentation, a molecular characterization of the pepper esterase gene (PepEST) that is highly expressed in the resistant response will be demonstrated as an example of development and industrial applications of versatile-usable genes of plant.

Synthesis of Pyto-patch as Silver Nanoparticle Product and Antimicrobial Activity (은나노 제품인 Pyto-patch의 제조공정 및 Pyto-patch의 고추 탄저병 방제효과)

  • The commercial product "pyto-patch" prepared as nano sized silver particle less than 5 nm, has effective antifungal activity against Collectotrichum gloeosporioides, Botrytis cinerea, Sclerotinia sclerotiorum in vitro. As a fungal growth inhibiton mechanism, it can reduce spore germination rate and mycelial growth. As s promising fungicide, Phyto-patch can control anthracnose effectively. The spore of C. gloeosporioides dipped in 5 ppm phyto-patch dilute suppressed germination rate to 13.2%, and mycelial growth stopped for 15 days at 10 ppm. The spore postinoculated on 10 ppm phyto-path smeared PDA surface could not germinate for 3 days and prohibit pathogen infection effectively. In field test, the anthracnose development of 4 ppm phyto-patch treated plot was less than 7% after 21 days compaired to 40% of it in untreated plot. In heavy rainfall season, pepper anthracnose effectivly contrrolled by regular 10 ppm phytopatch spraying every 7 days. The diseased pepper fruit decreased to 5.8% compaired to 94.6% in untreated plot. During drying period, the diseased pepper fruit havested in phyto-patch treated plot was 24.2%, but pepper fruit havested in untreated plot destroyed to 100% within 3 days. The nano silver particle coated on multching textile prevented late blight of pepper effectively and disease occurance delayed about month.

In vitro Fruit Assay for the Evaluation of Fungicide Activity Against Pepper Anthracnose (살균제 효과 검정을 위한 고추 탄저병의 실내 열매 검정법)

  • This study was performed to investigate the effect of conidial density, wetness period and temperature on conidial germination, appressoria formation and disease incidence. While there was not significantly correlated between conidial density and temperature, and conidial germination and appressoria formation, there was a significant correlation between those factors and disease incidence. The longer wetness period was, the higher the ratio of conidial germination, appressoria formation and the disease incidence was. The optimum conidial density, temperature and wetness period was $1{\times}10^6$ conidia $mL^{-1}$, $30^{\circ}C$ and 5 days, respectively. In case the wetness period was more than 5 days, the typical symptom was not found on pepper fruits because of the overgrowth of mycelia. Using this fruit assay method, which the pepper anthracnose pathogens were inoculated by spraying spore suspension on non-wounded or wounded pepper fruits, control effect of three fungicides were evaluated against pepper anthracnose by the protective and/or the curative application. Propineb showed high protective control activity, while it showed curative control activity on unwounded fruits, but did not showed curative control activity on wounded fruits. Tebuconazole, one of curative fungicide, showed higher control activity in non-wound inoculation than wound inoculation. Trifloxystrobin, one of strobilurin group, showed high both protective and control activity against anthracnose. In conclusion, we supposed that the newly developed in vitro pepper fruit assay can be used to evaluate antifungal activity of control agents against pepper anthracnose.

  • One of the most important vegetables consumed in Korea, chili peppers (Capsicum annuum) are widely cultivated around the world. Chili peppers have been severely damaged by anthracnose during their growth, so it is important to study prevention and resistance varieties. K1 anthracnose was inoculated against four cultivar of chili peppers that are resistant to anthracnose and one cultivar that is sensitive. The area of the disease that appeared over time was photographed and quantified through the program was analyzed. Through the ratio of the area of chili pepper fruit and the area of the bottle, the sensitive variety An-S showed weak reactions to anthracnose with about 40%, the resistant variety An-12R (23%), An-Tan (21%), and An-9R (19%), and PBC81 showed a strong response to anthracnose with about 11%. These quantitative value can be used as a basis for comparison in conducting resistance studies for new varieties.

Effect of pepper tree (Schinus molle) essential oil-loaded chitosan bio-nanocomposites on postharvest control of Colletotrichum gloeosporioides and quality evaluations in avocado (Persea americana) cv. Hass

  • Preventive and curative activity of postharvest treatments with chitosan nanoparticles (CS) and chitosan biocomposites loaded with pepper tree essential oil (CS-PEO) against anthracnose were evaluated on Avocado (Persea americana) cv. Hass artificially inoculated in rind wounds. After 10 days of storage significant preventive and curative activity against Colletotrichum gloeosporioides was observed with the absence of internal damage by applying CS and CS-PEO. Quality parameters like water losses and firmness changes were assessed on fruit treated. CS and CS-PEO were effective to reduce water losses and firmness losses.
