Occurrence of Bluish Green Mold of Pleurotus eryngii by Penicillium corylophilum (Penicillium corylophilum에 의한 큰느타리버섯(Pleurotus eryngii) 푸른곰팡이병의 발생)

  • Bluish green mold of Pleurotus eryngii caused by Penicillium was observed in Seungju on Feburary 4, 1999. The causal fungus was identified as Penicillium corylophilum based on its cultural and morphplogical characteristics. Conidiophores one-stage branched, terminating in a whorl of $2{\sim}4$ metulae. Metulae mostly $10{\sim}18{\times}2.5{\sim}3.0\;{\mu}m$. Phialides flask-shaped, $9{\sim}13{\times}2.2{\sim}2.8\;{\mu}m$. Conidia subglobose, $2.5{\sim}3.2{\times}2.5{\sim}3.0\;{\mu}m$. Colonies on Czapek agar mostly attaining a diameter of 21 mm within 7 days at $25^{\circ}C$. Colour blue-green soon becoming grey-green. Pathogenicity test by wound inoculation revealed that Penicillium corylophilum caused the same symptoms as observed in the field at $6{\sim}7$ days after inoculation.

Identification of Seed-borne Penicillium spp. on Gramineae Crops Based on Morphological Characteristics (형태적 특성에 의한 벼과작물 종자전염 Penicillium spp.의 동정)

  • A total of 81 isolates of Penicillium were isolated from postharvest seeds of barely, Job's-tears, maize, sorghum and rice from 1997 to 2003. Based on the morphological characteristics, they were identified as P. chrysogenum, P. citrinum, P. cyclopium, P. oxalicum, P. polonicum, P. purpurogenum and P. viridicatum. P. chrysogenum was detected from Job's-tears, rice and sorghum seeds, P. citrinum from maize seeds, P. cyclopium from sorghum seeds, P. oxalicum from barely, maize, sorghum and rice seeds, P. purpurgenum from maize, rice, sorghum seeds, P. viridicatum from Job's-tears, maize and rice seeds, P. polonicum from Job's-tears, maize, rice and sorghum seeds. Among these species, P. cyclopium, P. polonicum and P. purpurogenum were first reported in Korea. Especially, about 50% of the Penicillium isolates detected from the seeds were P. polonicum. Identification of the Penicillium species using morphological characteristics was difficult especially for the species belonging to the subgenus Penicillium such as P. polonicum.

Distribution and Characteristics of Penicillium spp. in Meju, aKorean Traditional Fermented Soybean Brick (전통 메주에서의 Penicillium spp.의 분포 및 특징)

  • Penicillium spp. are frequently found in meju, a Korean traditional fermented soybean brick. We isolated and identified 96 Penicillium spp. from 22 traditional meju, and their β-tubulin genes were sequenced for the genetic and taxonomic study. Penicillium Section Viridicata was the most commonly isolated group. Notably, we also isolated and identified Penicillium roqueforti, a crucial industrial strain employed in the fermentation of blue cheese. Additionally, certain strains exhibited relatively high protease and γ-glutamyl transpeptidase activities, suggesting that they might contribute to the development of kokumi flavor during meju fermentation. Interestingly, all eight Penicillium spp., including P. roqueforti, were found to possess both types of MAT1 genes. This intriguing finding suggests the feasibility of strain improvement through mating, thereby offering opportunities for industrial applications. Therefore, these studies pave the way for a deeper exploration of Penicillium's role in meju fermentation, potentially leading to the development of starters for producing plant-based cheese-flavored condiments.

A Study on Growth of Aspergillus versicolor and Penicillium polonicum within the beeswax treated paper (밀랍지 내에서의 Aspergillus versicolor와 Penicillium polonicum의 생장 특성 연구)

  • Paper composed mainly of organic materials and they can be used as nutrient by microorganisms. Therefore microorganisms are damaging the important documents and works of art irreversibly. Previous research reported that mycelium or spores of the fungi were observed on the wax of the volumes of King Sejong especially were heavily deteriorated by fungi. Fungal strains isolated from the annals of Joseon dynasty were identified as Biscogniauxia atropunctata, Aspergillus versicolor, Penicillium polonicum, Ceriporia lacerata, Irpex lacteus. Especially Aspergillus and Penicillium are able to grow on the substrates having a 7-8% moisture content, which is much lower than the general storage condition (RH55-60%) of the paper cultural properties. Moreover, they are known as fungi cause paper deterioration and discoloration. In this study, we selected Aspergillus versicolor and Penicillium polonicum among five strains, then biological aging has been executed for 30 days. The growth of Aspergillus versicolor and Penicillium polonicum in wax was lower than wax treated paper and Hanji. The growth of them in beeswax treated paper and Hanji both were excellent.

미생물비료 인산가용화균이 오이의 생산량에 미치는 영향

  • 난용성 인산염을 가용화 시키는 Penicillium sp. PS-113와 Lactobacillus sp. 를 이용하여온실에서 오이에 대한 생육 실험을 실시하였다. 실험 결과는 다음과 같다. Penicillium sp. PS-113 $5.0{\times}106cfu/ml$ + Lactobacillus sp. $6{\times}106cfu/ml $동시 처리구에서 평균 오이 길이는 대조구에 비하여 3cm 더 긴것으로 조사되었고, 오이 낱개의 무게는 310.6g으로 무처리구에 비하여 89.2g 이 증가하였으며, 오이 전체 생산량은 10681.7g로 무처리구와 비교해 볼 때 4517.3g이 많아 생산량이 73% 증가하였다. Penicillium sp. PS-113 $5.0{\times}105cfu/ml$ 단독처리와 Penicillium sp. PS-113 $5.0{\times}105cfu/ml$ + Lactobacillus sp. $6{\times}106cfu/ml$의 동시 처리구도 오이의 평균 무게와 길이에 있어 무처리구보다 약간씩 더 높았다. Penicillium sp. PS-113 $5.0{\times}105cfu/ml$ + Lactobacillus sp. $6{\times}106cfu/ml$의 동시처리구는 무처리에 비하여 오이의 수가 2개 적었으나 오이 전체 생산량은 무처리구에 비하여 Penicillium sp. PS-113 $5.0{\times}105cfu/ml$ 단독 처리구에서 53.7%, Penicillium sp. PS-113 $5.0{\times}104cfu/ml$ + Lactobacillus sp. $6{\times}106cfu/ml$ 동시 처리구에서 48.5% 증가되었다. 따라서 인산가용화균의 처리가 오이의 생산성을 증대시키는 것으로 나타났다.

Inhibitory effects of grapefruit seed extract(DF-100) on growth and toxin production of Penicillium islandicum (자몽종자 추출물(DF-100)이 Penicillium islandicum생육 및 독소 성분 skyrin생합성에 미치는 저해효과)

  • The anthraquionone mycotoxin, skyrin, is produced by Penicillium islandicum. DF-100 which was extracted from grapfruit seed extract and is a natural organic complex inhibited the biosynthesis of skyrin by Penicillium islandicum. This study was carried out to determine the potential of DF-100 to support Penicillium islandicum and skyrin production. DF-100 inhibited the growth of the fungus at 750ppm or less and caused complete inhibition of skyrin production at 500ppm or less. DF-100 appears to block the incorporation of emodinathrone into skyrin and an enzymatic step in the skyrin biosynthetic pathway which lies before skyrinanthrone.

Two Unrecoreded Species Belonging to Penicillium Section Exilicaulis in South Korea

  • Penicillium in section Exilicaulis is characterized by non-vesiculate monoverticillate and biverticillate stipes. Species in sect. Exilicaulis are commonly found in soil and plants in terrestrial environments; however, only a few species have been reported in Korea. To investigate the diversity of Penicillium sect. Exilicaulis, Penicillium species were isolated from terrestrial and marine environments. Based on sequence analyses of β-tubulin, calmodulin, and the second largest subunit of RNA polymerase II loci, 19 strains of Penicillium in sect. Exilicaulis were identified as P. citreonigrum, P. citreosulfuratum, P. corylophilum, P. menonorum, P. rubefaciens, P. velutinum, Penicillium sp. 1, and Penicillium sp. 2. Two of them, P. citreonigrum and P. citreosulfuratum, were confirmed to be new to Korea. Molecular phylogenies and detailed descriptions of the two unrecorded species are provided.

Penicillium from Rhizosphere Soil in Terrestrial and Coastal Environments in South Korea

  • Penicillium, the most common genus plays an important ecological role in various terrestrial and marine environments. However, only a few species have been reported from rhizosphere soil. As part of a project to excavate Korean indigenous fungi, we investigated rhizosphere soil of six plants in the forest (terrestrial habitat) and sand dunes (coastal habitat) and focused on discovering Penicillium species. A total of 64 strains were isolated and identified as 26 Penicillium species in nine sections based on morphological characteristics and the sequence analysis of β-tubulin and calmodulin. Although this is a small-scale study in a limited rhizosphere soil, eight unrecorded species and four potential new species have been identified. In addition, most Penicillium species from rhizosphere soil were unique to each plant. Penicillium halotolerans, P. scabrosum, P. samsonianum, P. jejuense, and P. janczewskii were commonly isolated from rhizosphere soil. Eight Penicillium species, P. aurantioviolaceum, P. bissettii, P. cairnsense, P. halotolerans, P. kananaskense, P. ortum, P. radiatolobatum, and P. verhagenii were recorded for the first time in Korea. Here, we provide the detailed morphological description of these unrecorded species.

A Study on the Effect of Lactobacillus spp. on the Growth and Citrinin production by Penicillium citrinum (Penicillium citrinum 생육과 citrinin 생성에 미치는 젖산균의 영향에 관한 연구)

  • ABSTRACT - This study was performed to investigate the possible effect of Lactobacillus spp. on the growth and citrinin production by Penicillium citrinum. Lactobacillus bulgaricus and Lactobacillus casei were grown with Pen. citrinum in modified APT broth containing 7% of glucose and incubated at $30^{\circ}C$ for 15 days. Four inoculation procedures were used; (a) Lactobacillus spp. and Pen. citrinum were grown alone(Pc, Lb, and Lc), (b) both organisms were added simultaneously(ST; Pc+Lb and Pc+Lc), (c) Lactobacillus spp. was grown 3 days, then conidia of Pen. citrinum were added(LbPc and LcPc), and (d) Pen. citrinum was grown 3 days, then Lactbacillus spp. was added (PcLb and PcLc). At 0, 3, 6, 9, 12, 15 days of incubation, the growth of each organism, pH and total acidity of broth, and content of citrinin were determined. Lactobacillus spp. and Pen. citrinum, when grown associatively, influenced the growth of each other. It was observed that slower growth of Pen. citrinum when in the presence of Lactobacillus spp. than when the mold grew alone. Production of citrinin by Pen. citrinum was markedly less in the mixed culture. No apparent growth and toxin production was observed when the Lactobacillus spp. was grown 3 days, then conidia of Pen. citrinum were added(LbPc and LcPc). The above results indicate that another microorganism or competing microflora in the culture can affect the behavior of Pen. citrinum.trinum.

Diversity and Plant Growth-Promotion of Endophytic Fungi Isolated from the Roots of Plants in Dokdo Islands (독도의 자생식물 뿌리에서 분리한 내생진균의 다양성과 생장촉진활성)

  • Endophytic fungi were isolated from the roots of plants growing naturally on the island of Dokdo. Plant samples, such as Miscanthus sinensis, Achyranthus japonica and Echinochloa crusgali were isolated from Dongdo, and those such as Honkenya peploides and Artemsia koidzumii were isolated from Seodo. Twenty one strains of endophytic fungi were isolated from these plants. To identify the strains, PCR (polymerase chain reaction) amplification of the partial ITS (Internal Transcribed Spacer) regions was done with universal primers ITS-1 and ITS-4 to determine the nucleotide sequence of the ITS regions. Of the strains isolated from Miscanthus sinensis, 75% were Penicillium sp. and 25% were Aspergillus sp. Fifty five percent of strains isolated from Achyranthus japonica were Penicillium sp., 30% were Aspergillus sp. and 15% were Zygorhynchus sp. Strains isolated from Echinochloa crusgali were Penicillium sp. (50%), Aspergillus sp. (12%), Giberella sp. (13%), Talaromyces sp. (9%) and Umbelopsis sp. (8%). Of the strains isolated from Honkenya peploides, 76% were Penicillium sp. and 24% were Pestalotiopsis sp. Strains isolated from Artemisia koidzumii were Penicillium sp. (81%) and Mucor sp. (19%). As a result of bioassay, Ec-3-1 strain isolated from Echinochloa crusgalli showed plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp. was the most abundantly distributed fungal strain in all plants used in this study.
