The aim of this study was to analyze the proteome expression of bovine satellite cells from longissimus dorsi (LD), deep pectoral (DP) and semitendinosus (ST) muscle depots during in vitro myogenic differentiation. Proteomic profiling by twodimensional gel electrophoresis and mass spectrometry of differentiating satellite cells revealed a total of 38 proteins that were differentially regulated among the three depots. Among differentially regulated proteins, metabolic proteins like lactate dehydrogenase (LDH), malate dehydrogenase (MDH) were found to be up regulated in ST, while alpha-enolase (NNE) in LD and DP depot satellite cells were down regulated. Also, our analysis found that there was a prominent up regulation of cytoskeletal proteins like actin, actincapping protein and transgelin along with chaperone proteins like heat shock protein beta 1 (HSPB 1) and T-complex protein 1 (TCP-1). Among other up regulated proteins, LIM domain containing protein, annexin 2 and Rho GDP-dissociation inhibitor 1 (Rho GDI) are observed, which were already proven to be involved in the myogeneis. More interestingly, satellite cells from ST depot were found to have a higher myotube formation rate than the cells from the other two depots. Taken together, our results demonstrated that, proteins involved in glucose metabolism, cytoskeletal modeling and protein folding plays a key role in the myogenic differentiation of bovine satellite cells.
In this paper, we propose a method to fabricate a patient-specific breast implant using MRI images and 3D scan data. Existing breast implants for breast reconstruction surgery are primarily fabricated products for shaping, and among the limited types of implants, products similar to the patient's breast have been used. In fact, the larger the difference between the shape of the breast and the implant, the more frequent the postoperative side effects and the lower the satisfaction. Previous researches on the fabrication of patient-specific breast implants have used limited information based on only MRI images or on only 3D scan data. In this paper, we propose an algorithm for the fabrication of patient-specific breast implants that combines MRI images with 3D scan data, considering anatomical suitability for external shape, volume, and pectoral muscle. Experimental results show that we can produce precise breast implants using the proposed algorithm.
Objectives: Various techniques of endoscopic thyroidectomy have been developed in thyroid resection since 1998 in the aspect of excellent cosmetic viewpoint. Of them, we evaluated our experiences and advantages of gasless axillary approach technique for resection of dominant thyroid nodules. Material and Methods: Twenty-nine cases of thyroid nodules were operated by the technique of gasless axillary approach during one year from December 2003 to December 2004. Twenty four patients underwent total lobectomy and five patients were partial lobectomy. Results: The operation time of first case took 300 minutes, however it became gradually shortened with case experiences down to 100-120 minutes. Pathologically, nodular hyperplasia was twenty cases, follicular adenoma five cases, papillary carcinoma three patients, and Hashimoto's thyroiditis one patient. There was no case of conversion to open thyroidectomy. Three cases of postoperative hoarseness were recovered spontaneously in 3 months. Hospital stay was four days for most patients. The cosmetic result was excellent without visible scar in anterior neck and chest. Conclusion: Endoscopic thyroidectomy via gasless axillary approach shows excellent result in cosmetic view point with hidden incision scar at axilla, and shorter hospitalization. However a question of longer operation time for dissection of the long plane over pectoral muscle is still remained.
To investigate the svnthyesis of muscle proteins during differentiation of chicken myoblast, cvtosolic and membrane fractions were used for both sodium dodecvl sulfate polvcrylamide gel eBectrophoresis and two-dimensional gel electrophoresis. An extensive cell fusion was observed in 4 day culture. In the protein pattern of the cvtosolic fraction from SDS-PAGE. several protein bands including 250 kDa and 46 kDa showed remarkable changes during culture. the protein of 46 kDa was the most prominent one ann its optical density was the highest in 5 day culture (OD = 1.30). In the membrane fraction, band of 19.8 kDa showed the highest absorbance with 0.93 OD at 12 hr after initial plating and decreased gradually thereafter to 0.23 in 5 nay culture. From the results of two-dimensional gel electrophoresis of cytosolic fraction, the 46 kDa spot was observed as ko separated forms from culture 2 nary culture, and the sixte of this spot was the largest in 5 nay culture. In the pattern of membrane protein, the extensive appearance of newiv synthesized Proteins was found in a naut culture, but no Prominent spot was observed throughout culture. From the results of the present clay, we found that, during myoblast differentiation, the most prominent proteins were bands of 46 kDa and 19.8 kDa in cvtosolic and membrane fraction, respectively, and the appearance of new proteins was initiated at 48 hr after initial plating, and the 46 kDa protein was predominant in the cytoplasm of late culture in which extensive cell fusion was observed.
Meat becomes brown and rancid during storage in the refrigerator and display in the case. Color changes, metmyoglobin formation and lipid oxidation are the important problems in the transportation / distribution of meat and retail display. The freshness of meat is determined by the sense of vision and smell. Since conventional method determining lipid oxidation is time consuming and destructive (it needs to homogenize meat with reagents, filtrate, time for reaction and read optical density using spectroscopy), more rapid and nondestructive technical tools are desired. The objective of this work was to evaluate near-infrared spectroscopy as an analytical tool for determining meat color, metmyoglobin formation and lipid oxidation. in beef, pork and chicken. Semitendinosus and longissimus thoracis muscles from six beef steers, biceps femoris and longissimus thoracis muscles from twelve LWD crossbred pigs, and superficial pectoral muscles from twenty-four broilers were used. About a 5-cm diameter and 1-cm thick sample (20.0g) was cut from the muscle and placed on plastic foam, over-wrapped with PVC film, and displayed under flourescent lights at 4 degrees C. during 10 days for beef and pork or 4 days for chicken. The spectra was measured by NIR systems Model 5500 Spectrophotometer using fiber optic scan at range of 400 - 1100 nm. Data were recorded at 2 nm intervals and 10 scans / 10 sec were averaged for every sample. Data obtained were saved as log 1/Re, where Re is the reflectance energy, and then mathematically transformed to second derivatives to reduce effects of differences in particle size. $L^{*}$, $a^{*}$ and $b^{*}$, and metmyoglobin formation were determined by conventional spectrophotometer using the integrating sphere unit. 2-Thiobarbituric acid reactive substances (TBARS) were measured for lipid oxidation. A multiple linear regression was used to find the equation which would best fit the data. The number of wavelengths used in the equation was selected based on the fewer number compared to the increasing multiple correlation and Decreasing standard error. (omitted)
Proteomics is a useful approach to know protein expression, post-translational modification and protein function. We investigated the protein expression pattern and identity in chickens fed with the tissue culture medium waste after harvest of Korean wild ginseng (TCM-KWG) (Panax ginseng). Two groups (n=60/group) of day old broiler chickens were administered with 0 (control) and 0.8% (treatment) TCM-KWG through drinking water. After 5 weeks, we examined the protein expression pattern of fibularis longus and superficial pectoral muscle by Two-dimensional electrophoresis analysis. Interestingly, TCM-KWG treatment significantly increased five spot's density, and markedly reduced five spot's density in the muscles. We identified 10 proteins (desmin, myosin light chain 1, heat shock 25 kDa protein, collapsin response mediator protein-2A, alpha enolase, vimentin, actin alpha 1, my023 protein, pyruvate kinase and troponin T) by the matrix-assisted laser desorption ionization time of flight (MALDI-TOF).
The disease occurred in cultured eel (Anguilla japonica) in a recirculatory culture system without any separated filtration apparatus. As the pond had a high level of nitrite with $60mg/{\ell}$, 1% NaCl was added to reduce nitrite toxicity to eel. The first outbreak was observed a week after the NaCl treatment and continued for 10 days. Accumulated mortality was about 0.2-0.5%. Affected fish ranged from 150-350 g were usually anorexic and exhibited yellow colour in the skin of the abdominal region and at the base of pectoral fins, as well as in the eyes. In a few individuals with severe symptoms, the lateral skin was also yellowish. The spleen, kidney, muscle and gall bladder were yellowish and the liver was pale-yellow colour but green on the posterior part. The gall bladder was shrunken without bile. Some abnormal erythrocytes such as "tear drop" cells (dacrocyte) were observed in peripheral blood smears stained with May-Grunwald Giemsa. Hematocrit values and hemoglobin contents in the jaundiced eel were significantly lower compared with apparently heathy eel. Severe haemosiderosis accompanied by erythrophagocytosis was found in the kidney and spleen. Haemosiderin deposits were observed in macrophages of the haematopoietic tissue of the kidney and in the splenocytes. But no significant alterations were found in the hepatic cells. In this study we report the first outbreak of jaundice in cultured eel in Korea. Pathological and hematological investigations suggested that severe hemolysis may resulted in jaundice in eel although the cause of hemolytic jaundice was not identified in this study.
Monochromatic green light-emitting diodes (LED) light stimuli influences the posthatch growth performance of chicks. This study was undertaken with the following objectives: i) to examine whether the green LED light stimuli induces an overheating effect by determining weight loss rate of fertile eggs during incubation period; ii) to look for the development of eyes and other primary organs at different ages of embryos and newly hatched chicks. Arbor Acres fertile broiler eggs (n = 480) were randomly assigned to 3 incubation groups and exposed to continuous white light, green light, or a dark environment (control) from the first day to 19 d of incubation. The light sourced from LED lamps with the intensity of 30 lx at eggshell level. The results showed that either green or white light stimuli during incubation did not significantly affect the weight loss rate of fertile eggs, hatching time, hatchability, chick embryo, or body weight (BW), the weight percentage of heart, liver, and eyes, as well as obvious systematic abnormalities in eye weight, side-to-side, back-to-front, or corneal diameter from 15 d of embryogenesis to 6 d of posthatch (p>0.05). Compared with the dark condition, green light stimuli during incubation tended to increase feed intake (p = 0.080), improved the BW gain of chicks during 0 to 6 day posthatch (p<0.05), and increased the percentage of pectoral muscle to the BW on 3- and 6-day-old chicks. In addition, embryos or chicks in green light had lower weight percentage of yolk retention on 19 d of embryogenesis and 1 d of posthatch in comparison to those in dark or white group (p<0.05). These results suggest that providing 30 lx green LED light stimuli during incubation has no detrimental effect on the development of eyes, heart and liver of embryos and hatchlings, but does have potential benefits in terms of enhancement of the chick growth during the early posthatch stages. In addition, the fertile broiler eggs stimulated with 30 lx green LED light during incubation does not cause an overheating effect.
격렬한 운동유형으로 분류되는 비행이 습성화 되어 있는 조류를 대상으로, 인위적으로 비행을 제한시켰을 떼 나타나는 심근의 퇴행성 변화를 살펴봄으로써, 운동량 감소가 심근에 미치는 영향에 대하여 검토하고자 하였다. 서식환경의 제한을 위해 자연상태에서 서식하는 비둘기를 1 $m^3$ 공간에 2달 동안 서식시킨 군 (confined type)과 자연상태의 군 (wild type)으로 나누어 심근의 변화양상을 미세구조적, 세포화학적 방법 및 입체해석학적 방법으로 분석하였다. 체중은 confined type에서 증가되어 상대적으로 심장의 무게는 감소되었고, 흉근의 무게는 6.5% 감소 (p<0.05) 되었다. Confined type은 wild type과 비교해 볼 때, 미세구조적으로 더 적은 빈도의 심근의 파동형 변화와 지방소적의 침적이 관찰되었고, 근절의 길이는 길게 관찰되었다. Cytochrome oxidase의 활성을 알아보기 위해 실시한 세포 화학적 반응 결과, confined type에서 활성이 15% 감소되었다. 또한 입체해석학적 분석 결과, confined type의 관원섬유 체적밀도는 높게 나타났으나, 근형질내세망의 체적밀도와 사립체 내막의 표면밀도는 각각 23%, 25% 낮게 (p<0.05, p<0.01) 나타났다. 사립체의 체적밀도는 confined type에서 낮게 나타난 반면, 수적 밀도는 wild type과 비교하여 17% 높게 (p<0.05) 나타났다. 이러한 결과를 종합해보면 비행이라는 고에너지를 요구하는 격렬한 운동인 비행이 습성화 되어 있는 비둘기에서 비행을 제한시키면, 심근은 운동량의 감소에 따른 퇴행성 변화를 겪게 되므로, 운동의 지속성 여부는 심근기능유지에 매우 중요한 요소로 판단된다.
脊椎動物 網膜과 腦에 있어서 LDH isozyme pattern을 系統的으로 밝히기 위하여 cellulose acetate 電氣泳動法을 이용하여 실험한 결과 다음과 같은 結論을 얻었다. 1. 붕어와 잉어의 網膜과 腦의 LDH isozyme은 $LDH_1$과 $LDH_2$에 해당되는 부위에 彌滿된 단일 band가 觀察되었다. 잉어의 心臟, 胞筋, 肝臟, 胃의 LDH isozyme도 모두 다같이 퍼진 단일 band로 나타났다. 2. 도룡농과 개구리의 腦에 있어서 LDH isozyme pattern은 다르게 觀察되었다. 도룡뇽에서는 음극으로 移動된 단일 band가, 개구리에서는 $LDH_5$와 $LDH_4$ 두 개의 band가 검출되었다. 3. 유혈목이의 網膜과 腦에서는 다같이 하나의 band로 觀察되나 자라에서는 網膜과 腦의 isozyme이 다르다. 卽 자라의 網膜에서는 5個의 isozyme band가 觀察되었는데 그 농도는 $LDH_5, LDH_4, LDH_3, LDH_2$ 및 $LDH_1$의 순으로 낮아 졌다. 4. 닭과 사랑새의 網膜과 腦에 있어 LDH isozyme은 다같이 단일 band로 나타났다. 5. 집토끼, 개. 돼지, 소의 網膜에서도 포유류의 特徵的인 5個의 isozyme band가 검출되었고 mouse, albino rat, 관박쥐의 腦에서도 5個의 band가 검출되었다. 이러한 사실들로 미루어 보아 網膜과 腦의 LDH는 그 電氣泳動 이 대체로 비슷하여 두 器官은 유사한 pyruvate 代謝가 이루어지고 있는 것으로 추측된다.
