A Study on the Development of A Juice-clarifying Enzyme Preparation (과실쥬우스용 효소제의 개발연구)

  • In order to develop an enzyme preparation for clarification of fruit juices, a microbial strain having a strong pectolytic activity was selected and a crude enzyme preparation from this strain was examined for the effects in the preparation of grape juice and wine. The results are summarized as follows: 1) A strain of Aspergillus niger was selected as having the highest productivity of pectolytic enzymes among many species of Aspergillus and Rhizopus. 2) A pectolytic enzyme preparation was purified from this selected strain and the effects of pH and temperature on its enzyme activity and stability were investigated. 3) The use of the enzyme preparation brought about the increase in the free run yield and clarity of grape juice. 4) Whereas the use of the enzyme preparation did not exhibit any effect in the brewing of red wine, its use showed a good effect on the rates of filtration and clarity in the case of white wine.

  • Isolates of Penicillium expansum with reduced pathogenicity were arbitrarily selected among benomyl-resistant isolates in order to investigate relationship of their pectolytic enzyme acitivity with pathogenicity. In artificial medium, strongly pathogenic isolate $S_1$ and weakly pathogenic isolate $R_2$ produced considerable amonts of endo-polymethylgalacturonase, endo-polygalacturonase, pectin methyl-trans-eliminase, and polygalacturonate-trans-eliminase. No marked difference in enzyme activities was observed between two isolates. In apple medium, the activities of endo-polymethylgalacturonase and endo-polygalacturonase of isolate $S_1$ were over 6 times higher than those of isolate $R_2$. But pectin methyl-trans-eliminase and polygalacturonate-trans-­eliminase did not show a great difference. Activities of endo-polymethylgalacturonase and endo­polygalacturonase precipitated at 80-95% saturation of ammonium sulfate were highest, and addition of these enzyme solutions increased pathogenicity of weakly pathogenic isolates $R_{1-4}$.

  • Botryosphaeria dothidea causing apple fruit rot was cultured in pectin-polypectate mine­raI salts or apple mediumss, to investigate pectolytic enzyme production and activity. Exo-polygalactu­ronase(PG) and exo-polymethylgalacturonase (PMG) in apple medium showed maximum of activity to 6.4 and 7.2 units at six days of culture, respectively. Their maximum activity in pectin-polypectate mineral salts medium was 5.9 and 5.3 units at eight days of culture lower than in apple medium respectively. Endo-PG and endo-PMG in pecin-polypectate mineral salts medium were maximum of activity to 4.4 and 16.2 units at six and eight days of culture, respectively, but activities in apple medium were 3.2 and 6.7 units at eight days of culture. Activity of polygalacturonate-trans-­eliminase(PGTE) and pectinmethyl-trans-eliminase(PMTE) was higher in pectin-polypectate mineral salts medium than in apple medium. Fungal growth was maximum at six and eight days of culture in pectin-polypectate mineral salts and apple medium, respectively.

Juice Clarification with the Use of Polygalacturonase Produced by Ganoderma lucidum (Ganoderma lucidum이 생산하는 Polygalacturonase를 이용한 과즙청징)

  • Ganoderma lucidum produced the potent pectolytic enzymes for clarifying cloudy fruit juice. Among the purified polygalacturonases (endo- and exo-polygalacturonase), endo-polygalacturonase had a good effect on juice clarification. The optimum temperature and concentration of endo-polygalacturonase for the juice clarification were $40^{\circ}C$ and 4 unit/5 ml juice, respectively. The apple juice was almost completely clarified at $40^{\circ}C$ for 60 min. It was suggested that culture filtrate of Ganoderma lucidum or it's ammonium sulfate fraction should be used as a good source of pectolytic enzyme for juice clarification.

Optimization of Demucilage Process of Opuntia ficus-indica var. saboten Fruit using High Hydrostatic Pressure Enzyme Dissolution (고압 효소 액화 장치를 이용한 백년초 점질물 분해 공정의 최적화)

  • This study aimed to develop and optimize a demucilaging process of Opuntia ficus-indica var. saboten (OFI) fruit to increase its usability as functional food ingredient and food additive. Viscozyme and Novozym 33095 as multienzyme complex having a broad spectrum of carbohydrases and pectolytic enzymes, respectively, were used in enzymatic dissolution along with high hydrostatic pressure liquefaction. To optimize the liquefaction process using high hydrostatic pressure liquefying extractor, response surface methodology with 3-factor central composite design was employed with reaction factors such as temperatures (25, 32, 40, 48, and $55^{\circ}C$), pressures (20, 40, 60, 80, and 100 MPa), and times (15, 30, 45, 60, and 75 min). At optimum conditions ($25^{\circ}C$, 100 MPa, and 58.275 min) for high hydrostatic pressure liquefaction process, the processed OFI fruit juice was predicted to have viscosity at 2.917 poise, partly due to the release of free sugars such as fructose and glucose detected using HPLC-ELSA system. The results above suggests that the OFI fruit juice with decreased viscosity may be used for various manufacturing processes of food, beverage, ice cream, and cosmetics.

Distribution of Pectobacterium Species Isolated in South Korea and Comparison of Temperature Effects on Pathogenicity

  • Pectobacterium, which causes soft rot disease, is divided into 18 species based on the current classification. A total of 225 Pectobacterium strains were isolated from 10 main cultivation regions of potato (Solanum tuberosum), napa cabbage (Brassica rapa subsp. pekinensis), and radish (Raphanus sativus) in South Korea; 202 isolates (90%) were from potato, 18 from napa cabbage, and five from radish. Strains were identified using the Biolog test and phylogenetic analysis. The pathogenicity and swimming motility were tested at four different temperatures. Pectolytic activity and plant cell-wall degrading enzyme (PCWDE) activity were evaluated for six species (P. carotovorum subsp. carotovorum, Pcc; P. odoriferum, Pod; P. brasiliense, Pbr; P. versatile, Pve; P. polaris, Ppo; P. parmentieri, Ppa). Pod, Pcc, Pbr, and Pve were the most prevalent species. Although P. atrosepticum is a widespread pathogen in other countries, it was not found here. This is the first report of Ppo, Ppa, and Pve in South Korea. Pectobacterium species showed stronger activity at 28℃ and 32℃ than at 24℃, and showed weak activity at 37℃. Pectolytic activity decreased with increasing temperature. Activity of pectate lyase was not significantly affected by temperature. Activity of protease, cellulase, and polygalacturonase decreased with increasing temperature. The inability of isolated Pectobacterium to soften host tissues at 37℃ may be a consequence of decreased motility and PCWDE activity. These data suggest that future increases in temperature as a result of climate change may affect the population dynamics of Pectobacterium.

Production of Pectolytic Enzymes and Change of Pectic Substances from Apple Fruits Infected with Botryosphaeria dothidea (사과 겹무늬썩음병균(Botryosphaeria dothidea)에 의해 부패된 사과 과실에서 Pectin질 분해효소의 생산과 Pectin질의 변화)

  • Pectolytic enzymes were extracted in apple fruits rotted by Botryosphaeria dothidea, and their activities and change of pectic substances were investigated. Exo-polygalacturonase(exo-PG), exo-polymethylgalacturonase(exe-PMG), polygalacturonate-trans-eliminase(PGTE) and pectin-methyl-trans-eliminase(PMTE) were produced by the pathogen. Activities of exo-PG and exe-PMG extracted from rotten apple fruits were high to 21.15 and 24.65 units/mg protein in specific activity at seven days after inoculation, respectively. Activities of PGTE and PMTE showed 5.60 and 7.90 units/mg protein, respectively, but they were lower than those of the exo-type enzymes. Water-soluble and versene-soluble pectins were 11.50 mg/100 mg-AIS and 7.31 mg/100 mg-AIS at 14 days after inoculation, namely, they were increased by 4.23 and 2.16 mg/100 mg-AIS over those of sound apples, respectively. Total soluble pectic substances of rotten apple were 72.4% of total pectic substances and it was higher by 24.8% than sound apple. Insoluble pectic substance was notably decreased from 15.32 to 7.16 mg/100 mg-AIS according to progress of decay while total pectic substances were not changed remarkably.

  • Isoates with changed pathogenicity were selected among iprodione-resistant Alternaria mali to investigate any relationship between their pectolytie enzyme activity and pathogenicity. In artificial medium, strongly pathogenic isolates $S_1$, and $R_3$ showed higher enzyme activity than weakly pathogenic isolate $R_8$.Activity of endo-polymethylgalacturonase and end-o-polygalacturonase was more than 3 times. But activity of pectin methylesterase and pectin Iyase by isolaste $S_1$ was higher than those by $R_3$ and $R_8$ isolates. In apple medium dialyzed against distilled water, activity of enzyme by each isolate was increased but growth of each isolate was reduced. When iprodione was added to the medium, enzyme activity and growth of isolate $S_1$, were reduced but strongly pathogenic isolate .$R_3$ among iprodione-resistant ones showed increased enzyme activity except for exo-polygalacturonase in dialyzed apple medium.

Clarification of the Juice Extracted from Stored Apples by pH Adjustment (저장 사과로부터 착즙한 쥬스의 pH 조절에 의한 청징)

  • Clarification of the juice extracted from stored Fuji apples was studied with pectolytic enzyme and pH control, separately and in combinations. In the separated treatment, the clarity of the juice was increased with the treated enzyme amount. The juice adjusted pH to 3.5 with malic acid had the highest clarity in pH range from 3.5 to 5.0, but this juice was not acceptable because of high acidity. In combination of the two treatment, the clarity of the juice treated with the enzyme at pH 4.0 was higher than that of the juice without the enzyme, and the juice with ligh trasmittance of about 92% could be obtained at the pH by addition with one-third amount of enzyme which was used for clarification of the juice extracted from the fresh apples at harvesting season.

Studies on the Pectolytic Enzymes from Byssochlamys fulva II (Byssochlamys fulca가 생성하는 펙틴질 분해효소에 관한 연구 II)

  • Polygalacturonase of Byssochlamys fulva was purified and characterized. Specific activity increased from 2.21 units/mg protein to 10.47 units/mg protein through $(NH_4)_2SO_4$ treatment, SephadexG-100 gel filtration, and DEAE-Sephadex ion exchange chromatography. Divalent cations, such as $Ca^{++}\;and\;Cu^{++}$, increased polygalacturonase activity greatly. Added as $10^{-3}M$ concentration, $Ca^{++}$ ion enhanced enzyme activity 9.8folds. Optimum temperature was $50^{\circ}C$ and optimum pH was 5.0. Activation energy of reaction was 8.69 Kcal/mole. Michaelis-Menten $constant(K_M)\;and\;V_{max}$ of reaction were $6.27{\times}10^{-3}mole/l\;and\;2.85{$\mu}moles/min$. Polygalacturonase of Byssochlamys fulva preferred polygalacturonic acid to pectin as substrate and was presumed as endo-type on the basis of the relationship between viscosity reduction and substrate degradation. Molecular weight of polygalacturonase was estimated as 55,000.
