An acidic polygalacturonase (PG) from Aspergillus kawachii was produced by solid state fermentation employing a polyurethane foam support. The conditions used for the production of acidic PG were particle size of support (0.6 or 500 $mm^3$) and fermentation time. From the factors studied, the particle size had important influence on enzyme production. The best conditions for acidic PG production were $0.6\;mm^3$ particle size, 18 hr at $30^{\circ}C$ and initial pH of 5.0. In addition, pectin was extracted from citrus pomaces (grapefruit, lime, and tangerine) by acidic PG at $50^{\circ}C$ for 24 hr with citric acid solution. Infrared spectroscopy showed that lime pomace had more high-methoxylated (65%) endogenous pectin than was obtained than from grapefruit or tangerine pomaces. The enzymatically extracted pectin yield in dry basis (d.b.) for grapefruit and lime pectins were 6.95 and 4.25%, respectively. The citric acid solution alone also contributed to pectin extraction from citrus pomaces (7-9%, d.b.). Limited pectin extraction by acidic PG from tangerine pomace was most likely due to the presence of low-methoxylated endogenous pectin. The enzymatic method for pectin extraction using acidic PG from A. kawachii is a promising technique for releasing highly polymerized pectic substances from high-methoxylated lime or grapefruit pomaces.
To determine the functions of specific cell wall polysaccharides, polysaccharides of three mutants, mur3-1, mur3-2, and mur3-3, obtained from Arabidopsis wild type, underwent structural characterization. Upon sequential separation of pectins (RG-I and RG-II) and cross-linking glycans (xyloglucan, XG), only XG was affected by the mud mutation. Wild-type XG contained a considerable amount of fucose, whereas the fucose level in mur3 XGs was less than 20% that of wild type. Further analysis of XGs by matrix-assisted laser-induced/ionization time-of-flight (MALDI-TOF) mass spectrometry indicated that mud lines considerably or completely lost the fucosylated XG oligosaccharides such as XXFG and XLFG and the double-galactosylated oligosaccharide XLLG $^1H$-NMR spectroscopic analyses of the XG oligosaccharides from mur3-3 plant revealed the absence of fucose and a galactose level in the galactosylated side chain that was reduced by 40% compared to that of Arabidopsis wild-type plant. In contrast, 85% less fucose and a slight loss of galactose were observed in the mur3-1 and mur3-2 lines which show normal growth habit. Of the three Arabidopsis mur3 lines studied here, mur3-3 is disrupted by a T-DNA insertion in the exon of MUR3 which encodes XG-specific galactosyltransferase, and exhibits slight dwarfism. These results indicated that the T-DNA insertion at the MUR3 locus did not induce the complete loss of galactose in XG, and that galactose, rather than fucose, in the XG side chains made a major contribution to overall wall strength.
Fusarium root rot caused by the soil-borne fungus Fusarium solani is one of the most important fungal diseases of cassava in Thailand, resulting in high yield losses of more than 80%. This study aimed to investigate if the exogenous application of salicylic acid formulations (Zacha) can induce resistance in cassava against Fusarium root rot and observe the biochemical changes in induced cassava leaf tissues through synchrotron radiation based on Fourier-transform infrared (SR-FTIR) microspectroscopy. We demonstrated that the application of Zacha11 prototype formulations could induce resistance against Fusarium root rot in cassava. The in vitro experimental results showed that Zacha11 prototype formulations inhibited the growth of F. solani at approximately 34.83%. Furthermore, a significant reduction in the disease severity of Fusarium root rot disease at 60 days after challenge inoculation was observed in cassava plants treated with Zacha11 at a concentration of 500 ppm (9.0%). Population densities of F. solani were determined at 7 days after inoculation. Treatment of the Zacha11 at a concentration of 500 ppm resulted in reduced populations compared with the distilled water control and differences among treatment means at each assay date. Moreover, the SR-FTIR spectral changes of Zacha11-treated epidermal tissues of leaves had higher integral areas of lipids, lignins, and pectins (1,770-1,700/cm), amide I (1,700-1,600/cm), amide II (1,600-1,500/cm), hemicellulose, lignin (1,300-1,200/cm), and cellulose (1,155/cm). Therefore, alteration in defensive carbohydrates, lipids, and proteins contributed to generate barriers against Fusarium invasion in cassava roots, leading to lower the root rot disease severity.
밀감 가공부산물에서 추출한 펙틴의 특성을 조사한 결과 알콜불용성 고형물(ASS)의 함량은 albedo층이 18.1%로 가장 높고, pulp가 5.7%로 가장 낮았다. AIS 구성 다당류 중 펙틴은 pulp와 albedo층이 각각 40.5% 및 35.2%로 구성되어 있어 펙틴 추출용으로서 좋은 소재인 것으로 나타났다. 총 펙틴 함량은 부위에 따른 큰 차이를 보이지 않아 대체로 AIS에 대해 30% 내외 이었으나, 가용성 펙틴 중 염산 가용성 펙틴의 경우에는 flavedo층이 14.0%로 가장 높고, pulp가 4.4%로 가장 낮았으며, 염류 가용성 펙틴의 경우에는 pulp부위가 15%임 에 비해 과피는 7% 정도로서 부위에 따른 차이가 나타났다. 0.05N-HCI을 사용하여 $85^{\circ}C의$ 열탕에서 펙틴을 추출한 결과 60분의 가열처리로 90%이상의 펙틴이 용출되었으며, microwave의 경우 5분간 2회 조사시킴에 의해 90% 정도 추출되었으나 부위별에 따른 차이는 크지 않았고, 이들 펙틴의 에스테르화도에 있어서는 microwave 처리구의 것이 다소 높은 값을 나타내었다. 추출 펙틴의 중성 당 조성을 조사한 결과 rhamnose, arabinose, galactose, glucose 및 xylose로 구성되고 있고, 이중 galactose가 60~78% 정도를 차지한 반면 xylose는 5% 이하이었다. 펙틴의 분자량 분포를 Sepharose CL-4B를 이용, gel permeation chromatography로 조사한 결과 부위별에 따른 큰 차이는 보이지 않았으나 pulp의 경우 분리 피크가 2개로 나타나 pulp를 구성하는 펙틴은 분자량이 높은 것과 이보다 낮은 두 group으로 구성되어져 있음을 알 수 있었다.
배 생산자원의 충분한 이용과 부가가치 증대를 위한 개발을 목적으로 생장시기에 따른 부위별 페놀성물질과 과육의 펙틴 변화를 측정하였다. 배의 생장시기에 따라 과피, 과육, 과심부의 총페놀성물질의 함량은 감소하는 경향을 보여 생장초기 과피에서는 20.61-22.98mg/g, 과육에서는 0.87-1.23mg/g, 과심부에서는 6.39-37.96mg/g으로 함량이 가장 높았다. 동일한 품종내에서 대부분 시기에 걸쳐 과피중의 총페놀성물질의 함량이 가장 높았고 과육의 함량이 가장 낮았다. 생장초기 3품종과 각 부위에서 모두 arbutin, chlorogenic acid, epicatechin이 검출되었고 과실이 성숙됨에 따라 과피와 과심부에서는 caffeic acid, catechin 그리고 4-hydroxymethyl benzoic acid(4-HMBA)도 검출되었으며 그 중 arbutin이 가장 많은 양을 차지하였다. 배 과육의 총펙틴 함량은 알코올 불용성 고형분(AIS)의 감소와 함께 생장초기 약 6mg/g에서 급격히 감소하다가 그 이후로 서서히 감소하였다. 생장초기 hydrochloric acid-soluble pectin(HSP)의 함량이 3.21-3.45mg/g으로 가장 많았고 또한 과실의 생장에 따라 감소하였으며 AIS water-solubel pectin(WSP)의 증가가 가장 컸다. 따라서 배의 성숙된 과실에 비하여 미숙한 과실이, 과육에 비하여 과피가 기능성식품소재 개발에 더 유용할 것으로 생각되었다. 현재 미이용되고 있는 미숙과나 낙과 등의 과피를 이용하여 알부틴과 같은 고부가가치 물질을 추출하여 기능성 물질로의 이용을 위해 앞으로 추출 방법 등의 연구를 수행할 예정이다.
청 매실을 수확한 후 LDPE 필름 포장재에 넣어 상온에서 일정 기간 보관할 때 포장재의 두께가 청매실의 조직 연화에 미치는 영향을 조사하기 위하여, 20, 30, 40$\mu\textrm{m}$두께의 포장재에 청매실 500 g씩을 각각 넣고 8일간 보존하면서 경도의 변화 및 무기성분과 펙틴질 성분의 함량 변화를 정기적으로 측정하여 무포장구의 경우와 비교하였고, 세포벽의 미세구조의 변화를 주사전자현미경으로 관찰하였다. 저장 중 매실의 연화와 관련된 경도와 펙 틴질의 감소 및 가용성펙틴 중 Ca 함량의 감소는 서로 밀접한 관계가 있음을 알 수 있었고, LDPE 30 포장재에 저장된 시료가 8일 동안 무포장구나 다른 포장재에 저장된 시료에 비 해 각종 펙틴질 성분 및 무기 성분의 변화와 그에 따르는 경도 변화가 가장 적었고 가용성 펙틴의 분자량 분포도 초기 상태와 가장 유사하였으며 주사전자현미경에 의하여 관찰된 외 형적인 상태도 LDPE 30 포장재에 저장된 시료가 저장전의 상태를 가장 잘 유지함을 확인할 수 있었다.
추출한 감과실의 세포벽에 polygalacturonase, ${\beta}-galactosidase$ 및 이들의 혼합한 효소액을 in vitro에서 처리하여 세포벽 구성 비섬유성 중성당의 변화를 연구, 검토하였다. 세포벽 구성 비섬유성 중성당의 변화는 효소 처리구에서 무처리 보다 많았으며, polygalacturonase 처리구에서는 rhamnose, xylose, galactose 등이 감소하였고, 혼합효소 처리구에서는 arabinose, galactose, rhamnose, xylose 등이 감소하였으며, ${\beta}-galactosidase$ 처리구에서 arabinose, galactose 등이 감소되었다. Pectin의 비섬유성 중성당은 모든 효소처리구에서 rhamnose, galactose, arabinose가 현저히 감소하였으며, 특히 polygalacturonase 처리시에 보다 현저하게 감소하였다. Hemicellulose I의 경우 polygalacturonase 처리구는 rhamnose, arbinose, xylose의 함량이 무처리구에 비해 높았으며, ${\beta}-galactosidase$ 처리구에서는 rhamnose와 xylose의 함량은 높았고, arbinose, mannose, galactose는 감소하였다. 혼합효소 처리구에서는 xylose, mannose, galactose가 높은 반면 arbinose와 galactose는 낮았다. 한편 hemicellulose II에서 비섬유성 중성당의 변화는 polygalacturonase 처리구에서 xylose와 glucose가 감소하였으나 다른 처리구에서는 뚜렷한 변화가 없었다. 이상의 결과를 종합하여 볼 때 polygalacturonase는 pectin을 분해함으로써 arbinose, galactose, rhamnose를 유리하고, ${\beta}-galactosidase$는 galactan과 arabinogalactan을 분해하여 galactose와 arabinose를 유리시키는 것으로 사료된다.
사과 겹무늬썩음병균 Botryosphaeria dothidea에 의해 부패된 사과과실에서 pectin질 분해효소를 추출하여 그들의 활성과 pectin 성분의 변화를 조사하였다. 본 병원균은 exo-polygalacturonase(exo-PG), exo-polymethylgalacturonase(exo-PMG), polygalacturonate-trans -e1iminase(PGTE)와 pectinmethyl-trans-eliminase(PMTE)를 생산하였다. 부패된 사과 과육에서 exo-PG와 exo-PMG는 접종 후 7일째 specific activity가 각각 21.15 및 24.65 units/mg protein으로 높게 나타났다. PGTE와 PMTE의 활성은 7일째 각각 5.60과 7.90 uints/mg protein으로 나타났으나 exo-type의 효소보다는 그 활성이 낮았다. 수용성 pectin은 부패가 진행됨에 따라 14일째에 11.50 mg/100 mg-AIS이었고, versene-soluble pectin 역시 7.31 mg/100 mg-AIS로 나타나 건전과와 비교하여 각각 4.23 및 2.16 mg/100 mg-AIS 증가하였다. 부패과의 총 가용성 펙틴 함량은 총 pectin의 72.4%로서 건전과와 비교하여 24.8% 더 높았다. 불용성 pectin 함량은 부패가 진행됨에 따라 15.32 mg/100 mg-AIS에서 7.16 mg/100 mg-AIS로 현저하게 감소하였으며, 총 pectin 함량은 큰 변화가 없었다.
Acidic endo-polygalacturonases are the major part of pectinase preparations and extensively applied in the clarification of fruits juice, vegetables extracts, and wines. However, most of the reported fungal endo-polygalacturonases are active and stable under narrow pH range and low temperatures. In this study, an acidic endo-polygalacturonase (EPG4) was purified and characterized from a mutant strain of Penicillium oxalicum. The N-terminal amino acid sequence of EPG4 (ATTCTFSGSNGAASASKSQT) was different from those of reported endo-polygalacturonases. EPG4 displayed optimal pH and temperature at 5.0 and 60-70℃ towards polygalacturonic acid (PGA), respectively, and was notably stable at pH 2.2-7.0. When tested against pectins, EPG4 showed enzyme activity over a broad acidic pH range (>15.0% activity at pH 2.2-6.0 towards citrus pectin; and >26.6% activity at pH 2.2-7.0 towards apple pectin). The Km and Vmax values were determined as 1.27 mg/ml and 5,504.6 U/mg, respectively. The enzyme hydrolyzed PGA in endo-manner, releasing oligo-galacturonates from PGA, as determined by TLC. Addition of EPG4 (3.6 U/ml) significantly reduced the viscosity (by 42.4%) and increased the light transmittance (by 29.5%) of the papaya pulp, and increased the recovery (by 24.4%) of the papaya extraction. All of these properties make the enzyme a potential application in the beverage industry.
박물관 및 도서관 등에 소장되어 있는 지류 유물은 균류, 곤충, 설치류 등과 같은 다양한 생물학적 열화요인들에 의해 비가역적인 손상을 입을 수 있다. 특히 습도가 높은 조건에서는 균에 의한 열화가 발생되기 쉬우며, 균류는 종이의 주성분인 셀룰로오스 및 사이징제, 전분, 아교 등의 탄수화물과 단백질 성분을 영양원으로 하므로 종이의 노화를 야기하게 된다. 현재까지 박물관과 도서관 등에서 지류 유물에 생물학적 열화를 발생시킬 수 있는 미생물종으로 약 300여 종이 동정되었으며, 그 중에서도 Aspergilli (30%)와 Penicilli (30%)가 셀룰로오스의 주요 열화 균으로 알려져 있다. 균류에 의한 생물학적 손상을 방지하기 위한 가장 좋은 방법으로서 습도, 온도, 산소 등 미생물의 생육 조건을 조절, 억제 또는 차단하는 방법을 들 수 있다. 따라서 본 연구에서는 산소흡수제를 이용하여 미생물의 생육조건 중 산소를 조절하여 Aspergillus versicolor 과 Penicillium polonicum 균에 의한 생물열화 방지 효과를 분석하였다. 연구 결과 산소흡수제 처리가 2종의 균에 의한 생물학적 열화 억제 효과가 있음을 확인하였다.
