An acidic polygalacturonase (PG) from Aspergillus kawachii was produced by solid state fermentation employing a polyurethane foam support. The conditions used for the production of acidic PG were particle size of support (0.6 or 500 $mm^3$) and fermentation time. From the factors studied, the particle size had important influence on enzyme production. The best conditions for acidic PG production were $0.6\;mm^3$ particle size, 18 hr at $30^{\circ}C$ and initial pH of 5.0. In addition, pectin was extracted from citrus pomaces (grapefruit, lime, and tangerine) by acidic PG at $50^{\circ}C$ for 24 hr with citric acid solution. Infrared spectroscopy showed that lime pomace had more high-methoxylated (65%) endogenous pectin than was obtained than from grapefruit or tangerine pomaces. The enzymatically extracted pectin yield in dry basis (d.b.) for grapefruit and lime pectins were 6.95 and 4.25%, respectively. The citric acid solution alone also contributed to pectin extraction from citrus pomaces (7-9%, d.b.). Limited pectin extraction by acidic PG from tangerine pomace was most likely due to the presence of low-methoxylated endogenous pectin. The enzymatic method for pectin extraction using acidic PG from A. kawachii is a promising technique for releasing highly polymerized pectic substances from high-methoxylated lime or grapefruit pomaces.
To determine the functions of specific cell wall polysaccharides, polysaccharides of three mutants, mur3-1, mur3-2, and mur3-3, obtained from Arabidopsis wild type, underwent structural characterization. Upon sequential separation of pectins (RG-I and RG-II) and cross-linking glycans (xyloglucan, XG), only XG was affected by the mud mutation. Wild-type XG contained a considerable amount of fucose, whereas the fucose level in mur3 XGs was less than 20% that of wild type. Further analysis of XGs by matrix-assisted laser-induced/ionization time-of-flight (MALDI-TOF) mass spectrometry indicated that mud lines considerably or completely lost the fucosylated XG oligosaccharides such as XXFG and XLFG and the double-galactosylated oligosaccharide XLLG $^1H$-NMR spectroscopic analyses of the XG oligosaccharides from mur3-3 plant revealed the absence of fucose and a galactose level in the galactosylated side chain that was reduced by 40% compared to that of Arabidopsis wild-type plant. In contrast, 85% less fucose and a slight loss of galactose were observed in the mur3-1 and mur3-2 lines which show normal growth habit. Of the three Arabidopsis mur3 lines studied here, mur3-3 is disrupted by a T-DNA insertion in the exon of MUR3 which encodes XG-specific galactosyltransferase, and exhibits slight dwarfism. These results indicated that the T-DNA insertion at the MUR3 locus did not induce the complete loss of galactose in XG, and that galactose, rather than fucose, in the XG side chains made a major contribution to overall wall strength.
Fusarium root rot caused by the soil-borne fungus Fusarium solani is one of the most important fungal diseases of cassava in Thailand, resulting in high yield losses of more than 80%. This study aimed to investigate if the exogenous application of salicylic acid formulations (Zacha) can induce resistance in cassava against Fusarium root rot and observe the biochemical changes in induced cassava leaf tissues through synchrotron radiation based on Fourier-transform infrared (SR-FTIR) microspectroscopy. We demonstrated that the application of Zacha11 prototype formulations could induce resistance against Fusarium root rot in cassava. The in vitro experimental results showed that Zacha11 prototype formulations inhibited the growth of F. solani at approximately 34.83%. Furthermore, a significant reduction in the disease severity of Fusarium root rot disease at 60 days after challenge inoculation was observed in cassava plants treated with Zacha11 at a concentration of 500 ppm (9.0%). Population densities of F. solani were determined at 7 days after inoculation. Treatment of the Zacha11 at a concentration of 500 ppm resulted in reduced populations compared with the distilled water control and differences among treatment means at each assay date. Moreover, the SR-FTIR spectral changes of Zacha11-treated epidermal tissues of leaves had higher integral areas of lipids, lignins, and pectins (1,770-1,700/cm), amide I (1,700-1,600/cm), amide II (1,600-1,500/cm), hemicellulose, lignin (1,300-1,200/cm), and cellulose (1,155/cm). Therefore, alteration in defensive carbohydrates, lipids, and proteins contributed to generate barriers against Fusarium invasion in cassava roots, leading to lower the root rot disease severity.

The amount and characteristics of pectin in the albedo and flavedo layers of the citrus peels, and those of the pulp were investigated. Alcohol insoluble solid(AIS) content was the highest in albedo layer(18.1%), and the lowest in pulp(5.7%). The pulp and the albedo layer showed a potential pectin sources as containing pectins of 40.5% and 35.2% of the total polysaccharides of the pulp and the albedo layer, respectively. Total pectin contents were about 30% of the AIS and showed comparatively constant values among the byproducts. Hydrochloric acid soluble pectin contents were the hightest in the flavedo layer, 14.0%, and the lowest in the pulp, 4.4%. Over 90% of the total pectin could be extracted after 60min with 0.05N HCI at $85^{\circ}C.$ Microwave treatment reduced the extraction time significantly ; a comparable extraction yield was acquired after 10min with microwave treatment. The degree of esterification of the extracted pectin also increased with microwave treatment. Neutral sugars in the hydrolysate of the pectin were rhamnose, arabinose, galactose, glucose and xylose. No differences in molecular weight distribution of the pectin were found between the albedo and flavedo layers. Pectin of the pulp showed different molecular weight distribution from that of the peels.
The changes in phenolic compounds and pectin content were investigated during the growth of 3 cultivars (Hosui, Niitaka and Chuwhangbae) of Asian pear (Pyrus pyrifolria) fruits. The amounts of total phenolic compounds in peel, flesh and core was 20.61-22.98mg/g, 0.87-1.23mg/g and 6.39-37.96mg/g during early growth, respectively, and decreased with pear growth. Arbutin, chlorogenic acid and epicatechin were detected in each part of all three cultivars during early growth; of these, arbutin content was the highest. Caffeic acid, catechin and 4-hydroxymethyl benzoic acid were detected in the peel and core during the ripening process. The total pectin content in the flesh of Hosui, Niitaka, and Chuwhangbae cultivars decreased from 5.93mg/g, 5.99mg/g and 5.40mg/g to 1.07mg/g, 1.60mg/g and 1.63mg/g, respectively. Of the soluble pectins, the hydrochloric acid-soluble pectin content was the highest, 3.21-3.45mg/g, and decreased during growth.

Effect of the thickness of packaging film on the ripening of mature-green mume (Prunus mume Sieb. et Zucc) fruit was investigated by measuring physicochemical changes of the fruit during storage. Fruits were packaged using low density polyethylene (LDPE) films with thicknesses of 20, 30, and 40$\mu$m and stored at the room temperature. The physicochemical properties such as contents of various pectic substances, molecular weight distribution of soluble pectic substances, and surface image of the fruit were determined during storage of 8 days. In general, regardless of the thickness of the films applied, a content of water-soluble pectin (WSP) in the fruit was increased during storage, but both contents of HCI- soluble pectin (HSP), and Ca and Mg in total alcohol-insoluble solids were decreased. Sephacryl S-300 gel filtration study revealed that fractions of high molecular weight HSP and WSP decomposed into lower molecular weight pectins during storage. The scanning electronic microscope also identified a significant structural change of the fruit skin over the storage time. It could be concluded from the results that fruits packaged with LDPE 30${\mu}{\textrm}{m}$ film maintained the highest physicochemical quality of green mume fruit during storage.

This paper was performed to investigate the changes of non-cellulosic neutral sugars composition in cell wall of persimmon fruit by treatment of cell wall degrading enzyme in vitro. Rhamnose, xylose and galactose in cell wall by polygalacturonase treatment, arabinose, galactose and rhamnose in cell wall by mixed enzyme treatment and arabinose and galactose in cell wall by ${\beta}-galactosidase$ treatment decreased, respectively. Noncellulosic neutral sugars of pectins extracted cell wall by enzyme treatments decreased and those by polygalacturonase treatment decreased remarkably. Rhamnose, arabinose and xylose in hemicellulose I of cell wall by polygalacturonase treatment were higher than those of untreated, and rhamnose and xylose in that by ${\beta}-galactosidase$ treatment were higher but arabinose, mnnose and galactose decreased. Xylose, mannose and glucose in that by mixed enzyme treatment were higher than those of untreatment and arabinose and galactose decreased. Contents of total non-cellulosic neutral sugars in hemicellulose of untreatment, and contents xylose, and glucose in hemicellulose II of cell wall by polygalacturonase treatmet decreased but those of other treatments were not changed.
Pectolytic enzymes were extracted in apple fruits rotted by Botryosphaeria dothidea, and their activities and change of pectic substances were investigated. Exo-polygalacturonase(exo-PG), exo-polymethylgalacturonase(exe-PMG), polygalacturonate-trans-eliminase(PGTE) and pectin-methyl-trans-eliminase(PMTE) were produced by the pathogen. Activities of exo-PG and exe-PMG extracted from rotten apple fruits were high to 21.15 and 24.65 units/mg protein in specific activity at seven days after inoculation, respectively. Activities of PGTE and PMTE showed 5.60 and 7.90 units/mg protein, respectively, but they were lower than those of the exo-type enzymes. Water-soluble and versene-soluble pectins were 11.50 mg/100 mg-AIS and 7.31 mg/100 mg-AIS at 14 days after inoculation, namely, they were increased by 4.23 and 2.16 mg/100 mg-AIS over those of sound apples, respectively. Total soluble pectic substances of rotten apple were 72.4% of total pectic substances and it was higher by 24.8% than sound apple. Insoluble pectic substance was notably decreased from 15.32 to 7.16 mg/100 mg-AIS according to progress of decay while total pectic substances were not changed remarkably.
Acidic endo-polygalacturonases are the major part of pectinase preparations and extensively applied in the clarification of fruits juice, vegetables extracts, and wines. However, most of the reported fungal endo-polygalacturonases are active and stable under narrow pH range and low temperatures. In this study, an acidic endo-polygalacturonase (EPG4) was purified and characterized from a mutant strain of Penicillium oxalicum. The N-terminal amino acid sequence of EPG4 (ATTCTFSGSNGAASASKSQT) was different from those of reported endo-polygalacturonases. EPG4 displayed optimal pH and temperature at 5.0 and 60-70℃ towards polygalacturonic acid (PGA), respectively, and was notably stable at pH 2.2-7.0. When tested against pectins, EPG4 showed enzyme activity over a broad acidic pH range (>15.0% activity at pH 2.2-6.0 towards citrus pectin; and >26.6% activity at pH 2.2-7.0 towards apple pectin). The Km and Vmax values were determined as 1.27 mg/ml and 5,504.6 U/mg, respectively. The enzyme hydrolyzed PGA in endo-manner, releasing oligo-galacturonates from PGA, as determined by TLC. Addition of EPG4 (3.6 U/ml) significantly reduced the viscosity (by 42.4%) and increased the light transmittance (by 29.5%) of the papaya pulp, and increased the recovery (by 24.4%) of the papaya extraction. All of these properties make the enzyme a potential application in the beverage industry.

Paper cultural heritages in museums and libraries are deteriorated by many biological factors like as fungi, insects, bacteria and rodents and get irreversibly damaged. Especially, paper components like as cellulose, hemicellulose, lignin, pectins, tannins, proteins and mineral additives are good nourishment for microorganism. Through some studies on fungi causing the aging of paper materials, Aspergilli (about 30%) and Penicilli (more than 30%) are the most common among 300 different kinds of microorganism that caused the biological aging of paper cultural heritages in museums and libraries. At present, various treatments are attempted to control the biodeterioration by these fungi. Especially, it is focused on the control of environmental factors such as humidity, temperature and oxygen. In this study, the oxygen absorbent was used to control oxygen, one of the these favorable conditions during the biological aging of Hanji by Aspergillus versicolor and Penicillium polonicum and then the effect on prevention in aging by this treatment was investigated. In result, the oxygen absorbent treatment had the good effect on prevention in aging during the biological aging by two species of fungi.
