한산 모시의 역학적 특성 및 태에 관한 연구(제1보) (Mechanical Properties and Fabric Handle of kansan Bamie (Part I))

  • Ramie is one of the traditional fabrics in Korea, and very comfortable fabric for summer clothes because it has a high moisture-absorbing and transporting property. Futhermore ramie is very popular and Koreans prefer its handle for summer clothes. The kansan ramie has better quality as fibers and can be weaved as fine fabrics which are famous as kansan Fine ramie. Even though the good quality of kansan ramie has known widely, very few research work have been carried out on kansan ramie in the field of textile science. In this study, the analysis of the physical and chemical characteristics of Hansan ramie was conducted by using two different kinds of Hansan ramie: Hansan Fine ramie and kansan Coarse ramie. In addition, the same experiment was held on the one kind of chinese ramie to be compared with those of Hansan ramie. The following results were obtained from this experimental study. By the analysis of chemical composition of ramie, the similar chemical composition (a -cellulose: 83∼85%, pectin substances: 2.81∼ 3.01%) were found from all of the ramie fabrics used in this study. It has shown that Hansan coarse ramie has the highest toughness value and wrinkle recovery angle among the samples used in this study. From the result of KES-F system, it was found that Hansan Coarse ramie which is composed with the thicker yarns has the highest value on the bending properties, 2HG and surface properties. The primary hand value was also calculated by KN-203 LDY and value of Koshi was shown as the order of kansan coarse ramie> Chinese ramie> kansan fine ramie, and Hansan fine ramie had shown the highest Numeri and Fukurami value among the 3 samples used in this study.

Isolation of a Novel Freshwater Agarolytic Cellvibrio sp. KY-YJ-3 and Characterization of Its Extracellular ${\beta}$-Agarase

  • A novel agarolytic bacterium, KY-YJ-3, producing extracellular agarase, was isolated from the freshwater sediment of the Sincheon River in Daegu, Korea. On the basis of Gram-staining data, morphology, and phylogenetic analysis of the 16S rDNA sequence, the isolate was identified as Cellvibrio sp. By ammonium sulfate precipitation followed by Toyopearl QAE-550C, Toyopearl HW-55F, and MonoQ column chromatographies, the extracellular agarase in the culture fluid could be purified 120.2-fold with a yield of 8.1%. The specific activity of the purified agarase was 84.2 U/mg. The molecular mass of the purified agarase was 70 kDa as determined by dodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The optimal temperature and pH of the purified agarase were $35^{\circ}C$ and pH 7.0, respectively. The purified agarase failed to hydrolyze the other polysaccharide substrates, including carboxymethyl-cellulose, dextran, soluble starch, pectin, and polygalacturonic acid. Kinetic analysis of the agarose hydrolysis catalyzed by the purified agarase using thin-layer chromatography showed that the main products were neoagarobiose, neoagarotetraose, and neoagarohexaose. These results demonstrated that the newly isolated freshwater agarolytic bacterium KY-YJ-3 was a Cellvibrio sp., and could produce an extracellular ${\beta}$-agarase, which hydrolyzed agarose to yield neoagarobiose, neoagarotetraose, and neoagarohexaose as the main products.

Delaware 포도에서 Gibberellin 처리와 봉지씌우기가 과립의 성숙과 품질에 미치는 영향 (Effects of Gibberellin Application and Bagging on Ripening and Quality in 'Delaware' Grape Berries)

  • This experiment was carried out to clarify the effects of gibberellin(GA) aplication and bagging on repeening and quality in 'Delaware' grape berries. Treatments are 4 plots(2X2 factorial experiment); GA, GA+bagging, bagging and control. The clusters were dipped twice in 100 ppm GA with GA treatment : 10 days before and after the full bloom. The results obtained as follows: 1. GA treatment made the seedless grape berry reduced in the fresh weight but it hastened the ripening period about 2 weeks. 2. Total soluble solid(TSS), viscosity and pH value of berry juice increased with maturation. The concentration of TSS and viscosity were higher in GA treatment plot than GA non-treatment. 3. Berry-hardness, titratable acidity and alcohol inslouble solid(AIS) decreased with maturation. Expically berry-hardness and AIS decreased more greatly in GA non-treatment than GA treatment. 4. The concentration of anthocyanin increased with ripening but pectic substance didn't fluctuate nearly. These of anthocyanin and pectin were higher in GA non-treatment plot than GA treatment. 5. By analysis of factorial experiment GA treatment was highly significant with the $^{o}$Brix/Acidity ratio, juice viscosity and AIS, but high negatively, significant with berry-hardness and berry fresh weight. And it was significant with T S S and negatively, titratable acidity. Bagging was significant with $^{o}$Brix/Acidity ratio and AIS content, but negatively, titratable acidity. 6. Qualitative characters were high correlated with the $^{o}$Brix/Acidity ratio in simple correlation but direct effect by the path-coefficient analysis didn't coincide with simple correlation. The direct effect of pH was large and juice viscosity, the next. And that of berry-hardness was negligible but, AIS, small negatively.

콩잎 물김치의 숙성과정 중 이화학적 변화 (The Physicochemical Change of Soybean-Leaf Water Kimchis during Fermentation)

  • This study was attempted to investigate the physicochemical changes during soybean-leaf water kimchis fermentation by adding wheat flour in cooking water. Soybean-leaf water kimchis with five different levels of wheat flour in cooking water(0%;A, 5%;B 10%;c, 15%;D, 20%;E) were tested for rhological parameters, fine structural changes, sensory evaluations and chemical analysis. The composition of the soybean-leaf was moisture (80.9%), protein(6.8%), fat(0.6%), ash(2.1%) and alkalinity (+14.9). The amount of reducing sugars of sample A and other samples were 0.75% and 1.08∼1.4% in the initial fermentation stage, but decreased to 0.3 and 0.43∼0.50% in the later fermentation stage, respectively. The pH of sample A decreased from 5.17 to 4.72 during the initial fermentation. On sample B, C, D and E, pH's decreased rapidly during the initial fermentation, but they did not change much in the later fermentation stage. The pectin contents of all samples decreased during 2nd and 4th day of fermentation, then the change was slow. The amount of hemicellulose. cellulose and lignin in terms of the DNF and ADF were varied from 4 to 33%, but the contents of them did not greatly changed during the fermentation. The sensory evaluation showed that both B and C samples had the good score in sweety taste, roasted nutty taste, and the ease of swallowing measured as chewiness. From these results, the optimum soybean-leaf water kimchis can be prepared when 200g soybean-leaf, 1000$m\ell$ water, 15g garlic, 3% red pepper powder and 5 ∼0% wheat flour were fermented at 20$^{\circ}C$ for 2 days.

섬유소-펙틴 분해력이 있는 새로운 Aspergillus tubingensis의 분리와 특성 규명 (Isolation and Characterization of a Novel Aspergillus tubingensis with a Hydrolyzing Activity of Cellulose-pectin Complex)

  • 대전광역시 근교의 야산과 들판 등지에서 썩은 나뭇잎, 짚, 흙을 채취하여 각각을 배양한 다음 Congo red test에 의해 cellulase 활성을 보이는 균주를 선별하였다. Genomic DNA를 분리한 후 PCR을 수행하여 DNA sequence를 Gene Bank를 통해 분석한 결과 A. tubingensis로 밝혀졌다. 이것을 배양하여 상등액을 crude enzyme으로 사용하여 온도와 pH를 달리하면서 효소의 활성정도를 측정하였다. 대조균주로 A. oryzae KCTC 6291를 이용하였고, 본 연구를 통하여 분리한 균주인 A. tubingensis가 생산하는 cellulase는 A. oryzae의 cellulase에 비하여 각각 다른 온도와 pH에서 높은 안정성을 보여주었다. A. tubingensis는 각각의 온도에서 활성의 정도가 비슷했으며, 45$^{\circ}C$, 55$^{\circ}C$에서 높은 활성을 나타내고 있지만, 고르게 활성이 나타났다. 또한 pH 12.0에서 가장 높은 활성을 보여 주었고, pH 2.0, 3.0, 4.0에서는 양쪽 모두 거의 활성이 없었으며, 중성, 염기성에 대해서 활성에는 큰 변화가 없었다. 따라서, 분리 동정한 A. tubingensis는 온도와 pH에서 고르게 활성을 나타내므로 생균제로 활용할 수 있는 범위가 클 것으로 여겨진다.

에틸렌 및 Elicitor처리에 의한 아기장대풀의 키틴 가수분해 효소 유도 (Induction of Arabidopsis thaliana Chitinase by Ethylene and Elicitor Treatment)

  • 키틴 가수분해 효소와 베타-1,3-글루카네이즈는 식물체의 주요 방어효소로 여겨지고 있다. 본 논문에서는 아기장대풀의 잎조직으로부터 고정된 현탁배양주를 만들고 이어서 에틸렌과 유인제 (elicitor) 처리에 의한 키틴 가수분해 효소유도양상을 분석하였다. 키틴 가수분해 효소활성은 $^3$H으로 표지된 키토산을 기질로 한 radiochemical 분석방법이나 담배식물체의 키틴 가수분해 효소를 대상으로 얻어진 항체를 이용하여 Western 분석방법을 사용하였다. 이 결과 에틸렌과 유인제 처리 공히 48시간후에 가장 활성이 높게 나타남을 관찰하였다. 또한 씨앗, 새싹, 식물체의 잎, 뿌리 등에서의 활성을 조사하여 뿌리에서 키틴 가수분해 효소 활성이 높음을 확인하였다.

궐련지의 섬유종류가 담배 연기성분에 미치는 영향 (The comparative analysis of smoke components delivered from cigarette papers manufactured by flax and wood pulp)

  • This study was conducted to evaluate the effect of cigarette papers, flax and wood, on the delivery of mainstream smoke. The main components of cigarette papers were cellulose, hemicellulose, lignin, and pectin. Lignin contents, known as precursor of smoke's phenolic compounds, of the flax and wood cigarette papers were 5.8% and 10.6%, respectively. The pyrolysis products of cigarette papers were similar by the profile of total ion chromatogram. But, the area % of some components, such as 1,3-cyclopentanedione, 3,5- dimethyl cyclopentane-1,2-dione, 2-hydroxy-3-methyl-2-cyclopentenone, dihydro-2(3H)-furanone, 3-methyl-2(5H)-fruanone, and 5-methyl-2-furaldehyde delivered through pyrolysis of the flax cigarette paper were higher than that of wood cigarette paper. Otherwise, the area % of some components, such as 2-methyl-cyclopentene-l-one), 2,3-butanedione, 2-cyclopentene-l-one, and 5-hydroxy-2-methyl-furaldehyde, 2-furaldehyde delivered through pyrolvsis of the wood cigarette paper were higher than that of flax cigarette paper. To identify the difference between two cigarette papers, we used the cigarette column filled with the cut cigarette paper instead of the cut tobacco leaf. The amounts of semi-volatile fraction delivered from flax cigarette paper was more than that of wood cigarette paper. But, by using the cut tobacco, there was no big difference of delivery amount between flax and wood cigarette papers. Also, aroma of TPM by collecting from brening cut tobacco wrapped in flax and wood papers showed a different pattern by the electonic nose system. Although the difference between two cigarette papers by using the cut tobacco was smaller than that of cut cigarette paper, this result indicated that the fax and wood had the different effects on the delivery of smoke components as shown in the sensory test results.

Relationship between the Methane Production and the CNCPS Carbohydrate Fractions of Rations with Various Concentrate/roughage Ratios Evaluated Using In vitro Incubation Technique

  • The objective of the trial was to study the relationship between the methane ($CH_4$) production and the Cornell Net Carbohydrate and Protein System (CNCPS) carbohydrate fractions of feeds for cattle and the suitability of CNCPS carbohydrate fractions as the dietary variables in modeling the $CH_4$ production in rumen fermentation. Forty-five rations for cattle with the concentrate/roughage ratios of 10:90, 20:80, 30:70, 40:60, and 50:50 were formulated as feed samples. The Menke and Steingass's gas test was used for the measurement of $CH_4$ production. The feed samples were incubated for 48 h and the $CH_4$ production was analyzed using gas chromatography. Statistical analysis indicated that the $CH_4$ production (mL) was closely correlated with the CNCPS carbohydrate fractions (g), i.e. CA (sugars); $CB_1$ (starch and pectin); $CB_2$ (available cell wall) in a multiple linear pattern: $CH_4=(89.16{\pm}14.93)$ $CA+(124.10{\pm}13.90)$ $CB_1+(30.58{\pm}11.72)$ $CB_2+(3.28{\pm}7.19)$, $R^2=0.81$, p<0.0001, n = 45. Validation of the model using 10 rations indicated that the $CH_4$ production of the rations for cattle could accurately be predicted based on the CNCPS carbohydrate fractions. The trial indicated that the CNCPS carbohydrate fractions CA, $CB_1$ and $CB_2$ were suitable dietary variables for predicting the $CH_4$ production in rumen fermentation in vitro.

Pectic-Oligoshaccharides from Apples Induce Apoptosis and Cell Cycle Arrest in MDA-MB-231 Cells, a Model of Human Breast Cancer

  • Background: The effects of plant products on cancer cells has become a field of major importance. Many substancesmay induce apoptosis in anti-cancer treatment. Pectins, a family of complex polysaccharides, and their degradation products may for exasmple exert apoptotic effects in cancer cells. Apples and citrus fruits are the main sources of pectin which can be applied for anti-cancer research. The present study concerned an intact form of pectic-oligoshaccharide named pectic acid (poly galactronic acid). Materials and Methods: Inhibition of cell proliferation assays (MTT), light microscopy, fluorescence microscopy (acridin orange/ethidium bromide), DNA fragmentation tests, cell cycle analysis, annexin PI and Western blotting methods were applied to evaluate apoptosis. Results: The results indicated that pectic acid inhibited cell growth and reduced cell attachment after 24h incubation. This did not appear to be due to necrosis, since morphological features of apoptosis were detected with AO/EB staining and cell cycling was blocked in the sub-G1 phase. Annexin/PI and DNA fragmentation findings indicated that apoptosis frequency increased after 24h incubation with pectic acid. In addition, the data showed pectic acid induced caspase-dependent apoptosis. Conclusions: These data indicate that apple pectic acid without any modification could trigger apoptosis in MDA-MB-231 human breast cancer cells and has potential to improve cancer treatment as a natural product.

A Culture-Based Study of the Bacterial Communities within the Guts of Nine Longicorn Beetle Species and their Exo-enzyme Producing Properties for Degrading Xylan and Pectin

  • In this study, bacterial communities within the guts of several longicorn beetles were investigated by a culture-dependent method. A total of 142 bacterial strains were isolated from nine species of longicorn beetle, including adults and larvae. A comparison of their partial 16S rRNA gene sequences showed that most of the bacteria constituting the gut communities can typically be found in soil, plants and the intestines of animals, and approximately 10% were proposed as unreported. Phylogenetic analysis demonstrated that the bacterial species comprised 7 phyla, and approximately half were Gammaproteobacteria. Actinobacteria were the second most populous group (19%), followed by Firmicutes (13%) and Alphaproteobacteria (11%). Betaproteobacteria, Flavobacteria, and Acidobacteria were minor constituents. The taxonomic compositions of the isolates were variable according to the species of longicorn beetle. Particularly, an abundance of Actinobacteria existed in Moechotypa diphysis and Mesosa hirsute, which eat broadleaf trees; however, no Actinobacteria were isolated from Corymbia rubra and Monochamus alternatus, which are needle-leaf eaters. Considerable proportions of xylanase and pectinase producing bacteria in the guts of the longicorn beetles implied that the bacteria may play an important role in the digestion of woody diets. Actinobacteria and Gammaproteobacteria were the dominant xylanase producers in the guts of the beetles.
