To evaluate the effect of dietary fiber on the protein utilization, Sprague-Dawley rats were fed diet containing 15% or 30% of pectin and 15% or 30% ${\alpha}-cellulose$. Control group was fed fiber free diet. The animals were fed and libitum for 5 weeks. Weight gain was less in the rat fed a pectin supplemented diet than those fed ${\alpha}-cellulose$ supplemented or control diet. Furthermore, weight gain decreased more by the addition of 30% pectin than 15% pectin level. The rats fed ${\alpha}-cellulose$ or pectin shwoed a decreasing tendency of food efficiency ratio compared to the control group. The rats fed a diet containing pectin showed an increasing tendency of the liver weight compared to the control group and those fed cellulose. The rats fed a diet containing pectin showed a decreasing tendency of hepatic protein content compared to those fed cellulose or control group fed fiber free diet. The rats fed diet containing pectin(15%, 30%) showed remarkable decreased activity of liver xanthine oxidase compared with those fed ${\alpha}-cellulose$ or the control group. These results suggested that the pectin may be alter the absorption of protein in intestinal lumen.
The effect of dietary pectin, tangerine pulp meal, propionate, lactate or fumarate on cholesterol (C) and triacylglycerol(TG) levels in the serum and liver, and the effect of pectin on dietary C absorption were studied in a series of three experiments. Mature female Sprague Dawley rats were fed a control diet or diets containing 5% pectin, 5% tangerine pulp meal, 3% propionate, 3% lactate 3% fumarate, or 10% pectin. Serum total C levels were lower(p<0.05) in rats fed the diet containing 5% pectin than in control rats after a 4-week feeding period(93.8 vs 119.2mg/100mL). Serum HDL, LDL+VLDL C levels were not different among diet groups. Liver total C level was also lower(p<0.05) in rats fed the diet containing 5% pectin than in control rats, but liver TG level was not influenced by diet. Dietary propionate, lactate or fumarate did not reduce serum C, indicating that propionate is not a regulator of serum C. However, dietary pectin(10%) increased fecal excretion of dietary C(or its metabolites) more than 70% over a control value. Our data indicate that dietary pectin reduces serum and liver C levels by increased fecal secretion of dietary C, but not by its fermentation product propionate or other gluconeogenic substrates. Pectin is a natural polysaccharide and biopolymer that serves as a structural component of plant tissues' primary cell walls. Pectin is primarily composed of D-galacturonic acid linked by α-1, 4-glycosidic linkage and is further classified by the ratio of esterified galacturonic acid groups known as degree of esterification (DE). Pectin that contains more than half of its carboxylate units as methyl esters is known as a high methyl (HM) ester. Conversely, pectin that has less than half of its carboxylate units as methyl esters is known as a low methyl (LM) ester. Pectin has various bioactive properties, including anticancer, anti-inflammatory, antioxidant, antidiabetic, anticholesterol, antitumoral, and chemopreventive properties. Moreover, pectin is a useful biopolymer in biomedical applications. Biomedical engineering, which is founded on research aimed to improve the quality of life using new materials and technologies, is typically classified according to the use of hydrogels, nanofiber mats, and nanoparticles. This paper reviews the progress of recent research into pectin-based biomedical applications and the potential future biomedical applications of marine-derived pectin.
High-ester pectin was demethylated by the treatments of HCl alone and a combination of HCl and $NH_4OH$. The low-ester pectin prepared were analyzed for chemical composition and the pectin gels were evaluated for firmness by sag values and strength by puncture stress. Gels made from HCl demethylated sample showed brittle, weak and poor elastic characteristics while the $HCl-NH_4OH$ treated samples generally resulted in a smooth and elastic gels except those samples having very low content of ester group or acid amide group. Statistical analysis showed that significant correlations were found between sag values and ester content or molecular weight, and puncture stress and ester content, acid amide groups or molecular weight. The equations derived for sag, puncture stress and sag/puncture stress from chemical data could be useful for prediction of some of the physical properties of low-ester pectin gel.

The changes of methoxy content. compositional sugars. degree of esterification (DE) and pectic substances such as LMP (low methoxy pectin), HMP (high methoxy pectin) IAP (ionically associated pectin) and CBP (covalently bounded pectin) of Fuji apple were investigated during maturity. The contents of LMP+HMP and IAP+CBP in alcohol insoluble solid(AIS) were 70% and 20%. respectively. These pectic substances were increased slightly during maturity. The reductions of hexose and uronic acid were Iron 20-50% and from 2-4%, respectively, but the inclosed of pentose was from 1-9% in LMP, HMP, IAP and CBP during maturity of Fuji apple. The galactose was decreased significantly among hexose. Whereas the relative ratio of uronic acid in HMP and CBP was increased.
Cell wall components were decreased during maturation and postharvest of persimmon fruits. Contents of pectin and alkali-soluble hemicellulose were increased during maturity, but those of acid-soluble hemicellulose were decreased. Contents of pectin and alkali-soluble hemicellulose were decrease in soft persimmon, whereas acid-soluble hemicellulose was increased remarkably. Cellulose contents were increased during maturation and this tend was notable in soft persimmon. Contents of cell wall polysaccharides per 100g-fresh weight were decreased. Contents of total pectin and insoluble pectin were increased during maturation but decreased in soft persimmon. Content of water-soluble pectin was increased during maturation and postharvest.
This study was attempted to determine the pectin contents of various fruits(Cherry, strawberry, plum, peach, apricot, apple, crabapple) and to investigate the changes in the properties of pectin and pectin gel during crabapple ripening. Methoxyl content, molecular weight and intrinsic viscosity were measured in ripened and unripened crabapples. In order to determine the characteristics of pectin gel, percent sag was measured by exchange Ridgelimeter, hardness and textural characteristics of jellies were measured by a penetrometer and a texturometer. The experimental results are summarized as follows: 1. The pectin contents of various fruits were 1.16% for cherry, 3.17% fir strawberry, 4.29% for plum, 5.51% for apricot, and pectin contents of peaches and apples varies depends on varieties from 4.54% to 5.61% to 6.52, respectively. The pectin contents of unripened and ripened crabapple wee 9.21% and 9.84% respectively, indicating that pectin content increased with fruit maturity. 2. The methoxyl contents, which was 6.61%~6.95%, was very low and it increased significantly during fruit ripening. Further, a decrease in acetyl content, intrinsic viscosity, molecular weight occurred during the ripening of crabapples. 3. The percent sag of crabapple jellies increased with fruit maturity from 5.26% to 5.79%, and the corresponding jelly grade decreased in ripened crabapple. 4. The hardness of jellies determined by a penetrometer was 79.15 pp.U. for ripened crabapple jelly, 90.62 pp.U. for unripe ned crabapple jelly. 5. In the textural characteristics of gels measured by a texturometer, the gels made from unripened crabapple were shown to have the highest values in hardness, cohesiveness, adhesiveness and gumminess.
Drug delivery systems that are based on pectin have been studied for colon specific delivery using the specific activity of colon microflora. The aim of this study was to design a novel method of manufacturing pectin microspheres without oils and surfactants and to investigate the potential use of the pectin microspheres as an oral colon-specific drug carrier. The pectin microspheres were successfully formed using the spray drying method and crosslinking with calcium chloride. From the crosslinked pectin microspheres, indomethacin (IND) release was more suppressed than its release from non-crosslinked microspheres. In a low pH (pH 1.4) environment, the pectin microspheres released IND at an amount of about 18${\pm}$2% of the total loaded weight for 24 h while the release rate of IND was stimulated at neutral pH (pH 7.4). IND release from the pectin microspheres was increased by the addition of pectinase. The results clearly demonstrate that the pectin microspheres that were prepared by the spray drying and crosslinking methods are potential carriers for colon-specific drug deliveries.

Oriental pear was used as treatment of asthma, control of blood pressure tonic medicine vasoactio, diabetes in oriental medicine. In this study, it was investigated that pear pectin effects on cardiovascular system as blood pressure and renin and atrial natriuretic peptide (ANP) in plasma, cardiac hypertrophy. The experiments were performed on Sprague-Dawley rats, 2K1C hypertension model was prepared by constricting the left renal artery with a sliver clip. Animals were then divided into four groups, 5 mg/kg, 10 mg/kg, 15 mg/kg and the control, pear pectin and apple pectin solutions were supplied with them. The blood pressure was more decreased in pear pectin 10 mg/kg than in apple pectin. The plasma ANP was decreased in pear pectin 10 mg/kg, and renin was increased in same concentration of drug. Cardiac hypertrophy had a tendency to decrease in pear pectin 15 mg/kg, but was not statistically significant compared to control group.
Oral drug delivery system using pectin gel was developed for colon-targeting of peptide drug. BSA(bovine serum albumin)-loaded pectin and pectin-alginate beads were prepared for drug release properties in vitro. Morphological studies by electron microscopy indicated that pectin and pectin-alginate beads were spherical in shape and approximately 1.0 mm. In order to find the suitable beads, effects of cross-linking agents (calcium chloride or zinc acetate) and drying temperature of beads were investigated. Drug release decreased with concentration of cross-linking agents and drying temperature. For colonic drug delivery from pectin and pectin-alginate beads, pectin degradable enzymes were added at 5 hrs from the beginning of drug release. After addition of enzymes, drug release was suddenly increased against free enzymes. Therefore, pectin and pectin-alginate beads can be promised as useful drug release carriers for colon-targeted delivery.
