NF-κB and Therapeutic Approach

  • Since NF-${\kappa}B$ has been identified as a transcription factor associated with immune cell activation, groups of researchers have dedicated to reveal detailed mechanisms of nuclear factor of ${\kappa}B$ (NF-${\kappa}B$) in inflammatory signaling for decades. The various molecular components of NF-${\kappa}B$ transcription factor pathway have been being evaluated as important therapeutic targets due to their roles in diverse human diseases including inflammation, cystic fibrosis, sepsis, rheumatoid arthritis, cancer, atherosclerosis, ischemic injury, myocardial infarction, osteoporosis, transplantation rejection, and neurodegeneration. With regards to new drugs directly or indirectly modulating the NF-${\kappa}B$ pathway, FDA recently approved a proteasome inhibitor bortezomib for the treatment of multiple myeloma. Many pharmaceutical companies have been trying to develop new drugs to inhibit various kinases in the NF-${\kappa}B$ signaling pathway for many therapeutic applications. However, a gene knock-out study for $IKK{\beta}$ in the NF-${\kappa}B$ pathway has given rise to controversies associated with efficacy as therapeutics. Mice lacking hepatocyte $IKK{\beta}$ accelerated cancer instead of preventing progress of cancer. However, it is clear that pharmacological inhibition of $IKK{\beta}$ appears to be beneficial to reduce HCC. This article will update issues of the NF-${\kappa}B$ pathway and inhibitors regulating this pathway.

Analysis of Information Adoption Pathways and Strategies of Raising Information Adoption of Farmers (농업인의 정보 수용경로 분석 및 정보수용 제고 방안)

  • This study analyzed the characteristics of diverse information adoption pathways per pathway of information composition category including 'information perception', 'information assessment', and 'information adoption' and examined information adoption characteristics per pattern. In addition, differences among agricultural researchers and farmers per information adoption stage were analyzed. According to these analyses, as for farmers' information adoption pathways, while the formal extension type and the situational reaction diffusion type used the direct pathway of 'information perception ${\rightarrow}$ information adoption,'the agriculturist connection type and the systematic approach type did not use the direct pathway of 'information perception ${\rightarrow}$ information adoption' but, instead, adopted information by moving from the 'information perception ${\rightarrow}$ information assessment' pathway to the 'information assessment ${\rightarrow}$ information adoption' pathway, with information assessment as the intermediation. In the case of farmers, information adoption was the highest in comparison with information assessment and information perception in all patterns whereas, in the case of agricultural researchers, the results were exactly opposite those of farmers. By suggesting an information adoption pathway appropriate to each pattern, this study sought to enhance information adoption.

The Gene Expression Profile of LPS-stimulated Microglial Cells

  • This study was conducted to evaluate the inflammatory mechanisms of LPS-stimulated BV-2 microglial cells. The inflammation mechanism was evaluated in BV-2 cells with or without LPS treated using the Affymetrix microarray analysis system. The microarray analysis revealed that B cell receptor signaling pathway, cytokine-cytokine receptor interaction, Jak-STAT signaling pathway, MAPK signaling pathway, Neuro-active ligand-receptor interaction, TLR signaling path-way, and T cell receptor signaling pathway-related genes were up-regulated in LPS stimulated BV-2 cells. Selected genes were validated using real time RTPCR. These results can help an effective therapeutic approach to alleviating the progression of neuro-in-flammatory diseases.

Pathway Crosstalk Analysis Based on Protein-protein Network Analysis in Ovarian Cancer

  • Ovarian cancer is the fifth leading cause of cancer death in women aged 35 to 74 years. Although there are several popular hypothesis of ovarian cancer pathogenesis, the genetic mechanisms are far from being clear. Recently, systems biology approaches such as network-based methods have been successfully applied to elucidate the mechanisms of diseases. In this study, we constructed a crosstalk network among ovarian cancer related pathways by integrating protein-protein interactions and KEGG pathway information. Several significant pathways were identified to crosstalk with each other in ovarian cancer, such as the chemokine, Notch, Wnt and NOD-like receptor signaling pathways. Results from these studies will provide the groundwork for a combination therapy approach targeting multiple pathways which will likely be more effective than targeting one pathway alone.

  • The static approach of representing metabolic pathway diagrams offers no flexibility. Thus, many systems adopt automatic graph layout techniques to visualize the topological architecture of pathways. There are weaknesses, however, because automatically drawn figures are generally difficult to understand. The problem becomes even more serious when we attempt to visualize all of the information in a single, big picture, which usually results in a confusing diagram. To provide a partial solution to this thorny issue, we propose J2dpathway, a metabolic pathway atlas viewer that has node-abstracting features.

Effect of Critical Pathway on Diabetes Mellitus (당뇨병 표준진료지침 적용 효과에 관한 연구)

  • Background : The aim of this study was to examine the effectiveness of the critical pathway to diabetic patients who were admitted in the hospital. Methods : For establishment of critical pathway for diabetic patients, we organized the multidisciplinary care team. During 5 months, 31 diabetic inpatients were applied the critical pathway and the results were compared with 11 diabetic inpatients who were treated with conventional way. We assessed the patients satisfaction, knowledge on the disease, compliance of medical regimen, length of stay(LOS) and hospital cost. We used the computer program SAS for statistical analysis. Data were summarized with mean, and analyzed using t-test repeated measures ANOVA and Wilcoxon rank sum test. Results : Length of stay(LOS) was remarkably shortened in critical pathway group compared with the conventional treatment group($7.6{\pm}1.23$ vs $12.0{\pm}4.73$, p<0.000). Although LOS was significantly shortened in critical pathway group, patients satisfaction was much higher than conventional treatment group. There were no significant difference of knowledge on the disease, compliance of medical regimen and blood glucose level between two groups at discharge. Hospital cost of each patients also significantly decreased in critical pathway group(890,000 won vs 1,280,000 won, p<0.05). Conclusion : These results showed that establishment of critical pathway for diabetic inpatients with team approach is the one of the way to improve the quality of diabetic patients management and to enhance the efficiency of hospital management.

Studies on Gene Expression of Yukmijihwang-tang using High-throughput Gene Expression Analysis Techniques (대규모 유전자 분석 기법을 이용한 육미지황원의 유전자 발현 연구)

  • Yukmijihwang-tang(YM) is a noted herbal prescription in Chinese and Korean traditional medicines, and it has been known to reinforce the vital essence and has been widely used for a variety of disease such as stroke, osteoporosis, anti-tumor, and hypothyrodism. Regarding its traditional use, YM has been known to reinforce the Yin (vital essence) of liver and kidney. Also it has been known to reinforce nutrition and biological function in brain. Recently, studies suggested that YM increase antioxidant activities and exert the protective effect against oxidant-induced liver cell injury. We investigated the high-throughput gene expression analysis on the Yukmijihwang-tang administrated in SD rats. Microarray data were validated on a limited number of genes by semiquantitative RT-PCR and Western blot analyses. The recent availability of microarrays provides an attractive strategy for elaborating an unbiased molecular profile of large number of genes in drug discovery This experimental approach offers the potential to identify molecules or cellular pathways not previously associated with herbal medicine. Total RNA from normal control brain and Yukmijihwang-tang administrated brain were hybridized to microarrays containing 10,000 rat genes. The 52 genes were found to be up-regulated(twice or more) excluding EST gene. The nine genes were found to be down-regulated(twice or more) excluding EST gene. Gene array technology was used to identify for the first time many genes expression pathway analysis that arecell cycle pathway, apoptosis pathway, electron transport chain pathway, cytoplasmic ribosomal protein pathway, fatty acid degradation pathway, and TGF-beta signaling pathway. These differentially expressed genes pathway analysis have not previously been iavestigated in the context of herbal medicine efficacy and represent novel factors for further study of the mechanism of herbal medicine efficacy.

Inhibitors of AKT Signaling Pathway and their Application

  • The AKT signaling pathway is a highly regulated cell signaling system that forms a network with other cell signaling pathways. Hence, the AKT signaling pathway mediates several important cellular functions that include cell survival, proliferation, cell migration, and et cetera. Irregularities that led overactive AKT signaling have been linked to many diseases such as cancer and metabolic-associated diseases. Hence, modulating the overactive AKT signaling pathway via inhibitor is a tantalizing prospect for treatment of cancer and metabolic-associated diseases. Two inhibitors of the AKT signaling pathway will be presented in this symposium: 1) Bisleuconothine A (BisA), a bisindole alkaloid that inhibit autophagy and 2) Ceramicine B (CerB), a limonoid that inhibit adipogenesis. The first topic is on a bisindole alkaloid, BisA and its mechanism in inducing autophagosome formation in lung cancer cell line, A549.(1) Since most autophagy inducing agents generally induce apoptosis, we found that BisA does not induce apoptosis even in high dose. BisA up-regulation of LC3 lipidation is achieved through mTOR inactivation. The phosphorylation of PRAS40, a mTOR repressor was suppressed by BisA. This observation suggested that BisA inactivates mTOR via suppression of PRAS40 phosphorylation. Interestingly, the phosphorylation of AKT, an upstream regulator of PRAS40 phosphorylation was also down-regulated by BisA. These findings suggested that Bis-A induces autophagosomes formation by interfering with the AKT-mTOR signaling pathway. The second topic is on CerB and its mechanism in inhibiting adipogenesis in preadipocytes cell line, MC3T3-G2/PA6.(2,3) CerB inhibits the phosphorylation of protein kinase B (AKT) at the Thr308 position but not the Ser473. Consequently, the phosphorylation of FOXO3 which is located downstream of AKT is also inhibited. Considering that FOXO3 is an important regulator of PPARγ which is a key factor in adipogenesis, CerB may inhibit adipogenesis via the AKT-FOXO3 signaling pathway. Taken together, both BisA and CerB highlighted the potential of AKT signaling pathway modulation as an approach to induce autophagy and inhibit the formation of fat cells, respectively.

Meaning of Housing through Oral Life History of Korean Chineses in Harbin, China : Focused on experiences of housing structure type and pathway approach (생애구술을 통해 본 중국 할빈 지역 조선족의 주거의 의미 : 주거유형 경험과 경로접근을 중심으로)

  • This research was designed to explore the meaning of housing among Korean Chinese in Harbin, China. In particular, the meaning of housing was examined by using the pathway approach. Utilizing qualitative research methods, this study administered the in-depth interview on the oral history of an individual life, and the 5 elderly persons in their 60s and 70s participated in the individualized interviews that were conducted from May 28 to 31 in 2010. The main findings of meaning of housing were as follows; 1. Similarly to the meaning of housing in 1970s and 1980s in Korea, house was viewed as both a shelter for family members and relatives and a place for their comfort. 2. Prior to multi-story residences, Harbin had only 3 different forms of single-story houses available; Chinese style with Kang and soil room(地室), Korean style with 'Ondol', and Russian style with open floor and Pechka, The promotion at work enabled participants to move to multi-story residences, their moving time varied from 1970 to 1991, and the residential moving determined their current housing status. 3. Multi-story residences were available around 1970s, floor-heating system was introduced from 1990s, and high-rise apartments were built from 1998. Korean Chinese(朝鮮族) weren't satisfied with the spatial composition of individual units embedded into the Chinese culture, especially, entrance, kitchen, bathroom and veranda. 4. Based on assimilation through socialism, adaptation to socialist society and capitalist acculturation, the lifestyles of the interviewees were categorized into five types - capitalist-proactive(Ms. KS), socialist-pragmatic(Ms. J), socialist-inducive(Ms. KY), family centered-conservative(Ms. L), and socialist-adaptive(Ms. P). This study implies that housing-related services for Korean Chinese are necessarily provided so as to embrace their life style and cultural identity in housing design, and further studies need to be explored.

Advanced estimation and mitigation strategies: a cumulative approach to enteric methane abatement from ruminants

  • Methane, one of the important greenhouse gas, has a higher global warming potential than that of carbon dioxide. Agriculture, especially livestock, is considered as the biggest sector in producing anthropogenic methane. Among livestock, ruminants are the highest emitters of enteric methane. Methanogenesis, a continuous process in the rumen, carried out by archaea either with a hydrogenotrophic pathway that converts hydrogen and carbon dioxide to methane or with methylotrophic pathway, which the substrate for methanogenesis is methyl groups. For accurate estimation of methane from ruminants, three methods have been successfully used in various experiments under different environmental conditions such as respiration chamber, sulfur hexafluoride tracer technique, and the automated head-chamber or GreenFeed system. Methane production and emission from ruminants are increasing day by day with an increase of ruminants which help to meet up the nutrient demands of the increasing human population throughout the world. Several mitigation strategies have been taken separately for methane abatement from ruminant productions such as animal intervention, diet selection, dietary feed additives, probiotics, defaunation, supplementation of fats, oils, organic acids, plant secondary metabolites, etc. However, sustainable mitigation strategies are not established yet. A cumulative approach of accurate enteric methane measurement and existing mitigation strategies with more focusing on the biological reduction of methane emission by direct-fed microbials could be the sustainable methane mitigation approaches.
