Isolation of Antifungal Substances by Bacillus amyloliquefaciens IUB158-03 and Antagonistic Activity against Pathogenic Fungi (Bacillus amyloliquefaciens IUB158-03이 생산하는 항진균물질의 분리와 항균활성)

  • For the control of pathogenic microorganisms, Bacillus spp. were isolated from diseased pepper fruits in Korea. Among them, Bacillus sp. IUB158-03 showed high inhibitory effect on mycelial growth and spore germination of C. gloeosporioides and Botrytis cinerea. The strain was identified as B. amyloliquefaciens IUB158-03 based on its physiological, biochemical characteristics and Microlog analysis. The highest level of antifungal substances by B. amyloliquefaciens IUB158-03 were obtained when the bacterium was cultured in medium containing 2% soluble starch, 3% yeast extract, 0.5% tryptone, 0.5% $NH_4H_2PO_4$, and 1% NaCl (pH 6.0) at $25^{\circ}C$ for 72 hrs. The antifungal substances were purified by butanol extraction, silica gel column chromatography, preparative thin layer chromatography, and high performance liquid chromatography. The purified antifungal substance was confirmed $R_f$ 0.27 by TLC. This substance exhibited antifungal activity against Fusarium solani, Rhizoctonia solani, Botrytis cineria, Alternata alternaria of plant pathogenic fungi and Trichophyton mentagrophytes, Epidermophyton floccosum, Cryptococcus neoformans of human pathogenic fungi.

First Report: Diversity of Endophytic fungi Possessing Antifungal Activity Isolated from Native Kougoed (Sceletium tortuosum L.)

  • Forty-three (n = 43) endophytic fungi with different morphologic characteristics were from a medicinal plant Sceletium tortuosum, were utilized to investigate their antifungal effectiveness against pathogenic fungi. All fungal isolates exhibited antifungal activity against one or more pathogens in the dual culture test whereas only 33 fungal culture filtrates (77%) showed decent antifungal effect. Fusaria and Aspergillus were the dominate genus that displayed significant antifungal activity. Isolates GG02, GG09, ND15, and ND17 showed the broadest spectrum of antifungal activity. Furthermore, culture filtrate of Fusarium sp. DR08 exhibited a broad range of antifungal activity against all the pathogens. The results suggest endophytic fungi isolated from medicinal plant might be a source of novel bioactive molecules. To the best our knowledge, this is the first report on endophytic fungi isolated from native kougoed exhibiting antifungal activity against plant fungal pathogens.

  • Two neolignan compound, magnolol $(5,5^{l}-diallyl-2,2^{l}-dihydroxybiphenyl, 1)$ and honokiol $(5,5^{l}-diallyl-2,4^{l}-dihydroxybiphenyl, 2)$ were isolated from the stem bark of Magnolia obovata and evaluated for antifungal activity against various human pathogenic fungi. Compound 1 and 2 showed significant inhibitory activities against Trichophyton mentagrophytes, Microsporium gypseum, Epidermophyton floccosum, Aspergillus niger, Cryptococcus neoformans, and Candida albicans with minimum inhibitory concentrations (MIC) in a range of $25-100{\mu}g/ml$. Therefore, compound 1 and 2 could be used as lead compounds for the development of novel antifungal agents.

  • Screening tests against six plant pathogenic fungi were performed in order to develop biopesticides. Actinomycetes were used to discriminate Bacillus thuringiensis for wide use as a microbial pesticide. From more than 100 actinomycetes tested, twelve strains showed potent antifungal activities. We report in vivo screening results from fermentation broths of these twelve strains and identification of the strain taxa.

  • 2-Arylthio-, 2-arylthio-5-methoxy-, 2, 3-bisarylthio-juglones and 2, 3-bisarylthio-5, 8-dimethoxy-1, 4-naphthoquinones were newly systhesized for the evaluation of antifungal activities. These derivatives were prepared by methylation of juglone and 2, 3-dichloro-5, 8-dihydroxy-1, 4-naphthoquinone. and by resioselective nucleophilic subsitution with arylthiols. All compounds were tested in vitro for their growth inhibitory activities against pathogenic fungi by the standard method. The MIC values were determined by comparison to flucytosine as a fungicidal standard agent. In general. In general. most juglone derivatives shows in vitro antifungal activities. Among them. 2-arylthio-5-methoxy-juglones showed most potent antifungal activities against all pathogenic fungi.

Effects of Compost Tea Making from Differently Treated Compost on Plant Disease Control

  • Antifungal activity of compost teas was evaluated in vitro and in vivo experiments. In vitro test, fourteen compost teas were produced using five different composts and through six different methods for compost tea production. Eleven pathogenic fungi were used as indicators of antifungal activity of compost teas. In vivo test, one of the compost teas used in vitro test was sprayed to cucumber leaves which were infected with powdery mildew in order to evaluate antifungal activity of compost tea. From the results of the tests, it was found that all compost teas used this study showed antifungal activity and the proper application of compost teas against pathogenic fungi for obtaining optimal effects was important to know.

  • A lichen-forming fungus was successfully isolated by discharged spore method from Korean lichen (Hetero-dermia sp.) and cultivated in pure culture. The isolate JR0012 inhibited mycelial growth of several plant-pathogenic fungi. Mycelial growth of the four Pythium spp. tested was completely inhibited. Potato dextrose broth was found to be the medium favorable for large-scale production of antibiotics from the isolate. Anti-fungal substances produced in axenic culture were partially purified. This is the first report in Korea of lichen-forming fungus successfully isolated and which exhibited strong antifungal activity against plant-pathogenic fungi, especially the four Pythium spp..

Lycorine: A Potential Broad-Spectrum Agent Against Crop Pathogenic Fungi

  • A screening test showed that lycorine exhibited significant antifungal activity against 24 pathogenic crop fungi at concentrations of 500 ${\mu}g/ml$ and 100 ${\mu}g/ml$, respectively. Fusarium graminearum was selected for antifungal mechanism studies by observing its mycelial morphology and investigating the variations in its conductivity. In addition, the substance absorption and metabolism of F. graminearum were explored. The mechanism was revealed as being one by which lycorine destroyed the cellular membrane and further influenced substance absorption and cell metabolism.

Paenibacillus polymyxa and Burkholderia cepacia Antagonize Ginseng Root Rot Pathogens

  • To isolate rhizobacteria exhibiting antifungal activities for for five pathogenic fungi (Sclerotinia sclerotiorum, Fusarium solani, Collectotricum gloeosporides, Fusarium oxysporum, and Botrytis cinerea) which cause damage to Ginseng root in Ginseng grown fields, four soils were collected from Cheorlwon gun, in Korea. From 4 soils, a total of 160 bacterial strains were isolated by dilution plate method. Among 160 strains, 32 strains showed antifungal activities for one or more pathogens. From 32 strains, three strains exhibited antifungal activities for all pathogens. These are two Burkholderia cepacia (ATCC 25416 and ET 13) and one Paenibacillus polymyxa (ATCC 842). These potent antifungal strains showed high identities (99% using 16S-rRNA sequencing).

  • 2.5-Disubstituted-6-arylamino-4.7-benzimidazolediones were synthesized and tested for in vitro antifungal activities against pathogenic fungi. The 2-aryl-6-arylamino-5-chloro-4.7-benzimidazolediones were prepared by nucleophilic substitution on 2-Aryl-5.6-dichloro-4.7-benzimidazolediones with appropriate arylamines in good yields. TIte synthesized 4.7-benzimidazolediones were tested in vitro for their growth inhibitory activities against pathogenic fungi by the standard method. The MIC values were determined by comparison to llucytosine as a fungicidal standard agent. The most active potential among the 4.7-benzimldazoledione series was found for 6-arylamino-2-(2-pyridyl)-4.7-benzimidazolediones. which showed generally good activities against all tested Candida apecies and A. niger.
