The mouse bioassay and high performance liquid chromatography (HPLC) post-column oxidation method are different methods of quantifying paralytic shellfish poisoning toxins. In this study, we compared their ability to accurately quantify the toxicity levels in two types of field sample (oysters and mussels) with different toxin profiles for routine regulatory monitoring. A total of 72 samples were analyzed by both methods, 44 of which gave negative results, with readings under the limit of detection of the mouse bioassay ($40{\mu}g/100g$ saxitoxin [STX] eq). In 14 oysters, the major toxin components were gonyautoxin (GTX) 1, -2, -3, -4, -5, decarbamoylgonyautoxin-2 (dcGTX2), and decarbamoylsaxitoxin (dcSTX), while 14 mussels tested positive for dcSTX, GTX2, -3, -4, -5, dcGTX2, neosaxitoxin (NEO), STX, and dcSTX. When the results obtained by both methods were compared in two matrices, a better correlation ($r^2=0.9478$) was obtained for mussels than for oysters ($r^2=0.8244$). Additional studies are therefore needed in oysters to investigate the differences in the results obtained by both methods. Importantly, some samples with toxin levels around the legal limit gave inconsistent results using HPLC-based techniques, which could have a strong economic impact due to enforced harvest area closure. It should therefore be determined if all paralytic shellfish poisoning toxins can be quantified accurately by HPLC, and if the uncertainties of the method lead to doubts regarding regulatory limits.
마비성 패류 독소(Paralytic shellfish poisoning, PSP)는 유해 조류에 의해 생성되며, 독소에 노출된 수산물을 섭취하였을 때 중독이 발생한다. 수산물 중 PSP를 검출하는 표준 시험법인 Mouse bioassay (MBA)는 낮은 검출한계와 동물 윤리 문제로 대체 시험법의 개발 필요성이 대두되고 있다. 이러한 대체 시험법 중, PSP가 신경 세포막의 Na+ 채널을 차단하는 기전을 이용한 마우스 뇌신경 모세포종 세포 기반 시험법(Neuro-2a assay)의 표준화를 위한 노력이 대두되고 있다. Neuro-2a assay의 원리는 Neuro-2a 세포주에 Na+/K+ ATPase 억제제인 Ouabain(O)과 Na+ 채널 활성화제인 Veratridine (V)을 처리하여 과도한 Na+ 유입으로 인한 세포사멸을 유도한 상태에서, Na+ 채널 억제제인 PSP를 처리하게 되면 Na+ 유입이 차단되어 세포가 생존하는 것을 측정하는 것이다. 본 연구에서는 PSP 검출을 위한 Neuro-2a assay를 국내 연구 환경에 맞게 다양한 매개변수를 개선하여 최적 시험법을 확립하고자 하였다. 고려한 매개변수들은 세포밀도, 배양 조건 및 PSP 처리 조건 등으로, 그 결과는 아래와 같다. 초기 세포밀도는 40,000 cells/well로, 세포 배양시간 및 처리시간은 각각 24시간으로 설정하였다. 또한 최적 O/V 농도는 500/50 μM로 설정하였다. 본 연구에서 PSP 중 Saxitoxin (STX)에 대해서 O/V 처리가 된 상태에서 S자형 용량-반응 그래프가 도출되는 8가지 농도(368~47,056 fg/μl)를 확인하였고, Neuro-2a assay의 실험실 간 변동성 비교를 통해, 실험의 적정성 확인을 위한 5가지 Quality Control Criteria와 실험 데이터의 신뢰가능 범위(Data Criteria) 6가지를 설정하였다. 확 립된 조건으로 Neuro-2a assay를 진행한 결과 반수영향농도(EC50) 값은 약 1,800~3,500 fg/μl로 나타났다. 실험실 간 변동성 비교 결과, Quality Control Criteria 값 및 Data criteria 값의 변동계수(coefficients of variation (CVs))가 1.98~29.15% 범위로 산출되어 실험의 적정성 및 재현성이 확인되었다. 본 연구를 통해 우리나라에서 활용할 수 있는 PSP 검출용 Neuro-2a assay 시험법의 최적 조건 및 5가지 Quality control 기준을 제시하였고, PSP 중 대표적인 독소인 STX을 대상으로 Neuro-2a assay를 실시한 결과 유의한 EC50 값을 산출할 수 있었으며, 향후 국내 수산물을 대상으로 MBA를 대체할 수 있는 PSP 검출법으로 활용될 것으로 기대된다.
1993년 진해만 일원의 마비성패독 발생의 원인규명을 위한 모니터링의 일환으로 원인생물의 독생산과 성분조성을 조사하기 위하여 양식장 인근해역의 저서 휴면포자를 발아시켜 분리한 무균주의 독성분을 분석하였다. 분리된 전체주중에서 수정리산 (St. 1) 5주, 욱곡리산 (St. 2) 3주 및 대곡리산 (St. 4) 11주의 독조성 및 독함량을 비교하였을 때, 각 지점별 평균 독량은 약 54-70 fmol/cell 높은 량을 나타내었고, 동일 휴면포자의 clone 분리주 뿐만 아니라 전체 분리주에서 개체별 독함량의 차이가 크게 나타났다. 독조성은 C1/C2 (epiGTX8/GTX8), GTX1/GTX4 및 neoSTX가 주요 구성성분을 이루었고, GTX2/GTX3, GTX5, C4, dcSTX 및 STX성분은 미량 또는 산발적으로 출현하였다. 주요 성분중에서 neoSTX는 $5-54mol\%$로 변동이 컸으나, 전체 분리주의 절반은 출현을 보이지 않아 이 지역에서 조성이 다른 개체군의 출현이 시사되었다 한편, 상대적으로 독성이 강한 GTX1-4 및 neoSTX와 같은 Carbamate군의 성분이 세 정점에서 각각 $57\%,\;54\%$ 및 $67\%$의 높은 평균치를 나타내어 이 지역에서의 높은 독화율과 독화 가능성의 잠재력이 큰 것으로 시사되었다.
마비성 패류독(Paralytic Shellfish Poison, PSP)에 의하여 독화된 패류의 유효이용에 대한 자료를 제공하고자 독화된 진주담치의 중장선 균질육, 0.1N HCI로 추출한 조독소 용액, 중장선으로부터 정제한 gonyautoxin(GTX) group 과 saxitoxin(STX) group 등 4가지 형태의 PSP 독소에 대한 내열성을 조사하였다. 독화된 진주담치의 중장선육 균지액, 산추출 조독소액, GTX group, STX group, STX group 등 4종류의 반응속도 상수는 $120^{\circ}C$에서 $3.28{\times}10^{-2},\;1.20{\times}10^{-2},\;5.88{\times}10^{-2},\;2.58{\times}10^{-2}$이었으며 4종류의 독소 중 0.1 N HCl로 추출한 조독소용액의 D-value가 가장 높았다. 반응속도 상수를 이용한 살균 온도 산정에 있어서, 최초 독력이 $200\;\mu\textrm{g}/100g$인 독화된 진주 담치육의 경우, 독력을 마비성 패류독 규제치인 $80\;\mu\textrm{g}/100g$으로 감소시키는데는 $90^{\circ}C$에서는 약 129분, $100^{\circ}C$에서는 약 82분, $110^{\circ}C$에서는 약 48분, $120^{\circ}C$에서는 약 28분 걸렸다. 이러한 결과는 최초 독력이 $200\;\mu\textrm{g}/100g$인 패류의 경우, 통조림 살균공정 후 잔존 독력이 규제치인 $80\;\mu\textrm{g}/100g$ 이하로 감소시키는 데에는 현재의 살균조건($115^{\circ}C$, 70분) 으로는 충분하다는 것을 입증한다.
마비성 패독(paralytic shellfish poisoning, PSP)에 의한 중독은 와편모조류(Dinoflagellates)가 생성하는 saxitoxin (STX)이 이매패류 등의 먹이활동에 의해 축적되고 이를 사람이 섭취함으로써 발생한다. 최근 분석기술의 발전으로 와편모조류가 STX외에도 gonyautoxin (GTX) group 및 N-sulfo carbamoyl toxins (C toxin) group 등 다양한 유사체들을 생성하는 것으로 보고되고 있다. 이에 CODEX, EFSA에서는 STX외 유사체의 안전관리를 위해 STX 및 유사체를 STX group으로 관리하고자 하는 움직임을 보이고 있다. 국내의 경우도 STX 유사체를 생성하는 조류의 발생이 이미 보고되고 있으며 실제 홍합에서 유사체의 오염사례도 소수 보고되고 있다. 따라서 국제적인 움직임에 발맞추어 국내에서도 STX 및 유사체의 group 관리를 위한 준비가 필요할 것으로 사료된다. 본 연구에서는 STX 및 유사체의 체계적인 모니터링 및 안전관리의 기반을 마련하고자 STX 및 유사체의 이화학적 특성, 생성조류, 국내외 발생현황, 독성 및 상대독성계수, 분석법, 오염현황 및 관리현황에 대한 폭넓은 검토를 수행하고자 하였다.
In Gamak Bay, Paralytic Shellfish Poisoning (PSP) was first detected from seafoods in 2003, however the toxin source is unknown yet. In this study, we report potential PSP producers of toxic dinoflagellates, describing morphology and abundance of cysts isolated from surface sediment of Gamak Bay. The most abundant type in these cysts was characterized with ellipsoidal and transparent wall identical to Alexandrium catenella and/or A. tamarense. Germination experiment of the cysts revealed that all motile cells germinated were morphologically identified as A. tamarense. This result suggests that A. tamarense may relate to PSP contaminations in Gamak Bay. Moreover, bottom water temperature in Gamak Bay is favorable for germination of A. tamarense cysts. Further studies are required to carry out the PSP monitoring for preventing the risk of PSP events that may outbreak in future at Gamak Bay.
$Na^+$ 전극도 개구리 방광막으로 만들어진 조직센서를 이용하여 복어독 (TTX), 조개독 (STX), 마비성조개독 (PSP) 등의 $Na^{+}$ 챈널 차단물질을 측정하였다 본 연구는 연속적으로 $Na^{+}$ 챈널 차단물질을 측정할 수 있는 조직 센서를 이용하여 한국산 한방약의 재료와 건조 및 생김 (Porphyra yezoensis)의 $Na^+$ 챈널 차단물질의 존재여부를 측정하였다. 본 센서는 한방약 재료 및 해조류 (김 등)와 같이 현재 일반적으로 사용되고 있는 마우스법으로 측정이 불가능한 fg 단위의 매우 적은 양의 $Na^{+}$ 챈널 차단물질까지 측정이 가능하였다. 본 바이오센서는 $Na^{+}$ 챈널 차단물질의 모니터링 및 해양환경감시에 응용 가능하리라 기대된다.
The annually outbreak of paralytic shellfish poisoning (PSP) were caused by toxic dinolagellate A. tamarense and A. catenella in Korea. The purpose of this study were to investigate the distribution of PSP-causative organisms, A. tamarense and A. catenella and their species classification. Sediment (Saemangeum, the south open sea) and water samples (southeastern coast) were sampled to establish clonal isolates in 2003. After isolation and purification, strains were cultured under $17^{\circ}C$, f/2 media, 14:10=L:D cycle. PST analysis and species identification were performed by HPLC-FD method and specific DNA probe, respectively. Thirty-ons strains were isolated from the Saemangeum reclamation, southeastern coast including Jinhae Bay and south open sea. PSTs were detected in all cultured strains. In eight strains from south offshore, major toxin components are GTX5, C1/2 and minors are GTX3/4, dcGTX3, neoSTX. Sixteen strains from south coastal area have GTX1/4, neoSTX, C1/2 as major toxin components and GTX2/3 as minors. Seven strains from the Saemangeum reclamation have GTX5, C1/2 as major toxin components and GTX1/2/3/4 as minors. Thus, among eight south offshore isolates, four A. tamarense have more toxic (38.31~l19.16 fmol.$cell^{-1}$) than A. catenella (3.78~13.13 fmol.$cell^{-1}$). With the previous results of different toxin composition, toxin components and toxin contents, .it is toxin profile that could used to diagnosis of regional toxic population and geographical distribution of both A. tamarense and A. catenella and their toxigenic strains.

The dinoflagellate Alexandrium tamiyavanichii Balech, a producer of toxins causing paralytic shellfish poisoning (PSP), has recently been considered as one of main organisms responsible for toxication of shellfish in Japan. In this study, A. tamiyavanichii was subjected to a molecular phylogenetic analysis inferred from 28S rDNA D1-D2 sequences and a species-specific LSU rRNA-targeted oligonucleotide DNA probe was designed to identify A. tamiyavanichii using the whole cell-FISH (fluorescence in situ hybridization). The sequences of the 28S rDNA D1-D2 region of A. tamiyavanichii showed no difference from A. cohorticular AF1746l4 (present name A. tamiyavanichii) and formed a distinct clade from the 'tamarensis species complex'. The probe, TAMID2, reacted specifically with A. tamiyavanichii cultured cells, without any cross-reaction with other species belonging to the same genus, including A. tamarense, A. catenella, A. affine, A. fraterculus, A. insuetum and A. pseudogonyaulax. In a test of cross-reactivity with a field sample, TAMID2 reacted consistently with only A. tamiyavanichii, indicating that the present protocol involving the TAMID2 probe might be useful for detecting toxic A. tamiyavanichii in a simple and rapid manner.

The marine dinoflagellate genus Alexandrium has been recognized as the most representative toxic phytoplankton on account of production of paralytic shellfish poisoning (PSP) throughout the world. PSP producers, generally A, tamarense and A. catenella, within the genus Alexandrium have caused high level intoxicauon of fisheries products and even death of human. In addition, more recent increasing of geographical range of this deleterious species has given rise to alarming tension. The study presented here aimed construction of the molecular phylogenetic relationships through sequences-determination from 16 morphotypic species (containing newly sequenced 3 morphotypic species, A. tamiyavainchii, A. fraterculus and A. pseudogonyaulax) in LSU rDNA D1-D2 and 12 morphotypic species (containing newly sequenced 6 - morphotypic species, A. catenella, A. tamiyavanichii, A. fraterculus, A. affine, A. insuetum and A. pseudogonyaulax) in SSU rDNA region, and the sequences were subjected to comparative-analysis in respect to regional population using functionally expressed rDNA genus and pseudogenes. And we discussed on genetic differentiation between A. tamarense and A. catenella together with putative PSP divegence of the genus Alexandrium. The results of phylogenetic analysis showed the robust monophyletic 14 distinct classes of A. tamarense, A. excavatum, A. catenella, Tasmanian A. tamarense, A. affine (and/or A. concavum), Thai A. tamarense, A. tamiyavanichii, A. fraterculus, A. margalefii, A. andersonii, A. ostenfeldii, A. minutum (and/or A. lusitanicum), A. insuetum, and A, pseudogonyaulx clade. A. fraterculus and A. tamiyavanichii were sister relationship and they were positioned independently between A, affine cluster and those of A. margalefi, A. andersonii, A. ostenfeldii, A. minutum and A. insuetum. A. pseudogonyaulax appeared to be an ancestral taxon among Alexandrium.
