AN IMMUNOHISTOCHEMICAL LOCALIZATION OF TENASCIN IN PERIODONTAL POCKET TISSUES (치주낭 조직내 tenascin의 분포에 관한 면역조직화학적 연구)

  • To determine the effect of tenascin on forming periodontal pocket and pseudopocket, the ginival tissues were surgically obtained from the patients with adult periodontitis(10) and non-inflammatory phenytoin-associated gingival hyperplasia(5). The excised tissue specimens were fixed in neutral formalin for $6{\sim}24$ hours, embedded with paraffin, sectioned at 4-6m in thickness, mounted on glass slides coated with 3-aminopropyltriethoxysilane(Sigma Chemical Co., St. Louis, MO, U.SA.) and immunohistochemically processed by Avidin-Biotin peroxidase complex method for the localization of tenascin, using monoclonal mouse anti-human tenascin antiboday(Chemicon-International Inc., Temecula, CA, U.S.A., 1: 5,000) as the primary antibody. Regardless of periodontal pocket and pseudopocket, tenascin was localized along the connective tissue subjacent to basement membrane of gingival epithelium, and strong positive reactivity was obviously noted in the papillary projections of gingival connective tissue. The results suggest that tenascin may affect the development of papillary projections and the proliferation of epithelial cells.

Investigating the Frequency of the ERCC1 Gene C8092A Polymorphism in Iranian Patients with Advanced Gastric Cancer

  • Background: Platinum compounds are the main drugs for treatment of advanced gastric cancer. Previous studies have shown that clinical outcome with platinum-based compounds depends on ERCC1 polymorphisms. The aim of this study was to investigate the frequency of a common polymorphism of ERCC1 gene (C8092A) in Iranian patients with advanced gastric cancer receiving platinum chemotherapy. Materials and Methods: Genetic analysis of the ERCC1 C8092A polymorphism was performed by the PCR - RFLP method using 50 paraffin-embedded tissue specimens. Results: Of the 50 cases, 32% of individuals showed CC genotype, 24% of them had CA genotype and 44% of patients had AA genotype. Conclusions: Based on the results, using of platinum-based chemotherapy would be expected to be specifically beneficial in only 32% of patients.

Expression of Glypican-3 is Highly Associated with Pediatric Hepatoblastoma: a Systemic Analysis

  • Objective: Glypican-3 (GPC3) is reported to be an oncofetal protein that is a useful diagnostic immunomarker for hepatoblastoma. However, the results are not inclusive. This study systemically investigated the association between expression of GPC3 and pediatric hepatoblastoma. Methods: Clinical studies evaluating the association were identified using a predefined search strategy. GPC3 immunohistochemistry was applied in the pathological diagnosis of hepatoblastoma using the monoclonal antibodies with formalin-fixed and paraffin-embedded specimens. Positive predictive rates for the association between expression of GPC3 and pediatric hepatoblastoma were calculated. Results: Specimens from four clinical studies which including 134 patients with pediatric hepatoblastoma tested by GPC3 immunohistochemistry were considered eligible for inclusion. Systemic analysis showed that, in all patients, pooled positive predictive rate of the association between expression of GPC3 and pediatric hepatoblastoma was 95.5% (128/134). Conclusion: This systemic analysis suggests that the expression of glypican-3 is highly associated with the diagnosis of pediatric hepatoblastoma.

Significance of Expression of Human METCAM/MUC18 in Nasopharyngeal Carcinomas and Metastatic Lesions

  • Human METCAM/MUC18, a cell adhesion molecule (CAM) in the immunoglobulin-like gene super family, plays a dual role in the progression of several epithelium cancers; however, its role in the nasopharyngeal carcinoma (NPC) remains unclear. To initiate the study we determined human METCAM/MUC18 expression in tissue samples of normal nasopharynx (NP), NPCs, and metastatic lesions, and in two established NPC cell lines. Immunoblotting analysis was used for the determination in lysates of frozen tissues, and immunohistochemistry (IHC) for expression in formalin-fixed, paraffin-embedded tissue sections of 7 normal nasopharynx specimens, 94 NPC tissue specimens, and 3 metastatic lesions. Human METCAM/MUC18 was expressed in 100% of the normal NP, not expressed in 73% of NPC specimens (or expressed at very low levels in only about 27% of NPC specimens), and expressed again in all of the metastatic lesions. The level of human METCAM/MUC18 expression in NPC tissues was about one fifth of that in the normal NP and metastatic lesions. The low level of human METCAM/MUC18 expression in NPC specimens was confirmed by a weak signal of RT-PCR amplification of the mRNA. Low expression levels of human METCAM/MUC18 in NPC tissues were also reflected in the seven established NPC cell lines. These findings provided the first evidence that diminished expression of human METCAM/MUC18 is an indicator for the emergence of NPC, but increased expression then occurs with metastatic progression, suggesting that huMETCAM/MUC18, perhaps similar to TGF-${\beta}$, may be a tumor suppressor, but a metastasis promoter for NPC.

The effect of the Ca-P coated DBBP on osseous regeneration in the rat calvarial bone defect (백서 두개골 결손부에서 Ca-P 피복된 이종골의 골재생 효과)

  • Purpose: This study was aimed to evaluate the effect of the deproteinated bovine bone powder (DBBP) coated with calcium phosphate (Ca-P) on osseous regeneration in the calvarial bone defect of rat. Materials and Methods : The DBBP (Control group, n=6) and the Ca-P coated DBBP (Experimental group, n=6) were grafted in the critical sized calvarial bone defect (8 mm) of rat weighing 250 g. The animals were sacrificed at 1, 4 week. The biopsy specimens were decalcified with 5%formaldehyde and embedded in paraffin. The rats were sacrificed at 8 week received tetracycline (1 week), calcein blue (4 week), and alizarin red (7 week), and the biopsy specimens were taken. The specimens were embedded in methylmethacrylate and ground to 10 ${\mu}m$ thin sections were made. All of the specimens were stained with H & E and Masson's trichrome and examined under light microscope. The specimens at 8 week were examined under fluorescent microscope. Results : In the Control group, the grafted DBBP was surrounded with connective tissue, and osteoblasts were observed partially around the grafted particles at 1 week. At 4 week, some osteoid was observed and, new bone formation was observed at the periphery of grafted materials at 8 week, In the Experimental group, some osteoid was seen at the periphery of the grafted Ca-P coated DBBP at 1 week, and osteoblast and newly formed bone were observed around the grafted materials. At 8 week, newly formed bone was observed at the periphery of the grafted materials. Conclusion: These results suggest that Ca-P coated DBBP group was more and faster than DBBP group in new bone formation and Ca-P could contribute to enhance bone formation in the critical sized calvarial bone defect of rat.

The Anatomical Identification-key of Ephedrae Herba and its adulterant (마황(麻黃)과 그 위품의 내부형태감별)

  • Objectives : Ephedra regeliana, although similar to the other E. sinica, E. intermedia, and E. equisetina, is not authorized by The Korean Pharmacopoeia and yet has also been imported under the name of 'Ma-Hwang'. The aim of this study was to perform a comparative anatomical evaluation of Ephedrae Sinicae Herba (ESH), Ephedrae Intermedicae Herba (EIH), Ephedrae Equisetinae Herba (EEH) and Ephedrae Regelianae Herba (ERH). Methods : Permanent paraffin-embedded specimens were produced using the paraffin-tertiary butyl alcohol (TBA) method, to visualize their anatomical features via light microscopy. Results : ESH and ERH transverse sections were in the shape of a long ellipse, while those of EIH and EEH were in the shape of a circle. These anatomical characteristics substantiated the results of external morphological examination. The sections revealed that each of the four samples showed significantly different cortexes. The cortex of ESH was wider than two thirds of the section, unlike that of the other samples which was relatively narrow. The xylems of ERH, EIH, and EEH demonstrated interconnectivity with other adjacent xylems, contrary to those in ESH. Therefore, the characteristic development of the xylems over time was used to distinguish ERH from the other. Furthermore, parenchymal morphology in the central cylinder was also a good criterion to differentiate between EIH and EEH, since EIH demonstrated parenchymal cells that were shaped like a rounded-triangle, whereas EEH parenchymal cells were chrysanthemum-shaped. Conclusions : The identification-keys established in this study would facilitate the accurate identification of microscopic features of the four varieties of 'Ma-Hwang'.

Lack of Association between Herpes Simplex Virus Type 2 Infection and Cervical Cancer - Taq Man Realtime PCR Assay Findings

  • Background: About one third of the human population suffer cancer during their lifetime and more than 20% of total morbidity is related to neoplasia. Cervical cancer is generally the most common cancer in developing countries and the second most common in women globally. The role of human papilloma viruses viruses in its induction is clear. However, the involvement of hepres simplex virus type 2 (HSV-2) is controversial. Therefore a survey was conducted of the prevalence of HSV-2 in patients with cervical cancer and also healthy people with sensitive and quantitative Taq Man real-time PCR assay. Materials and methods: Seventy six formaldehyde fixed paraffin embedded tissue specimens from patients with histologically proven history of cervical cancer as well as 150 control blocks were sectioned for deparaffinization and DNA extraction. Results: There was no HSV-2 DNA in our patient specimens but four control samples were positive, all with a history of hysterectomy. Conclusion: Considering the absence of any positive viral HSV-2 DNA in our patients and also the presence of four positive specimens among our controls, we did not find any relationship between the presence of HSV-2 DNA and cervical cancer.

IMMUNOHISTOCHEMICAL DETECTION OF GROWTH FACTORS AND EXTRACELLULAR MATRIX PROTEINS IN THE DEGENERATING TISSUES OF PRE-AND POSTNATAL HUMAN CLEFT LIP AND PALATE (태생 및 생후 구순.구개열에 나타나는 조직변성에 대한 성장인자와 세포외 기질 단백의 면역조직화학적 연구)

  • In order to elucidate the pathogenesis of cleft lip and palate, first of all, it is necessary to understand the developmental mechanisms of growth factors and extracellular matrix proteins in the tissues of cleft lip and palate. We have performed immunohistochemical studies on human cleft lip and palate tissues to elucidate the pathogenetic implications of cleft lip and palate. 16 specimens from postnatal human cleft lip and palate subjects and 17 specimens from autopsy of prenatal human cleft lip and palate were fixed in 10% buffered formalin, embedded in paraffin. The sections were routinely stained by hematoxylin and eosin, also stained by PAS, and followed by immunohistochemical stainings using the antiseras of growth factors and extracellular matrix proteins such as PCNA, S-100, c-erb-B2, MMP-3, MMP-10, HSP-70, transglutaninase-C, E-cadherin, VEGF, vWF. Both the prenatal and postnatal specimens of cleft lip and palate showed dysplastic proliferation of the basal cell layer, increased infiltration of melanocytes into mucosal epithelium, sebaceous gland hyperplasia ingrowing into the muscular tissue of lip and palate, and fatty infiltration into the submucosal deep connective tissue. The strong reactions of MMP-3 and HSP-70 were detected in the tissues of cleft lip and palate, especially increased in degenerating muscle bundles, while the immunostainings of PCNA and c-erb-B2 were weakly positive in the tissues of cleft lip and palate. These data suggest that the retrogressive tissue degeneration around the cleft areas persistently exist during the prenatal and postnatal period after cleft formation, and the sebaceous gland hyperplasia and fatty infiltration with the intense expression of MMP-3 and HSP-70 is closely related to the muscular degeneration around the cleft area.

A HISTOLOGICAL STUDY OF THE ROOT RESORPTION APPLYING TO INTERMITTENT AND CONTINUOUS FORCE FOR INCISOR INTRUSION OF DOG (성견의 전치부 INTRUSION시 간헐적 교정력과 지속적 교정력의 적용에 따른 치근 흡수에 관한 실험적 연구)

  • The purpose of this study was to investigate the root resorption pattern in incisors in dog under intrusive orthodontic loadings of various magnitude and duration. Intrusive forces were generated by closed coil springs. Force magnitudes were 15-30gm, 50-60gm and 80-110gm. Durations were continuous and intermittent. Intermittent duration was applied at intervals of 12 hours. The readjustment of the force was done every 4 days. The forces were maintained for 30 days. All specimens were decalfied, embedded in paraffin and stained with hematoxylin-eosin stain. Observations were made with light microscope. The following results were obtained; 1. The continuity of root surface was ceased in all, except intermittent forced teeth with 25gm. 2. The root resorptions, cementoclasts and cemental lacunae, were increased around periapical regions and the destructive scope of bone was deeply extended as the exerted orthodontic forces increased. 3. It was inspected that, under the same forces, root resorption and bone destruction were more deeply appeared in the continuously forced teeth than the intermittently forced ones. 4. All of the alveolar bone, showed direct and undermining bone resorptions.

HISTOPATHOLOGICAL EVALUATION OF PULP CAPPING AGENTS ON RATS MOLARS (치수복조제가 백서치수조직에 미치는 영향에 대한 병리조직학적 연구)

  • The purpose of this investigation was to study histopathological chronology and differences of the proprietary pulpcapping agents. One hundred eighty molars from thrity rats (Srague-Dawley species), weighing about 130gm, were divided into six groups. Cavities were prepared in their maxillary molars under intraperitoneal anesthesia with Secobarbital. The cavities in the right first and second molars were filled with Dycal$^{(R)}$ and the left ones were with Cavitec$^{(R)}$. Each group of rats were sacrificed at the intervals of 1, 3, 5, 7, 14 and 21 days following operation. The rats were decapituated, and the jaws were fixed in 10% neutral buffered formalin. Then the specimens were decalcified, embedded in paraffin or celloid, and sectioned at 6-8 ${\mu}$ in thickness through the cavities included and pulp mesiodistally. They were stained with Hematoxylin-Eosin and examined by lightmicroscope. The results were as follow: 1. The pattern of pulp healing was dependent upon the presence and character of the pulpcapping agents above. 2. Dentin bridge formation as a sign of pulp healing occurred in the 14 days after operation. 3. Dycal$^{(R)}$ reparation appeared to favor pulp bealing rather than Cavitec$^{(R)}$ preparation. 4. In the odontoblastic layer and pulp tissue specific vaculoes were showed at the 3, 5 and 7 days of the Dycal filling.
