Production of Glutathione by Candida sp. Mutant (Candida sp. 변이주에 의한 Glutathione 생산)

  • For the overproduction of glutathione, Candida sp. mutant was isolated by the treatment with U.V. light. The highest glutathione production of Candida sp. mutant was obtained after shaking culture for 48 hours in the cullture medium containing glucose 1.5%(w/v), yeast extract 4.0% (w/v), KH2PO4 0.04%(w/v), biotin 5 ng/ml, and L-cysteine 0.04%(w/v). The optimal pH and temperature for the glutathione production were pH 6.0 and 25C, respectively.

  • With the increase of the use of antibiotics and invasive procedures, infections caused by multidrug-resistant Acinetobacter baumannii(MRAB) are increasing. We screened the antibiotic producing strain B-51 for antibacterial activity against MRAB from the soils and studied the effects of culture medium on the antibiotic production of B-51. The medium conditions for maximum antibiotic productivity of B-51 was 2% glycerol, 0.5% soybean meal, 0.01% $CaCl_2$, 0.01% $MgSO_4{\cdot}7H_2O$ and 0.01% $KH_2PO_4$ at an initial pH of 6.0, at $30^{\circ}C$ for 76 h.

  • Distribution and in vitro growth characteristics of entomopathogenic Paecilomyces species were studied based on the specimens and isolates deposited in Entomopathogenic Fungal Culture Collection (EFCC), Kangwon National University, Chuncheon, Korea. Paecilomyces species were frequently collected from different mountains and valleys and were almost distributed uniformly. Collection period of Paecilomyces species ranged from July to September, with most of the collections at the end of September. P. tenuipes was mostly collected one among different Paecilomyces species. The optimum medium for mycelial growth of P. tenuipes was PDA, whereas in other media such as SDAY and HMA also produced similar growth. The optimum temperature and pH levels were $25^{\circ}C$ and 6.0~7.0, respectively. Best carbon and nitrogen sources were dextrose and tryptone, respectively, while $KH_2PO_4$ was the best mineral source for mycelial growth. Highest mycelial growth was observed when the C/N ratio was 20:1.

  • Agrocybe aegerita is Hymenomycetes fungus belonging to the order Agaricales and family of Bolbitiaceae. Much less is known about liquid culture of Agrocybe aegerita. Thus, the present study was to investigate the liquid cultural characteristics of Agrocybe aegerita my-celium. The optimal medium for the mycelial growth and density was ME medium, optimal tem-perature and initial pH were 25$\pm$1$^{\circ}C$ and 5.5, respectively. And optimal culture time for mycelial growth was 12 days. The modified optimal medium compositions were dextrin 3% (w/v), yeast extract 2% (w/w), MgSO$_4$0.05% (w/v), and KH$_2$PO$_4$0.15% (w/v). Under optimal culture conditions, the mycelial growth of modified optimal medium was higher than that of ME medium.

  • This study was conducted to isolate strain for the preparation of fermented antler (Cervus cornu parvum) and evaluate its physiological activities. The growth degrees of twenty-one samples from Bacillus sp., Lactobacillius sp. and mushroom strain on antler extract agar were evaluated in this study, and Bacillus subtilis KH-15, SCB-3, Cordyceps militaris, Phellinus linteus, Inonotus obliquus 26136, and Inonotus obliquus 26147 were selected. The fermented antler extract by C. militaris had relatively higher contents of total sugar (1619.3 ${\mu}g$/mL), uronic acid (302.0 ${\mu}g$/mL), sulfated-glycosaminoglycan (S-GAGs) (119.9 ${\mu}g$/mL) and sialic acid (21.6 ${\mu}g$/mL) than any other extracts. The anti-complementary activities of all fermented antler extracts were higher than non-fermented antler extract, and among these samples, fermented antler extract by C. militaris showed the highest anti-complementary activity (inhibition of 50% total complement hemolysis, $ITCH_{50}$; 50.1% at 1,000 ${\mu}g$/mL). The ability of fermented antler extract by B. subtilis KH-15 to scavenge 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical ($IC_{50}$; 4.97 mg/mL) was significantly the highest (p<0.05), whereas the extract from I. obliquus exerted significantly (p<0.05) high 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity ($IC_{50}$; 16.98 mg/mL) among all samples. The results of this study suggest that physiological effects including immuno-modulating and antioxidant activities of the antler may be increased through fermentation process.

  • Among microorganisms isolated from soil, YJ-66 strain was the best producer of flocculant and was examined for flocculating ability in the active carbon and CaCl2. YJ-66 strain was the best producer of flocculant and was examined for flocculating ability in the active carbon and CaCl2. YJ-66 strain was identified to be a species belonging to the genus Achromobacter. The optimum culture condition for production of bioflocculant with the isolated strain was for 72hrs at 3$0^{\circ}C$ and pH7.5. The favorable carbon, nitrogen sources and inorganic salts for production of the flocculant were sucrose, peptone, MgSO4 and KH2PO4, whose optimal concentrations were 2%. 0.067%, 0.1% and 0.1%, respectively. Addition of the carbon and inorganic salts significantly increased the production of flocculant. Compositions of optimized culture medium for bioflocculant production by Achromobacter sp. YJ-66 were 2% sucrose, 0.067% peptone, 0.1% MgSO4 and 0.1% KH2PO4 in initial pH 7.5 during at 3$0^{\circ}C$ for 72hrs.

  • Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.

  • This study was performed to obtain the scientific data an artificial culture of ear mushroom(Auricularia auricula). Liquid cultural of Auricularia auricula. mycelium was characterized with respect to temperature, pH, compositions of inorganic, carbon and nitrogen. The its optimum temperature and media pH were at $25^{\circ}C$ and pH 5.0~6.0 on the mycelial growth, respectively. The optimum inorganic compositions and concentration for mycelial growth were $MgSO_4$ 0.05%(w/v) and $KH_2PO_4$ 0.1%(w/v), respectively. The optimum carbon sources and nitrogen sources were black-sugar 2%(w/v) and soybean powder at 0.3~0.5%(w/v) with7 days optimum culture time.

  • The production of xylanase and $\beta$-xylosidase was investigated in submerged culture of Aspergillus niger NRC 107. The maximum production occurred when the pH was controlled at 6.0 during the fermentation. Among the various carbon sources investigated, corn-cob xylan (1.5%, w/v) yielded maximal production of the enzymes. The $NaNO_{3}$ was the most favorable nitrogen source for enzyme production and $KH_2P0_4$ concentration at 0.3%(w/v) was found to be optimum. Incorporation of wheat bran to the culture medium improved xylanase production. Addition of L( -) sorbose to the culture medium promoted the secretion of $\beta$-xylosidase. It was possible to increase the production of xylanase (39.43 units/ml) and that of $\beta$-xylosidase (4.2 unitslml) by submerged culturing the A. niger NRC 107 in the modified medium.

  • Nutritional characteristics and conditions for mycelial yield of Pleurotus ostreatus and Auricularia auricula-judae in shaking culture were investigated. Among the sugar substances, glucose and mannitol showed the good effect for mycelial yield of P. ostreatus, and mannitol and fructose were good for the mycelial yield of A. auricula-judae. Among the various organic acids, fumaric acid were good for the mycelial yield of P. ostreatus and A. auricula-judae. Among the nitrogen sources, peptone and urea showed the good result for mycelial yield of P. ostreatus, and peptone and casamino acid were good for mycelial yield of A. auricula-judae. Among the various amino acids, asparagin and threonine showed the good result for mycelial yield of P. ostreatus, and serine and threonine were good for mycelial yield of A. auricula-judae. Among the various vitamins, folic acid and thiamine were suitable for mycelial yield of P. ostreatus, and folic acid, inositol and riboflavin were suitable for mycelial yield of A. auricula-judae. Mycelial yield of P. ostreatus and A. auricula-judae were enhanced by the addition of $MgSO_4\;and\;KH_2PO_4$ at the concentration of 0.08 and 0.2% respectively. The optimum temperature and pH for mycelial yield were from $25\;to\;30^{\circ}C$ and pH 5.5 to 6.5 in P. ostreatus, and from $25\;to\;30^{\circ}C$ and pH 6.0 to 7.0 in A. auricula-judae.
