  • Imidazoline compounds have been developed in many industrial fields(eg. cosmetic, etc.). These materials can be used in almost all pH ranges and recently its use is successively increased because of nonirritative property. Since the concern of safety has enlarged at industrial application, naturally the use of amphoteric surfactants with new special properties has more. We report general properties of chemical structures of imidazoline surfactants effect.

Efficiency of methyl-esterified eggshell membrane biomaterials for intensified microalgae harvesting

  • This study investigates the use of methyl-esterified eggshell membranes (MESM) for the harvesting of microalgae species under various conditions. Eggshell membranes were esterified with HCl to impact polycationic characteristics. After methyl esterification, the negative surface charge property of eggshell membrane was changed from negative to positive for all pH values to improve microalgae sorption capacity. The harvesting efficiency of microalgae by MESM reached 78-99% for all pH ranges evaluated. In addition, a 150 mesh particle size and $10mg\;L^{-1}$ MESM dose were found to yield up to 98% microalgae harvesting. These results indicate that the high cationic charge of MESM strongly adsorbs the negatively-charged microalgae. MESM is biocompatible and can be applied to the harvest of microalgae.

  • Adsorption capacities of toxic heavy metal ions using as-received carbon(AC), single and double surface-modified activated carbon(OAC and DSMC) in wide pH ranges are extensively evaluated. Physical and chemical properties of surface-modified activated carbons are evaluated through BET analysis, surface acidity and oxides measurements. Based oil tile adsorption isotherms of Pb, Cd and Cr ions by AC, OAC and DSMC, the adsorption amount on DSMC was obviously higher than that on the other carbons. Breakthrough behaviors of ternary metal ions in a column packed with three kinds of carbon were also characterized with respect to the variations of the influent pH and concentration. The adsorption capacity of DSMC in a fixed bed stood a favorable comparison with that of as-received carbon.

  • $\alpha$-Galactosidase was partially purified from the culture filtrate of pichia guilliermondii by Mannobiose-Sepharose affinity column chromatography. The galactosidase exhibited maximum activity at pH 4.5 and 4$0^{\circ}C$, and was stable in the pH and temperature ranges of 4 to 5.5 and 30 to 6$0^{\circ}C$, respectively. The enzyme was inhibited by $Hg^{2+}$ and $Ag^{2+}$. The enzyme activity was ot affected considerably by treatment with other metal compounds. The enzyme hydrolyzed melibiose to galactose and glucose, raffinose to galactose and sucrose, and $Gal^{3}Man_{3}(6^{3}-$\alpha$-galactosyl-1,4-mannotriose)$ to galactose and mannotriose. On the contrary, it could not hydrolyze $Gal^{3}Man_{4}(6^{3}-galactosyl-1,4-$\alpha$-mannotetraose)$.

  • Chitosan is a natural polyelectrolytic compound. Researches of adsorption capacity using chitosan have been doing actively. We prepared bead type gel, simple modifier of chitosan, And then experimented adsorption test of heavy metals (Cd etc) using it. According to the result adsorption capacity of chitosan bead was five times higher than chitosan powder. Removal rate of cadmium resulted 90% over in the test that initial concentration of Cd was 100mg/L and bead dosage was 6g/100mL. Adsorption type of heavy metals was similar to general adsorption curve. And optical pH range was 4 - 10 in the adsorption test. In the experiments of other heavy metals (Pb, Zn, Cu, Mn) adsorption types had two stages, highly removal rate-stage at the short time (20minutes) and then slow rate-stage at the after. And removal efficiency at the variable pH ranges revealed relatively good.

Phytase Properties from Bifidobacterium animalis

  • Phytase activity was examined with various bifidobacterial strains cultured statically in MRS broth at $37^{\circ}C$ for 48 hr. Seven Bifidobacterium species showed mostly an intracellular phytase activity, though their specific activities were very low. The highest specific activity was found in Bifidobacterium animalis B33 strain, among 7 bifidobacteria tested. The specific activity was highest during the exponential growth phase. Carbohydrates and the concentration of phosphorus sources had an effect on the phytase activity and bacterial growth. Glucose was the most favorable carbohydrate for the phytase activity. Phytate inhibited the cell growth, and phytase activity decreased with increase of phytate concentration. The phytase activity was even higher in the static microaerophilic growth than that in anaerobic state, despite the stimulated growth in anaerobic growth. The optimal pH ranges were comparatively broad, but the optimal temperatures were $50^{\circ}C$ for all tested strains. The phytase activity was most active at pH 6.5 and $50^{\circ}C$ for B. animalis B33 strain.

  • Kimchi power made by kimchi fermentation and freeze-drying was added to Western foods of sauce, soup, pizza, cracker and bread. Flavor qualities of the processed fusionfoods were evaluated by analyzing volatile compounds and sensory evaluation. The optimum condition of kimchi fermented for the best flavor quality of freeze-drying was the ranges of pH4.0${\sim}$pH4.7 and acidity 0.5${\sim}$0.8. The number of volatile compounds identified from extracts of the freeze-dried kimchi powder was 24, which contained 7 alcohols, 5 esters, 5 acids, 3 sulfur-containing compounds, 2 aldehydes, 1 oxygen-containing and 1 other. Among the identified compounds, the most abundant compound was eugenol(39.40%), followed as the order of dodecanoic acid(4.91%), acetic acid(1.70%), methyl 2-propenyl disulfide(1.52%), hexanal(1.51%) and 2-pentylfuran(1.18%). These compounds affected the flavor quality of kimchi powder.

Optimal Conditions for the Mycelial Growth of Coprinus comatus Strains

  • The principal objective of this study was to acquire basic data regarding the mycelial growth characteristics for the artificial cultivation of Coprinus comatus. 12 URP primers were employed to evaluate the genetic relationships of C. comatus, and the results were divided into three groups. Among six kinds of mushroom media, MYP medium was selected as the most favorable culture medium for C. comatus. The optimal temperature and pH ranges for the mycelial growth of C. comatus were $23{\sim}26^{\circ}C$ and pH 6${\sim}$8, respectively. The carbon and nitrogen sources for optimal mycelial growth were sucrose and tryptone, respectively.

  • Seasonal changes of aquatic environmental factors, phytoplankton biomass and primary productivity were investigated in the Jido pond (a phytoplankton proliferating pond) from August 1982 to October 1983. Secchi disc transparency, pH, alkalinity and inorganic nitrogen concentration ranged 24~105cm, 7.5~10.6, 50~175mgCaCO3/l 0.1~4.0mgN/l, respectively. The minimum values of transparency, alkalinity and inorganic nitrogen concentration and the maximum value of pH were obtained during the phytoplankton proliferating season. The phytoplankton biomass changed in the range of 51~1146mgchl/m3 with considerable fluctuations but maintained fairly constant in winter. Themaximum and minimum rates of monthly carbon inflow (net primary production) of the phytoplankton community attained 1190gc/m2 in August 1982 and 68g/m2 in February 1983, respectively. The annual rates of inflow and outflow from August 1982 to July 1983 were 7.384 and 7.396kgc/m2, respectively. Turnover rate of phytoplankton carbon and efficiency of radiation of the phytoplankton community varied in the ranges of 60~130%/day (annual mean, 90%/day) and 0.9~11.2% (annual mean, 6.3%), respectively.

Isolation of Alkalophilic Bacillus sp. KJ-133 Producing Cyclomaltodextrinase and Its Enzyme Production (Cyclomaltodextrinase를 생산하는 Alkalophilic Bacillus sp. KJ-133의 분리와 효소생산 조건)

  • To produce and utilize microbial cyclomaltodextrinase being industrially useful, we isolated an alkalophilic Bacillus strain from soil which was capable of degrading cyclodextrins. The newly isolated strain was aerobic, gram-positive, spore-forming, motile, rod shape(0.2~0.4$\times$1.4~4.4 $\mu\textrm{m}$), and 35.8 mol% of DNA base composition. Based on its morphological, phisiological, and biochemical properties, it was identified as alkalophilic Bacillus sp. KJ-133 and cultivated well in the ranges of $30~40^{\circ}C$ and pH 8.0~9.0 . The cyclomaltodextrinase of the strain showed maximal production after 48h of cultivation at $37^{\circ}C$, and the activity was inhibited by Ag2+, Hg2+, Cu2+, and p-chloromercuribenzoate.
