Dissolution Characteristics of ph-Dependent Antacid Granules Agglomerated in High Speed Agitation Type Speed Agitation Type Granulator

  • Antacid granules were prepared by agglomeration and powder method in high speed agitation type granulator. The copmositions of the test antacids were sodium bicarbonate nad magnesium carbonate nad a coating material was powder of polyvinylacetal diethyl-aminocacetate (AEA) and an additive material was talc powder. The dissolution characteristics of base from the antacid granules were investigated to evaluate neutralization capacity of hydrochloric profile of base and neutralization behavior, the following results were obtained : The prepared granules showed a pH-dependent dissolution pattern of a base. The dissolution profile of a base was varied with addition of talc powder as well as coating amount of AEA. The relationship between the ratio of dissolution retarded time for 20% and 10% AEA. The relationship between the ratio of dissolution retarded time for 20% AEA coated granules $\theta_{20}/\theta_{10}$ and the diameter reduction of the granules was explained by the rate process of neutralization of hydrochloric acid.

  • The effects of pH on the sequential sorption/desorption of chlorinated phenols (2-chlorophenol, 2.4-dichlorophenol and 2,4,5-trichlorophenol) in HDTMA-montmorillonite were investigated by maintaining pH 4.85 or 9.15 in the sequential batch sorption and desorption experiments. The chlorinated phenols are hydrophobic ionizable orginic compounds; they can exist as either neutral (pH << pKa) or anionic (pH >> pKa) forms. Among the tested chlorinated phenols, 2,4,5-trichlorophenol showed the highest sorption affinity at pH 4.85 as expected by the $K_{ow}$ . Neutral speciation at pH 4.85 exhibited higher sorption affinity than anionic speciation at pH 9.15. Our results indicates that desorption of chlorinated phenols is strongly dependent on pH of the aqueous phase. Freundlich model was used to analyze the single-solute sorption/desorption results. The ideal adsorbed solution theory(IAST) was employed to predict the hi-solute sorption/desorption equilibria.

  • The efforts of Korean and Japanese radishes on the viability and interleukin (JL)-8 production by Helicobacter pylori were investigated in human gastric epithelial cell. Cell viability was significantly decreased when they were incubated with H. pylori toxin (p <0.05, p<0.01 and p<0.005). Co-incubation with Korean or Japanese radish increased H. pylori toxin-inhibited cell growth in a concentration-dependent manner. The production of IL-8 was greatly increased in H. pylori-infected gastric epithelial cell in concentration- and time-dependent manners. The increased production of IL-8 was significantly inhibited by Korean (p<0.05 and p<0.01) or Japanese (p<0.05) radishes $(5\~10mg/ml)$. These results indicate that Korean and Japanese radishes have protective effects on H. pylori-inhibited cell growth and H. pylori-induced gastric mucosal cell inflammation by suppressing the production of inflammatory cytokine (IL-8) from gastric epithelial cell.

EGCG Blocked Phenylephrin-Induced Hypertrophy in H9C2 Cardiomyocytes, by Activating AMPK-Dependent Pathway

  • AMP-activated protein kinase (AMPK) is a key regulator of energy metabolism. Previous studies have shown that activation of AMPK results in suppression of cardiac myocyte hypertrophy via inhibition of the p70S6 kinase (p70S6K) and eukaryotic elongation factor-2 (eEF2) signaling pathways. Epigallocatechin-3-gallate (EGCG), the major polyphenol found in green tea, possesses multiple protective effects on the cardiovascular system including cardiac hypertrophy. However, the molecular mechanisms has not been well investigated. In this study, we found that EGCG could significantly reduce natriuretic peptides type A (Nppa), brain natriuretic polypeptide (BNP) mRNA expression and decrease cell surface area in H9C2 cardiomyocytes stimulated with phenylephrine (PE). Moreover, we showed that AMPK is activated in H9C2 cardiomyocytes by EGCG, and AMPK-dependent pathway participates in the inhibitory effects of EGCG on cardiac hypertrophy. Taken together, our findings provide the first evidence that the effect of EGCG against cardiac hypertrophy may be attributed to its activation on AMPK-dependent signaling pathway, suggesting the therapeutic potential of EGCG on the prevention of cardiac remodeling in patients with pressure overload hypertrophy.

Synthesis and Swelling Kinetics of a Cross-Linked pH-Sensitive Ternary Copolymer Gel System (pH-민감성 삼성분계 공중합체 젤의 합성 및 팽윤 속도론)

  • A pH sensitive ternary copolymer gel was synthesized in the presence of ethylene glycol dimethacrylate (EGDMA) as a crosslinking agent through radical polymerization of vinyl acetate (VA), acrylic acid (AA) and methyl acrylate(MA) with a weight ratio of 1 : 1.3 : 1. A number of experiments were carried out to determine the swelling behavior of the gel under a variety of pH conditions of the swelling medium. As the pH of the swelling medium was changed from 1.0 to 8.0 at $37^{\circ}C$, the gel showed a shift in the pH-dependent swelling behavior from Fickian (n=0.3447) to non-Fickian (n=0.9125). The resulting swelling parameters were analyzed using graphical and statistical methods. The results showed that the swelling of the gel was controlled by the pH of the medium, i.e. $n=n_o{\exp}(S_{C}pH)$, where n is the diffusion exponent, $n_o(=28.9645{\times}10^{-2})$ is the pre-exponential factor and $S_C$(=0.1417) is pH sensitivity coefficient. The swelling behavior of the gel was also examined in aliphatic alcohols. The results showed that the rate of swelling increased with increasing number of carbon atoms in the alcoholic molecular chain.

Effect of the Amount of Pigment on the Color of Silk Dyed with Fermented Indigo Powder Dye (발효쪽 분말염료로 염색한 견직물의 색소 함량이 직물의 색상에 미치는 영향)

  • This research investigated the relationship between the amount of fixed pigment and the color of silk dyed with three types of fermented indigo powder dye under different temperatures and pH, by reduction or nonreduction method. Amount of fixed pigment was analyzed using the Ultimate 3000 HPLC-DAD instrument and the color of dyed silk was measured using the X-rite spectrocolorimeter. All silk samples dyed by reduction method showed PB color. The amount of indigotin fixation was dependent on the dyeing temperature and pH regardless of the indigotin composition in the dye. Indirubin was less dependent upon the dyeing condition in the reduction dyeing and its fixation was minimum level. Dyeing conditions which can maximize the indigotin fixation were $50^{\circ}C/pH$ 11 and $70^{\circ}C/pH$ 7 conditions in reduction dyeing. Color of silk showed more redness ($a^*$) thus higher PB color when the indigotin fixation was low and indirubin fixation was relatively high. Indirubin fixation was very low with slightly better fixation by nonreduction method. More reddish color was obtained by nonreduction dyeing, and by more alkaline dyebath.

  • Poloxamer-poly (acrylic acid) (PAA) interpenetrating polymer networks (IPNs) were prepared via matrix polymerization of acrylic acid with poloxamer prepolymer. The equilibrium s welling of poloxamer/PAA IPNs was determined in various pH medium. The swelling of poloxamer/PAA IPNs was more affected by pH difference compared with the swelling of homo PAA gel due to protonation and deprotonation of the PAA network, followed by reversible formation and dissociation of the interpolymer complex due to hydrogen bonding between acidic hydrogens and ether oxygens. Nonionic/anionic/cationic drugs were incorporated into IPN matriceds as a model drug and their release behavior was studied. Nonionic, drug revealed release patterns depending solely on pH dependent swelling kinetics. In contrast, the release of ionic drugs was significantly affected by ionic drug-polymer interaction as well as the swelling kinetics.

Inhibition of Cell-Cycle Progression in Human Promyelocytic Leukemia HL-60 Cells by MCS-C2, Novel Cyclin-Dependent Kinase Inhibitor

  • To elucidate the action mechanism of MCS-C2, a novel analogue of toyocamycin and sangivamycin, its effect on the expression of cell cycle-related proteins in the human myelocytic leukemia cell line HL-60 was examined using Western blotting and a flow cytometric analysis. MCS-C2, a selective inhibitor of cyclin-dependent kinases, was found to inhibit cell growth in a time- and dose-dependent manner, and inhibits cell cycle progression by inducing the arrest at G1 and G2/M phases, in HL-60 cells. The flow cytometric analysis revealed an appreciable arrest of cells in the G2/M phase of the cell cycle after treatment with MCS-C2. The HL-60 cell population increased gradually from 13% at 0 h, to 28% at 12 h in the G2/M phase, after exposure to $2{\;}\mu\textrm{M}$ MCS-C2. Furthermore, Western blot analysis demonstrated that MCS-C2 induced the cell cycle arrest at G1 phase through the inhibition of pRb phosphorylation. Hypophosphorylated pRb accumulated after treatment with $5{\;}\mu\textrm{M}$ MCS-C2 for 12 h, whereas, the level of hyperphosphorylated pRb was reduced. Thus, treatment of the cell with MCS-C2 suppressed the hyperphosphorylated form of pRb with a commensurate increase in the hypophosphorylated form.

Hemagglutination of Hantaan and Seoul Viruses (한탄바이러스와 서울바이러스의 혈구응집반응)

  • The hemagglutination activities of Hantaan virus and Seoul virus were demonstrated. The hemagglutinins were prepared by sucrose-acetone extraction method from suckling mouse brains infectecd with Hantaan and Seoul viruses. Hemagglutination of goose erythrocytes by these viral hemagglutinins was pH dependent in phosphate buffer system. Hantaan and Seoul viruses were distinguished by pH range of hemagglutination. 76/118 and 79/90 strains of Hantaan virus showed hemagglutination at the range of pH 5.75-6.4 and the optimal pH was 5.75 with the titer of 1:512 in 76/118 and 1:256 in 79/90. In contrast, KSNUSD 84/34 strain of Seoul virus revealed hemagglutination at the range of pH 6.2-6.4 and the optimal pH was 6.4 with the titer of 1 : 64.

Characteristics of Thiamine Uptake by the BeWo Human Trophoblast Cell Line

  • Little is known concerning the mechanisms responsible for the transplacental transfer of thiamine. So, the aim of this work was to characterize the placental uptake of thiamine from the maternal circulation, by determining the characteristics of $^3H$-thiamine uptake by a human trophoblast cell line (BeWo). Uptake of $^3H$-thiamine (50-100 nM) by BeWo cells was: 1) temperature-dependent and energy-independent; 2) pH-dependent (uptake increased as the extracellular medium pH decreased); 3) $Na^+$-dependent and $Cl^-$-independent; 4) not inhibited by the thiamine structural analogs amprolium, oxythiamine and thiamine pyrophosphate; 5) inhibited by the unrelated organic cations guanidine, N-methylnicotinamide, tetraethylammonium, clonidine and cimetidine; 6) inhibited by the organic cation serotonin, and by two selective inhibitors of the serotonin plasmalemmal transporter (hSERT), fluoxetine and desipramine. We conclude that $^3H$-thiamine uptake by BeWo cells seems to occur through a process distinct from thiamine transporter-1 (hThTr-1) and thiamine transporter-2 (hThTr-2). Rather, it seems to involve hSERT. Moreover, chronic (48 h) exposure of cells to caffeine ($1\;{\mu}M$) stimulated and chronic exposure to xanthohumol and iso-xanthohumol (1 and $0.1\;{\mu}M$, respectively) inhibited $^3H$-thiamine uptake, these effects being not mediated through modulation of the expression levels of either hThTr-1 or hSERT mRNA.
