The Chemical Kinetics for the Reaction of O(³P) with Ethylene

  • The kinetics of the reaction between O(3P) and C2H4 was investigated by measuring time-dependent concentrations of OH resulting from the reaction by using the LIF detection. Oxygen atoms were generated by titrating microwave discharged N2/He with NO to the chemiluminescent end point. The operating pressures in the flow reactor ranged from 5 to 15 torr and the mixtures consisted of He/O(3/P)/C2H4 in the approximate ratios from 100/1/0.1 to 100/1/1. The controlled residence time prior to the detection were estimated to be 0.8-17 ms at the reactor pressure of 7 torr. Experimentally determined profiles both in shape and magnitude were compared with the computed OH density for a specified set of experimental parameters, allowing us to arrive at a complete mechanism for the reaction of O(3P) with ethylene.

Effects of Rubus coreanus and Artemisia princeps Extracts on the Ultraviolet B-Induced DNA Damage Responses in HaCaT Cells (HaCaT 세포에서 자외선 B에 의해 유도된 DNA 상해반응에 대한 복분자와 쑥 추출물의 효과)

  • We investigated the effects of extracts from Rubus coreanus (RC) and Artemisia princeps var. orientalis (AP) on DNA damage response in ultraviolet B (UVB)-exposed HaCaT cells. Cell activity upon treatment for 24 h with RC or AP alone was similar to or greater than that of the nontreated control. When UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, cell activity increased in a concentration-dependent manner. Nuclear fragmentation analysis showed that postincubation with RC or AP decreased UVB-induced apoptosis by about 20% and 15%, respectively, of that in cells postincubated with growth medium. When UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, the level of cyclobutane pyrimidine dimer decreased in a concentration- dependent manner. Western blot analysis showed that treatment of cells not exposed to UVB with RC or AP alone did not significantly change the levels of phospho-p53 and GADD45 protein. Interestingly, when UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, phospho-p53 and GADD45 levels decreased in a concentration dependent manner. Our results suggest that RC and AP extract assist the survival of UVB-exposed cells in parallel with a decrease in levels of UVB-induced DNA damage and damage-response proteins, such as p53 and GADD45.

Resveratrol Induces Apoptosis in Primary Human Prostate Cancer Cells (Primary 인체 전립선 암세포에서 Resveratrol의 Apoptosis 유도 효과)

  • To evaluate resveratrol as a prostate cancer preventive material, we investigated its anti-proliferative and apoptotic effects in RC-58T/h/SA#4 primary human prostate cancer cells. Resveratrol significantly decreased the number of viable RC-58T/h/SA#4 cells in a dose- and time-dependent manner. Resveratrol showed cytotoxicity against RC-58T/h/SA#4, LNCaP, PC-3 human prostate cancer cells with $IC_{50}$ values of 245, 320 and $340\;{\mu}M$, respectively. However the cytotoxic potential of resveratrol against normal RWPE-1 cells was lower ($IC_{50}=982\;{\mu}M$). Resveratrol induced cell death as evidenced by the increased formation of apoptotic bodies, nuclear condensation, sub-G1 phase, and DNA fragmentation. Resveratrol activated initiator caspases 8, and 9 as well as effector caspase 3 in a dose-dependent manner. Furthermore, the general caspase inhibitor z-VAD-fmk significantly inhibited resveratrol-induced apoptosis compared to cells without treatment. These results clearly indicate that resveratrol-induced apoptosis was dependent on caspase activation. Further, resveratrol modulated the down regulation of Bcl-2 (anti-apoptotic), and Bid. However, the level of Bax (pro-apoptotic) remained unchanged. These results suggest that resveratrol induced apoptosis in RC-58T/h/SA#4 cells via a mitochondrial-mediated caspase-dependent pathway, suggesting therapeutic potential against prostate cancer.

Succinate Transport in Rabbit Renal Basolateral Membrane Vesicles (가토 근위세뇨관 Basolateral Membrane Vesicle에서 Succinate 이동 특성)

  • Properties of succinate transport were examined in basolaterat membrane vesicles (BLMV) isolated from rabbit renal cortex. An inwardly directed $Na^+$ gradient stimulated succinate uptake and led to a transient overshoot. $K^+,{\;}Li^+,{\;}Rb^+$ and choline could not substitute for $Na^+$ in the uptake process. The dependence of the initial uptake rate of succinate on $Na^+$ concentration exhibited sigmoidal kinetics, indicating interaction of more than one $Na^+$ with transporter Hill coefficient for $Na^+$ was calculated to be 2.0. The $Na^+-dependent$ succinate uptake was electrogenic, resulting in the transfer of positive charge across the membrane. The succinate uptake into BLMV showed a pH optimum at external pH $7.5{\sim}8.0$, whereas succinate uptake into brush border membrane vesicles (BBMV) did not depend on external pH. Kinetic analysis showed that a Na-dependent succinate uptake in BLMV occurred via a single transport system, with an apparent Km of $15.5{\pm}0.94{\;}{\mu}M$ and Vmax of $16.22{\pm}0.25{\;}nmole/mg{\;}protein/min$. Succinate uptake was strongly inhibited by $4{\sim}5$ carbon dicarboxylates, whereas monocarboxylates and other organic anions showed a little or no effect. The succinate transport system preferred dicarboxylates in trans-configuration (furmarate) over cis-dicarboxylates (maleate). Succinate uptake was inhibited by the anion transport inhibitors DIDS, SITS and furosemide, and $Na^+-coupled$ transport inhibitor harmaline. These results indicate the existence of a $Na^+-dependent$ succinate transport system in BLMV that may be shared by the other Krebs cycle intemediates. This transport system seems to be very similar to the luminal transport system for dicarboxylates.

Adsorption properties of activated carbon prepared from pre-carbonized petroleum coke in the removal of organic pollutants from aqueous solution

  • Activated carbon was prepared from pre-carbonized petroleum coke. Textural properties were determined from studies of the adsorption of nitrogen at 77 K and the surface chemistry was obtained using the Fourier-transform infrared spectrometer technique and the Boehm titration process. The adsorption of three aromatic compounds, namely phenol (P), p-nitrophenol (PNP) and benzoic acid (BA) onto APC in aqueous solution was studied in a batch system with respect to contact time, pH, initial concentration of solutes and temperature. Active carbon APC obtained was found to possess a high surface area and a predominantly microporous structure; it also had an acidic surface character. The experimental data fitted the pseudo-second-order kinetic model well; also, the intraparticle diffusion was the only controlling process in determining the adsorption of the three pollutants investigated. The adsorption data fit well with the Langmuir and Freundlich models. The uptake of the three pollutants was found to be strongly dependent on the pH value and the temperature of the solution. Most of the experiments were conducted at pH 7; the $pH_{(PZC)}$ of the active carbon under study was 5.0; the surface of the active carbon was negatively charged. The thermodynamic parameters evaluated for APC revealed that the adsorption of P was spontaneous and exothermic in nature, while PNP and BA showed no-spontaneity of the adsorption process and that process was endothermic in nature.

Stratum Corneum Exfoliation Effect with Hydroxy Acid according pH (pH에 따른 하이드록시 산(Hydroxy Acid)의 각질 박리 효과 연구)

  • Hydroxy acid has been used to enhance anti-aging and skin moisturization by peeling effect on the skin stratum corneum, and thus it has been widely used in topical products and cosmetic products. Among them, the effect that appears most effectively in a short period of time has been reported to be effected by the pH of the cosmetic formulations. However, there are many difficulties in use due to irritation caused by pH and concerns about side effects. The purpose of this study was to evaluate the effect of applying cosmetics with (1) varying concentrations, (2) types and (3) pH of hydroxy acid on human skin. 22 healthy adults were stained with DHA (dihydroxyacetone) and DC (dansyl chloride) on the forearm, and the skin exfoliation effect was measured after application of the test products. (1) The application of GA (glycolic acid) increased the desquamation by concentration dependent. (2) the test product prepared with neutral pH showed no exfoliation effect. In contrast, SA (salicylic acid) showed a statistically significant exfoliation effect at both acidic pH and neutral pH. (3) The neutral pH SA showed excellent exfoliation effect on bot DHA and DC stained stratum corneum. These results suggest that it is possible to manufacture safe cosmetics without damaging the skin barrier, providing an opportunity to use cosmetics that are expected to exfoliate to people, whose skin is sensitive to pH.

Determination of Chloramphenicol by Differential Pulse Polarography (미분 펄스 폴라로그래피에 의한 Chloramphenicol의 정량분석)

  • Chloramphenicol was studied by differential pulse polarography (DDP). A reduction peak which is dependent on pH of the solution appeared in the voltage range between zero and -1.50 volt vs. Ag/AgCl (sat. KCl) reference electrode. A plot of peak potentials (Ep) measured at room temperature (20$^{\circ}C$) vs. pH of the chloramphenicol solutions showed linear relationship changing slope (Ep/pH) at pH 8.9. The slope was -59.7 mV/pH in pH 2.7∼8.9 and -24.3 mV/pH in pH 8.9∼11.2, respectively. A log plot of peak currents (ip) vs. concentrations showed a linearity at the concentrations between 4.8 ${\times}$ 10$^{-7}$ M and 6.2 ${\times}$ 10$^{-5}$ M (0.16 ppm∼20 ppm) chloramphenicol in pH 8.0 ammonium buffer. Between the DPP method and the reference method measuring absorbance at 278 nm, the correlation coefficient was 0.996, which means an excellent linearity. The DPP method was able to detect degradation products of chloramphenicol in mild alkaline solution (pH = 8.0) more distinctly than the spectrophotometric method.

Biosorption of Cobalt by Methanotrophic Biomass (메탄자화균에 의한 코발트의 생물흡착)

  • The optimum pH range for biosorption of cobalt by methanotrophic bacteria was broadened to 6.0~12.0 which was compared to pH 10.5~11.5 of bios or bent-free control case. Removal efficiency of cobalt by methanotrophic biomass was pH dependent, but less sensitive than that of control. With 1.0 g biosorbent/L at initial solution pH 6.0. methanotrophic biomass took up cobalt from aqueous solutions to the extent of 170 mg/g biomass. As a result of scanning electron microscope(SEM) micrographs, cobalt removal by methanotrophic biomass seemed to be through adsorption on the surface of methanotrophic biomass and by exopolymer around the biomass. Optimum amount of methanotrophic biomass for maximum cobalt uptake was 1.0 g/L for initial 400 mg Co/L at initial pH 6.0. Removal efficiency of cobalt was slightly affected by ionic strength up to 2.0 M of NaCl and $NaNO_3$, respectively.

Poly(L-lysine) Based Semi-interpenetrating Polymer Network as pH-responsive Hydrogel for Controlled Release of a Model Protein Drug Streptokinase

  • With the aim of developing of pH-sensitive controlled drug release system, a poly(Llysine) (PLL) based cationic semi-interpenetrating polymer network (semi-IPN) has been synthesized. This cationic hydrogel was designed to swell at lower pH and de-swell at higher pH and therefore be applicable for achieving regulated drug release at a specific pH range. In addition to the pH sensitivity, this hydrogel was anticipated to interact with an ionic drug, providing another means to regulate the release rate of ionic drugs. This semi-IPN hydrogel was prepared using a free-radical polymerization method and by crosslinking of the polyethylene glycol (PEG)-methacrylate polymer through the PLL network. The two polymers were penetrated with each other via interpolymer complexation to yield the semi-IPN structures. The PLL hydrogel thus prepared showed dynamic swelling/de-swelling behavior in response to pH change, and such a behavior was influenced by both the concentrations of PLL and PEG-methacrylate. Drug release from this semi-IPN hydrogel was also investigated using a model protein drug, streptokinase. Streptokinase release was found to be dependent on its ionic interaction with the PLL backbones as well as on the swelling of the semi-IPN hydrogel. These results suggest that a PLL semi-IPN hydrogel could potentially be used as a drug delivery platform to modulate drug release by pH-sensitivity and ionic interaction.

Decomposition of Hexahydro-1,3,5-trinitro-1,3,5-triazine by Gamma Ray Irradiation (감마선 조사에 의한 hexahydro-1,3,5-trinitro-1,3,5-triazine(RDX)의 분해)

  • The purpose of this study was to evaluate the potential of a gamma ray irradiation to decompose hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) in an aqueous solution. The decomposition reaction of RDX by gamma ray irradiation was a first-order kinetic over the applied initial concentrations (10-40mg/L). The dose constant was strongly dependent on the initial concentration of the RDX. The removal of RDX was more efficient at pH below 3 and at pH above 11 than at neutral pH (pH 5-9). The required irradiation dose to remove 99% of the RDX (40mg/L) was 4, 8 and 1 kGy, at pH 2, 7 and 13, respectively. The dose constant was increased by two folds and over twelve folds at pH 2 and 13, respectively, when compared with that at pH 7. When an irradiation dose of 20 kGy was applied, the removal efficiencies of TOC were 80, 57 and 91% at pH 2, 7 and 13, respectively. Ammonia and nitrate were detected as the main nitrogen byproducts of RDX and formic acid was detected as an organic byproduct. The results showed that a gamma ray irradiation was an attractive method for the decomposition of RDX in an aqueous solution and it was found that a strong alkaline pH over 12 should be applied to the decomposition reaction of RDX.
