Relationship between Concentration of Phosphorus, Turbidity, and pH in Water and Soil under Aerobic and Anaerobic Conditions (혐기와 호기 상태의 물과 토양에서 pH, PO4-P, 탁도, T-P 농도 관계)

  • This research was carried out to elucidate the impacts of dissolved oxygen (DO) concentration to phosphorus release mechanism from soil into water in lakes. $PO_4-P$ contributed to the growth of the total phosphorus (T-P). $PO_4-P$ was steadily increasing because water was accumulating $PO_4-P$. T-P was closely related to turbidity, pH, and DO. We found that DO had decreased because DO was consumed in organic matter decomposition, and that the resulting anaerobic decomposition occurred whenever water had run out of DO. We also found that pH had decreased sharply by production of organic acid by the anaerobic decomposition and that T-P decreased because a decrease in pH removed turbidity by precipitation. T-P was dissolved without microbial decomposition. This mechanism was of great importance in lakes because phosphorus is released from soil into water.

Different Effects of Acidic pH Shock on the Prodiginine Production in Streptomyces coelicolor M511 and SJM1 Mutants

  • The changes in prodiginines productions caused by pH shock culture of Streptomyces coelicolor strains were estimated. In Streptomyces coelicolor M511, undecylprodiginine and streptorubin B productions increased 1.8-fold (37.22 mg/g) and 2.5-fold (18.61 mg/g), respectively, by pH shock (from 7.2 to 4.0). In contrast, this resulted in the significantly decreased prodigignines production in the redP deletion mutant SJM1; 3.7-fold for undecylprodiginine, 4.4-fold for streptorubin B, 5.2-fold for methylundecylprodiginine, and 6.4-fold for methyldodecylundecylprodiginine, respectively. RT-PCR analyses showed that, during pH shock, expression of redD, the transcription activator gene, was increased while the expression of fabH, the decarboxylative condensation enzyme gene in fatty acid biosynthesis, was decreased in both strains. The enhanced redD expression was in good accordance with the increased total prodiginines production of M511. However, for SJM1 mutant, the decrease of fabH expression occurred more strikingly, such that it became almost completely turned off during acidic pH shock culture. Therefore, a down-regulation of fabH was considered to be the cause of decreased amount of total prodiginines produced, although redD expression was high in SJM1 mutant.

Effect of pH Change on Vascular Smooth Muscle Contractility in Rat Superior Mesenteric Artery and Its Branches (쥐 상장간막 동맥과 그 분지에서 pH 변화가 혈관평활근 수축성에 미치는 영향)

  • Background: Extracellular and intracellular pH ($pH_o$ and $pH_i$), which can be changed in various pathological conditions such as hypoxia, affects vascular contractility. To elucidate the mechanism to alter vascular contractility by pH, the effects of pH on reactivity to vasocontracting agents, intracellular $Ca^{2+}$ influx, and $Ca^{2+}$ sensitivity in vascular smooth muscle were examined. Material and Method: Isometric contractions in rat superior mesenteric arteries (SMA) were observed. Intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) was recorded by microfluorometer using Fura-2/acetoxylmethyl ester in muscle cells. $pH_o$ was increased from 7.4 to 7.8 or decreased to 6.9 or 6.4. $pH_i$ was decreased by applying $NH_4^+$ or propionic acid or modulated by changing $pH_o$ after increasing membrane permeability using $\beta$-escin. Result: Decreases in $pH_o$ from 7.4 to 6.9 or 6.4 shifted concentration-response curve by norepinephrine (NE) or serotonin (SE) to the right and significantly increased half maximal effective concentration (EC50) to NE or SE. Increase in $pH_o$ from 7.4 to 7.8 shifted concentration-response curve by norepinephrine (NE) or serotonin (SE) to the left and significantly reduced EC50 to NE or SE. NE increased $[Ca^{2+}]_i$ in cultured smooth muscle cells from SMA and the increased $[Ca^{2+}]_i$ was reduced by decreases in $pH_o$. NE-induced contraction was inhibited by $NH_4^+$, whereas the resting tension was increased by $NH_4^+$ or propionic acid. When the cell membrane of SMA was permeabilized using ${\beta}$-escin, SMA was contracted by increasing extracellular $Ca^{2+}$ concentration from 0 to $10{\mu}M$ and the magnitude of contraction was decreased by a decrease in $pH_o$ and vice versa. Conclusion: From these results, it can be concluded that a decrease in $pH_o$ might inhibit vascular contraction by reducing the reactivity of vascular smooth muscle to vasoactive agents, $Ca^{2+}$ influx and the sensitivity of vascular smooth muscle to $Ca^{2+}$.

  • The biological effect of hyperthermia on the SCK tumor cells in vitro were analyzed in several respects. A comparision of the survival curve of SCK tumor cells in vitro and in vivo following hyperthermia demonstrated that the cytocidal effect of heating is far greater on the cells in vivo than on the cells in vitro. The pH change in the SCK tumor upon being heated at $43.5^\\circC$ started out at 7.05 and increased to 7.18 during the first 7 min of heating and then rapidly declined to 6.67 by 30 min. Contrary to the decrease in pH in the heated tumors, the pH in the muscle increased significantly when heated to $43.5-45.0^\\circC$. Following hyperthermia at $43.5^\\circC$ for 30 min, a maximum increase in the lactic acid content in the tumor and liver was observed at 1 hr and 3 hr, respectively. The increase in the tumor was followed by a gradual decrease below the control level, whereas the increase in the liver was maintained at quite a steady level for 24 hr. The hyperthermia at $43.5^\\circC$ for 1 hr exhibited a general tendency that high molecular proteins decrease markedly, whereas most of low molecular proteins increase. The most prominent change was that the heat shock protein 70K increased significantly along with other low molecular proteins in heat shocked cells.

  • Ozone alone, Ozone/GAC, Ozone/$H_2O_2$ and Ozone/GAC/$H_2O_2$ processes were introduced for treatment of humic acid, which is a representative refractory organic compound. $H_2O_2$ and GAC used as catalysts for experiment. The treatment efficiencies of humic acid in each process were analyzed for pH variation, DOC removal, and $UV_{254}$ decrease. $UV_{254}$ decrease in Ozone/GAC and Ozone/GAC/$H_2O_2$ processes were the highest with about 93%, and Ozone alone and Ozone/$H_2O_2$ processes were 88%. DOC removal in Ozone/GAC/$H_2O_2$ process was the highest with 71%. Removal by Ozone/GAC, Ozone alone, and Ozone/$H_2O_2$ processes were 66%, 39%, and 47%, respectively.

  • Cation exchange capacity of clay, white clay and zeolite was measured by the adsorption test for 3 different heavy metal (Cd, Cr, Zn) standard solutions whose concentrations were varied by 10, 20, 30 ppm and pH were varied by 3, 5, 7, 9, respectively. The adsorption rate of Cd and Zn increased with increasing pH and slowly increased with increasing pH above pH 5. However, adsorption rate of Cr did not increase with increasing pH. Especially, Cr adsorption rate of the mixture of clay and white clay at pH 5 showed an half decrease compared to that at pH 3. The adsorption rate of mixed heavy metal solutions was in the order of Cd, Zn > Cr; however, the order was changed by Fe>Pb, Cu>Cr>Zn>Cd in case of Cu, Fe and Pb addition.

Depletion of Nitrite by Lactic Acid Bacteria Isolated from Kimchi(II) (김치에서 분리한 유산균에 의한 아질산염 소모(II))

  • Depletion of nitrite by lactic acid bacteria isolated from Kimchi and the effects of pH and temperature on depletion of nitrite in Lactobacillus MRS broth were investigated. Depletion of nitrite by Lactobacillus plantarum and Lactobacillus sake was low in initial stage of growth but increased during growth at 15 and 2$0^{\circ}C$. L. plantarum and L. sake depleted than 90 and 75% of nitrite(250$\mu\textrm{g}$/$m\ell$) in 1 day of growth at 25 and 3$0^{\circ}C$, respectively, but depleted almost all of nitrite in 2 days. While the effect of Leuconostoc mesenteroides (150$\mu\textrm{g}$/$m\ell$) was lower compared to Lactobacillus(250$\mu\textrm{g}$/$m\ell$). In addition, even high concentrations(600 and 900 $\mu\textrm{g}$/$m\ell$) of nitrite was depleted at 2 days of growth by L. plantarum. pH of growing broth decreased as the increase of growth time and temperature, and depletion of nitrite in- creased as the decrease of pH of growing culture. However, pH of broth related with the decrease of depletion of nitrite at 25 and 3$0^{\circ}C$, but not at 15 and 2$0^{\circ}C$. Therefore, depletion of nitrite was greatly affected by growth temperature rather than by pH.

Physiological Relationship Between Thirst Level and Feed Intake in Goats Fed on Alfalfa Hay Cubes

  • The present study was carried out to measure changes of feed intake and thirst level caused by water deprivation in goats fed on dry feed and to elucidate the relationship between those two parameters. Water deprivation significantly (p<0.01) decreased cumulative feed intake and rate of eating at 30, 60, 90 and 120 min, respectively, after feed presentation. Cumulative feed intake, after completion of 2 h feeding, was reduced by about 20, 21 and 64 % due to water deprivation during feeding for 2 h (WD2), for 22 h (WD22) and for 46 h (WD46), respectively, compared to free access to water (FAW). Compared to the FAW, WD2, WD22 and WD46 increased thirst level by about 5, 5 and 9 times, respectively. Mean thirst level (X, g/30 min) was negatively correlated with cumulative feed intake (Y, g DM) after completion of 2h feeding (Y=1302-0.2 X, $r^2=0.97$, p<0.05). Water deprivation depressed plasma volume and there was a significant positive regression between plasma volume (X, ml) and cumulative feed intake (Y, g DM) after completion of 2h feeding (Y=-1003+0.6 X, $r^2=0.99$, p<0.01). Mean plasma osmolality (X, mOsmol/l) correlated significantly and negatively with cumulative feed intake (Y, g DM) after completion of 2h feeding (Y=27004-84.9 X, $r^2=0.95$, p<0.05). In conclusion, a decrease of feed intake during water deprivation is mainly due to an increase of thirst level quantitatively, and the act of feeding itself induces thirst more than the length of water-deprivation periods in goats fed on dry feeds. The present findings suggest that plasma osmolality and plasma volume which affect thirst level are involved in the decrease of feed intake in water-deprived goats.

Decrease efficiency of Offensive Odor from Pig Excreta by Yeast Strain, Pichia farinosa NASS-2 Isolated from Soy Bean Paste (된장에서 분리한 효모(Pichia farinosa NASS-2)의 돈분 악취감소효과)

  • The different microbial species were isolated from soy bean paste samples. A yeast strain NASS-2 was effective to decrease odor activity on pig excreata was identified as Pichia farinosa based on nucleotide sequences of 18S ribosomal DNA and Internal transcribed space (ITS). The extracellular fraction of P. farinosa NASS-2 was effective to decrease odor activity of pig excrements. Optimal medium component for decreasing order activity on odor material composed of soluble starch 2.0% (w/v) and yeast extract 0.8% (v/v). The decrease of odor material was maximum at $28^{\circ}C$ for 72 hours with pH 5.5. When the P. farinosa NASS-2 culture broth was treated to pig excrements, the removal efficiency was an average concentration with 1.38 ppm of ammonia gas.

Biological Characteristics of Calystegia japonica (메꽃의 생물학적(生物學的) 특성(特性))

  • The growth of Calystegia japonica Choisy as affected by pH, drying, burial, soil moisture and light intensity was determined. Germination of C. japonica rhizome was not affected by pH's ranging from 4.8 to 8.7,while the greatest growth after germination was obtained at pH 5.7. Drying longer than 14 h at $35^{\circ}C$ brought about a significant reduction in percent survival and the subsequent growth of C. japonica. A significant decrease in growth of C. japonica occurred when the rhizome was buried at 0 ㎝ and deeper than 8 cm. The greatest growth was obtained when the soil moisture content reached 40 to 60% of saturated soil. Increasing percent available light resulted in decrease in the plant height, but increase in the root length and dry weight.
