  • I studied about temperature and pH concentration having influence upon chlorophyll decrease on the Glucose culture of Chlorella variegata Beijerinck(211/10a). I cultured under $25^{\circ}C$(standard), $15^{\circ}C$ and $35^{\circ}C$ and compared with each other. The culture of $15^{\circ}C$ didn't have any large difference with the standard culture($25^{\circ}C$) but the culture of $35^{\circ}C$ had a large amount of chlorophyll decrease without carbohydrate accumulation, stimulation of cell division and nitrogen-deficiency. Chlorella variegata had optimum pH 6.5-7 and was a little weak in all phenomenon under pH 8 rather than under pH 6.5-7. Under pH 5 they had deep chlorophyll decrease without phephytin.

혐기적 산성조건하에서 젖산균의 생존과 그 특성 (Survivals of Lactic Acid Bacteria and its Characteristics under the Acidic and Anaerobic Condition)

  • We investigated the survival, $\beta$-galactosidase activity and cellular permeability of lactic acid bacteria such as Lactobacillus acidophilus ATCC 4356, Lactobacillus casei subsp. casei IFO 3533, Streptococcus thermophilus KCTC 2185, Lactobacillus delbrueckii subsp. lactis ATCC 4797, and Lactobacillus delbrueckii subsp. bulgaricus ATCC 11842 in anaerobic condition of pH 1.5-3.5 range. Numbers of all tested viable cells did not decrease at pH 3.5, but decreased rapidly at pH 1.5 and pH 2.5 during 2 hour incubation at modified EG medium. Immediately after 2 hour incubation, the decrease in population at pH 1.5 and pH 2.5 was about 6-8 and 5-7 log cycles/ml, respectively. Lactobacillus acidophilus ATCC 4356 showed the higest survival of all tested bacteria. The $\beta$-galactosidase activity from Lactobacillus delbrueckii subsp. bulgaricus ATCC 11842 and Streptococcus thermophilus KCTC 2185 decreased rapidly at pH 1.5 and 2.5, but there was a little decrease at pH 3.5. The cellular permeability that was measured by the leakage of intracellular materials increased with decrease of pH. These results suggest that the ingested lactic acid bacteria may be destroyed in contact with low pH of gastric acid.

  • Objectives: The information on the depth where pulse wave appears is as important as pulse waveform. The aim of this study was to classify pulse pattern using pressure-height(P-H) volume-curve by 5 applied pressure levels to find out the information on the depth of pulse and interpret the floating & sinking pulse in oriental medical pulse diagnosis. Methods: We used 3 dimensional pulse imaging analyser (DMP-3000, DAEYOMEDI Co., Korea), which measures radial pulse waveforms noninvasively by way of tonometric method at 5 applied pressure levels, and shows P-H volume-curves by applied pressure. 448 subjects were enrolled, pulse waveforms were measured and the P-H volume-curves were gained on the three locations of Chon, Kwan, and Cheok. Results: Gained P-H volume curves were classified into 3 types ; increase type, decrease type, and increase-decrease type. Increase-decrease type appeared more often on Chon and Kwan, while increase type appeared more often on Cheok. In a few cases, decrease-type appeared on Chon and Kawn, however it never appeared on Cheok. Conclusions: Through the classification of pulse by P-H volume-curve, we gained the information on the depth of pulse. We speculate the decrease type as floating pulse, the increase-decrease type as middle pulse, and the increase type as sinking pulse in oriental medical pulse diagnosis. After more researches on P-H volume-curve by applied pressure, the P-H volume-curve may be used as an important factor for pulse diagnosis.

환원액의 pH에 따른 인디고의 염색성 (The Dyeability of Indigo Depending on the pH of Reduction Bath)

  • Cotton fabric was dyed with indigo at different pHs, dye concentrations, and repeated dyeings. And the K/S value and dye uptake of the dyed materials at those conditions were determined. The decrease of pH during the reduction process of indigo stock solution was observed. As a result the decrease of pH of the dye stock solution depended on the dye concentration and initial pH. The highest K/S value was shown at fabric dyed at pH 11 and the relationship between K/S value and dye uptake depended on initial pH of reduction bath and the slope was lower as the pH increased to 13.

  • 본 연구에서는 보통강재인 SS 41 강을 열처리하여 얻은 M.E.F.복합조직강의 모상입경변화와 3.5% NaCl수용액의 pH변화 조건하에서 반복굽힘피로실험을 하여 부식 피로파괴에 미치는 영향에 대해 고찰하였다.

산성 발효 산물과 배양 pH가 Clostridium acetobutylicum의 유지 에너지에 미치는 영향 (Effects of Acidic Fermentation Products and Culture pH on the Maintenance Energy of Clostridium acetobutylicum)

  • In order to elucidate the acid tolerance mechanism of Clostridium acetobutylicum against organic acid, the maintenance energy with added butyrate at different pH was determined. Maintenance coeffecient in acidogenic chemostat was higher at pH 6.5 than at pH 5.5 showing that this organism is an acidophile. The addition of butyrate at pH 5.5 and different dilution rate caused linear decrease of the cell concentration though $Y_{ATP}$ did not decrease with increasing undissociated organic acid. $Y_{ATP}$ decreased by increasing the concentration of undissociated organic acid at pH 5.0 by the addition of butyrate. From these results it is hypothesized that the ATP conaumption for pH stat of acidophile C. acetobutylicum is increased at the circumstance with over 30mM of undissociated organic acid.cid.

물-시멘트비에 따른 시멘트계 재료의 pH 저하 저항성 평가 (Resistance to a pH Fall with Water-Cement Ratio in Cementitous Material)

  • 본 연구에서는 물-시멘트 비에 따른 시멘트계 재료인 시멘트 페이스트, 모르터 및 콘크리트의 pH 저하 저항성을 평가하였다. 우선 시멘트 페이스트에 대해 물-시멘트 비 30%, 40%, 50% 인 보통 포틀랜드 시멘트 페이스트의 산 증가에 따른 pH를 측정하였다. 또한, 페이스트의 pH 저하 저항성에 대한 골재의 영향에 대해 알아보기 위해서 콘크리트, 모르터, 페이스트의 pH 저하 저항성 측정 실험도 수행하였다. 실험결과, 산이 증가할수록 pH는 감소하였으며 특히 pH 10 정도에서 급격한 pH 저하가 발생되었다. 산 증가에 따른 pH 감소정도는 산중화능력으로 나타내었으며 시멘트 수화물에 의해 특정 pH에서 산중화능력이 물-시멘트비에 따라 높게 나타났다.

Response of Syntrophic Propionate Degradation to pH Decrease and Microbial Community Shifts in an UASB Reactor

  • The effect of pH on propionate degradation in an upflow anaerobic sludge blanket (UASB) reactor containing propionate as a sole carbon source was studied. Under influent propionate of 2,000 mg/l and 35℃, propionate removal at pH 7.5-6.8 was above 93.6%. Propionate conversion was significantly inhibited with stepwise pH decrease from pH 6.8 to 6.5, 6.0, 5.5, 5.0, 4.5, and then to 4.0. After long-term operation, the propionate removal at pH 6.5-4.5 maintained an efficiency of 88.5%-70.1%, whereas propionate was hardly decomposed at pH 4.0. Microbial composition analysis showed that propionate-oxidizing bacteria from the genera Pelotomaculum and Smithella likely existed in this system. They were significantly reduced at pH ≤5.5. The methanogens in this UASB reactor belonged to four genera: Methanobacterium, Methanospirillum, Methanofollis, and Methanosaeta. Most detectable hydrogenotrophic methanogens were able to grow at low pH conditions (pH 6.0-4.0), but the acetotrophic methanogens were reduced as pH decreased. These results indicated that propionate-oxidizing bacteria and acetotrophic methanogens were more sensitive to low pH (5.5-4.0) than hydrogenotrophic methanogens.

정수처리에서 pH 저감에 의한 응집효율향상에 관한 연구 (Improving Coagulation Performance with pH Preadjustment in Drinking Water Treatment)

  • This paper reports on a pilot scale comparison of PACS coagulation with and without pH preadjustment. The pH of the water was adjusted with carbon dioxide and sulfuric acid. Process performance was assessed on the basis of total organic carbon(TOC), UV absorbance, turbidity and disinfection by-product(DBP) precursors. Coagulation pH appeared to be a determining factor for maximum NOM removal. The optimum coagulation pH in order to decrease TOC and turbidity were pH 7. Preadjustment of pH 7 increased TOC removal to as much as 43, 47 percent with sulfuric acid and carbon dioxide. Moreover, coagulation at pH 7 caused a reduction in UV$_{254}$, THMFP and HAAFP compared to the baseline coagulation. For preadjustment of pH 7 with carbon dioxide, the percentage of TOC, UV$_{254}$, THMFP and HAAFP shows the reduction rate of 3.8, 0.5, 4.8, 9.4% comparing to the coagulation rendition using sulfuric acid. Acid addition to depress pH during coagulation decrease Langelier Saturation Index(LSI), potentially causing increase corrosion in water distribution systems. LSI for carbon dioxide and sulfuric acid at pH 6 was -2.3, -3.3. Therefore, carbon dioxide was more effective at controlling corrosion than sulfuric acid.

자가영양과 타가영양 조건하에서 Thiobacillus novellus의 생리 및 성장 (Growth and Physiology of Thiobacillus novellus under Autotrophic and Heterotrophic Conditions)

  • The growth of T. novellus in auto trophic and geterotrophic media was studied to determine the time required for cells to enter stationary phase and relative percentage of ribosomal proteins. When T. novellus was grown autotrophically, growth proceeded at a slow rate characteristic of autotrophs and did not enter log phase until the end of the first day. Logarithmic growth proceeded for 3-4 days at which time the cells entered the stationary phase. In particular, logarithmic growth was accompanied by decreasing pH of culture media and in the stationary phase the pH levelled off at 6.0, a decrease of 1.6 pH value compared to original pH of media. The pH decrease was greatest during log phase when cells oxidized thiosulfate to $H_{2}$$SO_{4}$. The doubling time was about 26h. In heterotrophic media growth proceeded at a much faster rate and cells entered stationary phase 20-22h after inoculation. The doubling time was 3h. The protein content of the ribosomes in T. novellus grown heterotrophically was 4.2% greater than those from the organism grown autotrophically.
