FeS 수용액 내 pH에 따른 5가비소의 반응 메커니즘 연구 (Mechanistic Study of FeS Reacted with Arsenate under Various pH Conditions)

  • Mackinawite (FeS), as a ubiquitous reduced iron mineral, is known as a key controller of redox reactions in anaerobic subsurface environment. The reaction of FeS with redox-sensitive toxic element such as arsenic is substantially affected by pH conditions of the given environments. In this study, the interaction of As(V) with FeS was studied under strict anaerobic conditions with various pH conditions. The pH-dependent arsenic removal tests were conducted under wide ranges of pH conditions and X-ray absorption spectroscopy (XAS) was applied to investigate the reaction mechanisms under pH 5, 7, and 9. The removal efficiency of FeS for As(V) showed the higher removal of As(V) under low pH conditions and its removal efficiency decreased with increasing pH, and no As(V) reduction was observed in 1 g/L FeS solution. However, XAS analysis indicated the reduction of As(V) to As(III) occurred during reaction between FeS and As(V). The reduced form of As(III) was particularly identified as an arsenic sulfide mineral (As2S3) in all pH conditions (pH 5, 7, and 9). As2S3 precipitation was more pronounced in pH 5 where the solubility of FeS is higher than in other pH conditions. The linear combination fitting results of XAS demonstrated that As(V) removal mechanism is concerted processes of As2S3 precipitation and surface complexation of both arsenic species.

음폐수의 중온 및 고온 산발효에서 초기 pH가 VFAs 생성에 미치는 영향 (The Effects of Initial pH on VFAs Production of Mesophilic and Thermophilic Acidogenic Fermentation for Food Waste Recycling Wastewater)

  • Batch cultivations were performed to evaluate the influences of the initial pH condition on mesophilic and thermophilic acidogenic fermentation with food waste recycling wastewater. In both conditions of mesophilic and thermophilic fermentation, TVFAs production rates were maximized at the initial pH 7 condition as 0.15 and 0.23 g TVFAs/L hr, respectively. And pH was also maintained stably between 6 and 7 during 72hr acidogenic cultivation at both conditions. However, predominant VFA components were different according to reaction temperature conditions. In mesophilic condition, propionic acid which has low conversion efficiency to methane was accumulated up to 1,348 mg/L while acetic and butyric acid were predominant in thermophilic condition. Therefore, thermophilic acidogenic fermentation was superior for the effective VFAs production than mesophilic condition. From the DGGE analysis, the band patterns were different according to the initial pH conditions but the correlations of the each band were increased in similar pH conditions. These results mean that microbial communities were certainly affected by the initial pH condition. Consequently, the adjustment of the initial pH to neutral region and thermophilic operation are needed to enhance acidogenic fermentation of food waste recycling wastewater.

침출 조건이 감잎차의 색 및 항산화 특성에 미치는 영향 (Effects of extraction conditions on color quality and antioxidant properties of persimmon (Diospyros kaki Thunb.) leaf tea)

  • 본 연구는 시판용 감잎분말을 이용한 침출차 제조 시 시료의 양 및 pH 조건이 감잎차의 색 품질과 항산화능에 미치는 영향을 알아보기 위해 수행되었다. 차 내 감잎의 양이 많을수록 차의 명도는 감소하고 적색도와 황색도는 증가하였다. 항산화 특성 관련 지표 분석 결과, 감잎 함량이 가장 높았던 감잎차(10 mg/mL)에서 항산화력(DPPH 라디칼 소거 활성: 0.138±0.000 mg AAE/mL, ABTS 라디칼 소거 활성: 0.908±0.003 µmol TE/mL, FRAP 환원력: 0.111± 0.004 mg AAE/mL)과 항산화 성분 함량(총폴리페놀 함량: 0.219±0.010 mg/GAE mL, 총플라보노이드 함량: 0.107±0.003 mg/CE mL)이 가장 높았다. 다음으로, 감잎차 내 시료의 양은 10 mg/mL로 고정하고, 침출 용매의 pH 조건을 pH 4에서 7 범위로 설정하여 색도와 항산화성을 분석하였다. 분석 결과, pH가 증가함에 따라 감잎차의 명도는 감소하고 적색도와 황색도는 증가하였다. 또한, DPPH 라디칼 소거 활성과 FRAP 항산화력은 pH가 낮을수록 높은 경향을 보였다. 일관되게, 가장 낮은 pH 4에서 다른 군에 비해 폴리페놀 함량과 플라보이드 함량이 가장 높은 것을 확인하였다. 전반적으로, pH 4 조건에서 제조한 감잎차의 항산화 활성이 다른 pH 조건 대비 더 우수하였고 폴리페놀 화합물 함량도 더 많았다. 동시에 색도는 더 밝으면서 붉은 정도와 황색 정도가 적었다. 결론적으로, 시료의 양을 높이면서 낮은 pH 조건에서 감잎차를 준비하는 것이 항산화 기능성 측면에서 더 나은 건강 효과를 나타내는 것으로 사료된다.

배양 조건에 따른 Pseudomonas sp.CH-414의 Phospholipid 생산능의 변화 (Phospholipid Production by Pseudomonas sp.CH-414 under Various Culture Conditions)

  • Pseudomonas sp. CH-414를 이용하여 통기속도 빛 pH 변화에 대한 균체량 벚 인지질 생산 변화에 대한 실험을 수행하였다. pH를 7.0에서 8.0으로 변 화시켰을 때는 인지질의 생산량뿐만 아니라 균체량 이 동시에 급격하게 감소하였으나, pH를 7.0에서 6 6.0으로 낮추어 주였을 경우 pH 7.0과 비교하여 인 지질이 약 20% 정도 증가하였으며 균체량의 변화는 없었다. 통기속도가 증가할수록 균체량은 증가하였으나 인지질의 생산은 변화가 심하였으며 3vvm에서는 인지질 농도의 급격한 감소를 보였다. 이상의 결과를 토대로 배양 30시간까지는 pH 7.0, 3vvm으로 배양한 후 그 이후부터는 pH 6.0, 1 vvm 으로 변화를 주어 배양하였을 때 균체량은 약 18.5g d dry cell weight/$$\ell$ 이었고 인지 질의 양은 약 $0.83g/\ell$(45mg/g cell)이였다.

Selective Homocysteine Assay with Cucurbit[7]uril by pH Regulation

  • We report the effect of pH on the supramolecular complexation of two biothiols, viz., homocysteine (Hcy) and cysteine (Cys), with cucurbit[7]uril (CB[7]). Under basic pH conditions, Cys did not complex with CB[7], whereas Hcy efficiently complexed with CB[7], as confirmed by 1H NMR spectroscopy and Ellman's reagent (5,5'-dithio-bis(2-nitrobenzoic acid), DTNB) assay. 1H NMR and Raman spectroscopic studies revealed that, in the absence of CB[7], Hcy auto-oxidized slowly (~36 h) to homocystine (HSSH) under basic pH conditions. However, the rate of Hcy oxidation increased by up to 150 fold in the presence of CB[7], as suggested by the DTNB assay. Thus, supramolecular complexation under basic pH conditions led to the formation of a HSSH-CB[7] complex, and not Hcy-CB[7]. The results indicate that Hcy is rapidly oxidized to HSSH under the catalysis of CB[7], which acts as a reaction chamber, in basic pH conditions. Our studies suggest that Hcy concentration, a risk factor for cardiovascular disease, can be selectively and more easily quantified by supramolecular complexation with CB [7].

생물학적 회분식 인 제거 공정에서 pH의 영향과 그래뉼 생성 (Influence of Different Operational pH Conditions and Granulation on Enhanced Biological Sequencing Batch Phosphorus Removal)

  • A sequencing batch reactor (SBR) was operated under different pH conditions to better understand the influence of pH to granulation in enhanced biological phosphorus removal systems. Granules from the SBR were also investigated using scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX). Considerable decreases in the amount of phosphorus released per substrate provided under anaerobic conditions and the content of biomass polyphosphate under aerobic conditions were observed when pH was changed from 7.5 to 7.0, followed by 6.5. Aerobic granulation was also observed at pH 7.0. A number of bacteria with the typical morphological traits of tetrad-forming organisms (TFOs) were observed at pH 7.0, including large members of cluster. Filamentous bacteria were also there in large numbers. The occurrence and growth of granules were further enhanced at pH 6.5. A SEM analysis showed that the aerobic granules had a compact microbial structure with shaperical shape and morphologically consisted of aggregates of small coccoid bacteria and filamentous bacteria encapsulated by extracellular polymeric substance. The main material ions identified by EDX moreover revealed that the structural materials for polyphosphate in the granules include phosphorus, potassium and calcium. Therefore, these results strongly suggested that PAOs are a dominant population in the microbial community of the aerobic granules.

치관보철용 Hydroxyapatite의 기계적 물성에 미치는 pH 및 반응온도의 영향 (Effect of pH and Reaction Temperature on Mechanical Properties of Hydroxyapatite for Dental prosthesis)

  • Hydroxyapatite powder were synthesized by precipitation method, varying pH, and reaction temperature. The powders were heated at 1,200$^{\circ}{\cdots}$ and 1,300$^{\circ}{\cdots}$ for fabrication of dental prosthesis. The results are as follows: Synthesized powder showed the smallest particle in size, under the conditions of pH 11 and reaction temperature 37$^{\circ}{\cdots}$. The hydroxyapatite was partially converted to $\alpha,\;\beta$-TCP at 1,200$^{\circ}{\cdots}$ and 1,300$^{\circ}{\cdots}$. Mechanical strength of sample was affected by such powder preparation conditions as pH and reaction temperature and sintering temperature. The mechanical strength of sample prepared under the same conditions was increased with increasing pH, reaction temperature and sintering temperature.

  • 참깨박 단백질의 기능개선을 위한 가수분해 조건을 검토한 결과 분리된 참깨박단백질에 대한 각 효소의 최적작용조건은 papain의 경우 pH 6.0, $60^{\circ}C$,효소 대 기질의 비는 기질량에 대해 3%, 기질농도 1.5%에서 최적이었으며, pepsin은 pH 2.0, $50{\sim}60^{\circ}C$, 효소의 농도는 기질량에 대해 3%, 기질농도 1%에서 최적이었다. 그리고 trypsin은 pH 9.0, $60^{\circ}C$, 효소의 농도는 기질량에 대해 1%, 기질의 농도 1%에서 가장 높은 효소작용을 보였다.

Ram Mating Behaviour under Different Social Conditions

  • The present study was conducted to investigate the attributes of ram sexual behaviour under different social conditions. Six intact rams and eight overiectomized ewes were used in this experiment. Ewes were artificially brought into oestrus. All mating traits were recorded every 10 sec during a 1 h mating session under five social conditions viz., ram exposed to single oestrus ewe, ram exposed to single oestrus ewe and audience ram, ram exposed to multiple oestrus ewes, ram exposed to single oestrus ewe with competitor ram and ram exposed to multiple oestrus ewes with competitor ram. Mean intensity of the mating activities during the 1 h mating session under five social conditions was compared. Either vocalization or leg kicking was found to be the predominant teasing activity under all five conditions. Sniffing differed significantly (p<0.05) between different social conditions. Vocalization, leg kicking, and leg kicking with vocalization did not differ significantly (p<0.05) between different social conditions. Mounts/h in the multiple oestrus ewe condition was higher and differed significantly (p<0.05) from other social conditions. Ejaculations/h for the experimental ram was higher when exposed to multiple oestrus ewes in comparison to other social conditions. Mounts/ejaculation was low in the audience ram condition in comparison to the other four social conditions.

  • This study was conducted to evaluate the effects of pH, using gametes, embryos and early development system of three sea urchins species, Hemicentrotus pulcherrimus, Anthocidaris crassispina and Scaphechinus brevis. As the result of performing effects of pH on early embryo development, the conditions of appropriate pH on formation of normal pluteus were pH 7.0-8.0 for H. pulcherrimus, A. crassispina and S. brevis. Otherwise, the conditions of pH 5.0, pH 6.0, pH 9.0 and pH 10.0 damaged the development of early embryos of each testing animal moderately or strongly.
