Production of Bioethanol by Using Beverage Waste (식음료폐기물을 이용한 바이오에탄올 생산)

  • Because beverage waste contains a lot of sugar, it can be used as a valuable resource for energy. But beverage waste is discharged through the water treatment. To prevent the waste of the energy resource, we produced bioethanol by using beverage waste in this study. In order to produce bioethanol, we added distillers stillage and NaOH for fermentation condition (nutrients and pH adjustment). As a results, ethanol concentration was 5.92 vol%. In contrast, ethanol concentration of blank (not added nutrients) was low and fermentation rate was very slow. Because components of the distillers stillage help the yeast growth, fermentation yield and rate was improved. Finally, we operated distillation and dehydration process by using fermented mash and produced fuel bioethanol (more than 99.5 wt%). We think that this results may provide useful information with application of commercial ethanol production using beverage waste.

Chemical/Electro-Chemical Method for Swine Wastewater Treatment (화학적/전기화학적 방법을 이용한 돈사폐수 처리)

  • In a batch mode treatment process, which electrolyzes the wastewater after derivation of N-P crystal formation and recovery, the characteristics of pollutant removal induced with the changes of loading rate and hydraulic retention time were studied. $MgCl_2$ was used as Mg source for the formation of struvite and the molar ratio of $MgCl_2$ to $PO_4^{3-}$ in influent was 1.3. When analyzing the average treatment efficiencies and removal characteristics obtained from four separate operations (Run I, II, III, IV), removal efficiencies of PO43- was not function of its loading rate. Under a condition of sufficient aeration and Mg source provided, over 88% of $PO_4^{3-}$ was eliminated by the formation of MAP without any pH adjustment, in spite of loading rate variation. An optimum-loading rate of NH4-N to achieve high removal efficiency was approximately $100g/m^3/d$. Below that loading rate, the removal of NH4-N was proportional to the loading rate into the system, and hence stable and high removal efficiency, over 90%, was achieved. However, when the loading rate increased over that rate, removal efficiency began to drop and fluctuate. Removal efficiency of TOCs was dependant upon the hydraulic retention time ($r^2$=0.97), not upon the loading rate. Stable and high color removal (94%) was obtained with 2 days of HRT in electrolysis reactor.

Trace-level Determination of N-nitrosodimethylamine(NDMA) in Water Samples using a High-Performance Liquid Chromatography with Fluorescence Derivatization (HPLC와 Fluorescence Derivatization 기법을 이용한 극미량 NDMA의 수질분석)

  • High-performance liquid chromatography(HPLC) and fluorescence derivatization were applied for a trace-level N-nitrosodimethylamine(NDMA) analysis of water samples. Fluorescence intensity was optimized with the excitation wavelength of 340 nm and the emission wavelength of 530 nm. pH adjustment after denitrosation was necessary to get a maximum intensity at pH between 9 and 12. Maximum intensity was found with a dansyl chloride concentration of 330 to 500 mg/L. Percentile error in the water sample analyses through solid phase extraction was 12-162% and 6-23% for the lower concentration level(10-200 ng/L NDMA) and the higher level(100-1000 ng/L NDMA), respectively, showing more discrepancy in lower level. However, the average ratios of estimated NDMA to the standard NDMA were close to 1 for both concentration ranges, presenting this HPLC method could detect from tens to hundreds nanograms NDMA per liter. Accurate determination of NDMA, which was injected to a wastewater effluent, revealed the selectivity of fluorescence derivatization for the target compound(NDMA) in the presence of complex interfering compounds. The HPLC with fluorescence derivatization may be applicable for determining NDMA of water and wastewater samples fur various research purposes.

CONDITIONS FOR PROCESSING OF MEATY TEXTURED FISH PROTEIN CONCENTRATE FROM ALASKA POLLACK AND MACKEREL (명태 및 고등어의 축육과 유사한 어육조직단백질 농축물의 가공조건)

  • For the effective utilization of the coastal fish resources in Korea, an investigation on the optimum processing conditions and the quality of a textured fish protein concentrate similar to the texture of animal meat has been carried out with the fish meat of Alaska pollack and mackerel. A noodle shaped product was prepared with the fish meat paste after the adjustment of pH and salt content. The product was soaked in $96\%$ ethyl alcohol to produce textured fish protein concentrate and then dried. The processing conditions were estimated with the rehydration capacity of the textured fish protein concentrate(FFC). The quality of the final product was evaluated with chemical composition, sensory test and texture measurement. The optimum pH and salt content of the fish meat for the processing of meaty textured FPC were 7.5 and $1.0\%$ respectively. The most effective soaking conditions were as follows:soaking time, 40 min. ; temperature of alcohol, 5 to $20^{\circ}C$;amount of alcohol, 4 times the weight of tile fish meat paste, number of soaking in alcohol, 4 times. The alcohol remaining in meaty textured FPC could be removed effectively by forced air drying. The yield and the contents of protein and lipid in the meaty textured FPC from Alaska pollack were $19.9\%\;84.3\%\;and\;0.5\%$ and those from mackerel were $29.8\%,\;78.1\%\;and\;3.6\%$ respectively. The content of essential amino acid in the meaty textured FPC from Alaska pollack and mackerel was not inferior to that of beef, textured soybean protein and FAO pattern. Beef meat can be substituted with the meaty textured FPC up to $50\%$ in processing meat balls withoutanysignificantlossinthetaste, ordor and texture.

Antimicrobial Effect of Caesalpinia sappan L. Extract on Foodborne Bacteria (식중독 세균에 대한 소목 추출물의 항균 활성)

  • To develop a natural antimicrobial agent, we investigated the antimicrobial activities of 13 species of edible herbal plant extracts against major Gram-positive foodborne bacteria. Among the 13 screened edible herbal plants, Caesalpinia sappan L. showed the highest antimicrobial activity. In the paper disc agar diffusion assay, Caesalpinia sappan L. extracts had strong antibacterial activities against most Gram-positive bacteria but did not have antibacterial activities against most Gram-negative bacteria. Minimum inhibitory concentrations of the ethanol extract were 0.06 mg/mL against Clostridium difficile and Listeria monocytogenes and 0.03 mg/mL against Staphylococcus aureus. Their inhibitory activities were not reduced by heat treatment or pH adjustment against C. difficile, L. monocytogenes, and S. aureus. Antimicrobial activities were higher in ethanol extract than in distilled water extract. These results support the potential use of Caesalpinia sappan L. ethanol extract as an antimicrobial agent or functional food components against Gram-positive bacteria.

Conditions of Water Activity of Raw Material and Adding Levels of Papain and Glucose for Processing Fermented Squid with Low Salt Concentrations (저염 오징어젓갈의 제조를 위한 원료어육의 수분활성도와 papain과 glucose의 첨가조건)

  • To develop methods to produce low salt fermented squid with rich flavor and acceptable shelf life, the optimum processing conditions such as water activity of raw material, amounts of NaCl, papain and gucose were investigated. Water activity of squid meat was adjusted to 0.94 (raw meat), 0.90 and 0.88 by cold air blast and each was salted with 3, 5, or 7% NaCl followed by fermenting at $10^{\circ}C$ for 6 weeks. Amino nitrogen was increased rapidly with high water activity and low NaCl concentration. As a result of organoleptic evaluation it was concluded that optimum conditions were to adjust water activity of raw material to 0.90 and to salt with 5% NaCl. When squid meat adjusting water activity to 0.90 was treated with 0.05, 0.1 and 0.5% papain and fermented at $10^{\circ}C$ for 6 weeks, SDS-PAGE pattern showed rapid breakdown of myofibrilar protein with increasing amounts of papain but the treatment with 0.1% enzyme was best organoleptically. pH values of squid meat added with 1 and 2% glucose were maintained lower than control (glucose 0%) but there were no significant differences between the two glucose treatments. Therefore, it was thought that adding of glucose might be extended shelf life of fermented squid with low salt concentration.

Effects of Amino Acids in Simple Phosphate-Free Media on Pregnancy Rate in Human In Vitro Fertilization and Embryo Transfer(IVF-ET) (Phosphate가 제거된 단순배양액 중 아미노산의 첨가가 체외수정시술 후 임신율에 미치는 영향)

  • The role of amino acids in culture media for IVF-ET was examined in a total of 76 cycles. Patients received clomiphene citrate (CC) followed by hMG or GnRH-a combined with gonadotropins (FSH/hMG) for controlled ovarian hyperstimulation. Severe male (<$4{\times}10^6$ motile sperm) or age factor (>39 y) patients were excluded in this study. Pregnancy was classified as clinical if a gestational sac or fetal cardiac activity was seen on ultrasound. No significant differences were found in age, duration of infertility, follicle size, the level of $E_2$ on the day of hCG injection, the mean number of oocytes retrieved, total motile sperm count, fertilization rate and the mean number of embryos transferred between bHTF (without amino acids) and mHTF (with amino acids) groups. However, total ampules of gonadotropins were higher (p<0.01) in mHTF group than bHTF group. Significantly (p<0.05) more clinical pregnancies were recorded in mHTF group (13/30) compared with bHTF group (9/46). The multiple pregnancy rates were 11.1% in bHTF group and 7.7% in mHTF group. There were one ectopic pregnancy in mHTF group and one heterotopic pregnancy in bHTF group. Abortion rates were 22.2% in bHTF group and 7.7% in mHTF, respectively. The ongoing pregnancy or livebirth rate was significantly (p<0.05) higher in mHTF group (12/30) than bHTF group (7/46). These results suggest that the addition of amino acids in culture media is essential for culture of zygotes in vitro and adjustment of energy substrates in phosphate-free culture media appears to be beneficial for human IVF-ET procedure.

Calcination Condition for Recovery of Calcium from Cuttle Bone and Characteristics of Calcined Cuttle Bone Powder (갑오징어갑으로부터 칼슘의 회수조건 및 소성 칼슘의 특성)

  • For the effective utilization of cuttle bone as a calcium powder, we examined calcination condition ($700^{\circ}C: 0\sim10\;hrs,\;800^{\circ}C:\;0\sim3\;hrs,\;900^{\circ}C:\;0\sim1\;hr\;and\;1,000^{\circ}C:\;0\sim30\;mins$) for recovery of calcium from raw cuttle bone powder (RCB) and characteristics of calcined cuttle bone powder (CCB) treated by optimal condition. During calcination of RCB, the yields was decreased, while total and soluble calcium contents and white index were increased up to constant calcination time ($8\;hrs\;at\;700^{\circ}C,\;2\;hrs\;at\;800^{\circ}C,\;45\;min\;at\;900^{\circ}C\;and\;20\;min\;at\;1,000^{\circ}C$). But, these after that almost unchanged. From these results, the optimal calcination conditions for recovery of calcium from RCB were revealed $8\;hrs\;at\;700^{\circ}C,\;2\;hrs\;at\;800^{\circ}C,\;45\;min\;at\;900^{\circ}C\;and\;20\;min\;at\;1,000^{\circ}C$. In the case of CCB treated for 2 hrs at $800^{\circ}C$, total calcium was about $70\%$, the major component was calcium oxide, and the structure consisted of porosity. The calcium solubility of CCB increased by 22 times compared to RCB. But, the pH of RCB was about 12.9. Therefore, for the effective utilization of RCB as a calcium powder, it requires a suitable modification operation for adjustment of pH ($pH\;2.0\~9.0$).

Vitamin D Deficiency and Related Factors in Patients at a Hospice (일개 호스피스 병동에서 비타민 D 결핍 현황 및 관련인자)

  • Purpose: Although vitamin D deficiency is more commonly found in cancer patient than in non-cancer patients, there have been little data regarding the prevalence of vitamin D deficiency in cancer patients at the very end of life. We examined vitamin D deficiency in terminally ill cancer patients and related factors. Methods: This study was based on a retrospective chart review of 133 patients in a hospice ward. We collected data regarding age, sex, serum 25-hydroxyvitamin D level, cancer type, physical performance, current medications and various laboratory findings. We investigated factors related to serum vitamin D levels after multivariate adjustment for potential confounders. Serum 25-hydroxyvitamin D<20 ng/mL was considered deficient and <10 ng/mL severely deficient. Results: Ninety-five percent of the patients were serum vitamin D deficient. Severe vitamin D deficiency was more common in male patients, non-lung cancer patients, $H_2$ blocker users and non-anticonvulsant users. Elevated levels of serum alanine aminotransferase (ALT) were also associated with low serum vitamin D levels. Multiple regression analysis showed that severe vitamin D deficiency was associated with male gender (aOR 3.82, 95% CI: 1.50~9.72, P=0.005), $H_2$ blocker users (aOR 3.94, 95% CI: 1.61~9.65, P=0.003) and elevated serum ALT levels (aOR 4.52, 95% CI: 1.35~15.19, P=0.015). Conclusion: Vitamin D deficiency was highly prevalent among terminally ill cancer patients. Severe vitamin D deficiency was more common in male patients, $H_2$ blocker users, and patients with elevated ALT levels.

Influence of Dietary Supplemental Germanium Colloid on Egg -Quality and Immune Response in Layers (산란계 사료에 게르마늄의 첨가가 계란품질 및 면역반응에 미치는 영향)

  • This study was conducted to investigate the effects of feeding germanium colloid(GC) on the egg quality and immune response in 1aying hens. One hundred forty four layers, 32-wk-old ISA Brown, were used in a 40-d experimental assay with a 7-d adjustment Period. Dietary treatments included 1) control (basal diet; CON), 2) 0.5ppm GC (basal diet + 0.Sppm germanium), 3) 1.0ppm GC (basal diet + 1.0 ppm germanium) . For overall Period, hen-day e99 Production tended to be increased as the concentration of GC in the diets was increased with significant difference (linear effect, p<0.01) . Laying hens fed 0.5ppm GC diet were significantly (P<0.01) lower in egg shell breaking strength than laying hens fed CON or 1.0 ppm GC diets. Egg shell thickness and yolk color were not influenced by GC supplementation. Laying hens fed 1.0ppm GC diet were h19her in egg yolk Index than laying hens fed CON and 0.5ppm GC diets with significant difference (quadratic effect, p<0.02) . As adding level of germanium colloid increased in the diets, the total serum cholesterol and triglyceride tended to be decreased. However, the effect of GC supplementation did not show significant in the levers of total serum cholesterol and triglyceride. Also, HDL- and LDL+VLDL-cholesterol in serum were not statistically different among the treatments. Red blood cell and white blood cell count in blood tended to be increased as the concentration of GC in the diets was increased with significant difference (P<0.01) . Hematocrit concentration in blood was successfully Increased by supplementation of GC (P<0.01) . In conclusion, although egg quality was not influenced by GC supplementation, supplementing GC Improved Immune status in laying hens.
