Development of Disposable Enzyme-linked Immunosensor Strip Platform (일회용 스트립형 효소면역센서용 플랫폼의 개발)

  • This study introduced the development of a strip type disposable enzyme-linked immunosensor platform for the detection of IgG. Strips of the strip sensor were fabricated by using commercial nitrocellulose filter membranes and a housing holder for the strips was manufactured by using a standard injection molding process for a plastic material. An IgG-urease conjugate was prepared and used for the competitive immune-binding with sample IgG. From the enzymatic reaction between the conjugated urease and urea added, ammonia was generated and caused a localized alkaline pH change on the immobilized antibody band which was coated onto the sensor strips. This pH increase subsequently caused a color change of the antibody band in the presence of a pH indicator, phenol red. Used in conjunction with a competitive immunoassay format, the intensity of the color produced is directly linked with the concentration of target analyte, IgG, and specific measurement of IgG in a lateral flow immunoassay format was achieved over the range 100 ppb to 2000 ppb IgG.

Study on the chemical environment for conformational change of i-motif DNA by atomic force microscopy cantilever (AFM 캔틸레버를 이용한 i-motif DNA의 구조 변화에 미치는 화학적 환경에 대한 연구)

  • Three-dimensional(3D) structure of specific DNA can be changed between two conformations under an external environmental transition such as pH and salt concentration variations. We have experimentally observed the conformational transitions of i-motif DNA using AFM cantilever bioassay. It is shown that pH change of a solvent induces the bending defleciton change of a cantilever functionalized by i-motif DNA. This indicates that cantilever bioassay enables the label-free detection of DNA structural changes upon pH change. It is implied that cantilever bioassay can be a de novo route to quantitatively understand the conformational transitions of biological molecules under environmental changes.

Development of control system for complex microbial incubator (복합 미생물 배양기의 제어시스템 개발)

  • In this paper, a control system for a complex microbial incubator was proposed. The proposed control system consists of a control unit, a communication unit, a power supply unit, and a control system of the complex microbial incubator. The controller of the complex microbial incubator is designed and manufactured to convert analog signals and digital signals, and control signals of sensors such as displays using LCD panels, water level sensors, temperature sensors, and pH concentration sensors. The water level sensor used is designed and manufactured to enable accurate water level measurement by using the IR laser method with excellent linearity in order to solve the problem that existing water level sensors are difficult to measure due to foreign substances such as bubbles. The temperature sensor is designed and used so that it has high accuracy and no cumulative resistance error by measuring using the thermal resistance principle. The communication unit consists of two LAN ports and one RS-232 port, and is designed and manufactured to transmit signals such as LCD panel, PCT panel, and load cell controller used in the complex microbial incubator to the control unit. The power supply unit is designed and manufactured to supply power by configuring it with three voltage supply terminals such as 24V, 12V and 5V so that the control unit and communication unit can operate smoothly. The control system of the complex microbial incubator uses PLC to control sensor values such as pH concentration sensor, temperature sensor, and water level sensor, and the operation of circulation pump, circulation valve, rotary pump, and inverter load cell used for cultivation. In order to evaluate the performance of the control system of the proposed complex microbial incubator, the result of the experiment conducted by the accredited certification body showed that the range of water level measurement sensitivity was -0.41mm~1.59mm, and the range of change in water temperature was ±0.41℃, which is currently commercially available. It was confirmed that the product operates with better performance than the performance of the products. Therefore, the effectiveness of the control system of the complex microbial incubator proposed in this paper was demonstrated.

  • It is well known that the family-mold has an advantage to reduce the cost for production and mold. However, defects are frequently occurred by over packing the smaller volume cavity during molding, especially when the family-mold has a volumetric difference between two cavities. In this study, the cavity-filling imbalance was confirmed by the temperature and the pressure sensors, and a variable-runner system was developed for balancing the cavity-filling. Experiments of balancing the cavity filling was carried out in the family-mold with the variable-runner system, and balancing the cavity-filling was confirmed by changing the cross-sectional area of a runner in the variable-runner system with the temperature and pressure sensors. The influence of the injection speed to the balancing-capability of the variable-runner system was also examined in the experiment.

  • Due to the high surface-to-volume ratio, the 3-dimensional(3D) nanostructures of metal oxides are regarded as the best candidate materials for the chemical gas sensors. Here we have synthesised flower-like 3D zinc oxide nanostructures through a simple hydrothermal route. Specific surface area of the 3D zinc oxide nanostructures synthesised in different pH values from 9.0 to 12.0 were evaluated by using a BET analyzer and the results were compared with that of a zinc oxide thin film fabricated by rf sputtering. Using interdigitated electrodes, superior CO gas sensing properties of the 3D zinc oxide nanostructures on the ZnO thin film to those of the ZnO thin film were demonstrated.

  • Over the past few decades, considerable research has been conducted on field-effect transistor (FET) biosensors; however, other than electrochemical sensors for pH, they have not reached the commercialization stage and still remain at the basic research level. Although several reports have been published on experiments with real biological samples, no reports exist of developments that have reached commercialization or finalized approval for use. In this paper, we explain the reason for the experiments of FET biosensors to induce spurious signals in an experimental setup and explain the existence of misunderstandings regarding the operating principle of FET biosensors owing to the spurious signals. Based on the thoughtful review of the results of previously published papers, we show that the electrochemical read-out principle of FET biosensors requires our intensive understanding of the interfacial potential between the solution and the sensor electrode for further progress in the FET biosensor research.

  • We perform intelligent substation construction of 154kV class with KEPRI. The intelligent substation is build in Gochang 765kV testing facility and consists of electronic instruments, digital control panel, remote monitoring and diagnosis system, and digital relay system. Rogowski coil type CT(RCT) and capacitive voltage divider(CVD) are applicable for instrument transformer of conventional type. Digital control panel(UP) replaces local panel which is drived by mechanical operation. For monitoring condition of GIS and TR, various sensors are used. In this paper, we report the synopsis and the progress state of project.

Highly Sensitive Colorimetric Formaldehyde Gas Sensors using Nylon Sheet and Dye (나일론 시트와 염료를 이용한 고감도 색변환 포름알데히드 가스 센서)

  • A colorimetric sensor was investigated to achieve a low-cost warning device for harmful gaseous formaldehyde (HCHO). The sensor is based on selective reactions between hydroxylamine sulfate and HCHO, leading to the production of sulfuric acid. The produced acid results in color-changing response through the acid-base reaction with dye molecules impregnated on a solid membrane substrate. For attaining this purpose, sensors were fabricated by drop-casting a dye solution prepared using different pH indicators on various commercially available polymer sheets, and their colorimetric responses were evaluated in terms of sensitivity and reliability. The colorimetric sensor using bromophenol blue (BPB) and nylon sheet was found to exhibit the best performance in HCHO detection. An initial bluish green of a sensor was changed to yellow when exposed to gaseous formaldehyde. The color change was recorded using an office scanner and further analyzed in term of RGB distance for quantifying sensor's response at different HCHO(g) concentrations. It exhibited a recognizable colorimetric response even at 50 ppb, being lower than WHO's standard of 80 ppb. In addition, the sensor was found to have quite good selectivity in HCHO detection under the presence of common volatile organic compounds such as ethanol, toluene, and hexane.

  • Smart materials capable of changing their characteristics in response to stimuli such as light, heat, pH, and electric and magnetic fields are promising for application to flexible electronics, soft robotics, and biomedicine. Compared with conventional rigid materials, these materials are typically composed of soft materials that improve the biocompatibility and allow for large and dynamic deformations in response to external environmental stimuli. Among them, smart magnetic materials are attracting immense attention owing to their fast response, remote actuation, and wide penetration range under various conditions. In this review, we report the material design and fabrication of smart magnetic materials. Furthermore, we focus on recent advances in their typical applications, namely, soft magnetic actuators, sensors for self-assembly, object manipulation, shape transformation, multimodal robot actuation, and tactile sensing.

An Implementation of DAQ and Monitoring System for a Smart Fish Farm Using Circulation Filtration System

  • A data acquisition and monitoring system was developed for an automated system of a smart fish farm. The fish farm is located in Jang Hang, South Korea, and was designed as circulation filtration system. Information of every aquaculture pool was automatically measured by pH sensors, dissolved oxygen sensors, and water temperature sensors and the data were stored in the database in a remoted server. Modbus protocol was used for gathering the data which were further used to optimize the pool water quality to predict the rate of growth and death of fish, and to deliver food automatically as planned by the fish farmer. By using JSON protocol, the collected data was delivered to the user's PC and mobile phone for analysis and easy monitoring. The developed monitoring system allowed the fish farmers to improve fish productivity and maximize profits.
