Determination of Thioglycolic acid in the presence of Copper(II) by Adsorptive Stripping Voltammetry (흡착 벗김 전압전류법에 의한 구리이온(II) 존재하에서 티오글리콜산의 정량)

  • Determination method of trace thioglycolate has been studied by adsorptive stripping voltammetry. Copper(II)-thioglycolate complex is adsorbed at the hanging mercury drop electrode and stripped during cathodic scan. Electrolyte was used pH 6.5 phosphate and pH 9.5 borate buffer solutions. Optimal conditions were a copper(II) concentration $1{\times}10^{-4}M$, an adsorption accumulation potential -0.2V, an adsorption accumulation time 60 sec and a scan rate 20mV/sec. A detection limit of $1{\times}10^{-9}M$ thioglycolate was obtained. The method was applied to the determination of thioglycolate in cold wave fluids and depilating creams.

  • Acetohydroxy acid synthase (AHAS) is one of several enzymes that require thiamine diphosphate and a divalent cation as essential cofactors. The active site contains several conserved ionizable groups and all of these appear to be important as judged by the fact that mutation diminishes or abolishes catalytic activity. Recently, we have shown [Yoon, M.-Y., Hwang, J.-H., Choi, M.-K., Baek, D.-K., Kim, J., Kim, Y.-T., Choi, J.-D. FEBS Letters 555 (2003), 185-191] that the activity is pH-dependent due to changes in $V_{max}$ and V/$K_m$. Data were consistent with a mechanism in which substrate was selectively catalyzed by the enzyme with an unprotonated base having a pK 6.48, and a protonated group having a pK of 8.25 for catalysis. Here, we have in detail studied the pH dependence of the kinetic parameters of the cofactors (ThDP, FAD, $Mg^{2+}$) in order to obtain information about the chemical mechanism in the active site. The $V_{max}$ of kinetic parameters for all cofactors was pH-dependent on the basic side. The pK of ThDP, FAD and $Mg^{2+}$ was 9.5, 9.3 and 10.1, respectively. The V/$K_m$ of kinetic parameters for all cofactors was pH-dependent on the acidic and on the basic side. The pK of ThDP, FAD and $Mg^{2+}$ was 6.2-6.4 on the acidic side and 9.0-9.1 on the basic side. The well-conserved histidine mutant (H392) did not affect the pH-dependence of the kinetic parameters. The data are discussed in terms of the acid-base chemical mechanism.

The pH as a Control Parameter for Oxidation-Reduction Potential on the Denitrification by Ochrobactrum anthropi SY 509

  • The pH as a control parameter for oxidation-reduction potential (ORP) was investigated through the denitrification by Ochrobactrum anthropi SY509 under non-growing condition. The optimal pH of nitrate reductase was 7.0, and the minimal ORP level was -250 mV for the denitrification under aerobic condition. In the case of anaerobic condition, the optimal pHs of nitrate and nitrite reductase were shifted to 10.0 and 9.0, respectively, and the minimal ORP levels of nitrate and nitrite reductase were decreased to -370 mV and -340mV, respectively. In the case of alkaline pH and anaerobic condition, the denitrification efficiency of nitrate was increased up to about 2-fold over that of neutral pH and anaerobic condition. Therefore, the combined control of pH and ORP in the anaerobic condition is shown to be an important parameter in the biological denitrification process.

Electrochemical Oxidation of Benzidine and Hydrazobenzene

  • The electrochemistry of benzidine and hydrazobenzene was studied in water-acetonitrile mixed solutions at various pHs and the results are reported. The cyclic voltammetric peak for the oxidation of benzidine shows a pH dependency of -62 mV/pH in the pH range of 0-3.5, no pH dependency between pH values of 3.5 and about 10.5, and of about -50 mV/pH between pH=10.50 and 14.0, indicating that oxidation mechanisms differ depending on the pH of the medium. However, the CV peak for the hydrazobenzene oxidation is shown to be independent of pH of the medium, suggesting that the proton is not involved in the rate limiting step of the electrochemical oxidation of hydrazobenzene to azobenzene. Results of in situ spectroelectrochemical experiments indicate that the oxidation products obtained during the oxidation of benzidine and hydrazobenzene depend on the result of dynamic equilibria taking place at various pHs.

  • The polarographic behaviors of Tl(I) was investigated in EDTA and NTA as supporting electrolytes for determination of Tl(I). A base solution containing 0.005 % gelatine, 0.1 M EDTA, and 0.05 M NTA was used. The halfwave potential of Tl(I) determined is -0.495 V v.s. S.C.E. in 0.1 M EDTA at pH 4.1 and -0.520V v.s. S.C.E. in 0.05 M NTA at pH 6.3. In this paper, the effects of gelatine concentration and pH values was investigated. The half-wave potentials of common elements was determined and compared with the half-wave potential of Tl(I).

Enhanced Production of Gamma-Aminobutyric Acid by Optimizing Culture Conditions of Lactobacillus brevis HYE1 Isolated from Kimchi, a Korean Fermented Food

  • This study evaluated the effects of culture conditions, including carbon and nitrogen sources, L-monosodium glutamate (MSG), and initial pH, on gamma-aminobutyric acid (GABA) production by Lactobacillus brevis HYE1 isolated from kimchi, a Korean traditional fermented food. L. brevis HYE1 was screened by the production analysis of GABA and genetic analysis of the glutamate decarboxylase gene, resulting in 14.64 mM GABA after 48 h of cultivation in MRS medium containing 1% (w/v) MSG. In order to increase GABA production by L. brevis HYE1, the effects of carbon and nitrogen sources on GABA production were preliminarily investigated via one-factor-at-a-time optimization strategy. As the results, 2% maltose and 3% tryptone were determined to produce 17.93 mM GABA in modified MRS medium with 1% (w/v) MSG. In addition, the optimal MSG concentration and initial pH were determined to be 1% and 5.0, respectively, resulting in production of 18.97 mM GABA. Thereafter, response surface methodology (RSM) was applied to determine the optimal conditions of the above four factors. The results indicate that pH was the most significant factor for GABA production. The optimal culture conditions for maximum GABA production were also determined to be 2.14% (w/v) maltose, 4.01% (w/v) tryptone, 2.38% (w/v) MSG, and an initial pH of 4.74. In these conditions, GABA production by L. brevis HYE1 was predicted to be 21.44 mM using the RSM model. The experiment was performed under these optimized conditions, resulting in GABA production of 18.76 mM. These results show that the predicted and experimental values of GABA production are in good agreement.

  • The rate of hydrolysis of insecticide, O,O-diethyl-O-(1-phenyl-3-trifluoromethylpyrazol-5-yl)phosphorothioate (Flupyrazofos) have been investigated in 25% (v/v) aqueous dioxane (${\mu}=0.1M$) at $45^{\circ}C$. The hydrolysis mechanism of flupyrazofos proceeds through the specific acid ($A_{AC}2$) catalysis below pH 4.0, specific base ($B_{AC}2$) catalysis above pH 11.0 and general acid & base ($B_{AC}2$) catalysis between pH 5.0 and pH 10.0 via trigonal-bipyramidal ($d^2sp^3$) intermediate as evidence by solvent effect ($|m|{\ll}|{\ell}|$), rate equation ($kt=ko+k_H+ [H_3O^+]+k_{OH}[OH^-]$) and product analysis. The half-life ($T\frac{1}{2}$) of hydrolytic degradation in neutral media at $45^{\circ}C$ was ca. 3 months.

Preparation and Clinical Application of pH-ISE(Ion Selective Electrode) Based on Tribenzylamine Ionophore (Tribenzylamine Ionophore를 이용한 pH-ISE의 제조 및 임상응용)

  • The pH-ISE(ion selective electrode) based on tribenzylamine as a hydrogen ion carrier was prepared and its electrochemical characterization was studied. It responded linearly to hydrogen ions in the range of pH 3.1 - pH 11.0 and the Nernstian slope showed 55.0 mV/pH (at $20{\pm}0.2^{\circ}C$), it also showed a fast response time of 8 sec. When it was directly applied to human blood(pH 6.0-8.5), we could get the same satisfying results. A good reproducibility and stability were shown with the precision of 2 mV (${\pm}0.1$). The pH-ISE based on tribenzylamine exhibited biocompatibility in clinical applications.

  • Purpose: The purpose of this study was to measure changes in the H-reflex and V wave under loading conditions (e.g. prone and standing position) and to investigate whether postural change would affect the H-reflex and V wave in post stroke hemiplegic patients. Methods: Thirty persons with hemiplegia resulting from stroke (20 males, 10 females) participated in this study. Electromyography (EMG) was used to electrically stimulate and record the soleus H-reflexes and V waves under various loading conditions. The normality of the distribution of each variable (H latency, $H_{max}/M_{max}$ ratio, $V_{max}/M_{max}$ ratio) was tested using the Kolmogorov-Smirnov test. The means of normally distributed continuous data were assessed by independent t-test (${\alpha}$=0.05). Results: There were statistically significant differences in $H_{max}/M_{max}$ ratio (p<0.01), $V_{max}/M_{max}$ ratio (p<0.01), H latency (p<0.01) among the prone and standing position. Conclusion: We found that the H-reflex and V wave in standing position was more active to weight bearing load than prone position.
