Regulation of Glyine max Ornithine Decarboxylase by Salt and Spermine

  • We examined the effect of CsCl and spermine on the induction of ornithine decarboxylase (ODC), a key enzyme in polyamine synthesis form Glycine max axes. Transcription of the ODC gene was induced by 0.1 and 1 mM of CsCl, and the amount of putrescine was increased 3.5-fold by 1 mM CsCl treatment. Spermine also induced the expression of the ODC gene in a die dependent manner. However, CsCl provoked an increase in the active phosphorylated ERK (pERK), a central element of the mitogen-activated protein kinase (MAPK) cascade. Our data demonstrates an interaction between the ODC induction and the MAPK signaling pathway, and suggests that the latter may be involved in cell signaling in salt-stressed plants.

Cardamonin inhibits agonist-induced vascular contractility via Rho-kinase and MEK inhibition

  • The present study was undertaken to investigate the influence of cardamonin on vascular smooth muscle contractility and to determine the mechanism(s) involved. Denuded aortic rings from male rats were used and isometric contractions were recorded and combined with molecular experiments. Cardamonin significantly relaxed fluoride-, phenylephrine-, and phorbol ester-induced vascular contractions, suggesting that it has an anti-hypertensive effect on agonist-induced vascular contraction regardless of endothelial nitric oxide synthesis. Furthermore, cardamonin significantly inhibited the fluoride-induced increase in pMYPT1 level and phenylephrine-induced increase in pERK1/2 level, suggesting inhibition of Rho-kinase and MEK activity and subsequent phosphorylation of MYPT1 and ERK1/2. This study provides evidence that the relaxing effect of cardamonin on agonist-induced vascular contraction regardless of endothelial function involves inhibition of Rho-kinase and MEK activity.

신경세포에서 sirtuin 1이 수상돌기 성장과 가시형성에 미치는 영향 및 기전에 관한 연구 (A Study on the Effects of Sirtuin 1 on Dendritic Outgrowth and Spine Formation and Mechanism in Neuronal Cells)

  • 우울증 발생 기전에 신경가소성의 손상이 관여한다. Sirtuin 1은 신경가소성의 조절에 중요한 역할을 담당하고 있다. 또한 mTORC1 신호전달의 활성화가 신경가소성을 향상시키는 것으로 알려져 있다. 본 연구는 sirtuin 1이 mTORC1 신호전달을 통해 수상돌기 성장과 가시형성에 미치는지 영향을 조사하였다. 덱사메타손이 처치된 신경세포와 정상 배양 신경세포에 resveratrol (sirtuin 1 활성제)과 sirtinol (sirtuin 1 억제제)을 각각 처치하였다. Western blot 분석법을 사용하여, sirtuin 1 발현 및 ERK1/2, mTORC1, p70S6K의 인산화 양을 분석하였고, 면역형광측정법으로 수상돌기의 길이와 가시밀도를 분석하였다. Resveratrol은 덱사메타손 환경에서 농도 의존적으로 sirtuin 1의 발현을 증가시켰으며, ERK1/2 (sirtuin 1의 하위 타겟), mTORC1, 그리고 p70S6K (mTORC1의 하위 타겟)의 인산화를 유의하게 증가시켰다. 또한 resveratrol은 수상돌기 성장과 가시밀도를 증가시켰다. 반면, sirtinol은 정상 배양액에서 sirtuin 1의 발현을 유의하게 감소시켰으며, ERK1/2, mTORC1, p70S6K의 인산화 양을 농도 의존적으로 유의하게 감소시켰다. 또한 sitinol은 수상돌기 성장과 가시밀도를 유의하게 감소시켰다. 신경세포에 sirtuin 1의 siRNA를 transfection시켜 sirtuin 1을 knockdown 시켰을 때, ERK1/2 및 mTORC1의 인산화 양이 감소하였을 뿐만 아니라, 수상돌기 성장과 가시밀도도 감소하였다. 본 연구는 sirtuin 1이 ERK1/2-mTORC1 신호전달을 통해서 수상돌기 성장과 가시밀도를 변화시켜 신경가소성을 조절한다는 것을 보여주었다.

MiR-30a-5p and miR-153-3p regulate LPS-induced neuroinflammatory response and neuronal apoptosis by targeting NeuroD1

  • Neurogenic differentiation 1 (NeuroD1) is an essential transcription factor for neuronal differentiation, maturation, and survival, and is associated with inflammation in lipopolysaccharide (LPS)-induced glial cells; however, the concrete mechanisms are still ambiguous. Therefore, we investigated whether NeuroD1-targeting miRNAs affect inflammation and neuronal apoptosis, as well as the underlying mechanism. First, we confirmed that miR-30a-5p and miR-153-3p, which target NeuroD1, reduced NeuroD1 expression in microglia and astrocytes. In LPS-induced microglia, miR-30a-5p and miR-153-3p suppressed pro-inflammatory cytokines, reactive oxygen species, the phosphorylation of c-Jun N-terminal kinase, extracellular-signal-regulated kinase (ERK), and p38, and the expression of cyclooxygenase and inducible nitric oxide synthase (iNOS) via the NF-κB pathway. Moreover, miR-30a-5p and miR-153-3p inhibited the expression of NOD-like receptor pyrin domain containing 3 (NLRP3) inflammasomes, NLRP3, cleaved caspase-1, and IL-1β, which are involved in the innate immune response. In LPS-induced astrocytes, miR-30a-5p and miR-153-3p reduced ERK phosphorylation and iNOS expression via the STAT-3 pathway. Notably, miR-30a-5p exerted greater anti-inflammatory effects than miR-153-3p. Together, these results indicate that miR-30a-5p and miR-153-3p inhibit MAPK/NF-κB pathway in microglia as well as ERK/STAT-3 pathway in astrocytes to reduce LPS-induced neuronal apoptosis. This study highlights the importance of NeuroD1 in microglia and astrocytes neuroinflammation and suggests that it can be regulated by miR-30a-5p and miR-153-3p.

Hesperetin Inhibits Vascular Formation by Suppressing of the PI3K/AKT, ERK, and p38 MAPK Signaling Pathways

  • Hesperetin has been shown to possess a potential anti-angiogenic effect, including vascular formation by endothelial cells. However, the mechanisms underlying the potential anti-angiogenic activity of hesperetin are not fully understood. In the present study, we evaluated whether hesperetin has anti-angiogenic effects in human umbilical vascular endothelial cells (HUVECs). HUVECs were treated with 50 ng/mL vascular endothelial growth factor (VEGF) to induce proliferation as well as vascular formation, followed by treatment with several doses of hesperetin (25, 50, and $100{\mu}M$) for 24 h. Cell proliferation and vascular formation were analyzed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and tube formation assay, respectively. In addition, cell signaling related to cell proliferation and vascular formation was analyzed by western blot. Furthermore, a mouse aorta ring assay was performed to confirm the effect of hesperetin on vascular formation. Hesperetin treatment did not cause differences in HUVECs proliferation. However, hesperetin significantly inhibited VEGF-induced cell migration and tube formation of HUVECs (P<0.05). Moreover, hesperetin suppressed the expression of ERK, p38 MAPK, and PI3K/AKT in the VEGF-induced HUVECs. In an ex vivo model, hesperetin also suppressed microvessel sprouting of mouse aortic rings. Taken together, the findings suggest that hesperetin inhibited vascular formation by endothelial cells via the inhibition of the PI3K/AKT, ERK and p38 MAPK signaling.

  • Lonicera japonica 에탄올 추출물의 처리에 의하여 LPS에 의해서 활성화된 RAW 264.7 세포에서 NO의 생성량과 TNF-${\alpha}$, IL-$1{\beta}$ 및 IL-6와 같은 염증성 사이토카인의 분비를 억제하였고, MAPK family인 ERK, p38 및 JNK의 인산화와 $I{\kappa}-B{\alpha}$의 분해를 억제하였다. LPS로 유도한 endotoxin shock 동물실험에서 Lonicera japonica 에탄올 추출물 20 mg/kg에서 LPS로 유도한 endotoxin shock에 대한 생존율을 3배 이상 증가시켰으며, 생존시간도 1.3~1.4배 증가시켰다.

Impaired Expression of MAPK Is Associated with the Downregulation of TNF-${\alpha}$, IL-6, and IL-10 in Mycobacterium abscessus Lung Disease

  • Background: Healthy individuals who develop nontuberculous mycobacteria (NTM) lung disease are likely to have specific susceptibility factors which can lead to a NTM infection. The aim of the present study was to investigate the mechanism underlying innate immune responses, including the role of mitogen-activated protein kinase (MAPK), in Mycobacterium abscessus lung disease. Methods: Extracellular signal-regulated kinase (ERK1/2) and p38 MAPK expression in monocytes from peripheral blood mononuclear cells were measured by Western blot analysis after stimulation by Mycobacterium avium in five patients with M. abscessus lung disease and seven healthy controls. A M. avium-induced cytokine assay was performed after inhibition of ERK1/2 and p38 MAPK pathways. Results: Mycobacterium avium induced p38 and ERK1/2 expression in monocytes from healthy controls and subsequently upregulated tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6, and IL-10 production. In monocytes from patients with M. abscessus lung disease, however, induction of p38 and ERK1/2 expression, and the production of TNF-${\alpha}$, IL-6, and IL-10 were significantly lower. Conclusion: Decreased activity of MAPK and cytokine secretion in monocytes from patients with M. abscessus lung disease may provide an explanation regarding host susceptibility to these uncommon infections.

간상피세포에서 BHT와 propyl gallate에 의한 gap junctional intercellular communication 억제 효과 (Inhibition of Gap Junctional Intercellular Communication in Rat Liver Epithelial Cells Induced by BHT and Propyl Gallate)

  • 본 연구에서는 정상 세포인 간상피세포를 이용하여 BHT와 PG가 GJIC에 미치는 효과 및 그 작용 기전을 살펴보았다. BHT와 PG를 WB-F344세포에 각각 1.0mM, 0.75mM 이상 처리한 결과, 세포 생육이 80%이하로 저하되어 세포 독성을 보였고, BHT와 PG의 GJIC에 대한 억제효과는 이보다 낮은 농도인 0.6mM, 0.1mM에서 나타났으며 처리에 따른 Cx43 단백질의 발현 및 인산화를 측정한 결과, BHT와 PG 모두 농도의존적으로 Cx43의 인산화가 증가하였고, Cx43의 구조적 변화와 관련된 MAP kinase는 주요 biomarker인 ERK, p38, JNK의 발현 및 인산화를 측정한 결과, 농도의존적으로 ERK와 p38의 인산화가 증가하는 것으로 나타났다. 이는 WB-F344세포에 BHT와 PG의 처리가 세포독성을 나타내기 전 농도에서 GJIC의 억제하였음을 의미하며 이는 식품첨가물의 안전성 평가에도 활용될 수 있을 것으로 기대된다.

Secretion of MCP-1, IL-8 and IL-6 Induced by House Dust Mite, Dermatophagoides pteronissinus in Human Eosinophilic EoL-1 Cells

  • The house dust mite (Dermatophagoides pteronissinus) is an important factor in triggering allergic diseases. The function of eosinophils, particularly in the production of cytokine or chemokine, is critical in understanding the pathogenesis of inflammatory diseases. In this study, we examined whether D. pteronissinus extract (DpE) induces the expression of monocyte chemotactic protein 1 (MCP-1)/CCL2, IL-8/CXCL8, and IL-6 that mediate in the infiltration and activation of immune cells and in its signaling mechanism in the human eosinophilic cell line, EoL-1. DpE increased the mRNA and protein expression of MCP-1, IL-8, and IL-6 in a time- and dose-dependent course in EoL-1 cells. In our experiments using signal-specific inhibitors, we found that the increased expression of MCP-1, IL-8, and IL-6 due to DpE is associated with Src family tyrosine kinase and protein kinase C $\delta$ (PKC $\delta$). In addition, the activation of extracellular signal-regulated kinase (ERK) is required for MCP-1 and IL-8 expression while p38 mitogen-activated protein kinase (MAPK) is involved in IL-6 expression. DpE induced the phosphorylation of ERK and p38 MAPK. PP2, an inhibitor of Src family tyrosine kinase, and rottlerin, an inhibitor of PKC $\delta$, blocked the activation of ERK and p38 MAPK. DpE induces the activation of ERK and p38 MAPK via Src family tyrosine kinase and PKC $\delta$ for MCP-1, IL-8, or IL-6 production. Increased cytokine release due to the house dust mite and the characterization of its signal transduction may be valuable in understanding the eosinophil-related pathogenic mechanism of inflammatory diseases.

Overexpression of CD44 Standard Isoform Upregulates HIF-1α Signaling in Hypoxic Breast Cancer Cells

  • Cluster of differentiation 44 (CD44), a cell surface receptor for hyaluronic acid (HA), is involved in aggressive cancer phenotypes. Herein, we investigated the role of the CD44 standard isoform (CD44s) in hypoxia-inducible $factor-1{\alpha}$ ($HIF-1{\alpha}$) regulation using MCF7 overexpressing CD44s (pCD44s-MCF7). When pCD44s-MCF7 was incubated under hypoxia, levels of $HIF-1{\alpha}$, vascular endothelial growth factor, and the $HIF-1{\alpha}$ response element-derived luciferase activity were significantly increased compared to those in the control MCF7. Incubation of pCD44s-MCF7 cells with HA further increased $HIF-1{\alpha}$ accumulation, and the silencing of CD44s attenuated $HIF-1{\alpha}$ elevation, which verifies the role of CD44s in $HIF-1{\alpha}$ regulation. In addition, the levels of phosphorylated extracellular signal-regulated kinase (ERK) was higher in hypoxic pCD44s-MCF7 cells, and $HIF-1{\alpha}$ accumulation was diminished by the pharmacological inhibitors of ERK. CD44s-mediated $HIF-1{\alpha}$ augmentation resulted in two functional outcomes. First, pCD44s-MCF7 cells showed facilitated cell motility under hypoxia via the upregulation of proteins associated with epithelial-mesenchymal transition, such as SNAIL1 and ZEB1. Second, pCD44s-MCF7 cells exhibited higher levels of glycolytic proteins, such as glucose transporter-1, and produced higher levels of lactate under hypoxa. As a consequence of the enhanced glycolytic adaptation to hypoxia, pCD44s-MCF7 cells exhibited a higher rate of cell survival under hypoxia than that of the control MCF7, and glucose deprivation abolished these differential responses of the two cell lines. Taken together, these results suggest that CD44s activates hypoxia-inducible $HIF-1{\alpha}$ signaling via ERK pathway, and the $CD44s-ERK-HIF-1{\alpha}$ pathway is involved in facilitated cancer cell viability and motility under hypoxic conditions.
