Nucleotide Sequences of Rep and CAT Proteins encoded by Chloramphenicol-Resistance Plasmid pKH7 (클로람페니콜 내성 플라스미드 pKH7의 Rep 단백질과 CAT 단백질의 염기서열 분석)

  • The nucleotide sequence of Xbal-Mbol fragment of pKH7, a chloramphenicol-resistant($Cm^{r}$) plasmid isolated from multidrug-resistant S. aureus SA2, has been determined. Xbal-Mbol fragment of pKH7 was found to contain two ORFs. One ORF encoded Rap and the other encoded CAT protein. The deduced amino acid sequences of Rep and CAT of pKH7 were compared to those of pUB112 and pC221. Comparisons revealed that there was one amino acid difference in CAT between pKH7 and pUB112. CAT of pKH7 exhibited 98.6% amino acid identity to that of pC221. In case of Rep proteins, a slightly lower homology of 96.4% and 86.7% in amino acid sequences was observed between pKH7 and pUB112 and between pKH7 and pC221, respectively.

  • The HIV-1 p24(202-221) sequence ETINNEEEWDRVHPV HAGP contains a B-cell epitope with the earliest immune response and the highest antibody titer against anti-mouse sera obtained by immunization with p24 antigens. A novel mouse monoclonal antibody (mAb) was generated against the immunodominant B-cell epitope of the HIV-1 p24 capsid protein, p24(202-221). BALB/c mice were immunized with the four branched multiple antigenic peptide (MAP) containing the HIV-1p24(202-221) sequence, and antibody-secreting hybridoma were produced by fusion of mouse splenocytes with P3X63Ag8.653, mouse myeloma cells. One clone which produced the antigen-specific mAb named KI-24 (Isotype IgG1, light chain: ${\kappa}$) was identified. mAb KI-24 was highly specific for both the p24(202-221) and p24 proteins when analyzed by ELISA and Western blotting. Since p24(202-221) also contains a cytotoxic T-lymphocyte epitope, this specfic peptide epitope and the monoclonal antibody with specific reactivity against the p24 protein and p24(202-221) can be used in peptide vaccine development and p24 antigen detection from HIV patients.

  • The nucleotide sequence of inducible chloramphenicol acetyl-transferase(CAT) gene isolated from a small plasmid pSBK203 of Staphylococcus aureus was determined. The base sequence shows that structural gene of pSBK203-CAT encodes a protein of 213 amino acids and has a leader region which encodes a short polypeptide of 9 amino-acids in its upstream. vertical bar /sup 35/S vertical bar-Methionine labelled CAT gene product in minicell showed almost same mobility with pC194-CAT of which molecular weight is 24Kdal on polyacrylamide gel electrophoresis. Predicted amino acid sequence of pSBK203-CAT has revealed a high degree of homology with the CATs of pC194 and pC221 than those of cat-86, Tn9 and proteus mirabilis PM13.

Adsorption of Dyes Reactive Blue 221, N Blue RGB and Acid Blue MTR on Two Different Samples of Activated Carbon

  • Removal of dyes Reactive Blue 221, N Blue RGB and Acid Blue MTR using two different samples of activated carbon by static batch method was studied. Experimental data on optical density of solutions at different concentrations ranging from 10 to 100 mg/L and of solutions after adsorption on activated carbon samples were measured. Calibration curves were plotted and the amount of dye $q_e$ adsorbed was calculated. The data was fitted to Langmuir and Freundlich isotherms for two different carbon samples and different concentration and pH values. Constants were calculated from the slope and intercept values of the isotherms. Coefficient of correlation $R_2$ and Standard Deviation SD were also noted. The data fitted well to the isotherms. Carbon sample $C_1$ showed higher potential to adsorb all the three dyes. Adsorption was higher at lower concentrations. Carbon sample $C_2$ showed better adsorption in acidic pH as compared to in alkaline pH. From the analysis of the data capacity of $C_1$ and $C_2$ to remove the dyes from water have been compared.

Rod Migration into the Posterior Fossa after Harms Operation : Case Report and Review of Literatures

  • C1 lateral mass and C2 pedicle (C1LM-C2P) fixation is a relatively new technique for atlantoaxial stabilization. Complications from C1LM-C2P fixation have been rarely reported. The authors report unilateral rod migration into the posterior fossa as a rare complication after this posterior C1-C2 stabilization technique. A 23-year-old man suffered severe head trauma and cervical spine injury after vehicle accident. He was unconscious for 2 months and regained consciousness. He underwent C1LM-C2P fixation for stabilization of type II odontoid process fracture described by Harms. The patient recovered without a major complication. Twenty months after operation, brain computed tomogram performed at psychology department for disability evaluation showed rod migration into the right cerebellar hemisphere. The patient had mild occipital headache and dizziness only regarding the misplaced rod. He refused further operation for rod removal. To our knowledge, this complication is the first report regarding rod migration after Harms method. We should be kept in mind the possibility of rod migration, and C1LM-C2P fixation should be performed with meticulous technique and long-term follow-up.

Arylation of Styrene by Palladium Acetate-Phosphine Complexes

  • When phenylation of styrene was carried out in the presence of Pd(OAc)2 and PPh3 in benzene, trans-stilbene was obtained in good yield (566%) with high selectivity (98%) under mild condition (55 ℃, 50 psi O2, 20 h). Since trans-stilbene could be produced not only from benzene but also from phenyl group of PPh3 by migration of its phenyl group to Pd, the competitiveness of benzene and the migratory aptitude of aryl group of triarylphosphine toward styrene has been investigated with various phosphines (PR3: P(p-C6H4CH3)3, P(p-C6H4OCH3)3, P(p-C6H4F)3, P(p-C6H4Cl)3, P(C6H5)3, P(C6H11)3, P(OC4H9n)3, P(CH2C6H5)3 and P(C6F5)3). The yield and selectivity toward trans-stilbene are increased as the basicity of the phosphines increases. The composition of arylated olefin from arylphosphine, in turn, increases as the electronegativity of the substituent on the aryl group of arylphosphines increases.

Effect of Monensin and Fish Oil Supplementation on Biohydrogenation and CLA Production by Rumen Bacteria In vitro When Incubated with Safflower Oil

  • An in vitro study was conducted to examine the effect of monensin or fish oil addition on bio-hydrogenation of $C_{18^-} unsaturated fatty acids and CLA production by mixed ruminal bacteria when incubated with safflower oil. Commercially manufactured concentrate (1%, w/v) with safflower oil (0.2%, w/v) were added to mixed solution (600 ml) of strained rumen fluid and McDougalls artificial saliva (control). Monensin $Rumensin^{(R)}$, 10 ppm, w/v, MO), mixed fish oil (0.02%, w/v, absorbed to 0.2 g alfalfa hay, FO) or similar amounts of monensin and fish oil (MO+FO) to MO and FO was also added into the control solution. All the culture solutions prepared were incubated in the culture jar anaerobically at $39^{\circ}C$ up to 12 h. Higher pH (p<0.047) and ammonia concentration (p<0.042) were observed from the culture solution containing MO at 12 h incubation than those from the culture solutions of control or FO. The MO supplementation increased (p<0.0001-0.007) propionate proportion of culture solution but reduced butyrate proportion at 6 h (p<0.018) and 12 h (p<0.001) of incubations. Supplementation of MO or MO+FO increased (p<0.001) the proportions of $C_{18:2}$. The MO alone reduced (p<0.022-0.025) the proportion of c9,t11-CLA compared to FO in all incubation times. The FO supplementation increased the proportion of c9,t11-CLA. An additive effect of MO to FO in the production of c9,t11-CLA was observed at 6 h incubation. In vitro supplementation of monensin reduced hydrogenation of $C_{18^-}$UFAs while fish oil supplementation increased the production of CLA.

  • Tetrahomodioxa p-phenylcalix(4)arene dihexylether(C/sub66/H/sub68/O/sub6/) has been synthesized and structurally characterized by X-ray diffraction. Reaction of tetrahomodioxa p-phenylcalix(4)arene with hexyl halide and NaH in DMF leads to the dihexyl derivatives, 7,13,21,27-tetraphenyl-29,31-dihexyloxy- 2,3,16,17-tetrahomo-3,17-dioxacalit(4)arenes. The crystal is orthorhombic, P2₁2₁2₁, a= 9.764(2), b=16.167(2), c=32.994(3) Å, V=5208(1) Å, Z= 4, Dc = 1.221 gcm/sup -3/. The structure was solved by direct methods and refined by full-matrix least squares. Refinement converged at R = 0.070 for 2009 observed reflections. This molecule has a C-1,2-alternate conformation with pseudo-centrosymmetry and has two pairs of opposite phenyl rings, which are approximately parallel to each other. The benzene rings A and B are up, and the rings C and D rings are down with respect to the plane of the macrocyclic ring.

  • Water in the supercritical state ($T{\geq}374^{\circ}C$, $p{\geq}221$ atm) is a good solvent for nonorganic pollutants, but it is extremely corrosive. Subcritical Water Extraction (SWE) is a very fast and an efficient method to extract nonpolar environmental pollutants adsorbed on the sediments and soils. Many nonpolar organic compounds are sufficiently soluble to be extracted to the water under subcritical conditions. Complete extraction of PCBs from the sediments and soils takes only a few minutes by applying SWE with the subcritical water at 50 atm and at $260^{\circ}C$.
