Characteristocs of the Somatotypes of Korean Farmer (한국 남녀 농업인의 체형 특성)

  • This study was to research the characteristics of the somatotype of Korean farmers. 836 Korean farmers(Male: 361 persons, Female: 475 persons) volunteered for this study. They volunteered from 9 different area such as Kumi, Damyang, Iksan, Injae, Chungju, Choongju, Haman, Whasung, and Whasoon. The reusults were as follows: 1. The subcutaneous body fat thickness were decreased as they aged(p<.05). 2. The thicker the subcutaneous body fat thickness of Korean farmer, the wider the circumference size by the Korean famer's physical parts(p<.05-p<.001). 3. There were no significant differences in the circumference size such as upper arm, chest, bust, waist, umbilical waist, hip, and calf between Korean farmers and Korean standard size. 4. Korean women farmer showed significantly differences in the upper arm circumference(p<.01), the chest circumference(p<.01), the bust circumference(p<.01), the waist circumference(p<.01), the umbilical waist circumference(p<.01), and the calf circumference(p<.05) from Korean women standard by ages. Especially, Korean women farmer had thicker the umbilical waist circumference(the 40's: 103.2mm, the 50's 52.3mm, the 60's 22.3mm, over 70's 22.1mm) than Korean women standard. 5. The 40's women farmer had thicker the upper arm circumference, the chest circumference, the bust circumference, the waist circumference, the umbilical waist circumference, and the calf circumference than Korean women standard. The 70's women farmer had skinner circumferences except the umbilical waist circumference than Korean women standard. 6. Korean farmers showed the crooked back shape, the broad-knee length and also front-bending knee shape as they aged.

Thermal Stability of the Major Color Component, Cyanidin 3-glucoside, from a Korean Pigmented Rice Variety in Aqueous Solution (한국산 유색미의 주요성분인 Cyanidin 3-glucoside의 수용액에서의 열안정성)

  • Thermal stability of the major color component, cyanidin 3-glucoside, isolated from Korean pigmented rice (Oryza sativa var. Suwon 415) were investigated to explore possible application of value-added natural colors as food additives. The anthocyanin showed red and blue color with maximum absorption peaks at 511 nm and 572 nm in acidic (pH 2.0) and alkaline (pH 9.0) buffer solutions, respectively, and the thermal degradation reactions were carried out with different temperature ranges at $50{\sim}95^{\circ}C$. Degree of degradation was determined with UV/Vis spectra which indicate characteristic absorption patterns with sharp isosbestic points at 350 nm (pH 2.0), and 275, 310, and 405 nm (pH 9.0). Thus the reaction follows simple first-order kinetics. The anthocyanin was very stable against heat at acidic pH and relatively stable at alkaline pH with half-life values of 50.3 hr and 0.6 hr at $70^{\circ}C$, respectively. The activation energies and Arrhenius frequency factors of the pigment were 26.9 kcal $mol^{-1}\;and\;6.0{\times}10^{11}\;s^{-1}$, at pH 2.0, and 15.2 kcal $mol^{-1}\;and\;1.4{\times}10^{6}\;s^{-1}$, pH 9.0, and respectively.

Discrimination of Geographical Origin for Herbal Medicine by Mineral Content Analysis with Energy Dispersive X-Ray Fluorescence Spectrometer (에너지분산형 X-선 형광분석기를 이용한 한약재의 무기질 분석 및 이에 의한 원산지 판별)

  • In this study, the macromineral content ratios of four herbal medicine samples(Saposhnikoviae Radix, Bupleuri Radix, Cnidii Rhizoma, and Astragali Radix) were analyzed to discriminate their geographical origins using an energydispersive x-ray fluorescence (EDXRF) technique. EDXRF is a rapid, non-destructive, and multi-elemental analysis technique. Initially, samples of both domestic and imported herbal medicines were pulverized, and then their macromineral contents, including P, S, K, and Ca, were analyzed using EDXRF. For the discrimination of their geographical origins, canonical discriminant analysis was carried out based on the estimated macromineral relative content ratios of the samples. According to the results, the discrimination accuracies were as follows: 93.3% for Saposhnikoviae Radix, 95.7% for Bupleuri Radix, 98.8% for Cnidii Rhizoma, and 87.5% for Astragali Radix. Overall, the results imply that this technique could be used as a standard method, to discriminate their geographical origins between domestic and imported herbal medicines.

Isolation and Characterization of Acinetobacter sp. BD5 Producing Lipolytic Enzyme (Lipolytic 효소를 생산하는 Acinetobacter sp. BD5 균주의 분리 및 특성)

  • A bacterium producing novel lipolytic enzyme was isolated from house sewage and identified as Acinetobacter sp. BD5 based on physiological characterization and 16S rDNA sequencing. The lipolytic activity of Acinetobacter sp. BD5 was tested using an EL agar medium and CE agar medium supplemented with 1% tributyrin and olive oil, respectively. The formation of a clear zone around the colony was detected by agar medium supplemented with 1% tributyrin and olive oil, respectively and Acinetobacter sp. BD5 formed powder-like zone around the colony on LB agar medium containing Tween 20. The quantitative lipolytic activity was determined by using p-NP butyrate as substrate. Acinetobacter sp. BD5 secreted the lipolytic enzyme during exponential growth phase, reaching a maximum amount after 6 hours of incubation. The lipolytic enzyme was found to be optimally active at $60^{\circ}C$ and retained more than 70% at $70-80^{\circ}C$. It displayed a high degree of activity in a pH of 7.0 to 10.6, with an optimal pH of 9.0.

Metabolic Elasticity and Induction of Heat Shock Protein 70 in Labeo rohita Acclimated to Three Temperatures

  • The metabolic response of Labeo rohita to thermal acclimation was assessed. Advanced fingerlings of L. rohita (average weight $31{\pm}1.4g$) were acclimated to 31, 33 and $36^{\circ}C$ compared with ambient temperatures ($26^{\circ}C$) for 30 days and different enzymes associated with stress response were estimated. Glycolytic enzyme-Lactate dehydrogenase, (LDH, E.C.1.1.1.27), TCA cycle enzyme-Malate dehydrogenase (MDH, E.C.1.1.1.37), Protein metabolizing enzymes-Aspartate amino transferase (AST, E.C.2.6.1.1) and Alanine amino transferase (ALT, E.C.2.6.1.2) of liver, gill and muscle, Gluconeogenic enzymes-Fructose 1,6 Bi phosphatase (FBPase, E.C. 3.1.3.11) and Glucose 6 phosphatase (G6Pase, E.C. 3.1.3.9) of liver and kidney were significantly (p<0.05) different with increasing acclimation temperatures. Heat Shock Protein-70 (HSP-70) was expressed in increasing intensity at 31, 33 and $36^{\circ}C$ but was not expressed at $26^{\circ}C$. Results suggest that higher acclimation temperatures enhance metabolism and L. rohita maintains homeostasis between $26-36^{\circ}C$ via an acclimation episode. Such adaptation appears to be facilitated by resorting to gluconeogenic and glycogenolytic pathways for energy mobilization and induction of HSPs.

CHIP promotes the degradation of mutant SOD1 by reducing its interaction with VCP and S6/S6' subunits of 26S proteasome

  • Previously we showed that CHIP, a co-chaperone of Hsp70 and E3 ubiquitin ligase, can promote the degradation of mutant SOD1 linked to familial amyotrophic lateral sclerosis (fALS) via a mechanism not involving SOD1 ubiquitylation. Here we present evidence that CHIP functions in the interaction of mutant SOD1 with 26S proteasomes. Bag-1, a coupling factor between molecular chaperones and the proteasomes, formed a complex with SOD1 in an hsp70-dependent manner but had no direct effect on the degradation of mutant SOD1. Instead, Bag-1 stimulated interaction between CHIP and the proteasome-associated protein VCP (p97), which do not associate normally. Over-expressed CHIP interfered with the association between mutant SOD1 and VCP. Conversely, the binding of CHIP to mutant SOD1 was inhibited by VCP, implying that the chaperone complex and proteolytic machinery are competing for the common substrates. Finally we observed that mutant SOD1 strongly associated with the 19S complex of proteasomes and CHIP over-expression specifically reduced the interaction between S6/S6' ATPase subunits and mutant SOD1. These results suggest that CHIP, together with ubiquitin-binding proteins such as Bag-1 and VCP, promotes the degradation of mutant SOD1 by facilitating its translocation from ATPase subunits of 19S complex to the 20S core particle.

New Korean record of the Banded Driftfish, Psenes arafurensis (PISCES: Nomeidae)

  • Psenes arafurensis, belonging to the family Nomeidae, is described based on four specimens (184.8-199.0 mm in standard length) collected from Korea. The species is most similar to the congeneric species P. maculatus and P. pellucidus, but differs from them in the number of lateral-line scales. (44-47 in P. arafurensis vs 67-70 in P. maculatus and 120 in P. pellucidus) and the number of vertebrae (31 in P. arafurensis vs 34-38 in P. maculatus and 40-42 in P. pellucidus). The present study is a new record of P. arafurensis with voucher specimens from Korean waters.

Changes of Chemical Composition during the Aging of Liquid Koji Kochujang (액체국에 의한 고추장 양조중의 성분 변화)

  • The three kinds of Kochujang were prepared with liquid koji equivalent to 30%, 50%, and 70% of brewing water. The qualities of the Kochujangs during aging at $25^{\circ}C$ for 4 months were compared with those of the control group which was made from solid koji. There were no significant differences in the contents of moisture, crude protein, crude fat, NaCl and ethyl alcohol between test groups. The contents of amino nitrogen, soluble nitrogen, reducing sugar and pH of each test group during Kochujang aging were high in the order of control group. 70% liquid koji group, 50% liquid koji group and 30% liquid koji group. The pH of control group was over 5.0 during aging period. The titratable acidity of Kochujang was high in the 50% liquid koji group and the 30% liquid koji group.

Nutrient Removal Hybrid Process to Use Suspended and Attached Growth Microorganisms and Apply the Contact and Stabilization Process (부유 및 부착성장 미생물을 이용한 접촉안정형 영양염류처리 하이브리드 공정)

  • Nutrient removal hybrid process to use suspended and attached growth microorganisms and apply the contact and stabilization process was process obtaining good results to HRT within 6 hours to dominate nitrifier and to promote separation and growth of autotrophs and heterotrophs to pack with EPS(Expanded Poly-Styrene) media in nitrification reactor. An average effluent quality of this process was below 5.2 mg/L, 7.3 mg/L, 4.9 mg/L as $BOD_5,\;COD_{Mn}$, SS concentration and 6.8 mg/L, 0.6 mg/L as T-N, T-P concentration. Also, An average removal efficiency of this process was 94.6%, 79.8%, 94.9% as $BOD_5,\;COD_{Mn}$, SS and 70.8%, 76.9% as T-N, T-P. 16S-rRNA analysis result of microorganisms attached to EPS media was researched Nitrosomonas and Nitrosococcus blown to ammonia-oxidizing bacteria cluster to include Gallionella and these microorganisms were researched to involve about 6% of biofilm attached EPS media. Consequently, this process was treated below 10 mg/L and 1.0mg/L as T-N, T-P concentration at short hydraulic retention time(about 6 hr) to dominate nitrifier.
