In our laboratory a R plasmid pKU10 was isolated from Pseudomonas and its characteristics were investigated. In this study, as a basic work to improve its utility as a cloning vehicle, restriction patterns of pKU10 were analyzed for other various restriction enzymes in addition to restriction evdonucleases previously examined. As a result, pKU10 DNA has two cleavage sites for ClaI and HpaI, and three sites for AvaI. The restriction map of pKU10 was supplemented with AvaI, ClaI, and HpaI. From the result of this experiment, the usefulness of PKU10 as a cloning vector in Pseudomonas will be enhanced by constructions of mini-plasmid or hybrid plasmids.
The pharmacokinetics of orally administered paclitlxel (50 mg/kg) was studied in six rabbits after 1hr pretreatment (2.0 mg/kg and 10 mg/kg) of tetramethoxyflavone or coadministration of (2.0 mg/kg, 10 mg/kg and 20 mg/kg) tetramethoxyflavone. The area under the plasma concentration-tine curve (AUC) and plasma concentration of paclitaxe1 coadministered with tetramethoxyflavone (10 mglkg) were increased significantly (p<0.05) compared with control. However, coadministration of tetramethoxyflavone (2 and 20 mg/kg) showed no significant effect on the pharmacokinetic parameters of paclitaxel. Pretreatment with tetramethoxyflavone significantly (p<0.05) increased the plasma concentration of paclitaxel. The area under the plasma concentration-time curve (AUC) and the peak concentration (C$_{max}$) of paclitaxel pretreated with tetramethoxyflavone were increased significantly (p<0.01, p<0.05) compared with control. The terminal half. life of paclitaxel pretreated with tetramethoxyflavone (2 mg/kg and 10 mg/kg) was significantly (p<0.05) prolonged compared with control. Pretreatment with tetramethoxyflavone (2.0 mg/kg, 10 mg/kg) significantly (p<0.01, p<0.05) increased the absolute bioavailability of paclitaxel compared with the control (154∼179%). On the basis of the results, it might be considered that tetramethoxyflavone may inhibit cytochrome P450 or P-glycoprotein efflux pump which are engaged in paclitaxel metabolism, result in increased AUC and t$_{1}$2/ of paclitaxel. However, further study should be conducted to clarify the roles of cytochrome P450 and P-glycoprotein on paclitaxel bioavailability with/or without tetramethoxyflavone. Hydrolysis reactions of S-phenyl-S-vinyl-N-p-tosylsulfilimine (VSI) and its derivatives at various pH have been investigated kinetically. The hydrolysis reactions produced phenylvinylsulfoxide and p-toluene sulfonamide as the products. The reactions are first order and Hammett ρ values for pH 1.0, 6.0, and 11.0 are 0.82, 0.45, and 0.57, respectively. This reaction is not catalyzed by general base. The plot of k vs pH shows that there are three different regions of the rate constants $(k_t)$ in the profile.; At pH < 2 and pH > 10, the rate constants are directly proportional to the concentrations of hydronium and hydroxide ion catalyzed reactions, respectively. The rate constant remains nearly the same at 2 < pH < 10. On the bases of these results, the plausible hydrolysis mechanism and a rate equation have been proposed: At pH < 2.0, the reaction proceeds via the addition of water molecule to sulfur after protonation at the nitrogen atom of the sulfilimine, whereas at pH > 10.0, the reaction proceeds by the addition of hydroxide ion to sulfur directly. In the range of pH 2.0-10.0, the addition of water to sulfur of sulfilimine appears to be the rate controlling step.
The effect of P. agarici and P. tolaasii causing the bacterial disease of mushrooms on the mycelial growth and fruitbody formation of F. velutipes was evaluated in laboratory. When the pathogenic bacteria was inoculated simultaneously with F. velutipes or 5 days after inoculation of F. velutipes, they significantly deterred both mycelial growth and fruitbody formation of F. velutipes in sawdust culture and showed strong inhibition under high population density. They appeared to be tender or milky in exhibiting symptom on F. velutipes by inoculating the concentration of $10^2{\sim}10^6$ of unit/g media, and their growth seemed to be stopped under $10^8\;cfu/g$ media. On $10^2\;cfu/g$ media of P. agarici and $10^4\;cfu/g$ media of P. tolaasii, there was no effect on the fruitbody yield of F. velutipes. P. tolaasii was more suppressive in the mycelial growth of F. velutipes than P. agarici, while on fruitbodies formation of F. velutipes, P. agarici showed slightly higher inhibition than that of P. tolaasii. When the bacteria was inoculated 10 days after inoculation of F. velutipes, both mycelial growth and fruit body formation were not affected nearly.
In this experiment, we examined the survival, fertilization, hatching times, and hatching rates of Far Eastern catfish Silurus asotus at pH ranging from 2 to 13 under laboratory conditions. Eggs could be fertilized at pH 3-12. In a hatching experiment, mortality was first observed at pH 13, when all fertilized eggs died within 8 min, followed by pH 2 (30 min), pH 12 (60 min), pH 3 (4 h), and pH 11 (5 h). Hatching only occurred at pH 4-10, with the highest hatching rate at pH 7 (52%) and the lowest at pH 10 (24%). Hatching rates in acid solutions were higher than in alkaline solutions, although the difference was not significant. Hatching was first observed at pH 10, beginning 27 h after fertilization and ending at the 31 h. A clear difference was observed between hatching times, ranging from 31 to 64 h and increasing in order with descending pH.

This paper proposes a balance-swap method for the dynamic economic load dispatch problem. Based on the premise that all generators shall be operated at valve-points, the proposed algorithm initially sets the maximum generation power at $P_i{\leftarrow}P_i^{max}$. As for generator i with $_{max}c_i$, which is the maximum operating cost $c_i=\frac{F(P_i)-F(P_{iv_k})}{(P_i-P_{iv_k})}$ produced when the generation power of each generator is reduced to the valve-point $v_k$, the algorithm reduces i's generation power down to $P_{iv_k}$, the valve-point operating cost. When ${\Sigma}P_i-P_d$ > 0, it reduces the generation power of a generator with $_{max}c_i$ of $c_i=F(P_i)-F(P_i-1)$ to $P_i{\leftarrow}P_i-1$ so as to restore the equilibrium ${\Sigma}P_i=P_d$. The algorithm subsequently optimizes by employing an adult-step method in which power in the range of $_{min}\{_{max}(P_i-P_i^{min}),\;_{max}(P_i^{max}-P_i)\}$>${\alpha}{\geq}10$ is reduced by 10; a baby step method in which power in the range of 10>${\alpha}{\geq}1$ is reduced by 1; and a swap method for $_{max}[F(P_i)-F(P_i-{\alpha})]$>$_{min}[F(P_j+{\alpha})-F(P_j)]$, $i{\neq}j$ of $P_i=P_i{\pm}{\alpha}$, in which power is swapped to $P_i=P_i-{\alpha}$, $P_j=P_j+{\alpha}$. It finally executes minute swap process for ${\alpha}=\text{0.1, 0.01, 0.001, 0.0001}$. When applied to various experimental cases of the dynamic economic load dispatch problems, the proposed algorithm has proved to maximize economic benefits by significantly reducing the optimal operating cost of the extant Heuristic algorithm.
Fifty-four PIC barrows were used to evaluate the effects of lower dietary lysine content and energy level on carcass characteristics and meat quality in slaughter pigs. Pigs were allotted to one of three treatments by body weight with six replicate pens in each treatment. The dietary treatments for body weights of 20-50 kg, 50-80 kg and 80-90 kg were as follows, respectively: control diet (digestible energy 14.22 MJ/kg, lysine/DE 0.67 g/MJ, 0.53 g/MJ and 0.42 g/MJ); a low lysine group (digestible energy 14.22 MJ/kg, lysine/DE 0.49, 0.38 and 0.30 g/MJ); and a low lysine-low energy group or low nutrient group (digestible energy 13.11 MJ/kg, lysine/DE 0.49, 0.38 and 0.30 g/MJ). The daily weight gain, daily feed intake and feed efficiency were calculated in the overall growth period (nearly 12 weeks). Meanwhile, carcass characteristics and meat quality were evaluated at 60 and 90 kg body weight respectively. During the overall growth trial, lowering dietary lysine and nutrient level both decreased weight gain (p<0.05) and feed efficiency (p<0.01). At 60 kg body weight, decreasing dietary lysine and nutrient level noticeably decreased dressing percentage (p<0.01) and back fat depth at last rib of PIC pigs (p<0.01), but enhanced marbling scores (p<0.10), intramuscular fat content (p<0.10) and water loss rate (p<0.01) of the longissimus dorsi muscle. At 90 kg body weight, lean percentage (p<0.01) was evidently reduced by both lowering lysine content and nutrient level in the diet. However, the shoulder back fat depth (p<0.05) and marbling scores of the loin eye muscle (p<0.05) were increased; Lowering dietary nutrient level could improve back fat depth of 10th rib (p<0.01) and last rib (p<0.01), intramuscular fat content (p<0.10), redness (p<0.01) and water loss rate of the loin eye muscle (p<0.05), but decrease loin area (p<0.05). Finally, when comparing the 60 kg and 90 kg slaughter weights, it was found that the shoulder back fat depth (p<0.01, p<0.10), 6th-7th rib (p<0.01, p<0.01), 10th-rib (p<0.01, p<0.01) and last rib back fat depth (p<0.01, p<0.01) of the low lysine and low nutrient group were all obviously increased comparing with the control group. Taken together, the results showed that decreasing dietary lysine content and nutrient level increased intramuscular fat content and water loss rate of longissimus dorsi muscle; On the other hand, both lowering dietary lysine and nutrient level markedly compensated to increase back fat deposition in the later finishing period (body weight from 60 to 90 kg) in contrast to the control group.
This study was conducted to investigate weight control behavior and obesity stress of college women. The data were collected from September 7 to September 13 of 2013 and the subjects were 213 college women in C city of Gyoung-nam Province. The data were analyzed by SPSS 20.0 program using descriptive statistics, t-test, ANOVA, Scheffe's test. The score of obesity stress was moderate($18.26{\pm}5.66$). The level of obesity stress was affected by various general characteristics and weight control behavior characteristics including economic status(F=3.99, p=0.020), figure recognition(F=14.41, p<.001), satisfaction of body figure(F=15.88, p<.001), interest of weight control(F=4.82, p=.001), control of food amount(F=2.41, p=.050), body figure compulsion(F=24.06, p<.001), experience of dieting (F=6.04, p<.001), sleeping hours(t=2.10, p=.036), desired losing weight (F=10.47, p<.001). Therefore, it is necessary to be considered these variables during development of body weight control programs for college women.
It is well-known that electrical field stimulation (EFS)-induced contraction is mediated by a cholinergic mechanism and other neurotransmitters. NO, ATP, calcitonin gene-related peptide (CGRP), and substance P are released by EFS. To investigate the purinergic mechanism involved in the EFS-induced contraction, purinegic receptors antagonists were used. Suramine, a non-selective P2 receptor antagonist, reduced the contraction induced by EFS. NF023 ($10^{-7}{\sim}10^{-4}M$), a selective P2X antagonist, inhibited the contraction evoked by EFS. Reactive blue ($10^{-6}{\sim}10^{-4}M$), selective P2Y antagonist, also blocked the contraction in a dose-dependent manner. In addition, P2X agonist ${\alpha}$,${\beta}$-methylene 5'-adenosine triphosphate (${\alpha}{\beta}MeATP$, $10^{-7}{\sim}10^{-5}M$) potentiated EFS-induced contraction in a dose-dependent manner. P2Y agonist adenosine 5'-[${\beta}$-thio]diphosphate trilithium salt ($ADP{\beta}S$, $10^{-7}{\sim}10^{-5}M$) also potentiated EFS-induced contractions in a dose-dependent manner. Ecto-ATPase activator apyrase (5 and 10 U/ml) reduced EFS-induced contractions. Inversely, 6-N,$N$-diethyl-D-${\beta}$,${\gamma}$- dibromomethylene 5'-triphosphate triammonium (ARL 67156, $10^{-4}M$) increased EFS-induced contraction. These data suggest that endogenous ATP plays a role in EFS-induced contractions which are mediated through both P2X-receptors and P2Y-receptors stimulation in cat esophageal smooth muscle.
In normal Fenton's reagent, the reductive mechanism of carbon tetrachloride (CT) with superoxide radical (${O_2}^-$.) was observed and the rate of ${O_2}^-$. production was investigated as a function of $H_2O$$_2$ concentration and pH. As pH was increased, the rate of 1-hexanol degradation was rapidly decreased from 90% (at pH 3) to 5% (at pH 11). On the other hand, more degradation of carbon tetrachloride was observed at higher pH regimes indicating Fenton's reaction is an oxidant-reductant co-existing system at neutral pHs. The rate of $O_2^{-}$ . production was observed at different $H_2$$O_2$ concentrations and at different pHs. The rate increased from (45.3$\pm$7.8) x $10^{-6}$ M/s to (151.0$\pm$26.2) x $10^{-6}$ M/s ($294mM H_2$$O_2$) at pH 11: the rate 3150 increased from (22.1$\pm$3.8) x $10^{-6}$ M/s at pH 7 to (151.0$\pm$26.2) x $^10{-6}$ M/s at pH 11 with 294mM $H_2$$O_2$, These results showed that Fenton's reagent could be applied at wide pH regimes. Especially, carbon tetrachloride, which can not be easily adsorbed to soils and then can be dissolved into groundwater causing a cancer, could be efficiently treated by Fenton's reagent.reagent.
